      • Analysis of Indoleamine 2-3 Dioxygenase (IDO) and EGFR Co-expression in Breast Cancer Tissue by Immunohistochemistry

        Background: To determine the amount of co-expression of IDO and EGFR in breast cancer patients. Materials and Methods:In order to obtain the distribution of co-expression of IDO and EGFR in breast cancer, we tested 110 breast cancer paraffin tissue blocks with immunohistochemical methods. Then we investigated the relationship between the diagnostic and pathologic characteristics (tumor size, lymph node status, histologic grade, the gene expression of ER, PR, HER2, p53, Ki67 and PCNA) with the situation of co-expression of IDO and EGFR by reviewing the medical records of 32 breast cancer patients. Results: Among 110 breast cancers, 32 cases demonstrated IDO and EGFR co-expression (29.1%), IDO and EGFR synchronous co-expression being found in 19.1% and asynchronous in 10.0%. Conclusions: IDO and EGFR were co-expressed in breast cancer, including synchronous and asynchronous co-expression. The results suggest that considering IDO and EGFR as two indicators for breast cancer treatment or prognosis analysis provides a potential option of individual treatment for the portion of breast cancer patients with co-expression of IDO and EGFR.

        Efficacy and Safety of the Safe Triangular Working Zone Approach in Percutaneous Vertebroplasty for Spinal Metastasis

        Objective: This study aimed to assess the technical feasibility, efficacy, and safety of the safe triangular working zone (STWZ) approach applied in percutaneous vertebroplasty (PV) for spinal metastases involving the posterior part of the vertebral body. Materials and Methods: We prospectively enrolled 87 patients who underwent PV for spinal metastasis involving the posterior part of the vertebral body, with or without the STWZ approach, from January 2019 to April 2022. Forty-nine patients (27 females and 22 males; mean age ± standard deviation [SD], 57.2 ± 11.6 years; age range, 31–76 years) were included in group A (with STWZ approach), accounting for 54 vertebrae. Thirty-eight patients (18 females and 20 males; 59.1 ± 10.9 years; 29–81 years) were included in group B (without STWZ approach), accounting for 57 vertebrae. Patient demographics, procedure-related variables, and pain relief as assessed using the visual analog scale (VAS) were collected at different time points. Tumor recurrence in the vertebrae after PV was analyzed using Kaplan–Meier curves. Results: The STWZ approach was successful from T1 to L5 without severe complications. Cement filling was satisfactory in 47/54 (87.0%) and 25/57 (43.9%) vertebrae in groups A and B, respectively (p < 0.001). Cement leakage was not significantly different between groups A and B (p = 1.000). Mean VAS score ± SD before and 1 week and 1, 3, 6, 9, and 12 months after PV were 7.6 ± 1.8, 4.2 ± 2.0, 2.7 ± 1.9, 1.9 ± 1.5, 1.7 ± 1.4, 1.7 ± 1.1, and 1.6 ± 1.3, respectively, in group A and 7.2 ± 1.7, 4.0 ± 1.3, 3.4 ± 1.6, 2.4 ± 1.2, 1.8 ± 1.0, 1.4 ± 0.5, and 1.7 ± 0.9, respectively, in group B. Kaplan–Meier analysis showed a lower tumor recurrence rate in group A than in group B (p = 0.001). Conclusion: The STWZ approach may represent a new, safe, alternative/auxiliary approach to target the posterior part of the vertebral body in the PV for spinal metastases.

        Quantitative Proteomic Analysis Reveals Impaired Axonal Guidance Signaling in Human Postmortem Brain Tissues of Chronic Traumatic Encephalopathy

        Chronic traumatic encephalopathy (CTE) is a distinct neurodegenerative disease that associated with repetitive head trauma. CTE is neuropathologically defined by the perivascular accumulation of abnormally phosphorylated tau protein in the depths of the sulci in the cerebral cortices. In advanced CTE, hyperphosphorylated tau protein deposits are found in widespread regions of brain, however the mechanisms of the progressive neurodegeneration in CTE are not fully understood. In order to identify which proteomic signatures are associated with CTE, we prepared RIPA-soluble fractions and performed quantitative proteomic analysis of postmortem brain tissue from individuals neuropathologically diagnosed with CTE. We found that axonal guidance signaling pathwayrelated proteins were most significantly decreased in CTE. Immunohistochemistry and Western blot analysis showed that axonal signaling pathway-related proteins were down regulated in neurons and oligodendrocytes and neuron-specific cytoskeletal proteins such as TUBB3 and CFL1 were reduced in the neuropils and cell body in CTE. Moreover, oligodendrocyte-specific proteins such as MAG and TUBB4 were decreased in the neuropils in both gray matter and white matter in CTE, which correlated with the degree of axonal injury and degeneration. Our findings indicate that deregulation of axonal guidance proteins in neurons and oligodendrocytes is associated with the neuropathology in CTE. Together, altered axonal guidance proteins may be potential pathological markers for CTE.

        Understanding Taiwanese Women's Decisional Experiences Regarding Prenatal Screening Procedures And Diagnostics: A Phenomenological Study

        Purpose: This study sought to understand Taiwanese women’s decisional experiences regarding prenatalscreening procedures and diagnostics. Methods: A hermeneutic phenomenological design guided semistructured interviews with 33 womenwho were 36 weeks pregnant. Data were collected between February and October 2016. Verbatimtranscripts were analyzed following hermeneutic circle to cocreate an understanding of Taiwanesewomen’s decision-making in prenatal screening and diagnostics. Results: Women’s existential experiences were derived from their decision-making process on prenatalscreening procedures and diagnostics for chromosomal aneuploidy. These decisional experiences werecaptured by four theme clusters and eight themes, which were inductively derived from 16 meaningunits: (1) accessing health information; (2) considering what was best for my baby; (3) consideringfamily finance; and (4) feeling anxiety posttest. Conclusion: Participants made informed choices on several prenatal screening procedures, ostensibly,based on their personal values and considerations. During the decision-making process, often-citedbenefits of genetic screenings were emphasized, but test limitations were often unheeded. A fundamentalneed for supportive information in decision making was further identified with recommendedstrategies. Hence, a revision of traditional genetic counseling approaches is recommended. As genomicstechnologies are increasingly available during antenatal services, women should be sufficiently educatedabout them to support decision making.

        Regioselective Enzymatic Procedure for Preparing 3'-O-stearoyl-6- azauridine by Using Burkholderia cepacia Lipase

        Previous studies on the lipase-mediated acylation of 6-azauridine with vinyl stearate in organic solvents revealed that while preparing a potential prodrug, 3'-Ostearoyl-6-azauridine, a lipase from Burkholderia cepacia showed high regioselectivity toward the second hydroxyl group. The most suitable reaction solvent, molar ratio of vinyl stearate to 6-azauridine, and reaction temperature were anhydrous acetone, 15:1, and 45°C, respectively. Under these conditions, the initial reaction rate, 3'-regioselectivity,and maximum substrate conversion were as high as 10.4mM/h, 86.0, and 99.0%, respectively.

        Optimal Configuration of Flexible Interconnection Devices for Transferring Photovoltaic Power in Active Distribution Network

        With the increasing penetration of distributed photovoltaic in distribution network, it is more difficult to control active distribution network (ADN). A flexible interconnection device (FID) can realize regional interconnection of the ADN through transferring power. However, the influence of installation position and number of FIDs on the ADN varies, it is necessary to analyze its operational planning model. In this study, first, a photovoltaic power and load forecasting model is established, followed by reduction of the typical scenarios. Second, the ADN operational planning model is established according to the line load balance index to analyze the coordination and determine the power transfer value of the FID in different installation positions and numbers for the ADN. Third, the net income index of the distribution network in the entire life cycle is established to analyze the economy of the ADN and determine the optimal FID installation location and quantity. Finally, numerous simulations and comparisons are carried out on the actual example system, and the results show that the FID can effectively transfer the active power, strengthen the ADN power supply level, and solve the problem of power supply imbalance caused by distributed photovoltaic access.

      • Expression of Fas/FasL in CD8<sup>+</sup> T and CD3<sup>+</sup> Foxp3<sup>+</sup> Treg Cells - Relationship with Apoptosis of Circulating CD8<sup>+</sup> T Cells in Hepatocellular Carcinoma Patients

        Aims: Dysfunction of the host immune system in cancer patients can be due to a number of factors, including lymphocyte apoptosis. Several studies showed that $Foxp3^+T$ cells take part in inducing this process by expressing FasL in tumor patients. However, the relationship between apoptosis, $CD8^+T$ cells and $Foxp3^+T$ cells in HCC patients is still unclear. The present study was designed to investigate the correlation between apoptosis levels and Fas/FasL expression in $CD8^+T$ lymphocytes and $Foxp3^+T$ cells in patients with HCC. Methods: $CD8^+T$ cells and $CD3^+Foxp3^+T$ cells were tested from peripheral blood of HCC patients and normal controls and subjected to multicolor flow cytometry. The expression of an apoptosis marker (annexin V) and the death receptor Fas in $CD8^+T$ cells and FasL in $CD3^+Foxp3^+T$ cells were evaluated. Serum TGF-${\beta}1$ levels in patients with HCC were measured by enzyme-linked immunosorbent assay. The relationship between apoptosis and Fas expression, as well as FasL expression in $CD3^+Foxp3^+T$ cells was then evaluated. Results: The frequency of $CD8^+T$ cells binding annexin V and Fas expression in $CD8^+T$ cells, were all higher in HCC patients than normal controls and the proportion of apoptotic $CD8^+T$ cells correlated with their Fas expression. Serum TGF-${\beta}1$ levels correlated inversely with $CD3^+Foxp3^+T$ cells. Conclusions: Fas/FasL interactions might lead to excessive turnover of $CD8^+T$ cells and reduce anti-tumor immune responses in patients with HCC. Further investigations of apoptosis induction in $Fas^+CD8^+T$ cells in vitro are required.

        Full-scale testing and modeling of the mechanical behavior of shield TBM tunnel joints

        For shield TBM (Tunnel Boring Machine) tunnel lining, the segment joint is the most critical component for determining the mechanical response of the complete lining ring. To investigate the mechanical behavior of the segment joint in a water conveyance tunnel, which is different from the vehicle tunnel because of the external loads and the high internal water pressure during the tunnel’s service life, full-scale joint tests were conducted. The main advantage of the joint tests over previous ones was the definiteness of the loads applied to the joints using a unique testing facility and the acquisition of the mechanical behavior of actual joints. Furthermore, based on the test results and the theoretical analysis, a mechanical model of segment joints has been proposed, which consists of all important influencing factors,including the elastic-plastic behavior of concrete, the pre-tightening force of the bolts and the deformations of all joint components, i.e., concrete blocks, bolts and cast iron panels. Finally, the proposed mechanical model of segment joints has been verified by the aforementioned full-scale joint tests.

        Bayberry Tannin as Stabilizer for the Synthesis of Highly Active and Reusable Heterogeneous Pd Catalysts and Their Application in the Catalytic Hydrogenation of Olefins

        In this study, the homogenous Pd nanoparticles (Pd NPs) were first prepared with bayberry tannin (BT) as the stabilizers. Subsequently, the obtained bayberry tannin-stabilized Pd nanoparticles (BT-Pd) were immobilized onto ${\gamma}-Al_2O_3$ to prepare heterogeneous ${\gamma}-Al_2O_3$-BT-Pd catalysts. Fourier Transformation Infrared Spectrum (FTIR) and X-ray Photoelectron Spectroscopy (XPS) analyses confirmed that the Pd NPs were well stabilized by the phenolic hydroxyl groups of BT. Transmission Electron Microscopy (TEM) observation indicated that the diameter of the Pd NPs can be effectively controlled in the range of 4.2-16.0 nm by varying the amount of BT. It is found that the ${\gamma}-Al_2O_3$-BT-Pd catalysts exhibit highly activity for various olefin hydrogenations. For example, the initial TOF (turnover frequency) of the ${\gamma}-Al_2O_3$-BT-Pd in the allyl alcohol hydrogenation is as high as $12804 mol{\cdot}mol^{-1}{\cdot}h^{-1}$. Furthermore, the ${\gamma}-Al_2O_3$-BT-Pd can be reused 5 times without significant loss of activity, exhibiting a superior reusability as compared with conventionally prepared ${\gamma}-Al_2O_3$-Pd catalysts.
