        담수환경에서 분리한 곰팡이의 세포외분해효소 활성탐색

        This study aimed to isolate and characterize fungi from freshwater environments in SouthKorea and evaluate their extracellular enzyme activities. Fungal strains were collected fromvarious freshwater sources and identified using phylogenetic analysis. The isolated fungiincluded known aquatic hyphomycetes and previously unreported species. Extracellularenzyme, including those of protease, amylase, lipase, cellulase, laccase, and chitinase,activities were evaluated. Among the isolated strains, several showed high enzyme activity,suggesting their potential role in organic matter decomposition in freshwater ecosystems. This research expands our knowledge of the diversity and enzyme activities of the fungi infreshwater environments, contributing to our understanding of their ecological roles.

        근시(近視)의 침·지압 치료에 관한 고찰 - 한국어권, 영어권, 중국어권을 중심으로 -

        Objectives The purpose of this study is to obtain knowledge and views on treatment of acupuncture and acupressure for myopia. Methods The key words such as 'myopia, acupuncture', 'myopia, acupressure', 'refractive errors, acupuncture', 'refractive errors, acupressure' are used for the research through PubMed. Total 13 studies among the searched 66 were analyzed, except duplicated studies and have little relation to myopia or acupuncture and acupressure. Results Total 13 studies were selected: RCT (5), NRCT (3), UCT (3), Review (2). RCT had two studies on treatment by stimulating auricular acupoints, one study on a treatment by stimulating auricular acupoints and meridian acupoints, one study on a treatment to stimulate auricular acupoints with medication, and one study on electroacupuncture. In NRCT, two studies were on treatment to stimulate auricular acupoints with medication and only one study was on analysis of effect according to intensity of acupuncture. UCT had two studies on acupuncture, and one study on treatment to stimulate auricular acupoints. In Review, there were one study on introduction of Chinese eye exercises, and one study on systematic review for effectiveness and safety of acupuncture. Conclusions The effect on treatment of acupuncture and acupressure for myopia had mostly positive results. However, still not many researches were done to conclude this found. Also, the design of the experiment should be more accurate with systematically designed experimental group and control group.

        Effect of direct current electrical stimulation on osteogenic differentiation and calcium influx

        Electrical stimulation is effective for bone healing. Several studies have shown that electrical stimulation affects cell migration and proliferation. This study examined whether electrical stimulation enhances the osteogenic differentiation of mouse clonal mesenchymal stem cells (cMSCs). Mouse cMSCs were exposed to electrical stimulation at three different voltages (1 V/cm, 0.5 V/cm, and 0.05 V/cm) for one hour per day. The applied electrical stimulation did not affect the cell viability. The alkaline phosphatase (ALP) activity was increased significantly at day 14 after electrical stimulation with 0.5 V/cm for one hour per day. The genes related to osteogenic differentiation were examined at days three, seven, and 14 after exposure to the three electrical stimuli. Osteocalcin (OC) expression was increased dramatically under all three-voltage conditions. These increases were closely related to the electrically stimulated increases in the intracellular calcium concentrations, assessed by Fluo-4 AM on day 14 after exposure to 0.05 V/cm for one hour per day. Furthermore, the expression of calcineurin/NFAT pathway-related genes was upregulated significantly after electrical stimulation. Thus, electrical stimulation enhances the osteogenic differentiation of mouse cMSCs by increasing the intracellular calcium concentration.

        Outbreak of Cucumber mosaic virus and Tomato spotted wilt virus on Bell PepperGrown in Jeonnam Province in Korea

        In August 2006, a severe disease incidence showing mosaic and/or necrotic symptoms on two bell pepper varieties including red-colored ‘Special’ and yellowcolored ‘Fiesta’ was observed in a greenhouse located in Gwangyang, Jeonnam province, Korea. To identify causal viruses, total RNAs were extracted from 11 fruit samples with and without symptoms. Specific oligonucleotide primers for Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Tomato spotted wilt virus (TSWV) and Pepper mild mottle virus (PMMoV) were designed based on the sequences available on GenBank. Database comparisons of the deduced amino acid sequences of each sequence produced 100% and 98% matches with nucleocapsid protein gene of TSWV (Acc. No. ABE11605) and coat protein gene of CMV (Acc. No. DQ018289), respectively, suggesting that the symptoms on bell pepper fruits might be caused by the infection of CMV and TSWV. To our knowledge this is the first report of necrotic as well as mosaic virus disease on bell pepper fruits by the infection of CMV and TSWV in Jeonnam province, Korea.

        First Report of Chrysanthemum (Chrysanthemum morifolium) Crown Rot Caused by Fusarium solani in Korea

        In August 2010, a severe crown rot was observed on chrysanthemum (Chrysanthemum morifolium Ramat.,variety Sinro) in several greenhouses located at Damyang and Muan, Jeonnam province, Korea. Three isolates (EML-CHS1, -CHS2, and -CHS3) of Fusarium were isolated from the affected plants and identified based on morphological characteristics and rDNA internal transcribed spacer (ITS) sequence analysis. Sequence analysis by BLAST indicated that EMLCHS1,-CHS2 and CHS3 were closest to a Fusarium species, F. solani with > 99% sequence similarity. Pathogenicity tests were performed on chrysanthemum with spore suspensions containing 3.4 × 106 spores/ml using the dipping method. Ten days after inoculation, similar symptoms to those observed in the greenhouses were seen on the inoculated plants. The causal fungus was reisolated from the artificially inoculated basal stems,fulfilling Koch's postulates. To our knowledge, this is the first report of crown rot by Fusarium solani on chrysanthemum (Chrysanthemum morifolium) in Korea.

        Assessment of Cre-lox and CRISPR-Cas9 as tools for recycling of multiple-integrated selection markers in Saccharomyces cerevisiae

        We evaluated the Cre-lox and CRISPR-Cas9 systems as markerrecycling tools in Saccharomyces cerevisiae recombinants containing multiple-integrated expression cassettes. As an initial trial, we constructed rDNA-nontranscribed spacer- or Ty4- based multiple integration vectors containing the URA3 marker flanked by the loxP sequence. Integrants harboring multiple copies of tHMG1 and NNV-CP expression cassettes were obtained and subsequently transformed with the Cre plasmid. However, the simultaneous pop-out of the expression cassettes along with the URA3 marker hampered the use of Cre-lox as a marker-recycling tool in multiple integrants. As an alternative, we constructed a set of CRISPR-Cas9-gRNA vectors containing gRNA targeted to auxotrophic marker genes. Transformation of multiple integrants of tHMG1 and NNV-CP cassettes by the Cas9-gRNA vector in the presence of the URA3 (stop) donor DNA fragments generated the Ura- transformants retaining multiple copies of the expression cassettes. CRISPR-Cas9-based inactivation led to the recycling of the other markers, HIS3, LEU2, and TRP1, without loss of expression cassettes in the recombinants containing multiple copies of tHMG1, NNV-CP, and SfBGL1 cassettes, respectively. Reuse of the same selection marker in marker-inactivated S. cerevisiae was validated by multiple integrations of the TrEGL2 cassette into the S. cerevisiae strain expressing SfBGL1. These results demonstrate that introducing stop codons into selection marker genes using the CRISPR-Cas9 system with donor DNA fragments is an efficient strategy for markerrecycling in multiple integrants. In particular, the continual reuse of auxotrophic markers would facilitate the construction of a yeast cell factory containing multiple copies of expression cassettes without antibiotic resistance genes.

        Effectiveness of prophylactic calcium and vitamin D supplementation for preventing post-thyroidectomy hypocalcemia: a meta-analysis

        Background: Postsurgical hypocalcemia is the most common and troublesome consequence of thyroidectomy. We investigated the potential role of routine calcium or vitamin D supplementation in preventing postsurgical hypocalcemia. Methods: We searched MEDLINE and Embase for English-language publications using the keywords “calcium,” “vitamin D,” and “thyroid cancer.” The primary outcome was any postoperative hypocalcemia, and the secondary outcome was symptomatic hypocalcemia. Results: Four studies that included 381 patients were eligible for this meta-analysis. A random-effects model showed no significant difference in the occurrence of hypocalcemia between calcium/vitamin D treatment and placebo/no treatment. However, the occurrence of symptomatic hypocalcemia was lower in patients with calcium/vitamin D treatment. In the combined results, preoperative calcium and vitamin D supplementation were associated with a reduced incidence of symptomatic hypocalcemia. Conclusions: Our findings support the use of preoperative calcium and vitamin D supplementation in conjunction with routine postsurgical supplementation for patients after total thyroidectomy.

        Molecular characterization of the Saccharomycopsis fibuligera ATF genes, encoding alcohol acetyltransferase for volatile acetate ester formation

        Aroma ester components produced by fermenting yeast cells via alcohol acetyltransferase (AATase)-catalyzed intracellular reactions are responsible for the fruity character of fermented alcoholic beverages, such as beer and wine. Acetate esters are reportedly produced at relatively high concentrations by non-Saccharomyces species. Here, we identified 12 ATF orthologues (SfATFs) encoding putative AATases, in the diploid genome of Saccharomycopsis fibuligera KJJ81, an isolate from wheat-based Nuruk in Korea. The identified SfATF proteins (SfAtfp) display low sequence identities with S. cerevisiae Atf1p (between 13.3 and 27.0%). All SfAtfp identified, except SfAtf(A)4p and SfAtf(B)4p, contained the activation domain (HXXXD) conserved in other Atf proteins. Culture supernatant analysis using headspace gas chromatography mass spectrometry confirmed that the recombinant S. cerevisiae strains expressing SfAtf(A)2p, SfAtf(B)2p, and SfAtf(B)6p produced high levels of isoamyl and phenethyl acetates. The volatile aroma profiles generated by the SfAtf proteins were distinctive from that of S. cerevisiae Atf1p, implying difference in the substrate preference. Cellular localization analysis using GFP fusion revealed the localization of SfAtf proteins proximal to the lipid particles, consistent with the presence of amphipathic helices at their N- and C-termini. This is the first report that systematically characterizes the S. fibuligera ATF genes encoding functional AATases responsible for acetate ester formation using higher alcohols as substrate, demonstrating their biotechnological potential for volatile ester production.
