        압착식, 압출식 착유 대마 종실유의 색깔과 주요성분 비교

        대마[Cannabis sativa L.]는 오랫동안 섬유작물로 재배되었지만 종실의 기름 함량이 많고 불포화지방산 비율이 높아 식용유 원료작물로 유망하다. 오랫동안 대마 종실유를 이용해온 유럽의 여러 나라에서는 압출식으로 착유하는데, 이용 역사가 짧은 우리나라에서는 대마 종실유에 대한 정보 부족으로 소비자 기호도가 낮은 편이다. 본 연구는 압착식과 압출식으로 착유한 대마 종실유의 성분을 분석하여 소비자들에게 올바른 정보를 제공하기 위해 수행하였다. 압출식 착유기로 착유하면 착유량이 압착식에 비해 많았다. 착유 부산물인 유박은 압출식에서는 으깨어져 얇고 납작하게 배출되지만, 압착식은 종실이 다소 납작해졌으나 종실의 형상을 유지하였다. 압출식 착유 종실유는 압착식에 비해 Chlorophyll A, B 및 Carotenoid 함량이 높아 명도(L)는 낮고 녹색과 황색은 진하였다. 지방산 조성에서 Palmitic acid, Stearic acid, Oleic aicid, γ-Linolenic acid 비율은 압출식 착유 종실유가 압착식에 비해 다소 높았으나, Linoleic acid와 α-Linolenic acid는 낮았다. 대마 종실유는 총 토코페롤 함량이 많고 그 중 γ-Tocopherol 함량이 많은 것이 특징이었는데 압출식 착유 종실유가 압착식에 비해 다소 적었다. Hemp [Cannabis sativa L.] has been cultivated as a fiber crop for long history, but it was a good oil crop because its seed contain plenty of lipid which is high ratio of unsaturated fatty acid. Hemp seed oil was extracted with a extruding method in many countries including Europe. The color of oil extracted with extruding method is dark green which could be difficult to attract consumer’s interest in Korea because of insufficient information about hemp seed oil. This study was conducted to know correct information about hemp seed oils which were extracted with pressuring and extruding methods. In extruding method, seeds were crushed during the extracting process and discharged oil cake in shape of thin ribbon, but maintained seed shape although the seed were slightly flatted in pressuring method. Oil yield were higher in the extruding method in comparison with pressuring method. The oil have lower degree of lightness but higher degree of greenness and yellowness in the extruding method in comparison with pressuring method because of higher content of chlorophyll A, B and carotenoid. In fatty acid composition, the ratio of palmitic acid, stearic acid, oleic acid and γ-linolenic acid were higher but linoleic acid and α-linolenic acid were lower in the extruding method in comparison with pressuring method. The content of total tocopherol and γ -tocopherol were lower in the extruding method in comparison with pressuring method.

        자동비상제동 시스템의 안전성능평가

        This paper presents a performance evaluation procedure for advanced emergency braking (AEB) system. To guarantee the performance of AEB system, AEB test scenario should contains various driving conditions which can be occurred in real driving condition. Also, performances of each elements of AEB system, such as sensor, decision, human machine interface (HMI) and control, should be evaluated in various situations. For this, driving conditions, road types, environment, and elements of AEB system were introduced. Test scenario has been designed to represent the real driving condition and to evaluate the safety performance of AEB system in various situations. To confirm that the proposed AEB test scenario is realistic and physically meaningful, vehicle test have been conducted in two cases of proposed AEB test scenario: subject vehicle cut-out scenario and narrow street turn left scenario.

        미립자 응집반응을 이용한 C-reactive Protein의 면역측정법에 관한 연구

        환자의 복수와 늑막액으로부터 p-diazonium phenylphosphorylcholine(DPPC) coupled Separose-4B affinity chromatography와 hydroxylapatite chromatography를 실시하여 C-reactive protein(CRP)를 분리, 제정하였다. 정제된 CRP를 토끼에게 면역화하여 항혈청을 얻고 affinity chromatography를 하여 면역항체(IgG)를 분리하였다. 분리된 면역항체를 미립자에 감작시킨 후 미립자 응집반응에 의하여 3분내에 CRP를 측정할 수 있는 간이 면역측정법을 개발하였다. 본 연구에서 개발된 CRP측정법의 검출범위는 0.5~20㎎/dl이며, 임상 시험 결과 0.7~2.9㎎/dl에서는 강한 응집반응을, 5.0~13.2㎎/dl에서는 약한 응집반응을 보였고 28㎎/dl이상에서는 항원 과잉으로 인한(zone of Ag excess phenomenon) 위음성을 나타냈다. 74명의 환자 혈청을 대상으로 CRP의 농도를 조사한 결과 평균치는 3.8㎎/dl이었으며 대부분의 환자에서는 10㎎/dl 이하의 농도로 존재하였다. 그러므로 1차판정시 음성을 나타낸 시료라도 혈청을 5~10배정도 희석하여 재분석한다면 오차없이 CRP를 검출할 수 있었다. 환자 혈청을 검체로 하여 본 연구에서 개발한 면역측정법과 현재 수입 시판중인 프랑스의 B사 제품과 일본의 I사 제품을 비교한 결과 좋은 상관관계를 보였다. 이와 같은 평가분석을 통하여 볼 때 본 연구에서 개발한 간이 면역측정법은 사용이 비교적 간편하며 신빙성이 있어 CRP를 스크리닝 하는데 효과적임을 알 수 있었다. The C-reactive protein(CRP) from ascitic and pleural fluid was purified using calcium dependent affinity chromatography of CNBr activated Sepharose-4B covalently coupled to p-diazonium phenylphosphorylcholine(DPPC) and hydroxylapitite chromatography. Polyclonal antibody was prepared from rabbit by immunizing the purified CRP. Specific immunoglobulin G was isolated using affinity chromatography and coupled to microparticles. A sensitive microparticle-based immunoassay was developed to measure CRP within 3 mins. The detection range was between 0.5㎎/dl and 20㎎/dl in serum, showing strong response in the range of 0.7~2.9 ㎎/dl, weak response in 5.0~13.2 ㎎/dl and zone phenomenon over 28㎎/dl. The average value of CRP in 74 samples was 3.8㎎/dl and most of the values were lower than 10㎎/dl. The CRP values of serum samples were determined by our microparticle-based immunoassay, and were compared with those obtained using the other commercial products(B Co., France and I Co., Japan). Good correlations were shown between the values obtained by our developed mi-croparticle-based immunoassay system and those by other commercial products. All performance characteristics evaluated make our developed microparticles-based immunoassay suitable for a simple, rapid, and reliable screening of CRP in serum.

        자율주행 자동차 임시운행 허가를 위한 안전 성능 평가 시나리오

        This paper presents an evaluation scenario of safety performance for extraordinary service permission of autonomous vehicle driving on a motorway. Based on advanced driver assistance system (ADAS) which is already mass-production, an autonomous vehicle driving on motorway is tested on the public roads and also getting close to mass-production. Before the autonomous vehicle tested, the safety of autonomous driving system should be evaluated based on a proper test scenario. Prior to develop the test scenario, this paper reviews the licensing standards for an autonomous vehicle in California and Nevada, and the international regulations of each ADAS. To develop the scenario, the driving conditions of motorway are categorized into five modes and fundamental evaluation requirements of elements of autonomous driving system are derived. An evaluation scenario, which represents the real driving conditions, has been developed to assess the safety of autonomous vehicle. This scenario has validated by computer simulation using model predictive control (MPC) based autonomous driving algorithm.

        재조합 preS2 peptide 에 대한 단일 클론항체 생산과 그를 이용한 면역 친화성 column 제조에 관한 연구

        Three monoclonal antibodies specific to the recombinant preS₂ peptide were prepared using preS₂-β-galactosidase as an immunogen. All three were classified as immunoglobulin M. One of the three monoclonal antibodies (040-01) was precipitated by 6% PEG and used for the preparation of immunoadsorbent column. This immunoaffinity column was utilized to purify recombinant preS₂ peptide which was solubilized and extracted from sonicated E. coli cells. For the storage of this specific antibody, 0.1 M glycine-HCl buffer (pH 2.5) gave the best result. Other buffers tested in this study caused a significant decrease in antibody reactivity. The binding capacity of an immunoadsorbent column for preS₂ peptide was determined using preS₂-ABEI-H conjugate.

      • Production of anti-$preS_2$, peptide monoclonal antibodies and preparation of anti-$preS_2$, immunoaffinity column

        $PreS_2-{\beta}$-galactosidase 융합단백질을 면역원으로 하여 재조합 $preS_2$ peptide에 대한 단일 클론항체 3종류를 생산하였으며 모두 IgM class로 확인되었다. 그 중 040-01 단일 클론항체를 6% PEG로 처리하여 대장균에서 발현되는 재조합 $preS_2$ peptide를 정제시키기 위한 면역 친화성 column 제조에 이용하였다. Elution buffer로는 0.1M-glycine-HCl(pH 2.5)이 다른 종류의 buffer에 비해 IgM 항체 활성도를 가장 안정되게 유지시켰다. $preS_2$ peptide에 대한 면역 친화성 column의 정제력은 $preS_2$-ABEI.H conjugate를 이용하여 결정하였다.         Creatine Kinase Isoenzyme 의 분리와 정제에 관한 연구

        Bovine creatine kinase MM was purified from bovine heart muscle, through homogenization, ethanol precipitation, DEAE-cellulose chromatography, two step of gel filtration chromatography and affinity chromatography, Human CK-MM wa spurified from fetal human heart muscle through homogenization, ethanol precipitation, and DEAE-cellulose chromatography. Specific activities of bovine and human CK-MM were presented at an activity of 801 U/㎎ and 369 U/㎎, respectively. CK-BB from bovine brain and human fetal brain tissue were purified by homogenization, ethanol precipitation and DEAE-cellulose chromatography. Specific activities of bovine and human CK-BB appeared to 702 U/㎎ and 615 U/㎎, respectively. Ck-MB isoenzyme was synthesized by recombining each single subunit of MM and BB which were prepared by treating with 8 M guanidine-HCl, and its activity was identified on agarose gel electrophoresis.
