Genes Regulating the ABORTED MICROSPORES (AMS)-Mediated Male Sterility Networks in Melon (Cucumis melo L.)

        Ling Wang,Dong-yang Dai,Xia Wu,Yun-yan Sheng,Peng Ji,Dan-dan Li,Fan Zhang,Di Wang 한국원예학회 2021 원예과학기술지 Vol.39 No.5

The male sterile plants have higher heterosis in the production of hybrid seeds. The ABORTED MICROSPORES (AMS) gene has been demonstrated to be a candidate gene for ms-5. However, the genetic mechanism underlying AMS-mediated male sterility (MS) regulatory networks in melon (Cucumis melo L.) is still not clearly understood. In the present study, we used transcriptome sequencing analysis, yeast hybridization technology, quantitative real-time polymerase chain reaction (qRT-PCR), and bioinformatics analyzed to systematically investigate the AMS-mediated MS regulatory networks in melon. A set of 15 proteins interacting with AMS, including the C. melo L. Zinc Ribbon protein 1 (CmZR1) gene, was identified using the yeast one-hybrid (Y1H) system and further confirmed using the yeast two-hybrid (Y2H) assay. The interaction of the CmZR1 protein with the C. melo L. Pectin Methylesterase Inhibitor 1 (CmPMEI1) protein was identified and further verified by the glutathione S-transferase (GST) pull-down technique. Bioinformatics analyzed the physical and chemical properties, gene structure, and kinship of the melon PMEI family. We proposed a partial regulatory network for melon MS in which the interaction of CmPMEI1 protein with CmZR1 protein regulates the expression of the AMS gene for pollen abortion. These findings provide important information for increasing the understanding of the molecular mechanism of the MS regulatory network in melon.

Impact of Cu2O doping on high dielectric properties of CuO ceramics

        Dan-Dan Wang,Feng-Zi Zhou,Jing-Xiao Cao,Li-Ben Li,Guo-Ling Li 한국물리학회 2017 Current Applied Physics Vol.17 No.5

Single-phase CuO ceramic samples were prepared with the starting nano powders of CuO þ xCu2O (x ¼ 0, 1, 3, 7% in mole ratio) via solid state reaction method, and characterized by X-ray diffraction, Raman scattering and scanning electron microscopy. For all the samples, the temperature dependences of dielectric constants and losses were measured at the frequencies of 102, 103, 104, 105 and 106 Hz, respectively. With increasing doping content of Cu2O, a strong correlation is demonstrated at given temperature and frequency between the measured dielectric constants and the unit-cell volumes of CuO. The strong correlation is argued in terms of the change in densities of Cu]O defects (e.g. Cu/O vacancies and/or interstitial Cu impurities) due to Cu2O doping, which is supported by the formation energies of Cu]O defects and the corresponding unit-cell volume from first-principles calculations. The high dielectric constant (~103e105) of CuO ceramic is therefore attributed to the reduction in resistance due to Cu/O defects in the grain by Maxwell-Wagner mechanism.

Experimental and theoretical studies of drug-polymer interactions to control the drug distributions in dissolving microneedles

Jing Ling Liu,Yun Hao Feng,Xiao Peng Zhang,Dan Dan Zhu,Li Qin Zhang,Xin Dong Guo 한국공업화학회 2020 Journal of Industrial and Engineering Chemistry Vol.84 No.-

Dissolving microneedles (DMNs) have attracted extensive attention for transdermal drug delivery inrecent years. Generally, the skin elasticity impedes DMNs to fully insert into skin thus optimizedfabrication methods are needed to improve delivery efficiency. Here, we investigated the formulations ofDMNs as the main influencing factors during the microneedle preparation process. Through the selectionof the matrix materials and the optimization of the ratio and formulae, the DMNs can meet therequirements of convenient and effective drug delivery. For DMNs prepared with polyvinyl alcohol (PVA)as matrix material, the 25 wt% concentration of PVA was beneficial to prepare MNs, which MNs was ableto focus drug on the part of MN tips. However, for DMNs fabricated with hyaluronic acid (HA), theincreasing material concentration had a weaker ability to concentrate drug in MNs and increasedthe difficulty of preparing MNs. The computer simulation analysis was consistent with the results of theexperiment, indicating the feasibility of modelling to investigate the preparation technology of DMNsthat offer appropriate, convenient and highly efficient drug delivery.

Seismic performance of the concrete-encased CFST column to RC beam joints: Analytical study

        Dan-Yang Ma,Lin-Hai Han,Xiao-Ling Zhao,Wei-Biao Yang 국제구조공학회 2020 Steel and Composite Structures, An International J Vol.36 No.5

A finite element analysis (FEA) model is established to investigate the concrete-encased concrete-filled steel tubular (CFST) column to reinforced concrete (RC) beam joints under cyclic loading. The feasibility of the FEA model is verified by a set of test results, consisting of the failure modes, the exposed view of connections, the crack distributions and development, and the hysteretic relationships. The full-range analysis is conducted to investigate the stress and strain development process in the composite joint by using this FEA model. The internal force distributions of different components, as well as the deformation distributions, are analyzed under different failure modes. The proposed connections are investigated under dimensional and material parameters, and the proper constructional details of the connections are recommended. Parameters of the beam-column joints, including material strength, confinement factor, reinforcement ratio, diameter of steel tube to sectional width ratio, beam to column linear bending stiffness ratio and beam shear span ratio are evaluated. Furthermore, the key parameters affecting the failure modes and the corresponding parameters ranges are proposed in this paper.

      • Understanding the Mapping Principle of One Syllable One Character as a Predictor of Word Reading Development in Chinese

        Dan Lin,Ling-po Shiu,Yingyi Liu 한국아동학회 2016 Child studies in Asia-Pacific context Vol.6 No.2

Speech-print mapping awareness is defined as the awareness of the principles underpinning how speech sound is matched to print symbols. Chinese is unique in that it follows the one syllable one character mapping principle. The present study examined the predictive power of speech.print mapping awareness in young children’s word reading. Seventy-four Hong Kong children from the first and second kindergarten years were tested with phonological awareness, visual skills, syllable-level mapping awareness, and Chinese reading ability at Time 1. Chinese reading abilities were tested again 1 year later. It was found that syllablelevel mapping awareness predicted Chinese word reading abilities 12 months later. Further, it seemed that the link of syllable mapping to Chinese reading is particularly significant for beginning readers. The findings suggest that understanding the language-specific speech. print mapping principle is critical for reading acquisition at the early stage of reading development.

Core–shell structure g-MnO2-PANI carbon fiber paper-based flexible electrode material for high-performance supercapacitors

        Xuxia Ling,Guoliang Zhang,Zhu Long,Xuefeng Lu,Zhibin He,Jie Li,Yuheng Wang,Dan Zhang 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.99 No.-

Good conductivity is the key to ensuring high electrochemical performance, and the uniformity of theactive material directly affects the electrical conductivity. This paper proposes a novel modified stepwisein situ polymerization method to prepare composite materials, and then uses the K/S value method tocharacterize the uniformity of polyaniline distribution. Specifically, the study presents a papermakingwet-lay technique to prepare a carbonfiber paper substrate (CP) that is light-weight,flexible, andfoldable while also having low internal resistance and high porosity. The g-MnO2 is wrapped with PANI toform a core–shell structure, and a modified stepwise in situ polymerization method is adopted to depositthe core–shell structure uniformly on the CP substrate by physical adsorption. This process yields aflexible electrode material for supercapacitors, called g-MnO2-PANI-CP electrode. Results show thatthe g-MnO2-PANI-CP electrode has a high specific capacitance of 642.5 F g 1 at a current density of 1A g 1, and also has a high energy density of 114.2 Wh kg 1 at a power density of 798.6 W kg 1. In addition, the electrode retains 81.3% of the initial capacitance after 5000 cycles, demonstrating high cyclingperformance.

      • Role of Neoadjuvant Chemotherapy in the Management of Advanced Ovarian Cancer

Zhao, Dan,Wu, Ling-Ying,Wang, Xiao-Bing,Li, Xiao-Guang Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.6

Objective: To analyze efficacy of neoadjuvant chemotherapy for advanced ovarian cancer. Materials and Methods: A total of 107 patients with advanced ovarian cancer undergoing cytoreductive surgery were divided into a neoadjuvant chemotherapy group (n=61) and a primary debulking group (n=46) and retrospectively analyzed. Platinum-based adjuvant chemotherapy was applied to both groups after cytoreductive surgery ande overall and progression-free survival times were calculated. Results: No significant difference was observed in duration of hospitalization ($20.8{\pm}6.1$ vs. $20.2{\pm}5.4$ days, p>0.05). The operation time of neoadjuvant chemotherapy group was shorter than the initial surgery group ($3.1{\pm}0.7$ vs. $3.4{\pm}0.8$ h, p<0.05). There were no significant differences in median overall survival time between neoadjuvant chemotherapy group and surgery group (42 vs. 55 months, p>0.05). Similarly, there was no difference in median progression-free survival between neoadjuvant chemotherapy group and surgery group (16 vs. 17 months, p>0.05). The surgical residual tumor size demonstrated no significant difference between initial surgery and neoadjuvant chemotherapy groups (p>0.05). Multivariate analysis showed that more than 3 cycles of regimen with neoadjuvant chemotherapy was associated with more resistance to chemotherapy compared with patients without receiving neoadjuvant chemotherapy (OR: 5.962, 95%CI: 1.184-30.030, p<0.05). Conclusions:Neoadjuvant chemotherapy can shorten the operation time. However, it does not improve survival rates of advanced ovarian cancer patients.

Ginsenoside Rg5 overcomes chemotherapeutic multidrug resistance mediated by ABCB1 transporter: in vitro and in vivo study

        Sen-Ling Feng,Hai-Bin Luo,Liang Cai,Jie Zhang,Dan Wang,Ying-Jiang Chen,Huan-Xing Zhan,Zhi-Hong Jiang,Ying Xie 고려인삼학회 2020 Journal of Ginseng Research Vol.44 No.2

Background: Multidrug resistance (MDR) to chemotherapy drugs remains a major challenge in clinicalcancer treatment. Here we investigated whether and how ginsenoside Rg5 overcomes the MDR mediatedby ABCB1 transporter in vitro and in vivo. Methods: Cytotoxicity and colon formation as well as the intracellular accumulation of ABCB1 substrateswere carried out in MDR cancer cells A2780/T and A549/T for evaluating the reversal effects of Rg5. Theexpressions of ABCB1 and Nrf2/AKT pathway were determined by Western blotting. An A549/T cellxenograft model was established to investigate the MDR reversal activity of Rg5 in vivo. Results: Rg5 significantly reversed ABCB1-mediated MDR by increasing the intracellular accumulation ofABCB1 substrates without altering protein expression of ABCB1. Moreover, Rg5 activated ABCB1 ATPaseand reduced verapamil-stimulated ATPase activity, suggesting a high affinity of Rg5 to ABCB1 bindingsite which was further demonstrated by molecular docking analysis. In addition, co-treatment of Rg5 anddocetaxel (TXT) suppressed the expression of Nrf2 and phosphorylation of AKT, indicating that sensitizingeffect of Rg5 associated with AKT/Nrf2 pathway. In nude mice bearing A549/T tumor, Rg5 and TXTtreatment significantly suppressed the growth of drug-resistant tumors without increase in toxicitywhen compared to TXT given alone at same dose. Conclusion: Therefore, combination therapy of Rg5 and chemotherapy drugs is a strategy for the adjuvantchemotherapy, which encourages further pharmacokinetic and clinical studies.
