The present study was performed to investigate the effect of in-vitro maturation and in-vitro fertilization time in Korean Native Cow(KNC) oocytes on in-vitro development and to examine whether the duration of fertilization time of matured oocytes affects the blastocyst(BL) formation. There was no significant difference in cleavage and 8-cell stage rates among the treatment groups, but BL and BL/8-cell rates were significantly higher(P<0.005) in 18hr maturation(IVM18) group(31.0±5.7 and 82.0±5.1%) than 22 and 24hr maturation(IVM24) group. As the time of in-vitro fertilization time increased, the fertility rates was increased but the BL and BL/8cell rates were decreased. The proportion of BL formed on day 7 and 8 was significantly higher(P<0.05) in IVM18:IVF16 group(100%) than in IVM24:IVF24 group(55%). These results suggest that effective in-vitro production of KNC oocytes need to proper IVM and IVF times, and two conditions must be considered simultaneously.

The visual data processing system of man is a highly complex and exceedingly versatile system. It is man's primary sensory input and as such, has received considerable attention and investigation from almost all scientific disciplines. It is the purpose of this study to combine the engineering techniques of system analysis with the principles and techniques of the study of physiological optics to better understand the biological control system involved in positioning the eye to follow a moving target. The primary emphasis of this study concerns the records of saccadic eye-movement responses to step and pulse-step stimuli. A study on the mechanism of saccadic eye-movement is important in that not only does it allow one the benefit of predicting eye motion for a given target motion but more importantly, adds funda-mental knowledge to the much more complex system, of which it is an integral part, the visual process itself.

After knitted cotton fabric was finished silicone quaternary ammonium salt of antibacterial agent, effects of treatment by nonionic and anionic surface active agent and salt-methanol rinsing on antibacterial properties were investigated. When laundered with anionic surface active agent, cation of quaternary ammonium, antibacterial functional radical from the first laundry was compete masked with anion of surface active agent, and antibacterial properties was decreased extremely after ten times of laundry. But, as a result of milk test, the effect of antibacterial properties was known as positive comparing with unfinished fabric.

The extractability and stability of ginkgoflavonolglycosides under presence of several cellulase preparations were investigated. The enzymes used were macerosin, cellulase C and cellulase NC. The content variation of the glycosides was measured with HPLC method, using caffeic acid as an internal standard. The methanol extract of ginkgo leaf, containing the total flavonolglyco-sides of 4.46%, was used for the content comparison. By extraction with the enzymes, each or mixed, the peak levels of all the glycosides began to decrease after 1 or 2 hours. After 24 hour extraction, most of the glycosides were degraded to minor components. The flavonolglycosides in ginkgo leaf were also hydrolysed simply by the water extraction. After 24 hour extraction with water at 40°C, the peak levels of major glycosides were distinctly decreased. Rutin was hydrolysed by enzyme treatment or by ginkgo leaf itself. As a result, it was concluded that the commercially available cellulases and the ginkgo leaf itself contain the activities of β-glycosidase and α-rhamnosi-dase. Kaempferol-3-0-(6' -0-p-coumaroylglucosyl)-rhamnoside and four other ginkgo flavonolglycosides were not hydrolysed under the same condition."
Study of the Interaction Between HSA and Oligo-DNA Using Total Internal Reflection Ellipsometry

Techniques of quantitative analysis are very important for studies of the interactions between bio-molecules in the field of biotechnology and drug development. The total internal reflection ellipsometry system (TIRE) is an attractive label-free procedure for the quantitative analysis of biomolecules because it combines the analytic ability of ellipsometry and the high surface sensitivity of surface plasmon resonance. In this work, we have used TIRE to study the optical properties of an aquatic monolayer of human serum albumin (HSA) and oligo-DNA. Also, we have monitored the adsorption and the interaction processes of protein layers.
Anti-Inflammatory Activities of Biapigenin Mediated by Actions on p38 MAPK Pathway

Biapigenin is a naturally occurring biflavonoid found in Selaginella tamariscina. This study aimed to investigate the anti-inflammatory activity of biapigenin and its mechanisms of action, and to identify possible target proteins. Biapigenin suppressed nitric oxide (NO) production, as well as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and macrophage inflammatory protein (MIP)-2 cytokine release. Moreover, biapigenin completely inhibited IL-1β, inducible nitric oxide synthases (iNOS), and MIP-2 mRNA expression and partially suppressed MIP-1 and TNF-α expression. Lipopolysaccharide (LPS)-induced upregulation of p38 mitogen-activated protein kinase (MAPK) phosphorylation was significantly reduced by biapigenin. Fluorescence titration experiment revealed that biapigenin bound to p38 with a high binding affinity of 2.57 × 106 M−1. These results suggest that biapigenin demonstrates anti-inflammatory activities by the regulation of phosphorylation of p38 MAPK pathway. Our results strongly suggest that biapigenin may be a potent biflavonoid inhibitor of p38 MAPK and have therapeutic applications in the treatment of inflammatory diseases.

Quercetin is a major dietary flavonoid found in onions, apples, tea, and red wine, and potentially has beneficial effects on disease prevention. We carried out this study to investigate the effect of quercetin on UVB-induced matrix metalloproteinase-1 (MMP-1) expression in human keratinocyte HaCaT cells and to further understand the mechanisms of its action. The anti-inflammatory activity of quercetin was investigated and quercetin significantly suppressed the NO production in LPS-stimulated RAW264.7 mouse macrophages. Post treatment of quercetin decreased UV irradiation-induced phosphorylation of JNK, p38 MAPK, and ERK by 91%, 21%, and 17%, respectively. MMP-1 is mainly responsible for the degradation of dermal collagen during the aging process of human skin and quercetin suppressed the UVB-induced MMP-1 by 94%. Binding studies revealed that quercetin binds to p38 with high binding affinity (1.85 × 106 M−1). The binding model showed that the 4′- hydroxy groups of the B-ring of quercetin participated in hydrogen bonding interactions with the side chains of Lys53, Glu71, and Asp168 and the 5-hydroxy group of the A-ring formed a hydrogen bond with the backbone amide of Met109. The major finding of this study shows that quercetin inhibits phosphorylation of JNK, p38 MAPK, and ERK pathway leading to the prevention of MMP-1 expression in human keratinocyte HaCaT cells. Therefore, our findings suggested the potentials of quercetin as a skin anti-photoaging agent.
