        연속배양 체제에서의 침출수 미생물에 의한 6가 크롬이온의 환원

        침출수 미생물에 의한 크롬 6가이온의 3가이온으로의 환원을 회분식 배양 및 실험실 규모의 연속배양체제에서 연구하였다. 회분식 배양에 있어서 다양한 범위는 침출수 (10~60%)와 glucose (50~200 mM)를 첨가한 최소배지에서 초기 농도 $20mg\;L^{-1}$의 크롬(VI)을 첨가한 경우 72시간내에 최대 90%의 크롬(VI)이 침출수 미생물에 의하여 환원되었다. 이때 glucose의 첨가는 크롬환원을 촉진시켰으나, 배지내 침출수 비율의 증가는 오히려 크롬환원을 저해하였다. 후자의 경우는 침출수에 존재하는 황산이온이 크롬이온과 경쟁적 저해를 일으키기 때문인 것으로 추정되었다. 연속배양 실험은 124일간 상온에서 크롬(VI)의 농도를 $5{\sim}65mg\;L^{-1}$로 증가시키면서 수행하였으며, 수리학적 체류시간이 36시간일 때 $5{\sim}50mg\;L^{-1}$의 크롬농도 범위에서의 환원은 거의 100%에 달하는 것으로 나타났다. 평균 81.2%의 환원효율을 보인 101~124일 구간에서 계산된 specific reduction rate는 $3.492mg\;g^{-1}\;protein\;h^{-1}$였다. 이상의 결과는 침출수에 존재하는 미생물이 크롬은 함유한 다양한 침출수 또는 피혁폐수의 크롬 무독화과정에 유용하게 쓰일 수 있다는 가능성을 제시해 주는 것이다. Reduction of hexavalent chromium to its trivalent form by leachate microorganisms was studied in batch and bench-scale continuous stirred tank reactor. The inoculum was a culture of microorganisms in leachate and capable of providing up to 90% chromate reduction during 72 h batch assay with $20mg\;Cr(VI)\;L^{-1}$ in minimal media containing different levels of leachate (10 to 60%) and glucose (50 to 200 mM). Addition of glucose increased the efficiency of chromate reduction, but adverse effect was observed with increase of leachate probably due to the competitive inhibition between chromate and sulfate ions. The continuous culture experiment was conducted for 124 days using synthetic feed containing different levels of chromate (5 to $65mg\;L^{-1}$) at room temperature. With a hydraulic retention time of 36 h, chromate reduction efficiency was mostly 100% when Cr(VI) concentrations in the reactor were in the range of 5 to $50mg\;L^{-1}$ Specific rate of Cr(VI) removal was calculated as $3.492mg\;g^{-1}\;protein\;h^{-1}$ during the period of 101~124 days from the start-up which showed 81.2% of average reduction efficiency. The results indicate the potential application of using leachate microorganisms for detoxification of hexavalent chromium in various chromium-contaminated wastewater from landfill or tannery sites.

        공공발주기관의 ERP 적용효과 평가방안 - 사례연구중심으로

        기업의 생산성, 품질향상 및 원가절감 등을 위해 공공기업에서도 ERP시스템을 도입하여 활용중이다. 본 논문은 ERP 도입 후 업무프로세스 개선 정도를 분석하여 ERP 도입 성과를 제시하였다. 이를 위해 K사를 대상으로 업무프로세스 개선이 어느 정도 향상 되었는지 설문을 통하여 자료를 수집하고, AHP기법을 이용하여 평가 방안을 제시하고 분석하였다. 설문을 통해 각 업무 분야별 가중치를 결정하고, 업무개선정도를 평가하기 위해 업무효율성, 업무처리 투명성, 정보공유도 향상, 사용편의성에 대해 ERP 도입 후 효과를 평가하였다. K사가 ERP 도입 초기임을 감안하면 ERP도입의 성과가 발현되는 것으로 판단할 수 있다. 프로세스 개선효과가 가장 크게 나타난 분야는 인사관리로 직원들이 실질적으로 활용 가능한 업무프로세스들이 포함되어 있어 가장 높은 평가를 받았다. 그러나 업무개선정도 지표인 정보공유도 향상은 상대적으로 낮은 평가를 받음으로써 추후 ERP시스템 보완이 필요한 것으로 판단된다. 이와 같이 본 연구에서는 사용자 중심의 주관적인 평가를 AHP기법을 이용하여 ERP 도입성과를 정량화하여 보여줌으로써 공공발주기관의 ERP 도입 성과를 측정 평가하는데 도움을 줄 것으로 기대한다. The public corporations have already implemented ERP to improve productivity, quality, and cost saving. This paper presents the result of business process improvement based on implementation of ERP. Therefore, this paper performed case study of K corporation. Survey was conducted to define the degree of improvement and evaluation method was presented using AHP. The weights for each business area is assigned based on AHP. Then ERP is evaluated in efficiency, work transparency, information sharability, and easy of use. It is meaningful achievement considering its short term implementation. The human resource management shows a relatively larger improvement than other management processes. However, information sharability shows relatively low improvement than others. This quantified result of user-oriented subjective evaluation on ERP implementation will contribute to evaluate the impact of ERP for owner organizations.

      • 유처리제 PRP?瑛? 효능에 대한 연구

        To examine whether PRP, which is a oil spill response product, extensively enhances the biodegradation of crude oil, the bench-scale batch culture test was carried out. Although the initial rate of the crude oil degradation of PRP added sample was increased with a increase of incubation temperature, it was lower than that of control sample. This result was due to the volatilization of low molecular weight components in crude oil, and was confirmed using the headspace gas analysis. Because the PRP interacted with the crude oil to form a PRP/crude oil mixture that was semisolid and relatively stable, volatile hydrocarbons were trapped and the crude oil degradation was hindered by limited oxygen in aggregate during the experiment. After 5 weeks of incubation at 5, 10, 15 and 20°C, the removal rates of the crude oil(1 g/L) in sample were 35, 40, 55 and 80%, respectively. The CO2 produced from the mineralization of crude oil was measured using the Biometer flask method during 2 months. The addition of PRP did not have any enhancing effect on the evolution CO₂ in the crude oil degradation, but it increased the mineralization in the diesel oil degradation. It can be concluded that PRP could not accelerate the degradation rate of crude oil significantly, but it has a high oil-adsorbing capacity.

        중소벤처기업의 경영컨설팅 프로세스가 조직역량과 경영성과에 미치는 영향

        Every country is trying its own best to boost its economy with the advent of global economic downturn in recent years. Korea is promoting small and medium sized venture businesses through creative economy, with the purpose of creating jobs and developing its economy. The researcher`s goal was to find out how business consulting process, supported by governments, associations and educational institutions, affects business performance of small and medium sized business owners. The subject for this research was companies that have received business consulting service for the five years (from 2008 to 2012). The survey was conducted on the internet and mobile devices from 24 June to 5 July 2013. As a result, business consulting process for small and medium sized venture businesses has positive effect on both financial and non-financial performance. It was also found that business consulting process has positive effect on their organizational capabilities, which in turn contribute to financial and non-financial performance. Below are some practical points the researcher would like to suggest based on theoretical findings listed above: First, from an academic perspective, previous research has usually dealt with forms and patterns of consulting as a way of providing business consulting service. Instead, this study proved that organizational capabilities of a company can be improved through a business consulting process. Second, from a business perspective, this research will be a useful guide for organizations in conducting consulting services. Based on these findings, the researcher is convinced that this research could become a good basis for training and instruction materials for small and medium sized venture businesses consulting.

        사상성 진균 Aspergillus nidulans에서 forkhead 유전자인 fkhF의 구조와 기능 분석

        Genome analysis of a model filamentous fungus, Aspergillus nidulans, revealed that there are six putative forkhead genes. Among them, fkhF (AN8949.2) showed A. nidulans-specific. fkhF gene is located in chromosome VII and composed of 2,337 bp coding region for 778 amino acid. Since little is known about the involvement of the forkhead proteins in the developmental process of the filamentous fungi, including A. nidulans, we generated a deletion mutant of fkhF gene and analyzed. Deletion of fkhF resulted in less-dense conidiophore formation in a solid culture. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhF deletion mutant produced conidiophores and conidia under the submerged culture, suggesting that the fkhF gene is involved in repression of inappropriated induction and maturation of asexual developmental process but not in sexual development. 사상성 모델 진균인 Aspergillus nidulans에 존재하는 forkhead 유전자들의 구조와 기능을 체계적으로 연구하기 위해 유전체 분석을 통해 forkhead 도메인을 가지고 있는 6개의 forkhead 유전자를 발견하였다. 이들 중 4개의 유전자들은 다른 진균에서 발견되는 forkhead 유전자들과 전반적인 유사성이 있었으나, 2개의 유전자들은 다른 종에서는 보존되어있지 않은 A. nidulans 특이적인 유전자들이었다. A. nidulans 특이적 forkhead 유전자들 중 하나인 fkhF(AN8949.2) 유전자는 염색체 7번에 위치하고 있고, ORF는 2,337 bp로 구성되어 있으며 778개의 아미노산을 암호화하고 있는 것으로 추정되었다. 예상되는 FkhF 단백질의 N-말단 부위에 보존된 forkhead 도메인을 가지고 있었으며, 이 유전자의 기능을 분석하기 위해 유전자제거 돌연변이 균주를 제조하였다. fkhF 유전자 결손돌연변이주는 유성분화 유도 조건 등을 포함한 여러 고체배지 배양 조건에서 유성분화에는 영향을 미치지 않았으나 무성포자병(conidiophore)의 생성 밀도와 성숙도에 영향을 주는 것으로 관찰되었고, 야생형과 달리 진탕배양시에 무성포자병을 형성함이 관찰되었다. 이는 fkhF 유전자가 A. nidulans의 부적절한 무성분화를 억제하고 정상적인 무성포자 성숙과정에 관여하는 유전자라는 것을 보여준다.

        보문 : Aspergillus nidulans forkhead 유전자 fkhE의 구조와 기능 분석

        A homothallic filamentous fungus Aspergillus nidulans has been used as the a model organism for studying growth and development for eukaryotic system. Various studies about specific transcription factors have been performed for elucidating the molecular mechanisms of growth, asexual and sexual developmental processes. Among them, the fkhE gene (AN2025.3) is located in chromosome VII and contains an ORF encoding 718 amino acid polypeptide intervening with two short introns. The cDNA sequencing revealed that at least four types of alternative splicing events were occurred when the fkhE gene was transcribed. The putative FkhE polypeptide contains a conserved forkhead domain and a bipartite nuclear localization signal at it`s N-terminus and C-terminus, respectively. Deletion of fkhE resulted in impaired conidiophore formation in a solid medium. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhE deletion mutant produced conidiophores and conidia under the submerged culture, indicating that the fkhE gene is involved in asexual developmental process similar to the fkhF gene.

        순창군 장류로부터 분리된 황국균의 동정 및 특성

        In this study, we attempted to isolate fungi from soybean fermented foods produced in Sunchang County and to identify Aspergillus oryzae from fungal isolates. Ten fungal isolates were identified with β-tubulin gene. According to the sequences of β-tubulin gene, ten fungal isolates were identified as A. oryzae/flavus complex. For further identification of the ten of fungal isolates, omtA gene, one gene of the aflatoxin biosynthesis gene cluster, was sequenced and the sequences were compared with those of A. oryzae and A. flavus strains from the GenBank database. In addition, identification of the ten fungal isolates was further confirmed using the PCR amplicon of norB and cypA intergenic region, in which a deletion was recognized relative to A. flavus and A. parasiticus. The amplicon size of the ten fungal isolate strains was smaller than those of A. flavus and A. parasiticus, but the same as that of the reference A. oryzae strain. These results indicated that the ten isolates should be identified as A. oryzae. The protease activity in rice koji made with 6, 13, 17, 27, 37 and 38 of strain, respectively was twice higher than that in control. The kojis made with nine of the A. oryzae isolates, respectively, did not produce aflatoxin, suggesting that the strains could possibly be used as starters for soybean products.
