돼지의 마이코플라스마성 폐렴(mycoplasmal pneumonia of swine:MPS)은 돼지를 사육하고 있는 모든 지역에 상재화되어 있어 경제적인 손실이 크므로 pig monitoring system에서는 폐렴병변과 일당증체량(mean daily gain:MDG), 도축일령(days to slaughter:DS) 등의 성장지수(growth indicators:GI)와의 상관관계를 비교 분석할 필요가 있다. 이 실험에서는 1996년 6월부터 1997년 2월까지 돈육 수출을 위해서 도축된 354두의 돼지를 대상으로 MPS 병변과 GI간의 상관관계를 폐병변 점수법(scoring method: SM)과 폐병변 중량법(weight method: WM)으로 생산성을 분석한 결과 다음과 같이 유의성 있는 결과를 얻었다. 1. 354두 중 267두(76.1%)의 폐에서 MPS의 특징적인 병변이 확인되었다. 폐엽에 따른 병변형성 빈도는 right cranial lobe(RCL)와 left cranial lobe(LCL)에서 각각 80.1%, 73.4%로 매우 높게 나타나 cranial lobe이 MPS의 주 병변형성 폐엽임을 알 수 있었다. 2. 354두 중에서 일령, 생체량, 개체구분 등이 정확히 이루어진 146두로부터 MPS병변과 GI간에 유의성 있는 상관관계가 성립되었으며, 이로부터 얻어진 회귀분석(regression analysis)를 통해서 볼 때 폐병변이 약 10% 증가함에 따라서 폐병변접수법(SM)에 의한 경우에는 일당증체량(MDG)이 28.8g이 저하되고 도축일령(DS)은 9.7일이 지연되었으며, 폐병변중량법(WM)이 경우 30.7g이 저하되고 10.3일이 지연되는 것으로 나타났다. 한편, MPS 병변의 Mean±SD은 SM의 경우 4.75±7.75, WM의 경우 5.62±8.66로 큰 차이를 보이지 않았다. 3. SM과 WM 방법을 회귀방정식으로 비교한 결과 매우 높은 상관관계(R²=0.89, P<0.0001)를 나타냄으로써 도축장에서는 SM을 적용하는 것이 적합하다고 판단되었다.
전자스핀공명법(ESR)을 이용한 방사선조사 조제유류의 판별

Electron Spin Resonance (ESR) spectroscopy has been used to detect the presence of radiation-induced free radicals in biological samples since the mid 1950s and to irradiate foods containing cellulose, crystalline sugar, and bone. Therefore, we analyzed the ESR spectrum of irradiated infant formula and its ingredients in this study. Samples were irradiated with 2 different radiation sources of 60Co gamma rays and electron beams (EBs), and the absorbed doses were 0, 1, 3, 5, and 7 kGy. ESR measurements were performed under normal atmospheric conditions using a JEOL JES-FA100 spectrometer equipped with an X-band bridge. Irradiated infant formula showed anunsymmetrical spectrum (g1=2.0050, g2=2.0006); in contrast, non-irradiated samples showed asymmetrical spectrum. The ingredients of irradiated samples showed a multi-component ESR signal in glucose and lactose and a singlet-type spectrum in milk powder (g=2.0050). R2 of the dose-response curve showed a fine linearity of over 0.95 across the entire sample. We also compared the spectra of identical samples irradiated with 60Co gamma rays and EBs, because EBs can be used for food irradiation in foreign countries, although this is not permitted in Korea. However, we could not find any significant differences according to the types of radiation source. Thus, ESR spectroscopy can be used to detect irradiated infant formula and several types of primary ingredients in this formula.

A monoclonal antibody(MAb), KCT-23ER, with specificity for E-rosetted T cells of Korean native cattle was prepared by cell hybridization of myeloma P3/NS-1/1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with E-rosetted lymphocytes. The isotype of KCT-23ER to T lymphocytes was mouse $IgG_{2b}$. KCT-23ER was reacted with 53.6% to peripheral blood lymphocytes and with 67.8% to nylon wool nonadherent blood lymphocytes. And it was reacted with 72.2%, 59.2% and 35.3% to thymocytes, prescapular lymph node cells and splenocytes, respectively. Immunocytological reactive rates to E-rosetted and non-E-rosetted cells were 72.5% and 22.4%, respectively. These results indicated that KCT-23ER reacted to E-rosetted cells was one of the MAb for investigate of $CD_2$ receptor positive cell subset in the Korean native cattle.

This study was undertaken to develop the monoclonal antibody(MAb) for lymphocytes of Korean native cattle by the cell hybridization of myeloma P3/NS-1/ 1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with nylon wool column eluted peripheral T lymphocytes of Korean native cattle. The isotype of MAb KCT-14 against T lymphocyte was mouse $IgG_1$. KCT-14 positivity of mononuclear cells(MNC) from peripheral blood lymphocytes, nylon wool nonadherent and adherent-lymphocytes was 41.7%, 58.4% and 22.6%, respectively. And that of mesenteric lymph node-, spleen and thymus-MNC was 43.3%, 40.2% and 33.6%, respectively. Immunoperoxidase staining of frozen tissue sections showed that the MAb positive cells were located in the medulla of the thymus and in the paracortical area and the mantle zone of the germinal center in the lymph nodes. These results indicated that KCT-14 was one of the MAb for investigate of T lymphocyte subpopulations in the Korean native cattle.

In this study, the most preferred trans fatty acid analysis methods, AOAC 996.06 and the Korea Food and Drug Administration official method, were reviewed and modified to apply to dairy products and dairy products imported into the Republic of Korea for evaluating trans fatty acid (TFA) content. The Rose-Gottlieb method for total fat analysis was validated with accuracy and precision parameters by analyzing infant formula standard reference material provided by the National Institute for Standards and Technology. The accuracy and precision data satisfied the CODEX guidelines. TFAs were analyzed with a resolution of 1.5 for 45 min using the modified oven temperature program. This modified method was applied to 45 dairy products from 11 countries. Average TFA contents in these imported dairy products ranged from 0.1 to 5.4 g per 100 g product. The majority of dairy products imported into the Republic of Korea were cheeses. TFA contents in the cheeses were 0.1 to 2.4 g per 100 g cheese. TFA contents in other dairy products were 1.7 to 5.4 g per 100 g product. These TFAs content variations can be explained by the trans fatty acids naturally present in ruminant milk formed by bacterial bio-hydrogenation in the rumen of cows and the different vegetable fat used as ingredients in the final products. 본 연구는 유가공품에 적용할 수 있는 트랜스 지방산 분석조건을 확립하고 우리나라로 수입되는 유제품의 트랜스 지방산 함량 수준을 확인하기 위해 실시되었다. 조지방 추출은 Rose-Gottlieb법으로 조제분유 표준인증물질을 분석하고 그 인증값과 비교하였던 바, CODEX 가이드에 부합되는 정확도를 나타냈다. 유제품의 트랜스 지방산 분석을 위해 가스크로마토그래프 오븐 온도 상승 조건을 다양하게 변형하여, 트랜스 지방산들이 양호한 분해능을 보이면서 분석되는 적합한 온도 조건을 제시하였고, 최적화된 분석법을 사용하여 우리나라로 수입되는 유가공품(13개 유형, 45제품)에 적용하였다. 수입되는 유제품의 트랜스 지방산 함량은 제품 100 g 당 0.1-5.4 g 범위를 나타내었다. 가장 많이 수입이 되는 치즈류는 0.1-2.4 g/100 g 제품(평균값 1.1 g/100 g, n=35)으로 확인된 반면, 치즈를 제외한 유가공품은 1.7-5.4 g/100 g 제품 수준이었다. 이러한 트랜스 지방산 함량은 반추동물의 제1위 미생물에 의해서 생성되어 우유 중에 존재하는 트랜스 지방산 함량수준이며, 원료로 사용되는 우유의 트랜스 지방산 함량이나 부가적으로 첨가되는 식물성 지방에서 유래되는 트랜스 지방산의 함량에 따라 검사대상 유제품에서 함량의 차이는 편차가 있을 것으로 평가하였다
2010년 국내산 및 수입 축산물의 잔류 동물용 의약품 탐색 조사

The Korean National Residue Program consists of three sampling plans for domestic and imported foods of animal origin: monitoring, surveillance/enforcement and exploratory testing. Monitoring and surveillance/enforcement testing programs are routinely implemented by 17 Provincial Veterinary Services for domestic products and two regional offices of Animal, Plant and Fisheries Quarantine and Inspection Agency (QIA) for imported products, respectively. The exploratory testing is designed to test substances which are not included in the list of monitoring and enforcement testing programs controlled by headquarter of QIA. In 2010, the exploratory testing was carried out in domestic and imported foods of animal origin for 24 veterinary drugs including florfenicol, clavulanic acid, four quinolones (nalidixic acid, difloxacin, marbofloxacin, orbifloxacin), two anthelmintics (closantel, levamisole), two sedatives (azaperone, carazolol), six glucocorticoids (dexamethasone, betamethasone, flumethasone, prednisone, prednisolone, methylprednisolone), eight non-steroidal anti-inflammatory drugs (phenylbutazone, paracetamol, carprofen, flunixin, ketoprofen, meloxicam, tolfenamic acid, acetylsalicylic acid). In the total of 1,153 domestic samples, only florfenicol was detected from 17 pig muscles at levels of 0.2~614 ng/g. Of 17 positive pig muscles, 16 samples were non-violative and one sample was violative. In the total of 1,065 imported samples, florfenicol was detected at 0.4 ng/g in one pork. Also, flunixin was detected at 22 ng/g in one beef.

Oysters are a highly consumed seafood throughout Korea, but they have a short shelf life because they support rapid microbial growth due to their of high moisture content and fragile muscle tissue. We examined natural food additives including lactic acid bacteria fermentation powder, rosemary extract, and lemon juice for their ability to preserve raw oyster Crassostrea gigas quality. Samples were stored at 4℃, and microbiological and physicochemical analyses were conducted. Among the natural additives tested, lemon juice was the most effective. Lemon juice was thus applied at different concentrations (50-300 ppm) to quantitatively assess its effect on total viable cell count, pH, glycogen, soluble protein, and turbidity. 200 ppm was confirmed to be optimal, and is projected to extend shelf life by 2 days compared to the control group.
