        DFT방법을 이용한 Bis(μ-oxo)dicopper-enzyme의 수소이동반응 연구

        금속은 생명현상에 필수적인 효소반응에서 핵심적인 역할을 수행한다. 따라서 금속효소 안에서 일어나는 반응에 대한 구체적인 메커니즘은 많은 과학자의 오랜 관심사였다. Methane MonoOxygenase (MMO)는 메탄을 메탄올로 산화시키는 반응을 일으키는 효소이며 최근 Tolman등은 MMO의 모델로서 Bis(μ-oxo)dicopper Complexes의 Hydroxide Transfer 반응에 대한 몇 가지 가능한 메커니즘을 제시하였다. 이후 Hydrogen Transfer와 Hydroxide Rebound의 2단계로 이루어지는 메커니즘이 이론적으로 제시 되었다. 본 연구에서는 Bis($\mu$-oxo)dicopper Complexes 의 반응단계 중 첫 번째 단계인 Hydrogen Transfer 반응과정의 반응물과 전이상태, 생성물의 구조를 최근에 개발된 M06계열의 M06, M06L, M06-2X를 비롯한 여러 가지 DFT방법을 이용하여 계산하였다. M06/6-31G(d,p)/LANL2DZ방법을 사용하여 계산된 반응물의 구조가 실험에서 얻은 XRD구조와 가장 잘 일치했으며 기저함수의 크기에 따라 전이상태의 구조, 활성화 에너지 및 반응에너지에 큰 차이를 보였다. Metals have often played important roles to some enzymatic reactions that are essential to biological processes. Therefore many scientists have studied the reaction mechanisms of catalytic reactions in metaloenzymes for many years. Methane MonoOxygenase (MMO) is an enzyme that oxidize methane to methyl alcohol. Recently Tolman et al. studied a model reaction for MMO, which is a hydroxide transfer reaction in Bis-($\mu$-oxo)-dicopper complex, and suggested several possible mechanisms. Later a two-step mechanism, which is hydrogen transfer followed by hydroxide rebound, was proposed from theoretical studies. In this study we calculated the reactant, product, and the transition state structures, and energetics of the first hydrogen transfer reaction using various DFT methods including recently developed the MO6 family of DFT, namely, MO6, MO6L, and MO6-2X. We found that the M06/6-31G(d,p)/LANL2DZ method reproduce the experimental XRD structure of reactants very well. The TS structures, barrier heights, and reaction energies depend very much on the size of the basis sets.

      • Zygotic Expression of c-myc Gene in Mouse Early Embryos: Functional Role of c-myc Promoter

        c-myc proto-oncogene은 여러 세포들의 분화와 형질전화에 뿐만 아니라 정상세포의 분열조절에도 관여한다고 알려져왔다. 특히 생쥐의 초기배아에서 c-myc mRNA가 발현되고 antisense c-myc oligomer의 미세주입에 의해 배발생이 억제된다는 연구결과는 c-myc이 초기배아의 발생 및 분열에 관여하는 것을 시사한다. 그러나 최근까지 초기배아에 존재하는 c-myc promoter의 기능적 활성화에 관한 연구는 미진하였다. 이를 위하여, c-myc promoter와 대장균의 lacZ 유전자를 결합시킨 두 종류의 vector(pcmyc-Gall, pcmyc-Ga12)를 만들어 수정란의 전핵에 미세주입한 후, 배 발생에 따른 c-myc promoter의 활성화를 lacZ 유전자의 산물인 $\beta$-galactosidase 에 의한 X-gal 염색으로 조사하였다. 미세주입된 초기 배아는 2세포기 배아를 포함하는 여러 발생단계에서 $\beta$-galactosidase 의 활성을 보였다. 이는 c-myc 유전자가 배아의 게놈유전자로부터 발현되며, 또한 궁극적으로 초기 배아의 발생과정에 중요한 역할을 하고 있음을 시사하고 있다. The c-myc proto-oncogene is Involved In the control of normal cell proliferation and differentiation of many cell lineages. Although it has heen suggested that c-myc may play an important role in the mammalian early development, it Is unclear whether the embryonic c-myc mRNA is originated from zygotic gene expression or stored maternal message. Thus, we have construded expression vectors, In which the 5, flanking sequences including c-myc promoter region and a large non-coding exon I are fused 'sith E. coli lacZ gene that encedes $\beta$-galactosldase as a reporter. As c-myc exon I contains a modulatory sequence, we designed t, vo types of vectors (pcmyc.Gall and pcmyc-Ga12) to examine the role of exon I in c-myc expression. The former contains the complete exon I and the later has a deletion in 40 bp of modulator sequence located In the exon I of c-myc These vectors were microInjected into fertilized one-cell embryos and $\beta$-galactosidase activity was examined by X-gal staining during early embryogenesis. $\beta$-galactosidase activity derived from c-myc promoter was decreased at two-cell stage. The expression level directed by pcmyc- Ga12 was similar to that of pcmyc-Gal1, indicating that the medulatory sequence in exon I may not be Involved at least In the regulation of embryonic c-myc expression. In summary, the present study indicates that the c-myc promoter is functional at the early stage embryo, and the regulation of c-myc expression is under the control of "zygotic" clock of preimplantation mouse embryos.e embryos.

      • 의료협력과 국제 의료관광 네트워크에 관한 연구 - 센텀 메디컬 파트너스를 중심으로 -

        Purpose. Medical tourism is recently becoming a new industry with great growth potential. The South Korean government is shifting medical tourism from simple cultural tourism to a high value-added industry with a new paradigm. Methods. The government has been providing positive support and marketing policies since the introduction of the article concerning foreign patient attraction to the medical law in 2009, and various types of medical institutions around the country has participated actively in medical tourism by themselves or in cooperation of government bodies and made increasingly greater performance. Results. This study obtained the following results. The medical institutions in Korea have been making efforts to see more development and profitability in diverse ways, including medical tourism for foreign patients and the advance of the Korean medical institutions into foreign markets. However, many local governing bodies and medical institutions participating in medical tourism around the country have primarily focused on examination and treatment on the basis of foreign patients' visit to South Korea and rarely built a medical network with other countries directly for medical tourism. This study presents a case of building a local medical network and a network for international medical tourism successfully on the basis of the local medical association, CMP, which has been formed naturally in Busan. The success factors for CMP included 1) enthusiasm of the official in charge; 2) the medical level, the service level, and open-mindedness of participant medical institutions; 3) cost efficiency due to executive office management with no costs, no conflicts, and constant partnership; 4) security of non-competitive expertise for participants; 5) local factors of CMP; 6) participation of good agencies; 7) reinforcement of participation networks; and 8) post facto management and local doctor management. Conclusions. Its positive effects included patient introduction and greater profitability on an internal basis as well as construction of the collaboration system with the institutions related to medical tourism and confidence. However, there are some limitations: it is still difficult to predict performance due to the short period of their activities, and it is necessary to continue to observe their constant activities since a single medical association was involved.

The organized characteristics of this study is as follows. 1. 'The harmony of nature (heaven, earth and human)' is the base of medicine. Because medicine control life directly and all life is 'the son of nature'. 2. 'The harmony of Eum and Yang' is the essential power of universe and the key of metabolism of all life including human, animals and plants. Because 'the nature of universe' and all 'Life field' is 'electro-magnetic field'. 3. The Convergence of Oriental medicine and Western medicine orients 'the harmony of Eum and Yang'.

        저자들은 발열, 체중감소 등의 증상과 흉부 X-선 사진상 폐경화의 소견이 있었던 환자에서 폐의 병변이 역형성 거대세포 림프종으로 추측되는 1례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Anaplastic large cell lymphoma(ALCL) is a recently recognized large cell lymphoma characterized by large pleomorphic cells, with prominent nucleoli, expressing the CD30(Ki-1) antigen. It is often misdiagnosed as metastatic carcinoma, malignant histiocytosis or Hodgkin's disease. Primary pulmonary ALCL is extremely rare although it frequently involves extranodal sites, e.g. skin, bone, gastrointestinal tract, soft tissue and lung. We report a case of ALCL suspected as primarily involving lung.

        한국 애니메이션 스토리텔링 전략 연구

        본고는 1990년대 이후 발표된 한국 애니메이션을 중심으로 스토리텔링 전략의 성과와 한계를 파악하고, 그것을 기반으로 실천적인 논의를 활성화시킴으로써 한국 애니메이션 스토리텔링 전략을 생산적으로 탐구하기 위한 것이다. 이것은 애니메이션 스토리텔링의 변별적 자질과 각각의 구현 매체별 특성을 전제로 향유를 활성화시킬 수 있는 스토리텔링 전략이 침체된 한국 애니메이션이 진일보할 수 있는 결정적 기제가 된다는 사실에 기인한 것이다. 애니메이션의 변별성을 바탕으로, 스토리텔링의 다양한 부면을 효과적으로 반영하고 전략적으로 접근하기 위해서는 논의를 세분화하여, 1) 애니메이션 스토리텔링 원천소스 개발, 2) 애니메이션 스토리텔링 전환(adaptation) 전략, 3) 애니메이션 스토리텔링 전략, 4) 애니메이션 스토리텔링 리터러시(literacy)의 네 가지 관점에서 1990년대 이후 한국 애니메이션 스토리텔링 전략을 탐구하였다. 한국 애니메이션 스토리텔링 전략이 보다 생산적인 양상을 드러내기 위해서는 1) 애니메이션 스토리텔링에 대한 변별적 인식의 강화, 2) 스토리텔링을 통한 향유의 활성화 유도, 3) TV용과 극장용 등 매체별로 적합한 스토리텔링 전략 마련, 4) 소스 개발, 장르 간 전환, 구현전략, 리터러시 등의 유기적인 시스템 구축, 5) 수평적/수직적 One Source Multi Use가 강화될 수 있는 스토리텔링 기획 등이 필수적으로 요구된다. 애니메이션 스토리텔링의 실천적 전략을 중심으로 미시적인 전략은 장르, 매체, 향유자 등에 대한 섬세한 파악을 통하여 매우 구체적인 차원에서 논의되어야 할 것이다. This paper aims at understanding storytelling strategy"s result and limitation based on Korean animation released since 1990 and researching storytelling strategy of Korean animation productively, as enhancing the practical argument based on it. That is due to the fact storytelling strategy, which can revitalize enjoyment, is a keyword to enhance stagnant Korean animation, assuming discriminative disposition of animation storytelling and each specific quality by medium. Based on specific character of animation, for reflecting various part of it effectively, and approaching it strategically, I subdivided the argument and tried to research the storytelling strategy of Korean animation released since 1990 by from followed 4 angles. 1) Developing the original source of animation storytelling. 2) Strategy to adapt animation storytelling. 3) Animation storytelling strategy 4) Animation storytelling literacy. Followed are also necessities for storytelling strategy of Korean animation to be prosperous in more productive condition. 1) Understanding animation storytelling discriminately. 2) Trying to actualize enjoyment with storytelling, 3) Planning appropriate storytelling strategy discriminately by medium such as for TV, or theater. 4) Constructing organic system such as source development, adapting to each genre, strategy to realize, and literacy. 5) Planning storytelling to enhance parallel and vertical One Source Multi Use. Based on practical strategy of animation storytelling, micro strategy should be discussed so concretely, while researching genre, medium, and enjoyer.

        합성섬유 여재를 충진한 고속여과장치에 의한 탁수처리에 관한 연구

        Laboratory study was undertaken to pursue the filter performance of a micro-filter module employing highly porous fiber media under a high filtration rate (over 1,500 m/day), faster than that of any conventional filter process. The effects of filtration rate, head loss, raw water turbidity, and filter aid chemicals on filter performance were analyzed. In spite of the extremely high filtration rate, the filter achieved an attractive efficiency, reducing the raw water turbidity by over 80%. As with other filter systems, the filter aid used (PAC in this study) greatly affected the performance of this particular fiber filter. Long term repetitive runs were additionally carried out to confirm the reproducibility of the filter performance. Finally, a comparison was carried out with other high rate filter systems which are either being tested for use in experimental studies, or are already commercially available.

        서사를 활용한 문화콘텐츠 간 One Source Multi Use 활성화 방안 연구

        This article is about researching the strategy for activating One Source Multi Use(OSMU) utilizing storytelling. OSMU is defined as the marketing activity, such as diversifying windows of one source, adapting genre, selling concerned products, and effect of creating brand, for maximizing value added in this article. On that base it is to be examined to activate OSMU with storytelling, dividing parts as to windowing, genre adaptation, merchandising, and creating brand strategically. The most suitable windowing should be found by the advent of new media and the change of revenue structure, and the strategic compression and expansion of storytelling is to be observed to actualize windowing in the process. The order and scope should be decided by discrimination of genre, embodying media, enjoyer, and the condition and scale of market in adapting genre with storytelling. It is also needed acutely to merchandise with systemic interrelation between macro contents and micro contents and plan, develop storytelling based on creating brand. The practical strategy is to be prepared by concrete case-study at this point of time, when the demand on strategy for activating OSMU using storytelling is increasing rapidly.
