        국내산 더덕의 Tangshenoside I과 Lobetyolin 정량분석

        Deoduk (Codonopsis lanceolata) has a complex chemical composition that includes polyphenols, saponins, amino acids, and other unidentified compounds. The contents of tangshenoside and lobetyolin are considered as standard of quality evaluation of Deoduk. In this study, an ultra-performance liquid chromatography (UPLC) method was developed for the quantitative determination of the two marker constituents, tangshenoside and lobetyolin. The methods for determining the standards of quality were validated by measuring their linearity, specificity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using UPLC. Reversed-phase UPLC analysis was conducted quantitatively to identify individual tangshenoside and lobetyolin in Deoduk extracted with 50% (v/v) aqueous ethanol. We used 21 samples to carry out quantitative analysis of tangshenoside and lobetyolin. Based on their dry weights, the levels of tangshenoside and lobetyolin were 0.36~3.54 mg/g, 0.24~1.29 mg/g, respectively. These results will be valuable as basic data for standardization of Korean Deoduk.

        새싹 더덕의 항산화 활성

        The purpose of this study was to determine the content of polyphenols and flavonoids, vitamin C, and antioxidant activity for the extract from the Deodeok sprout. To accomplish this, the Deodeok sprout whole (CLS-W), above ground part (leaf, stem, CLS-L), and root (CLS-R) were individually extracted using 70% ethanol. The highest levels of total polyphenols and total flavonoids were observed in the Deodeok sprout extract CLS-L2. Similarly, antioxidant activities resulting in radical scavenging activities increased significantly in the extract of CLS-L2. In conclusion, these results indicate that Deodeok sprouts can be used as a viable, new natural antioxidant source.

        배추 유전자원의 항산화 효과

        This experiment was conducted to assess the high antioxidant activity varieties of Chinese cabbage (Brassica rapa L. ssp. pekinensis) from the 55 accessions. The antioxidant activity of Chinese cabbage were determined by the TPC, TFC, DPPH, ABTS, and chlorophyll, carotenoid contents. The TPC and TFC showed a range of 1.21~4.61 mg GAE/g DW, 0.18~3.09 mg CE/g DW. The DPPH and ABTS assay were in the range of 0.65~4.36 and 1.42~6.91 mg ascorbic acid equivalent (ASCE)/g DW, respectively. The UPLC analysis was performed quantitatively to identify chlorophyll and carotenoid in the Chinese cabbage extract. The levels of the total chlorophyll and total carotenoid were 86.60~1,235.91, and 75.86~490.11 ㎍/g, respectively. The comprehensive differences in the total and individual chlorophyll contents have also been observed among different varieties. These results will be valuable as basic data for the standardization of Chinese cabbage.

        국내산 녹차의 카테킨 함량

        녹차의 성분은 폴리페놀, 알칼로이드, 아미노산 및 기타 화합물로 구성되어 있고 카테킨과 카페인의 함량은 녹차의 품질 평가 기준으로 알려져 있다. 본 연구에서는 ultra performance liquid chromatography(UPLC)를 이용하여 녹차의 지표성분 epicatechin, epigallocatechin, epicatechin gallate, epigallocatechin gallate 및 카페인을 동시 분석하였다. 최적 추출조건으로는 25% 에탄올을 이용하여 온도 25°C, 1시간 추출하였고, UPLC를 이용한 빠르고 간편한 분석조건을 확립하고 정확성, 정밀성, 직선성, 특이성 등 분석법의 유효성을 검증하였다. 지역별로 녹차 잎을 추출하여 UPLC로 카테킨 화합물 함량을 분석한 결과 총 카테킨의 함량(mg/g, 건조 중량)은 제주도 168.87 mg/g> 경상남도 156.75 mg/g> 전라남도 148.22 mg/g 순으로 존재하였고 약 7.3~26.1 mg/g의 카페인이 함유되어 있다. 지역별 생육환경에 따라 함량 차이가 나타났으며, 지역별 품질관리를 위한 객관적이고 과학적인 근거를 제시하여 국내에서 재배되는 녹차의 품질평가에 대한 하나의 기준이 될 것으로 기대된다. The chemical composition of green tea is complex and includes polyphenols, alkaloids, amino acids, and other unidentified compounds. The contents of catechins and caffeine are considered a standard of green tea quality evaluation. In this study, an ultra performance liquid chromatography (UPLC) method was developed for the quantitative determination of five marker constituents in green tea; epicatechin, epigallocatechin, epicatechin gallate, epigallocatechin gallate, and caffeine. Reversed-phase UPLC analysis was conducted quantitatively to identify individual catechins and caffeine in teas extracted with 25% (v/v) aqueous ethanol. Content (mg/g, dry weight) of tea catechins decreased in the following order: Jeju-do 168.9 mg/g> Gyeongsangnam-do 156.8 mg/g> Jeollanam-do 148.2 mg/g. Based on their dry weights, the green teas contained 7.3 to 26.1 mg/g of caffeine. These results will be valuable as basic data for standardization of Korean green tea.

        지역별 오미자의 리그난 함량 및 분석법 검증

        본 연구는 재배지역별 오미자의 리그난 함량을 조사하기 위해 수행되었다. HPLC를 사용하여 특이성, 직선성, 검출한계(LOD), 정량한계(LOQ) 및 정확도를 결정함으로써 리그난의 분석법이 검증되었다. 리그난 중에는 schizandrin이 3.85~6.60 mg/g으로 가장 풍부했고 그다음 gomisin N이 2.69~4.38 mg/g으로 많았다. Schizandrin A와 gomisin A는 0.60~2.04, 0.86~1.63 mg/g이었고, gomisin G, gomisin H, gomisin J는 0.5 mg/g 미만이었다. 오미자의 리그난 화합물 7종 함량은 9.04~14.90 mg/g이었다. 재배지역별 오미자는 각 리그난의 함량은 다르지만 전체 함량에는 큰 차이가 없다. 오미자 표준화를 위한 기초자료로 유용할 것으로 기대된다. This study examined the lignans (schizandrin, schizandrin A, gomisin A, gomisin G, gomisin H, gomisin J, and gomisin N) contents in the fruit of Omija (Schisandra chinensis) according to the cultivation region. The methods for determining lignans were validated by measuring the specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using HPLC analysis. Among the lignans, schizandrin was the most abundant (3.85∼6.60 mg/g) followed by gomisin N (2.69∼4.38 mg/g). Schizandrin A and gomisin A were approximately 0.60∼2.04 and 0.86∼1.63 mg/g, respectively, and gomisin G, gomisin H, and gomisin J were less than 0.50 mg/g. The contents of seven lignans of Omija were 9.04∼14.90 mg/g. Omija showed different contents of lignans according to the cultivation region, but there was no significant difference in the total contents. These results will provide valuable basic data for the standardization of Korean Omija.

        암갈색그물버섯(Boletus umbriniporus)에서 분리한 indole alkaloid

        Mushrooms are valued as a nutritional food and also as an important source of useful medicinal components. They produce various secondary metabolites which have interesting biological activities and unique chemical structures. As part of our ongoing investigation on chemical constituents and bioactive components of Korean native mushrooms, compound 1, an indole alkaloid, was isolated from the fruiting body of Boletus umbriniporus. B. umbriniporus is characterized by its yellow flesh, which changes to pallid blue when exposed to air, and its chemical constituent has not been reported. Chemical structure of compound 1 was determined to be flazin on the basis of ESI-mass, $^1H$ NMR, $^{13}C$ NMR, $^1H-^1H$ COSY, HMQC, and HMBC analysis. This compound was isolated from the fruiting body of B. umbriniporus for the first time. 우리나라 자생 버섯에 함유된 화학적 구성성분을 조사하던 중 암갈색그물버섯에서 indole alkaloid 화합물을 분리하고 mass 및 NMR 분석을 통하여 화학구조를 flazin으로 동정하였다. 비록 Flazin 화합물이 젖비단그물버섯에서 분리되어 보고된 적은 있으나 본 연구에 의하여 암갈색그물버섯에도 flazin 성분을 함유하고 있음이 처음으로 밝혀졌다.

        더덕 추출물과 용매 분획물의 항산화 및 티로시나아제 저해활성

        The purpose of this study was to investigate the antioxidant activity and tyrosinase inhibitory activity of Codonopsis lanceolata 50% ethanol extract, and its solvent fractions (n-hexane, ethyl acetate (EA), n-butanol, water). The main components of the EA fraction were qualitatively analyzed using UPLC Q-ToF/MS. Additionally, a quantitative analysis was performed using UPLC. As a result, the total polyphenol content was 113.36 mg gallic acid/g in the EA fraction, which contained the largest amount of the C. lancolata solvent fractions. Also EA showed the highest antioxidant activity than other fractions. The IC<sub>50</sub> of DPPH(2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity was 0.03 mg/mL and the IC<sub>50</sub> of ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate)] radical scavenging activity was 0.049 mg/mL. The EA fraction showed tyrosinase inhibitory activity than other fractions and especially inhibited monophenolase oxidase reaction higher than diphenolase oxidase reaction. The monophenolase oxidase inhibited 55% when the concentration of the EA fraction was 0.25 mg/mL. As a result of Q-ToF/MS analysis, it was confirmed that tangshenoside I and lobetyolin were the main components of EA fraction. Thus, these results suggest that C. lanceolata may be used as a potent source of cosmetic agents.

        더덕(Codonopsis lanceolata Trautv.)순 추출물의 다양한 생리활성

        본 연구에서는 기능성식품 소재로서 더덕순을 활용하고자 다양한 생리활성을 평가하였다. 생장길이별 더덕순 시료(CLS-A~D) 70% 에탄올 추출물의 총 폴리페놀 및 총 플라보노이드 함량은 각각 580.82~719.56 ㎎ GAE/100 g 및 287.45~398.52 ㎎ CE/100 g 범위였으며 생장길이가 길어질수록 감소하는 경향을 보였다. DPPH와 ABTS 라디칼 소거능을 평가한 결과 CLS-B 시료에서 각각 622.87, 436.98㎎ AA eq/100 g을 보여 가장 높은 항산화 활성을 보였고 생장길이가 길어질수록 항산화 활성은 감소하는 경향을 나타냈다. 또한, 더덕순의 tyrosinase, α-glucosidase 및 elastase 효소 저해 활성을 평가한 결과 모두 CLS-B 시료에서 가장 우수한 저해 효과를 나타내었다. 본 연구를 통해 더덕순의 새로운 기능성식품 소재로서 넓은 활용 가능성을 확인했으며, 추후 더덕순의 주요 기능 성분에 대한 정성 및 정량분석과 건강식품 소재화 연구가 추가로 필요할 것으로 생각된다. This study was undertaken to evaluate the physiological activities for subsequent utilization of Deodeok (Codonopsis lanceolata Trautv.) buds as a new functional food. Deodeok buds (CLS-A∼D) were classified according to the growth length. The total polyphenol and flavonoid contents of the 70% ethanol extracts ranged from 580.82 to 719.56 ㎎ GAE/100 g, and 287.45 to 398.52 ㎎ CE/100 g, respectively. The contents tended to decrease with increasing growth length of Deodeok buds. The DPPH and ABTS radical scavenging activity of CLS-B were determined to be 622.87 and 436.98 ㎎ AA eq/100 g, respectively, showing the highest antioxidant activity. Similar to other contents, increasing growth length resulted in decreased antioxidant activity. Moreover, tyrosinase, α-glucosidase, and elastase enzyme inhibitory activities of Deodeok buds also showed best inhibitory effect in CLS-B. Considering the result of this study, we confirm the possibility of wide application of Deodeok buds as a new functional food material.

        채취시기별 보검선인장 줄기의 항산화, 항당뇨 및 항알츠하이머 활성평가

        In this study, we assessed antioxidant, anti-diabetic, and anti-Alzheimer activities of Opuntia ficus-indica var. saboten (OFI) at harvest time. OFIs were cultivated December 2015~November 2016 in Jeju island. The 70% ethanol extracts of OFI were used to investigate total polyphenol and flavonoid contents, antioxidant(DPPH and ABTS radical scavenging assay), anti-diabetic(yeast α-glucosidase and rat α-glucosidase inhibition assay), and anti-Alzheimer(Acetylcholinesterase and butyrylcholinesterase inhibition assay) activities. Total polyphenol and flavonoid contents of OFIs were 17.40~23.11 μg garlic acid/mg Ex and 2.17~6.22 ug (+)-catechine/mg Ex, respectively. DPPH and ABTS radical scavenging activity of OFIs were 131.98~184.90 mg ascorbic acid(AA) eq/100 g and 63.60~101.83 mg AA eq/100 g, respectively. In the anti-diabetic and anti-Alzheimer activities, 70% ethanol extracts of OFI exhibited moderate inhibition activity, compared to control (acarbose and beberine). Total polyphenol and flavonoid contents, antioxidant, anti-diabetic, and anti-Alzheimer activities were no significant differences by season, respectively. Therefore, information on comparative biological evaluations of OFI may be a beneficial in exploring functional food and drug development.

        보육시설 이용부모가 인식한 보육교사 이미지

        본 연구는 인간의 주관적 영역인 느낌, 태도, 의견, 선호 그리고 신념 등의 본질적 가치를 객관적으로 측정하는 Q 방법론을 이용하여 보육시설 이용부모들이 인식하고 있는 보육교사에 대한 이미지 유형을 탐색하였다. 보육교사의 이미지 유형을 파악하고 유형별 특성 및 유형별 차이점을 분석하고자, 총 29개의 Q 표본과 19명의 P 표본을 선정하였다. 수집된 자료는 QUANL PC 프로그램으로 처리하였으며, 요인분석을 실시하였고, 요인회전방식은 배리맥스를 이용하였다. Q 방법론을 적용한 결과를 보면 보육교사의 이미지 유형이 3개가 도출되어 보육교사에 대한 인식이 다양하게 존재함을 확인할 수 있었다. 만 최근 몇 년간 끊임없이 발생하는 아동학대 사례로 인해 보육교사들에 대한 부정적인 인식과 이미지가 강하게 나타난 것으로 보인다. 따라서 보육교사에 대한 사회적 인식을 정상화하고, 믿고 맡길 수 있는 보육제도를 공고화하기 위한 정책적, 실천적 과제를 제시하였다. 본 연구가 객관적이고 학술적인 자료로 활용되기를 기대한다. This research experimented child care teacher’s image based on child care center parent’s cognition using Q methodology, which describes human’subjective perception through objective measures. 29 Q-samples and 19 P-samples were selected to classify the types of child care teacher’s impression, and analyze the differences between each type. QUANL-PC program was used for principle component analysis, and varimax rotation was used for factor analysis. Result from Q methodology demonstrate that child care teachers have highly diverse impression with three different types. Some negative impression appears to be the result of child abuse in the child care center over recent years. This paper gives an overview of the political and practical tasks to increase positive impression of child care teachers for firm and stable child care system. This research expects to be an objective and academic reference for future study.
