Reduction of allergenicity of irradiated ovalbumin in ovalbumin-allergic mice

<P><B>Abstract</B></P><P>Egg allergy is one of the most serious of the immediate hypersensitivity reactions to foods. Such an allergic disorder is mediated by IgE antibodies stimulated by T-helper type 2 (Th2) lymphocytes. This study was undertaken to evaluate changes of allergenicity and cytokine profiles by exposure of irradiated ovalbumin (OVA), a major allergen of egg white, in the OVA-allergic mice model. OVA solutions (2mg/ml in 0.01M phosphate buffered saline (PBS) were gamma-irradiated to 50 and 100kGy. The allergenicity in the OVA-allergy-induced mice model was remarkably reduced when challenged with irradiated OVA. Cultures of spleen cells harvested from OVA-sensitized mice showed a significant decrease in Th2 cytokine levels of ILs-4 and -5 with a concomitant increase in Th1 cytokine levels of IL-12 when co-cultured with irradiated OVA. However, IFN-<I>γ</I> level decreased dependant on the radiation dose of co-cultured OVA. The levels of IgEs and Th2-cytokine were reduced dependant on the radiation dose. These data show that the irradiated OVA could downregulate the activity of Th2 lymphocytes in OVA-sensitized mice.</P>

Background: Excess proliferation and extracellular matrix (ECM) accumulation of mesenchymal cells such as vascular smooth muscle cells (VSMC) and glomerular mesangial cells cause chronic allograft nephropathy showing transplant vascular sclerosis and glomerulosclerosis. Mycophenolic acid (MPA) and rapamycin (RPM) are well known as strong inhibitors of VSMC proliferation, but their effects on the glomerular mesangial cells are not yet clearly understood. This study examined the effects of MPA or RPM on PDGF-induced proliferation and ECM accumulation in rat glomerular mesangial cells. Methods: Mesangial cells isolated from the glomeruli of Sprague-Dawley rats were cultured with DMEM containing 20% fetal bovine serum. Growth arrested and synchronized cells were administered with test drugs (MPA10 nM-10μM, RPM 0.1 nM-1μM) before the addition of PDGF 10 ng/mL. Cell proliferation was assessed by [³H]thymidine incorporation, collagen by [³H]proline incorporation, and fibronectin, ERK, and p38 MAPK by Western blot analysis. Results: PDGF increased mesangial cell proliferation by 4.64-fold. Compared to stimulated control, MPA above 500 nM and RPM above 10nM showed a significant inhibitory effect in a dose-dependent manner. The IC_(50) of MPA and RPM against PDGF-induced mesangial cell proliferation were around 500 nM and 100 nM, respectively. The collagen synthesis was also inhibited by MPA and RPM, but the fibronectin secretion was inhibited by MPA alone. The proliferation of mesangial cell correlated with activation of ERK and. p38 MAPK. MPA, but not RPM, inhibited ERK and p38 MAPK activation. Conclusion: This study demonstrated that MPA and RPM significantly inhibited PDGF-induced proliferation and ECM production in rat glomerular mesangial cells. The inhibitory effects of MPA, but not RPM, are correlated with ERK and p38 MAPK.
A Case of Acquired Brown Syndrome after Surgical Repair of a Medial Orbital Wall Fracture

A case of acquired Brown syndrome caused by surgical repair of medial orbital wall fracture is reported in the present paper. A 23-year-old man presented at the hospital with right periorbital trauma. Although the patient did not complain of any diplopia, the imaging study revealed a blow-out fracture of the medial orbital wall. Surgical repair with a calvarial bone autograft was performed at the department of plastic surgery. The patient was referred to the ophthalmologic department due to diplopia that newly developed after surgery. The prism cover test at distant fixation showed hypotropia of the right eye, which was 4 prism diopters (PD) in primary gaze, 20 PD in left gaze, while orthophoric in right gaze. Eye movement of the right eye was markedly limited on elevation in adduction with normal elevation in abduction with intorsion in the right eye present. Forced duction test of the right eye showed restricted elevation in adduction. Computerized tomography scan of the orbits showed the right superior oblique muscle was entrapped between the autografted bone fragment and posterior margin of the fracture. When repairing medial orbital wall fracture that causes Brown syndrome, surgeons should always be careful of entrapment of the superior oblique muscle if the implant is inserted without identifying the superior and posterior margin of the orbital fracture site.

Based on the development of designed technology for aluminum carbody, the prototype aluminum carbody has been constructed. All extrusion profiles required for the carbody has been produced and their quality has also been proven. For sound construction, welding technology to join aluminum extrusion profiles has been developed and jigs for precise assembly of blocks have been made. The aluminum carbody for urban subway train has been completed with the required chamber being set and the welding deformations being constrained by jigs. The safety of the carbody structure has also been proven by the static load test. And also, painting technology has been developed and the surface of the carbody has been pre-treated and painted. The developed technology to construct the aluminum carbody can be used in mass production of aluminum cars ordered by domestic and foreign customers.
설탕을 첨가한 Polyethylene Glycol 용액의 장정결 효과 및 복용 순응도

목적: PEG 용액은 안전하고 정결 효과가 우수하여 현재 임상 에서 대장내시경 전처치제로 가장 널리 사용되고 있지만, 특 유의 짠맛으로 환자의 순응도가 낮다는 제한점이 있다. 이번 연구는 이러한 단점을 보완하기 위해 표준 PEG 용액에 설탕 을 첨가하여 복용하도록 한 후 대장 정결 효과 및 환자 순응도 를 표준 PEG 용액을 복용한 군과 비교 분석하고자 하였다. 대상 및 방법: 2012년 1월부터 6월까지 강릉아산병원 소화기 내과에서 대장내시경을 시행받은 총 100명의 환자들에 대해 무작위 전향적 대조 연구를 진행하였다. 대장 정결은 표준 PEG 용액 4L 또는 설탕을 첨가한 PEG 용액 4 L 중 한 가지 로 무작위 배정하였다. 시술자 설문지를 통해 대장 정결도와 내시경의의 검사만족도를 조사하였고, 환자 설문지를 통해 순 응도를 조사하였다. 결과: 두 군 사이에 대장 정결도(4.2±2.0 vs. 4.1±1.5, p=0.783)와 내시경의의 검사만족도(8.2±1.8 vs. 8.5±1.3, p=0.253)는 유의한 차이가 없었으나 환자의 역겨움의 정도 는 설탕첨가 PEG군이 3.9±2.9로 표준 PEG군 6.4±2.3과 비교하여 볼 때 역겨움의 정도가 낮았으며(p<0.05), 같은 방 법으로 재검사하겠다는 응답도 설탕첨가 PEG군이 2.3±0.7 로 표준 PEG군 2.0±0.6보다 높았다(p<0.05). 전처치제를 복용하면서 느낀 주관적 불편감은 두 군 사이에 유의한 차이 가 없었다. 결론: 설탕을 첨가한 PEG 용액은 효과적인 대장내시경 전처 치 방법이며 표준 PEG 용액을 복용할 때 짠맛으로 인해 복용 의 어려움을 호소하는 환자에게 좋은 대안이 될 수 있을 것으 로 기대된다. Background/Aims: Although polyethylene glycol (PEG) solution is commonly used for colonoscopic bowel preparation because of its safety and effectiveness, its salty taste decreases patient`s compliance. The aim of this study was to compare the sugared PEG solution with the standard PEG solution in regard to the quality of bowel preparation and patient`s compliance. Methods: From January through June in 2012, 100 patients who underwent colonoscopy in Gangneung Asan Hospital were prospectively enrolled. They were randomly assigned to receive either standard PEG solution or sugared PEG solution. The quality of bowel preparation was assessed by a doctor`s questionnaire and the patient`s compliance was assessed by a patient`s questionnaire. Results: There was no significant difference in the quality of bowel preparation (4.2±2.0 vs. 4.1±1.5, p=0.783), and endoscopist`s satisfaction score (8.2±1.8 vs. 8.5±1.3, p=0.253) between two groups. However, The degree of disgust was lower in the sugared PEG group than the standard PEG group (6.4±2.3 vs. 3.9±2.9, p=0.000). The willingness to repeat same regimen was higher in the sugared PEG group than the standard PEG group (2.0±0.6 vs. 2.3±0.7, p=0.004). There was no difference in side effects between two groups. Conclusions: The sugared PEG solution as a bowel preparation method revealed a higher patient`s compliance and was effective as the standard PEG solution. When the patient dislike the taste and saltness of the standard PEG solution, the sugared PEG solution will be good alternative method.

<P>Histone deacetylase (HDAC) inhibitors represent a promising group of anticancer agents. Treatment of cancer cells with HDAC blockers, such as suberoylanilide hydroxamic acid (SAHA), leads to the activation of apoptosis-promoting genes. To enhance proapoptotic efficiency, SAHA has been used in conjunction with radiation, kinase inhibitors, and cytotoxic drugs. In the present study, we show that at the suboptimal dose of 250 muM, sulindac [2-[6-fluoro-2-methyl-3-[(4-methylsulfinylphenyl)methylidene]inden-1-yl]-acetic acid] significantly enhances SAHA-induced growth suppression and apoptosis of A549 human non-small cell lung cancer cells, primarily via enhanced collapse of the mitochondrial membrane potential, release of cytochrome c, and caspase activation. Furthermore, sulindac/SAHA cotreatment induced marked down-regulation of survivin at both the mRNA and protein levels and stimulated the production of reactive oxygen species (ROS), which were blocked by the antioxidant N-acetyl-l-cysteine. Overexpression of survivin was associated with reduced sulindac/SAHA-induced apoptosis of A549 cells, whereas suppression of survivin levels with antisense oligonucleotides or small interfering RNA further sensitized cells to sulindac/SAHA-induced cell death. Our results collectively demonstrate that sulindac/SAHA-induced apoptosis is mediated by ROS-dependent down-regulation of survivin in lung cancer cells.</P>

<P>The glutamate decarboxylase of γ-aminobutyric acid-producing Lactobacillus brevis 877G (LbGAD) was expressed in Escherichia coli. The optimal pH and temperature for the purified LbGAD activity were respectively determined to be pH 5.2 and 45 C. CaCl2 was shown to be a potent activator of this LbGAD activity. The kinetic parameters for LbGAD were a Km value of 3.6 mmol/L and a Vmax value of 0.06 mmol/L/min for L-monosodium glutamate.</P>

<P>A multifunctional assay with up-/down-conversion (UC/DC) nanohybrids which enables the detection and real-time tracking of hazardous molecules has been developed for use in the field of photoluminescence (PL) point-of-care testing due to its cost and convenience. In particular, innovative approaches such as dual or multimodal imaging and detection under only a single-photon pulse system are highly difficult owing to the issues of device simplification and miniaturization. In this work, we first demonstrated single-photon-driven UC/DC dual-modal PL nanohybrids and showed their high performance in <I>in vivo</I> mercury detection and real-time tracking in a mussel simultaneously. Specifically, UC/DC nanohybrids capable of being stimulated by a single photon were presented <I>via</I> a facile and versatile strategy by combining DC fluorophores for heavy metal ion screening with triplet-triplet annihilation upconversion (TTA-UC) nanocapsules for real-time tracking. By adopting the advantages of the structural transformation of DC fluorophores and highly stable TTA-UC nanocapsules, the outstanding monitoring performance of a standard heavy metal ion (<I>i.e.</I> Hg<SUP>2+</SUP>) was achieved by a dual-modal PL assay with nanohybrids, exhibiting ultra-sensitivity (under 1 nM) and high-selectivity. Interestingly, their application in the real world was also remarkable in screening and tracking of mercury in mussels. This single-photon-driven UC/DC convergence system will provide powerful analytical methodologies for target detection and real-time tracking <I>in vivo</I> and will attract widespread attention from researchers in the fields of PL nanomaterials and fluorophores.</P>
