SKF 525A가 흰쥐의 대동맥에서 Sodium nitroprusside, Acetylcholine, Pinacidil에 의한 이완반응에 미치는 효과

TEA, glibenclamide, L-NAME and SKF 525A-induced contraction were investigated using acetylcholine, sodium nitroprusside (SNP, NO donor) and pinacidil (ATP sensitive K+ channel opener) in rat abdominal and thoracic aorta. The relaxant effects of acetylcholine, SNP and pinacidil were not different in the abdominal aorta and in the thoracic aorta. Acetylcholine-induced relaxation was dependent on endothelial cell, but pinacidil was independent endothelia cell. In the presence of TEA, glibenclamide, L-NAME,mepacrine and SKF 525A, acetylcholine and SNP did not change, but pinacidil-induced relaxation was significantly reduced in presence of glibenclamide, which is ATP sensitive K+ channel blocker. SKF 525A, which is inhibitor of cytochrome P450 dependent epoxygenase, partially inhibited the pinacidil-induced relaxation. These results indicate that the pinacidil-induced relaxation may be mediated by ATP sensitive K+ channel and partially by EETS, which is produced by cytochrome P450 dependent epoxygenase.

The effects of evening primrose oil (EPO). Vit.C and Vit.E on nerve conduction velocity (NCV) in streptozotocin -induced diabetic rats were investigated. 5 weeks- diabetes caused 27±0.5 % reduction in sciatic motor conduction velocity as compared with normal control. Dietary supplement of 5% EPO. 125mg/kg Vit.C. and lg/kg Vit.E prevented significantly the development of the motor nerve conduction velocity deficit about 21±0.5, 21±0.5, 26±0.5% respectively. The total serum cholesterol and triglyceride were decreased by administration 5% EPO. Vit.C, and Vit.E as compared with diabetic control. However, the serum glucose was not decreased by administration 5% EPO. 125mg/kg Vit.C but the serum glucose was decreased by administration lg/kg Vit.E as compared with diabetic control.
Omeprazole-cholestyramine Resin 제제의 인체에 대한 약물동력학적 연구

We have performed the pharmacokinetic study of omeprazole- cholestyramine resinate, which has been developed to increase the stability of omeprazole, the well-known proton pump inhibitor, in an acidic condition. In the human experiments, AUC or T_max of oral administration of omeprazole-cholestyramine resinate is bioequivalent with omeprazole, but C_max was 20% higher in omeprazole-cholestyramine resinate than omeprazole. Omeprazole-cholestyramine resinate suppository showed higher C_max and shortened T_max, so it is expected to show a rapid absorption. These data suggest that omeprazole-cholestyramine resinate is more stable in acid and more rapidly absorbed than omeprazole.
