        New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.

        Purpose: This study aimed to know whether there is a comparison of perception, knowledge and health promoting behaviors about particular matter between nursing students and non-health departments college students. Methods: A quantitative research is used. Data were collected from August 1 to 19. The study sample was nursing students and non-health departments students. For the similar condition of samples, this study excluded whose first language is not Korean. Independent T-test and pearson’s correlation was used for this study. Results: A significant positive correlation did not existed between difference of major and perception(t=0.017, p=0.986), knwledge(t=0.652, p=.516) and health promoting behaviors(t=-0.114, p=.910) about particular matters. Conclusion: Based on the study results, there is no significant correlation of perception and actions about particular matters between nursing students and non-health department college students.

      • A Phase 3 Evaluation of Daclatasvir plus Asunaprevir in Treatment-Naive Patients with Chronic HCV Genotype 1b Infection

        Background/Aims: Treatment-naive GT 1b-infected patients from mainland China, South Korea and Russia were assessed for SVR at follow-up week 12 (SVR12) after receiving daclatasvir (60 mg, QD) and asunaprevir (100 mg, BID) (DCV+ASV). Methods: Patients were randomized 3:1 to receive DCV+ASV (24 weeks; immediate treatment [IM]) or 12 weeks of placebo followed by DCV+ASV (24 weeks; placebo-deferred treatment [PD]). The primary endpoint was to evaluate SVR12 in the IM arm to the historical rate for peginterferon/ribavirin (70%). Secondary endpoints included overall safety and safety comparisons between the treatment arms during the first 12 weeks. Results: 207 patients were randomized to IM (n=155) or PD (n=52); Asian (86%), female (59%), IL28B CC genotype (68%) and median age 49 (range 18-73) years; cirrhosis (13%), HCV RNA ≥6x106 IU/mL (53%). SVR12 in the IM arm was 92% and broadly unaffected by most baseline factors assessed (Figure); SVR12 was higher in patients without (96%) baseline NS5A-L31M/V or Y93H polymorphisms. There were 6 virologic breakthroughs, 6 relapses and 1 detectable HCV RNA at end-of-treatment in the IM arm. Safety was mostly comparable between the two arms during the first 12 weeks. The most frequent adverse events (AEs; ≥5%) during DCV+ASV (24 weeks) treatment in both arms were aminotransferase, bilirubin and INR elevations, hypertension, fatigue and respiratory tract infections; the most frequent treatment-emergent grade 3/4 laboratory abnormalities were aminotransferase (≤4.5%) and hematologic, lipase and total bilirubin abnormalities (≤2%); one patient (IM) discontinued DCV+ASV for aminotransferase elevations, nausea and jaundice (all reversible); one patient PD) discontinued DCV+ASV for a fatal AE unrelated to treatment. Conclusions: These data demonstrate that DCV+ASV is a highly efficacious and well tolerated treatment for treatment-naive HCV GT 1b-infected patients. Those treated immediately with DCV+ASV achieved a 92% SVR12 rate which was unaffected by factors known to attenuate response to interferon.

      • Studies on Cryopreservation of D-shaped and Umbo Larvae of Arkshel1(Scapharca broughtonii)

        The present study examined the possibility of cryopreservation of the D-shaped and umbo larvae of arkshell (Scapharca broughtonii), in terms of the survival rates after freezing and thawing. D-shaped and umbo larvae of arkshells were obtained from a shellfish farming on Yosu city. The average shell lengths were m and , respectively. Five cryoprotectants (CPAs), dimethyl sulfoxide (DMSO), glycerol, ethylene glycol (EG), propylene glycol (PG), and methanol, were tested at the concentrations of 1.5, 2.0 and 2.5 M. After larvae suspended in CPAs, cryoprotectants were loaded in 0.5 ml straws at a larval density of 50-100 larvae per straw, and epuilibrated for 10 and 20 minute at room temperature (), repectively. Straws were cooled at a rate of /min from to , held for 5 min at , and then cooled at /min to and equilibrated for 5 min followed by plunging in liquid nitrogen. After storage in liquid nitrogen for 1 day, straws were thawed in a water. As soon as straws were observed to melt, larvae were diluted with an equal volume of ASW and then washed twice with a large volume of ASW at an interval of 2 min to unload the CPAs. The results showed that after equilibration for 10 and 20 minute at room temperature, no larvae survived using methanol as CPAs, and it was observed that larval shells all open slightly, and larval flesh broke down and slopped over the shells. The highest survival rates (D-shaped larvae: 77.6%, umbo larvae: 59.3%) were obtained with 2M DMSO, and 1.5M glycerol yielded survival rates of 53.8% for D-shaped larvae and 37.5% for umbo larvae. The surviving D-shaped larvae showed active rotary motion and perfect membrane integrity and cytoplasmic normality, and the vigorous movement of veliger cilia was observed inside the closed shells. The breakdown of tissue occurred in the abnormal larvae, and the isolated cell often run out of shells.

      • Two S-wave gap symmetry for single crystals of the superconductor BaFe<sub>1.8</sub>Co<sub>0.2</sub>As<sub>2</sub>

        To clarify the gap structure of the iron-pnictide superconductors, we synthesized optimally doped single crystals of BaFe<SUB>1.8</SUB>Co<SUB>0.2</SUB>As<SUB>2</SUB>, which had a critical temperature, T<SUB>c</SUB>, of 23.6K. The initial M-H curve was used to find the lower critical field, H<SUB>c1</SUB>. The full range of the temperature dependence of H<SUB>c1</SUB> was explained by using a two S-wave gap symmetry. We estimate the two gap as Δ<SUB>1</SUB>(0)=1.64+/-0.2meV for the small gap and Δ<SUB>2</SUB>(0)=6.20+/-0.2meV for the large gap.

        SHM benchmark for high-rise structures: a reduced-order finite element model and field measurement data

        The Canton Tower (formerly named Guangzhou New TV Tower) of 610 m high has been instrumented with a long-term structural health monitoring (SHM) system consisting of over 700 sensors of sixteen types. Under the auspices of the Asian-Pacific Network of Centers for Research in Smart Structures Technology (ANCRiSST), an SHM benchmark problem for high-rise structures has been developed by taking the instrumented Canton Tower as a host structure. This benchmark problem aims to provide an international platform for direct comparison of various SHM-related methodologies and algorithms with the use of realworld monitoring data from a large-scale structure, and to narrow the gap that currently exists between the research and the practice of SHM. This paper first briefs the SHM system deployed on the Canton Tower, and the development of an elaborate three-dimensional (3D) full-scale finite element model (FEM) and the validation of the model using the measured modal data of the structure. In succession comes the formulation of an equivalent reduced-order FEM which is developed specifically for the benchmark study. The reducedorder FEM, which comprises 37 beam elements and a total of 185 degrees-of-freedom (DOFs), has been elaborately tuned to coincide well with the full-scale FEM in terms of both modal frequencies and mode shapes. The field measurement data (including those obtained from 20 accelerometers, one anemometer and one temperature sensor) from the Canton Tower, which are available for the benchmark study, are subsequently presented together with a description of the sensor deployment locations and the sensor specifications.

        On the limit cycles of aeroelastic systems with quadratic nonlinearities

        Limit cycle oscillations of a two-dimensional airfoil with quadratic and cubic pitching nonlinearities are investigated. The equivalent stiffness of the pitching stiffness is obtained by combining the linearization and harmonic balance method. With the equivalent stiffness, the equivalent linearization method for nonlinear flutter analysis is generalized to address aeroelastic system with quadratic nonlinearity. Numerical example shows that good approximation of the limit cycle can be obtained by the generalized method. Furthermore, the proposed method is capable of revealing the unsymmetry of the limit cycle; however the ordinary equivalent linearization method fails to do so.

        Effects of the Chicken Sex-linked Dwarf Gene on Growth and Muscle Development

        The aim of this study was to analyze the effects on growth and muscle development during the growing period of the sex-linked dwarf gene in the background of a Taiwan Country chicken strain, L2, selected for egg production. Eight crossbred males, heterozygous for the DW*DW mutation, were each backcrossed to six females of the L2 strain to produce two genotypes of BC females, either normal (DW*N+/-) or dwarf (DW*DW/-). The experiment included 251 normal and 207 dwarf pullets. The effect of the dwarf gene on body weight and shank length was highly significant from 2 weeks of age. The reduction of body weight by the dwarf gene reached 34.8% and 37.4% as compared to normal sibs at 16 and 20 weeks of age, respectively. Parameters of the growth curve were estimated: the age at inflection (TI) was higher in normal pullets (66.9 days) than in dwarf pullets (61.2 days). A significant effect of the dwarf gene on single muscle fiber cross-section area was found from 12 weeks of age onwards, whereas the dwarf gene had no effect on the total number of muscle fibers. Comparing the effect of the dwarf gene on shank length at different ages revealed an earlier effect on skeleton growth, observed from 2 weeks of age, than on muscle development, which was affected from 8 to 12 weeks of age.
