      • 하악골 결손부의 골형성에 미치는 Bioplant® HTR(Hard Tissue Replacement)의 영향

        The present study was performed to investigate the effect of HTR® (Hard Tissue Replacement) on osteogenesis in the mandibular bone defects. Eight adult male white rabbits weighing 2.5 to 3.0 kg were used. Four bone defects (8 mm in diameter and 4 mm in depth) were made at the both mandibular body. In the control group, the right mesial bone defect was filled with blood clot and spontaneously healed. In the DFDB group, the right distal bone defect was filled with xenogenic demineralized freeze-dried bone. In the HTR® group, the left mesial bone defect was filled with HTR®. In the HTR®-membrane group, the left distal bone defect was filled with HTR® and covered with BioMesh® membrane. The rabbits were sacrified at 2, 4, 6 and 9 weeks after the operation and microscopic examination was performed. Results obtained were as follows: In the control and DFDB groups, inflammatory cells and the fibrous connective tissue existed and the bone growth was slower than HTR® group by 6 week, and there was intervention of the soft tissue at 9 week. In the HTR® group, bone trabeculi extended between the HTR® particles without intervention of inflammatory cells and the connective tissue at 4 and 6 weeks. In addition, extensive osseous ingrowth into the HTR® particles was observed at 9 week. Bone formation was more active in the HTR® group than the control and DFDB groups. There was not obvious difference in the bone healing rate between the HTR® and the HTR®-membrane group. These results suggest that the HTR® promotes osteogenesis in the bone defects and the HTR® group has no difference in comparison with the HTR®-BioMesh® membrane group in bone healing.

      • 제통단이 퇴행성 슬관절염 환자의 삶의 질에 미치는 영향

        Objectives : to evaluate the effects of Jetongdan, the newly developed herbal medicine, on the quality of life in patients with osteoarthritis of knee Methods : Placebo-controlled, randomized clinical trial on the 80 patients with osteoarthritis of the knee was fulfilled. After enrollment, they took medication as they enrolled. And they were asked to answer the questionnaires (Korean Health Assessment Questionnaire (KHAQ), Lequesne's functional index (LF1), visual analogue scale (VAS)) and analysed with the erythrocyte sedimentation rate (ESR) at baseline, after 4 weeks and 8 weeks of medication. Results : Total KHAQ score, some categories of KHAQ like hygiene, activities were significantly improved after 8 weeks of medication. And the change of the KHAQ score was significantly correlated with the change of VAS. But LFI, VAS, ESR level was not significantly improved, and it was considered because of the short medication period and small size of study population. Conclusion : In conclusion, Jetongdan could improve the health-related quality of life in patients with osteoarthritis of knee. Further study in the large population, in long period were recommended.

      • 四君子湯, 四物湯, 十全大補湯 및 補中益氣湯이 cisplatin 投與로 誘發된 體重減少와 血液變化에 미치는 影響

        To investigate the side effects of cisplatin, We attempted to examine the effects of Sakunjatang(四君子湯), Samultang(四物湯), Sibjeondaebotang(十全大補湯) and Boojunhyikgitang(補中益氣湯), combined with cisplatin. For the purpose of clarifying their action, We observed the body weight, WBC, RBC, platelet and survival ratio in mice and rats. The results were as follows : 1. Administration of Sakunjatang significantly increased the body weight, WBC and RBC in the side effects of cisplatin. 2. Administration of Samultang did not increase the body weight, WBC, RBC and platelet in the side effects of cisplatin. 3. Administration of Sibjeondaebotang significantly increased th body weight, WBC, RBC and platelet in the side effects of cilplatin and prolonged their survival ratio. 4. Administration of Boojungyikgitang did not increase the body weight, WBC, and RBC, but significantly increased the platelet in the side effects of cisplatin. According to the above results it is recognized that the prescription of Boki(補氣) and Bokihyul(補氣血) have therapeutic effects in the side effects of cisplatin.

      • 6-(3,4-디클로로페닐)아미노-7-클로로-5,8-퀴놀린디온의 항진균작용 및 안전성 평가

        6-(3.4-Dichlorophenyl)amino-7-chloro-5.8-quinolinedione (RCK50) was tested for antifungal activities in mice systemically infected with Candida albicans. The therapeutic potential of RCK50 was also assessed in comparison with ketoconazole. RCK50 had ED_50 0.22±0.01 ㎎/㎏. Ketoconazole as a positive control had ED_50 6.00±1.70 ㎎/㎏. Intraperitoneally administered RCK50 at the ED_50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver. And administered RCK50 at the ED_50 for 14 days improved survival rates. The genotoxicities of RCK50 had been evaluated. RCK50 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. RCK50 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK50 has no genotoxic potential under these experimental conditions. Acute oral toxicity studies of RCK50 were carried out in ICR mice of both sexes. RCK50 did not show acute oral toxicities and LD_50 values were over 2.850 ㎎/㎏ in ICR mice.

        Inflammatory Cytokine Expression and Sebum Production after Exposure of Cultured Human Sebocytes to Ultraviolet A Radiation and Light at Wavelengths of 650 nm and 830 nm

        Background: The effectiveness of ultraviolet (UV) radiation, visible light, or infrared light therapy for the treatment of acne is the subject of ongoing scientific debate. Objective: This study was conducted to investigate changes in sebum production and the expression of inflammatory cytokines, matrix metalloproteinases (MMPs), and antimicrobial peptides (AMPs), following exposure of cultured human sebocytes to UVA radiation and light at wavelengths of 650 nm and 830 nm. Methods: Reverse transcription polymerase chain reaction assays were performed to measure the gene expression levels of inflammatory cytokines (interleukin [IL]-1β, IL-6, IL-8, and tumor necrosis factor-α), MMPs (MMP-1, MMP-3, and MMP-9), and AMPs (psoriasin, hBD-2, hBD-3, and LL-37) in cultured sebocytes after exposure to UVA radiation (2 J/cm2, 3 J/cm2, and 5 J/cm2) and light at wavelengths of 650 nm (14 J/cm2, 29 J/cm2, and 87 J/cm2) and 830 nm (5 J/cm2, 10 J/cm2, and 30 J/cm2). Expression of inflammatory cytokine proteins and sebum production were measured using enzyme- linked immunoassays and a lipid analysis kit, respectively. Results: Exposure of cultured sebocytes to UVA radiation and light at wavelengths of 650 nm and 830 nm did not show a significant increase in the expression of inflammatory cytokines, MMPs, or AMPs. Sebum production was not significantly decreased after exposure to UVA radiation and light at both wavelengths. Conclusion: We propose that UVA radiation, visible light, and infrared light can be used to target Propionibacterium acnes for the treatment of acne, without an increase in the expression of inflammatory biomarkers and sebum production.         Fabrication of Microfluidic Chips Containing Porous Membrane for Continuous Production of Lipid Nanoparticles

        다공성 막을 중간층으로 갖는 폴리디메틸실록산 미세유체칩을 이용하여 항암 약물인 독소루비신을 함유한 지질나노입자를 연속적인 공정으로 제조하였다. 불연속상으로는 지질, 독소루비신, 및 라우릭 산을 함유한 에탄올상을, 연속상으로는 증류수를 각각 시린지펌프를 이용하여 미세유체칩에 투입하였다. 작은 기공을 갖는 다공성막을 사용하고, 연속상의 유속이 높을 수록 작은 지질나노입자가 제조되었다. 라우릭 산은 지질나노입자의 콜로이드 안정성을 증가시켜 주었고, 독소루비신과 라우릭 산의 첨가는 HeLa 암세포주에서 높은 세포 독성을 나타내었다. 이러한 미세유체칩은 다른 약물은 물론, mRNA와 siRNA를 함유한 지질나노입자제조에 응용될 수 있다. A polydimethylsiloxane (PDMS) microfluidic device containing a porous membrane as a middle layer was fabricated to continuously produce lipid nanoparticles (LNPs). Ethanol with lipids and water were used as discontinuous and continuous phases. Doxorubicin and lauric acid were added in the ethanol phase for tumor therapy and LNP stability. The smaller LNPs were prepared at the higher flow rate of the continuous phase by using the membrane with smaller pore size. The LNPs with a size of 87.6±3.84 nm were obtained at 0.3 mL/min of the flow rate of the continuous phase by using the membrane with a pore size of 0.02 μm. The addition of lauric acid enhanced the colloidal stability of LNPs. In addition, the LNPs with doxorubicin and lauric acid exhibited lower viability for HeLa cells and enhanced colloidal stability of LNPs. The microfluidic device with a porous membrane can be used as a general platform for the continuous production of LNPs containing mRNA, siRNA, and other therapeutic agents.
