        유리체절제술 시 평면부 초음파분쇄술 후 각막내피세포의 장기 변화

        목적: 유리체절제술과 백내장수술을 동시에 시행 받았던 유리체절제술 시 평면부를 통한 초음파분쇄술의 술 후 2년째 각막내피세포 변화를 알아보고자 한다. 대상과 방법: 유리체절제술 시 평면부를 통한 초음파분쇄술을 시행 받은 환자 중 2년 이상 정기적으로 추적관찰하였던 환자 20안을 대상으로 후향적 의무기록을 분석하였다. 경면현미경을 이용하여 수술 전, 수술 후 1달, 3달, 6달, 1년, 2년째 측정한 각막내피세포 밀도(endothelial cell density, ECD), 세포면적변이계수(coefficient of variation, CV), 육각형세포율(hexagonality, HA)을 비교하였다. 결과: 수술 전 측정된 평균 ECD는 1,782 ± 623개/mm2, CV는 35.55 ± 3.03%, HA는 49.86 ± 5.60%였다. 수술 2년 후 측정한 평균 ECD는 1,722 ± 532개/mm2, CV는 35.50 ± 3.03%, HA는 53.00 ± 4.91%로 수술 전과 유의한 차이를 보이지 않았으며 (p=0.9731, 1.000, 0.997), 수술 후 1개월, 3개월, 6개월 및 1년의 시점에 측정한 값과도 유의한 차이를 보이지 않았다. 결론: 유리체절제술 시 평면부를 통한 초음파분쇄술은 수술 전 ECD가 적은 환자에게 수술에 따른 각막내피세포 손실을 최소화할 수 있는 선택 가능한 수술 방법임을 확인하였다. Purpose: To evaluate the long-term changes in cornea endothelial cell density (ECD) after pars plana vitrectomy (PPV) with fragmentation. Methods: Twenty patients (20 eyes) who underwent PPV with fragmentation and who were followed up for 2 years were enrolled in this retrospective study. The cornea ECD, coefficient of variation (CV), and hexagonality (HA) were calculated using a specular microscopy at 1, 3, 6, 12 months, and 2 years after surgery. Results: The preoperative mean ECD was 1,782 ± 623 cells/mm2, and the postoperative mean ECD did not significantly change at 1, 3, 6, and 12 months. Additionally, there were no significant changes in CV or HA. At 2 years after surgery, the mean ECD was 1,722 ± 532 cells/mm2, the mean CV was 35.50 ± 3.03%, and the mean HA was 53.00 ± 4.91%. There were no significant changes in ECD, CV, or HA preoperatively and postoperatively at 1, 3, 6, and 12 months. Conclusions: In this study, the mean ECD did not significantly decrease after PPV with fragmentation at 2 years after surgery. A PPV with fragmentation was an appropriate surgical procedure for patients with low cornea endothelial cell density.

        버섯균사체 배양기질로서의 손바닥선인장의 활용과 그 배양추출물의 기능적 특성

        본 연구는 버섯균사체의 배양 기질로서 손바닥선인장 열매의 활용 가능성을 타진하기 위하여 수행하였다. 5 종류(신령, 잎새, 노루궁뎅이, 차가, 상황)의 버섯균사체를 MEB배지와 손바닥선인장 열매 액체배지(CB)에서 각각 배양하여 에틸아세테이트로 추출한 후, 각 추출물의 수율, 총 폴리페놀 함량, 그리고 몇몇 생리활성을 분석하여 비교하였다. CB 배지에서 배양한 버섯 균사체 중에 잎새버섯 균사체가 추출 수율(0.4 g/l)과 총 폴리페놀 함량(22.7%)에서 가장 양호하였다. CB 배지에서 배양한 버섯 균사체 추출물들은 항산화 활성(DPPH 자유기 소거활성, superoxide의 소거활성, xanthine oxidase 억제 활성), LPS로 활성화시킨 RAW264.7세포에서의 NO 생성 억제 활성, 항균활성, 그리고 HL-60에서의 세포증식 억제 활성 모두가 MEB 배지에서 배양한 버섯균사체 추출물의 것과 비교해 현저하게 증진되어 있음을 확인할 수 있었다. 특히, 잎새버섯 발효추출물에서 DPPH 자유기 소거활성($IC_{50}=362.9{\mu}g/ml$), xanthine oxidase 저해활성(80% at $500{\mu}g/ml$), superoxide 소거활성(80% at $500{\mu}g/ml$), NO 생성 억제 활성($IC_{50}=43.1{\mu}g/ml$), 항균활성, 그리고 HL-60 세포 증식 억제 활성 등 분석된 모든 생리활성이 가장 높게 나타났다. 본 연구 결과는 손바닥선인장 열매가 버섯균사체의 기능성 증진을 위한 배양기질로서 활용될 수 있음을 시사하였다. This study was performed to know the potentialities of the fruits of Opuntia ficus-indica, as a medium for mushroom mycelial culture. Five mushroom mycelial (Agrocus blazei, Grifola frondosa, Hericium erinaceum, Innonotus obliquus, Phellinus linteus) were frown on the malt extract broth (MEB) and the cactus broth medium (CB). The submerged culture mixtures were extracted using equal volume of ethyl acetate, and their extract yields, total polyphenol contents, and some physiological activities were compared with each other Each extract from mycelial culture grown on CB medium showed remarkable enhancement in physiological activities compared with each counterpart grown on MEB. Among five mycelial cultures grown on CB medium, the extract yield and polyphenyl content were highest in the extract from Grifola frondosa (extract yield, 0.4 g/L and polyphenol content, 22.7%). Also, the extracts from Grifola frondosa showed the highest physiological activities, such as DPPH radical scavenging ($IC_{50}=362.9{\mu}g/ml$), xanthine oxidase inhibition (about 80% at $500{\mu}g/ml$), and superoxide radical scavenging (about 80% at $500{\mu}g/ml$), and NO production inhibition ($IC_{50}=43.1{\mu}g/ml$) in LPS-stimulated RAW 264.7 cells. This result suggests that the fruit of Opuntia ficus-indica can be used as a culture medium for improving the functional properties of various mushroom mycelia.

      • Graham Hough의 "Synthesis"理論 序設 : With the problem of criteria as the central figure criteria 문제를 中心으로

        Graham Hough's critical point of view focuses on "a critical synthesis". In short, a complete criticism must arrive at "synthesis" which can be created through the dialectical process between "moral theory" and "formal theory". He also emphasizes that the ultimate purpose of critical activity is "judgement of value". According to Hough's critical attitude, we come to arrive at the conclusion that general criteria for value-judgement may be found in "synthesis" theory. But he evades comment on this criteria. He only represents formal criteria-"integrity" "consonance", "radiance"-and moral criteria-"relevance to life" However, the consistency of Hough's attitude can be derived from the concept of "tension" that he unclearly suggests in "synthesis"theory. If a work of literature is a project, involving both moral and formal activity, the relation between the two sides will be one of tension. So to speak, there always exists "tension"or "a current of energy" in work. Although "tension" may be "strong" or "poor" or "balanced" in the literary work, "tension" can be one of the great criteria in critical in critical literary activity. One is apt to blame Hough's critical attitude as "ecleticism" or "disconsistency". In the chaotic age of criticism, however, it is worthy of his efforts to arrive at the close normative criticism without falling in onesided theory. Furthermore, his critical attitude is very helpful to us as students of literature, no to fall in some critical dogmatism.

      • 科學으로서의 批評 : 그 妥當性과 問題點 : With Northrop Frye's critical principle as the central figure Northrop Frye의 批評原理를 中心으로

        The appearance of New Criticism has brought about the new controversy whether criticism is science or art. In other words, modern critics have classified the function of criticism or its activity into "description" and "value-judgement," and have asserted opposing opinions about this problem. Frye asserts that criticism is science which is the structure of objective knowledge. In short, his critical attitude can be summarized as follow: if criticism is a structure of knowledge, and if knowledge and value are separate, then value cannot be a part of criticism. Finally, he takes the separatism between description and value-judgement. His critical attitude, however, does not accomodate all the basic and important problems of criticism because his principle is very simple and schematic. Krieger and Hirsch dispute Frye's separatism. Krieger insists on anti-separatism, citing Kant's priority of cognition. Hirsch also asserts the necessity of value-judgement, using Kant's aesthetic Eigenschaft. Especially, Hirsch admits the objectivity of value-judgement, referring to Kant's aesthetic universality. In a sense, it seems that he tries to harmonize with the extreme opposite attitudes - Frye's separatiam and Krieger's anti-separatism. If we admit Frye's separatism, firstly we must accept the great cultural sacrifices which abandon artistic quality of literature completely, man's unique aesthetic respones to literature, and its subject value. From this point of view, Frye's assumption-criticism is the structure of knowledge seems to commit "petitio principii. " Secondly, Frye's extreme inductive thought gives rise to the idea that criticism is the attempt of organizing our knowledge of only our past culture. We, however, cannot ignore the importance of deductive philosophy which helps to perceive the present and future culture in the history of human knowledge. However, Frye's critical attitude has some problems which were above mentioned, we cannot deny the significance of his separatism in the history of criticism. When we admit G. Hough's view of the aim of criticism a complete criticism is to "understand literary work rightly on its own plane, and that we may know the better from the worse," Frye's "criticism as science" becomes the first order of work in critical activity.

      • Development of a simple HPLC method for simultaneous determination of medimin E and α-tocopherol in swine serum

        A rapid and simple HPLC method has been developed for simultaneous determination of medimin E. a new a -tocopherol derivate and atocopherol in swine serum. The analytes were extracted with ethanol from 0.2 ml of swine serum and separated by HPLC on a C_(18) reversed-phase column using methanol as a mobile phase and UV detection a t 285 nm. The method was validated over a concentration range of 2-100 μg/ml with a correlation coefficient of 0.998 for medimin E and 0.2-10 μg/ml with 0.996 for a -tocopherol. The lower limit of quantification (LOQ) in serum were 2μg/ml for medimin and 0.2 μg/ml for a-tocopherol: intra- and interday coefficients of variation were ≤13.0% except for LOQ. Advantages of this validated assay include small samples volume. simplified serum extraction, excellent extraction recovery and fast run time. This method can be applied to analysis of medimin E and a-tocopherol in clinical pharma-cokinetics and biological activities studies.
