RAPD 분석을 통한 대황(大黃)과 종대황(種大黃) 감별용 SCAR 유전자 마커 개발

Objectives: Due to the morphological similarity and frequent occurrence of intermediate forms as well as morphological variations of aerial part, the correct identification between Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma is very difficult. To develop a reliable method for correct identification and improving the quality standards of Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma, we analyzed RAPD and developed SCAR marker. Methods: To amplify target DNA at the genomic level, 32 Operon 10-mer random primers were applied with four Rheum species, R. officinale, R. palmatum, R. tanguticum and R. undulatum. The nucleotide sequences were determined and species-specific primers were prepared depending on the species-specific RAPD amplicons after subcloned into the pGEM-Teasy vector. To develop the SCAR markers, species-specific PCR amplification and mutiplex-PCR were carried out using the single species-specific primer pairs and combinations of them, respectively. Results: We used RAPD analysis of four Rheum plant species to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed two SCAR markers that amplified 314 bp and 390 bp DNA fragments in only R. undulatum but not in R. officinale, R. palmatum, R. tanguticum and R. undulatum, for distinguishing Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. Conclusions: These genetic markers can be used for the efficient discrimination of plants species and commercial herbal medicines between Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma, to ultimately prevent indiscriminate distribution and prescription of these herbal medicines.

Although operations of implant in dentistry are expensive, operations are increasing every year. However, when operations failed, then it would rise to serious side effects such as malocclusion, falling off, fracture, infection and others. The model is fitness bridge of the set of fixture and alveolus are made by using a CAE software. We have obtained a combination bridge solution by using orthogonal array. Finally, we have obtained a optimum design solution by using orthogonal array.
외대잔대(Adenophora raoemosa J.Lee & S.Lee)개체군의 classification과 ordination 분석

This study was carried out to investigate vegetation structure and soil properties of Adenophora racemosa population distributed in Jeombongsan, Seoraksan, Odaesan and Dutasan in Gangwon-do, Korea. From August 2007 until September 2009, 2 m × 2 m quadrate was established in native area of Adenophora racemosa in order to record a dominants and coverage, and soil factors at 22 sites. It was found that the altitude in the distributed areas for Angelica gigas population was 800 m or more. Adenophora racemosa population was classified into Rhododendron mucronulatum dominant population, Geranium var. hirsutum dominant population, Carex siderosticta dominant population and Aruncus dioicus var. kamtschaticus dominant population. In the site of study, soil pH, electrical conductivity, soil organic matter, available phosphorous, and exchangeable potassimn, exchangeable calcimn, exchangeable magnesium, exchangeable sodium concentration and total nitrogen were ranged from 5.0~7.1, 0.06~0.65dS/m, 0.96~8.94%, 12.3~32.8mg/kg, 0.12~0.89cmol+/kg, 0.34~10.08cmol+/kg, 0.1~l.4cmol+/kg, 0.02~0.29cmol+/kg and 0~8.4% respectively. According to the results of DCCA, Adenophora racemosa population were distributed in the high available phosphorous and exchangeable potassium, and Rhododendron mucronulatum dominant population was situated on low available potassium and high slope degree, Geranium var. hirsutum dominant population was high altitude, electrical conductivity and exchange magnesium, and Carex siderosticta dominant population and Aruncus dioicus var. kamtschaticus were distributed on high total nitrogen, organic matter, available phosphorous.
피마자유와 양명아주 추출물을 원료로 하는 유기농업자재 유효성분의 열 안정성 평가

BACKGROUND: Castor oil and wormseed extract are important active ingredients for biopesticide, and ricinoleic acid in castor oil and three monoterpenes (ascaridole, carvacrol and p-cymene) in wormseed extract are known bioactive substances. However, their stabilities had not been studied, even though the stability was the core property for estimation of shelf-life of biopesticide. Aimed to investigate the thermal stabilities of the bioactive substances in castor oil and wormseed extracts. METHODS AND RESULTS: The contents of ricinoleic acid and three monoterpenes (ascaridole, carvacrol and p-cymene) were analyzed by gas chromatography (GC). The thermal stabilities of the bioactive substance were measured at 0℃, 23℃, 30℃, 40℃, 45℃and 54℃for 84 d. The half-lives of ricinoleic acid in biopesticides was ranged from 28.9 d to 57.8 d at 30℃, and the stability of pure castor oil were located in the range (t<sub>1/2</sub>=46.2 d for Indian product and 27.7 d for Korean product) at the same temperature. The half-lives of the total monoterpenes in biopesticides were ranged from 3.9 d to 27.7 d at 30℃. Among the monoterpenes, the stability ascaridole and p-cymene were decreased in acidic condition. All the bioactive substances showed similar stability on the different thermal conditions. CONCLUSION: The half-lives of most bioactive substance from castor oil and wormseed extracts were less than 100 d. To increase the stability of bioactive substance in biopesticide, stabilizing additives like antioxidant and oxygen remover should be considered to extend of the shelf-life.
