        흑미(Black rice)처리모의 물리적 성상과 형태학적 구조의 변화

        To examine the ingredients of natural hair dye, bleached hair in 7 level was dyed with henna of oxidation hair dye and natural hair dye on the market and then it was dyed with black rice ethanol extract processed by acid or alkali. The coloring, fastness of color and hair damage have compared and analyzed in terms of physical change, observation with the naked eye and microscope. As a result, hair processed by black rice in 3 ways is superb in coloring and fastness in terms of observation with the naked eye, brightness, redness and yellowness. Moreover, black rice-processed hair is considered to be better in thickness, tensile strength and the thickness of hair epidermis compared to oxidation hair dye and be less damaged; therefore, it is dyed peachblow and khaki so it is very practical in stylish dye.

        This study of executive management consulting services for cosmetic recognition (interest / utilization) identify and consulting industries and related institutions are intended to provide the basis. Survey on Current Status and consulting for Jeonju, Iksan, Gunsan, Kimje, Jeongeup the executive management of the beauty shop to 150 people were conducted September, 2011 3-2011 days until September 30, direct interviews fill his way through the survey was carried out statistical analysis using the SPSS WIN 18.0 program were analyzed.         Cystein 처리 후 산화제 반응시간이 컬형성과 컬유지력에 미치는 영향

        The aim of this study was evaluated the formation and sustainability of curl and hair damage minimization during the permanent waving. So that, sodium bromate and hydrogen peroxide as oxidant have been treated with cysteine on normal hair for 5-, 10-, 15- and 20- minutes and then the wave form, physicochemical properties and morphological change have been observed. As the results, optimal reaction time of oxidants has been confirmed as 20-minutes application of sodium bromate and 10-minutes application of hydrogen peroxide. When comparing two oxidants, hydrogen peroxide was excellent in formation rate of wave and sodium bromate was superb in curl sustainability. Considering the decrease in hair thickness was lower in sodium bromate than in hydrogen peroxide but was same in both 10-minutes treatment of sodium bromate and 20-minutes treatment of hydrogen peroxide. In addition, tensile strength was higher in treatment of hydrogen peroxide. When it comes to morphological observation of hair cuticle, hair damage turned out to be low because hydrogen peroxide with strong oxidizing capacity reacts to hair for a very short period of time. Therefore, it is considered that sodium bromate should be used to preserve elastic S-curl for a long time and to reduce the perm frequency and hydrogen peroxide should be treated to curtail the time required in permanent waving and to minimize hair damage.

        뷰티산업의 감성 서비스 요인별 고객만족도

        As emotion came to the fore in such fields of service industry as emotional management, emotional marketing, emotional engineering and emotional leadership in the 21st century, all the cultural fields also have attached great importance to the value of emotion, thus positioning the emotional trend as a new tide of times. Purpose of this study was to examine the literature on the emotional services of existing cosmetic service business and then present the components of emotional service marketing. In addition, the investigator examined the influence of the five emotional service components recognized by beauty salon users upon customer satisfaction and significance, in order to minimize customers` complaint and cope with customer psychology appropriately, thus providing help in establishing emotional service marketing strategies for customers` re-visitation. In order to concretely materialize consumers` emotional desires in the sales of tangible products and intangible services, it is necessary to examine and find out consumers` psychology. So, the investigator established the service areas of five senses as influential variables upon such dependent variables as satisfaction and preference. And the investigator carried out the analysis on satisfaction and significance as parameters and then statistically processed it through a questionnaire survey. Study findings showed that all the service areas of five senses (sight, hearing, taste, touch and smell) had statistically significant influence upon significance and preference. Among them, the sense of sight had the biggest influence upon significance and preference and it was followed by the sense of touch and the sense of smell in order. In addition, study findings showed that beauty salon customers recognized cosmetic services not only by its technologic aspects but also by their five senses` emotional aspects and regarded their five senses` satisfaction with services as their satisfaction with the beauty salon itself. Furthermore, their preferences of a certain beauty salon were influenced by their five senses` satisfaction with services as well.

        안토시아닌과 플라본을 함유한 흑미의 모발 염색력과 모발 보호 효과

        Long term and excessive exposure of chemical hair dyeing cause to hair damage, dermatitis and cancers. In this study, coloring effect, colorfastness and hair damage were investigated in two positive control dyes(oxidizing colorant, henna(Rawonia inermis) and 3 different prepared black rice extracts. Seven hair pieces were divided for normal, bleaching and treatment with oxidizing colorant, henna and 3 kinds of blackrice. Hair pieces and shafts of all pieces were measured color value, thickness and tensile strength and observed change of hair cuticles` structure before and after 30-times shampoo. As the result, the color revealed rose in oxidizing colorant treated hair piece, matt khaki in henna treated hair piece, red-violet in balck rice ethanol extract treated hair, bright red-violet in balck rice ethanol extract and acid treated hair, ash brown in black rice ethanol extract and alkali treated hair. All hair pieces were faded after 30-times shampoo, especially oxidizing colorant treated hair piece was the strongest. Not only thickness and cuticle thickness of hair shafts in henna treated hair shafts and 3 kinds of black rice treated hair shafts before and after 30-times shampoo were thicker, but also tensile strength was stronger compare to oxidizing colorant treated hair shafts. Damage of cuticles showed heavier in oxidizing colorant treated hair shaft than that of henna and 3 kinds of black rice extracts. In conclusion, treatment of black rice extract and acid/alkali can be useful for the natural hair colorant.

        Microbacterium sp. AL-210이 생산하는 levan fructotransferase의 효소활성에 중요한 아미노산의 동정

        [ $\beta$ ]-Fructofuranosidases, a family 32 of glycoside hydrolases (GH32), share three conserved domains including the W(L/M)(C/N)DP(Q/N), FRDPK, and ECP(D/G) motifs. The functional role of the conserved acidic residues within three domains of levan fructotransferase, one of the $\beta-fructofuranosidases$, from Microbacterium sp. AL-210 was studied by site-directed mutagenesis. Each mutant was overexpressed in E. coli BL21(DE3) and purified by using Hi-Trap chelating affinity chromatography and fast performance liquid chromatography. Substitution of Asp-63 by Ala, Asp-195 by Asn, and Glu-245 by Ala and Asp decreased the enzyme activity by approximately 100-fold compared to the wild-type enzyme. This result indicates that three acidic residues Asp-63, Asp-195, and Glu-245 play a major role in catalysis. Since the three acidic residues are present in a conserved position in inulinase, levanase, levanfructotransferase, and invertase, they are likely to have a common functional role as nucleophile, transition state stabilizer, and general acid in $\beta-fructofuranosidases$. 당 분해효소의 family 32 (GH32)에 속하는 $\beta-fructofuranosidase$는 3차구조를 근거로 볼 때 W(L/M)(C/N)DP(Q/N), FRDPK, 그리고 ECP(D/G) 부위를 포함하는 세 군데의 보전적인 영역을 가지고 있다. 이러한 $\beta-fructofuranosidase$ family에 속하는 Microbacterium sp. AL-210 유래 levan fructotransferase (LFTase)의 보전적인 산성 아미노산들의 역할이 특정위치 돌연변이법으로 검사되었다. 각각의 돌연변이체는 대장균인 E. coli BL21 (DE3)균주에서 발현되어 대량 생산되었고, 금속 친화 크로마토그래피법과 FPLC법으로 순수 정제되었다. wild-type LFTase의 효소의 활성은 0.74 unit 인 반면 네 개의 돌연변이체인 D63A, D195N, E245A, E245D 각각은 specific activity를 측정해 본 결과 원 균주와 비교해서 약 100배 정도 감소한 효소활성을 보여 주었다. 이로써 아미노산 변형의 target이 되었던 Asp-63, Aps-195, 그리고 Glu-245가 모두 효소 활성 및 기질과의 결합에 상당히 중요한 역할을 하고 있음이 판명되었다. 이러한 세 부위의 산성 아미노산들은 inulinase, levan fructotransferase와 invertase에 모두 보전적으로 위치 하므로 이들은 $\beta-fructofuranosidase$ family내 에서 공통된 역할을 할 것으로 사료된다.

        E. coli 발현 시스템에 의해 생산된 recombinant human bone morphogenetic protein-2의 정제와 생물학적 활성

        Purpose: Bone morphogenetic protein-2(BMP-2) has been shown to possess significant osteoinducitve potential. There have been attempts to overcome a limitation of mass production, and economical efficiency of BMP. The aim of this study was to produce recombinant human BMP-2(rhBMP-2) from E. coli in a large scale and evaluate its biological activity. Materials and Methods: The E.coli strain BL21(DE3) was used as a host for rhBMP-2 production. Dimerized rhBMP-2 was purified by affinity chromatography using Heparin column. To determine the physicochemical properties of the rhBMP-2 expressed in E. coli, we examined the HPLC profile and performed Western blot analysis. The effect of the purified rhBMP-2 dimer on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity and representing morphological change using C2C12 cell. Results: E. coli was genetically engineered to produce rhBMP-2 in a non-active aggregated form. We have established a method which involves refolding and purifying a folded rhBMP-2 dimer from non-active aggregates. The purified rhBMP-2 homodimer was characterized by SDS-PAGE as molecular weight of about 28kDa and eluted at 34% acetonitrile, 13.27 min(retention time) in the HPLC profile and detected at Western blot. The purified rhBMP-2 dimer stimulated ALP activity and induced the transformation from myogenic differentiation to osteogenic differentiation. Conclusion: rhBMP-2 was produced in E. coli using genetic engineering. The purified rhBMP-2 dimer stimulated ALP activity and induced the osteogenic differentiation of C2C12 cells.
