Background: Ceramide, a major class of integral hair lipid(IHL), may help determine the physicochemical properties of human hairs.Ceramidase(CDase) plays a crucial role in initiating the protective mechanism as it converts ceramide into nonapoptotic metabolites via the hydrolytic pathway. Objectives: The purpose of current study was to investigate the role of CDase in human hair follicles, especially its association with hair cycle. Methods: We observed that hair follicles express CDase at mRNA levels using real time RT-PCR. Furthermore, we examined whether ceramide accumulation induced by N-oleoylethanolamine(NOE), a competitive inhibitor of CDase, influences human hair growth, a change in hair cycle and proliferation/apoptosis-related molecular expression using immunofluorescence stains. Results: As the transition from anagen to catagen progressed, mRNA expression of aCDase and nCDase decreased. NOE inhibited the human hair growth and promoted transformation from anagen to catagen in organ cultured human hair follicles. When the hair follicles were treated with NOE, ki-67 decreased and TUNEL increased. This implies that the hair follicles treated with NOE experience decreased proliferation and increased apoptosis. Conclusion: According to the results above, it is speculated that the IHL of the hair, ceramide in particular, influences hair cycling. In addition, it could be deduced that ceramide hydrolytic effects of CDase have protective effects on growth of the hair follicle.
강원지역에서 10년간(1999~2008) 내원 환자 중 피부악성종양과 피부암전구증에 관한 임상적 고찰

Background: The incidence of cutaneous malignant tumors and premalignant lesions has recently increased according to extended life span, social and medical environment changes. However, a few clinical data have been reported on the survey of cutaneous malignant tumors and premalignant lesions in Gangwon province. Objective: To investigate the incidence and clinical patterns of cutaneous malignant tumors and premalignant lesions in Gangwon province. Methods: We reviewed 485 cases of cutaneous malignant tumors and premalignant lesions from January 1999 to December 2008, at the Department of Dermatology at Wonju Christian Hospital. Results: The average annual incidence of cutaneous malignant tumors among the total number of outpatients was 0.51% and that of premalignant lesions was 0.48%. Both incidences tended to increase with time in 10 year intervals. The most common cutaneous malignant tumor was basal cell carcinoma (47.98%), followed by squamous cell carcinoma (24.19%), extramammary paget`s disease (7.26%), metastatic cancer (5.65%), malignant melanoma (4.44%), and lastly, lymphoma (4.44%). Among premalignant lesions, actinic keratosis (74.68%) and Bowen`s disease (25.32%) were common. The mean age of onset was 67.53 years in cutaneous malignant tumors and 70.51 years in premalignant lesioins. The most common site for both, the cutaneous malignant tumors and premalignant tumors, was the face (63.97% and 81.43%, respectively). The ratio of men to women was 1.11:1 in cutaneous malignant tumors and 1:2.48 in premalignant lesions. Conclusion: The incidence of cutaneous malignant tumors and premalignant lesions has gradually increased in Gangwon province. This data will be made available for the study about the association of cutaneous malignant tumors and premalignant lesions. (Korean J Dermatol 2012;50(2):95~100)

A 52-year-old man presented with a brown to black nodule on his left cheek. Histological examination revealed nests of nevus cells throughout the upper and mid-dermis. An eosinophilic oval-shaped bony structure was found within the nevus. We report here on a case of nevus of Nanta combined with metastatic renal cell carcinoma. A cholesterol embolism is a systemic disease resulting from the occlusion of arteries by cholesterol crystals released from atheromatous plaques. It has been associated with inflammation, sheering forces from blood flow due to hypertension, aneurysm formation, infection, mechanical manipulation during surgery or vascular procedures, and anticoagulation. We report a case of a cutaneous cholesterol embolism in a 66-year old man who had purpuric skin lesions on both feet associated with diabetes mellitus, hypertension, and anticoagulation for a cerebral infarction. Milia are small, white to yellow, benign, superficial keratinous cysts. They are divided into primary or secondary cysts. Secondary milia are associated with disease, medication, and trauma. We report a case of a 45 year-old woman with grouped yellowish papules and brownish patches on her left ankle after bullous cellulitis. This case shows that bullous cellulitis may be one of the causes of secondary milia. (Korean J Dermatol 2011;49(9):840~842)

Background: Recent studies suggest that miR-137 functions as tumor suppressor in various tumors including colorectal cancer, glioblastoma multiforme, oral cancer, and squamous cell carcinoma of the head and neck. The silencing of miR-137 could be related with its abnormal promoter hypermethylation. The purpose of this study was to investigate the significance of MIR137 promoter methylation in lung cancer. Methods: Lung cancer cell lines were treated with DNA methyltransferase inhibitor (5’-Aza) and/or HDAC inhibitor (Trichostatin A) whether miR-137 could be reactivated. From paired tumor and adjacent non-tumor lung tissues (n=50), real-time RT-PCR for miR-137 expression and cyclin dependent kinase 6 (CDK6), quantitative methylation specific PCR for methylation analysis, and bisulfite modified DNA sequencing for validation were used for the study. Results: miR-137 was reactivated by treatment with 5’-Aza and/or Trichostatin A in lung cancer cell lines. miR-137 was significantly downregulated and CDK6 was significantly upregulated in lung tumor tissues compared with adjacent non-tumor tissues. Quantitative methylation specific PCR showed increased MIR137 promoter methylation in lung tumor tissues compared with adjacent non-tumor tissues, which was further validated with bisulfite sequencing. Conclusions: These results suggest that miR-137 has a role of tumor suppressor and its expression is regulated by promoter methylation. The decreased expression of miR-137 combined with increased expression of CDK6 could be associated with lung cancer carcinogenesis.

Background: Clinical usefulness of noninvasive diagnostic methods for the evaluation of liver fibrosis was not fully documented. We evaluated the accuracy of the FibroScan®, acoustic radiation force impulse (ARFI) and real time elastography (RTE) and the efficacy of the addition of the platelet and P2MS to the elastography for predicting liver fibrosis in nonalcoholic liver disease (NALD). Methods: FibroScan®, ARFI and RTE were performed simultaneously at the same day of liver biopsy in 70 patients with various NALD from October 2010 to March 2011. ARFI was performed with SIEMENS ACUSON S2000 ultrasound system and RTE with HITACHI HI VISION-Preirus and EUP-L52 linear probe (3-7MHz). The median values of 10 measurements in ARFI and RTE were calculated, respectively. We used platelet count and P2/MS [platelet count (109/L)]2/ [monocyte fraction(%)×segmented neutrophil fraction (%)] for predicting more accurately liver fibrosis. We defined Vs as velocity of sheer wave (m/s) in ARFI and Es as elasticity score in RTE. Results: From 70 patients, 10 had METAVIR scoring system F0, 4 had F1, 22 had F2, 12 had F3 and 22 had F4. Positive correlations between each methods and fibrosis were found in FibroScan®, ARFI, RTE, P2MS, Vs/PLT, Vs/P2MS, Es/PLT, and Es/P2MS. In the diagnosis of significanct fibrosis (≥F2), the area under the receiver-operating characteristic curve (AUROC)s by FibroScan, ARFI and RTE are 0.689, 0.687 and 0.554. The AUROCs by Vs/PLT and Es/PLT are 0.860 and 0.769. In addition, the AUROCs by P2MS, Vs/P2MS and Es/P2MS are 0.781, 0.778 and 0.746. Comparing with AUROC of each methods for predicting fibrosis (≥F2), Vs/PLT is superior to ARFI (p=0.0018) and Es/PLT is also superior to RTE (p=0.015). Es/P2MS is superior to RTE (p=0.0316), but Vs/P2MS is not significantly different to ARFI (p=0.1491). Moreover, Vs/P2MS is not superior to Vs/PLT (p=0.0084) and not significantly different to Es/PLT (p=0.5481). Conclusion: Both FibroScan® and ARFI are useful noninvasive modalities for the assessment of liver fibrosis. Vs/P2MS and Es/P2MS can be more helpful for predicting liver fibrosis in NALD than elastography alone, but are not superior to Vs/PLT and Es/PLT.

Research has been conducted in many fields for the zero energy of domestic buildings. Among them, the development of insulation has become an essential element. Accordingly, researches are being made to improve the performance of organic insulating materials, and PF boards having the lowest thermal conductivity among organic insulating materials have been in the spotlight. However, problems have arisen due to the problems of durability of insulation materials such as PF boards and past acidification, and the durability of insulation materials is deteriorated when moisture or water enters due to crack gaps during the insulation of the basement layer or the external insulation method. In regard to the durability of the insulation, when the organic insulators of different kinds were placed in water, the pH was weakly basic in all organic insulation materials except PF, and the PF was about 4 pH. As a result, the PF should be continuously reviewed, and further analysis should be carried out to determine what causes acidification.
