      • 담낭질환의 Neuron Specific Enolase 반응세포 출현에 관한 면역조직화학적 연구

        A total 50 patients who had undergone cholecystectomy for cholecystopathy were included in the study to observe the staining pattern of the cells and distribution of antihuman Neuron Spectific Enolase (NSE) antibody by avidin-biotin immunoperoxidase method. The studies consisted of 31 cased of chronic cholecystitis with lithiasis, 9 cases of adenoma and 10 cases of carcinoma including 6 cases of moderately differentiated adenocarcinoma, 3 cased of poorly differentiated adenocarcinoma, and 1 case of small cell carcinoma of combined type. The results were as follows : 1. In the chronic cholecystitis with lithiasis, nine of 31 cases (29%) with the NSE immunoreactive cells, arranged in single of grouped fashion, are focally observed in the covering surface epithelium. 2. The NSE immunoreactivities of carcinoma is more stronger than that of chronic cholecystitis and adenoma. 3. The coarse granular intracytoplasmic strong positive reaction of small cell carcinoma is diffusely observed in noeplastic small cell and glandular epithelium. Therfore, authors suggested that two elements arise from a common primitive cells of endodermal derivation capapble of differentiation along two cell-endocrine ans epithelial.

      • Cadmium과 Di-(2-ethylhexyl) Phthalate에 의한 고환 독성 : 아연과 Apoptosis의 역할 The Role of Zinc and Apoptosis

        Cadmium (Cd) and di-(2-ethylhexyl) phthalate are used widely and distributed ubiquitously in the environment, which are well-known reproductive toxicants. DEHP and/or Cd causes the apoptosis, degeneration of spermatogenic cells, and massive necrosis, resulting in testicular atrophy. Zinc is an essential metal in spermatogenesis, the zinc concentration in testis is changed in DEHP- or Cd-treated animals. However, the role of zinc or apoptosis in testicular toxicity by DEHP or Cd has not been understood well. In this study, Sprague-Dawley male rats were given DEHP (2 g/kg, 5 ml corn-oil, PO) or Cd (CdCl₂2 mg/kg, 4 ml saline, sc) for 1, 2, 7 and 14 days. Control rats were given vehicle corresponding to each group. At day 1, 2, 7, and 14 after administration of DEHP or Cd, the morphological findings including apoptosis in testes were observed under the light microscope, and the zinc concentrations in testes were determined by atomic absorption spectrometry with flame method. Fas and FasL mRNA in testes were analyzed by RT-PCR method at 12 hours, 1, 7, 14 days after adminstration of DEHP or Cd. Absolute and relative testicular weight were lower in DEHP- or Cd-treated rats than control. DEHP or Cd produced morphological changes in testes, including apoptosis, degeneration, necrosis and massive loss of spermatogenic cells, resulting in testicular atrophy. The zinc concentration in testes was not different at day 1 after administration of DEHP, but was decreased at later times. Testicular zinc was lower at day 1, 2 in Cd-treated rats than control, but was higher at 7, 14 days after Cd administration. The levels of Fas and FasL mRNA were highly expressed in testes after administration of DEHP or Cd. These results suggest that the change of zinc level in testes might be a secondary effect of DEHP- or Cd-induced testicular toxicity, and Fas signal system might be play an important role in pocessing of apoptosis in testes by DEHP or Cd.

        Extremely Low Frequency Magnetic Field is an Environmental Stress Factor by Exerting Oxidative Stress

        The previous study reported the biological effect of magnetic field exerted by acting on endocrine and anti-oxidant system. The present study aims to study whether ELF-MF (extremely low frequency magnetic field) affects the physiological endocrine systems such as thyroid and whether ELF-MF affects the defense system against oxidative stress when it alters the function of thyroid. Finally, we correlate the effects of MF on oxidative stress, and adrenal and thyroid with an environmental stress factor. We exposed sham or MF to rats for 5 or 25 days. After the exposure, we determined pain sensitivity, level of TSH, $T_3$ and free $T_4$ in plasma. We also assayed in whole brain, lipid peroxidation, the activity of enzymatic anti-oxidant defense including superoxide dismutase(SOD) and glutathione peroxidase (GPx), and non enzymatic defense such as reduced or oxidized glutathione contents. MF induced the hypersensitivity to thermal stimuli with the reduction of latency. $T_3$ and $T_4$ levels were also increased by the exposure of MF. In addition, we observed the rise of MDA level in rat brain by MF although the MF did not change superoxide dismutase and glutathione peroxidase activity. The effect of MF on both reduced and oxidized glutathione results in decrease in reduced or oxidized glutathione in whole brain. In every experiment, there was no significant difference in MF influence between short term (5 days) and long term (25 days) exposure. Taken together, MF exposure affects the thyroid hormonal control in brain. The elevated thyroid hormone acts on brain, leading to hyper-utilization of oxygen. This phenomenon may be correlated with oxidative stress resulting from MF exposure. In conclusion, we suggest that MF exposure may be an environmental stress by exerting oxidative stress.

        Synergic Effect of Trimebutine Combined with Mosapride on Gastrointestinal Dysfunction and Visceral Pain Induced in Stress Models

        The present study was undertaken to determine whether combined treatment with prokinetic trimebutine and mosapride has a synergic effect on gastrointestinal motility and visceral pain associated with gastrointestinal dysfunction. To develop effective gastroprokinetic agents with greater potencies than trimebutine or mosapride for the treatment of gastrointestinal tract disease, a mixture of trimebutine and mosapride was designed and prepared. In the present study, treatment with trimebutine alone showed a dose-dependent effect on propelling movements of normal small and large intestine in mice, whereas mosapride effected only small intestine motility. Co-administration of trimebutine with mosapride, a well-established prokinetic drug, produced a synergistic influence on normal small intestine motility, but demonstrated an unclear effect on large intestine motility, with a slight tendency to reduce the propelling time. In a stress model, the small and large intestine motilities were significantly decreased. The reduction of intestine motility was restored to a normal level and the restoring effect was more pronounced in the combined treatment with trimebutine plus mosapride than treatment with trimebutine or mosapride alone. Furthermore, treatment with trimebutine plus mosapride significantly decreased acute visceral pain which was not controlled by trimebutine or mosapride alone. These data suggest that combination therapy with trimebutine plus mosapride has a synergic effect on small and large intestine motility and visceral pain control in gastrointestinal disorders

        Adrenal Ganglioneuroma Treated by Laparoscopic Surgery

        Adrenal ganglioneuromas are rare benign tumors arising from the neural crest tissue of the sympathoadrenal nervous system. Because most of these tumors are hormonally silent and patients with them are often asymptomatic, they are often identified incidentally in patients undergoing abdominal imaging studies for unrelated reasons. Diagnosis of this entity is very difficult to make, with definitive diagnosis being based on histological examination. Surgical resection may be both diagnostic and therapeutic. A technique for laparoscopic adrenalectomy of benign adrenal tumors has recently been developed. If the tumor is excised with an adequate resection margin, the postoperative prognosis is very good. In this report, we describe two cases of adrenal ganglioneuroma that were treated using a laparoscopic resection procedure, and we also present a review of the pertinent literature.

        In vivo Tracking of Mesenchymal Stem Cells Labeled with a Novel Chitosan-coated Superparamagnetic Iron Oxide Nanoparticles using 3.0T MRI

        <P>This study aimed to characterize and MRI track the mesenchymal stem cells labeled with chitosan-coated superparamagnetic iron oxide (Chitosan-SPIO). Chitosan-SPIO was synthesized from a mixture of FeCl<SUB>2</SUB> and FeCl<SUB>3</SUB>. The human bone marrow derived mesenchymal stem cells (hBM-MSC) were labeled with 50 µg Fe/mL chitosan-SPIO and Resovist. The labeling efficiency was assessed by iron content, Prussian blue staining, electron microscopy and in vitro MR imaging. The labeled cells were also analyzed for cytotoxicity, phenotype and differentiation potential. Electron microscopic observations and Prussian blue staining revealed 100% of cells were labeled with iron particles. MR imaging was able to detect the labeled MSC successfully. Chitosan-SPIO did not show any cytotoxicity up to 200 µg Fe/mL concentration. The labeled stem cells did not exhibit any significant alterations in the surface markers expression or adipo/osteo/chondrogenic differentiation potential when compared to unlabeled control cells. After contralateral injection into rabbit ischemic brain, the iron labeled stem cells were tracked by periodical in vivo MR images. The migration of cells was also confirmed by histological studies. The novel chitosan-SPIO enables to label and track MSC for in vivo MRI without cellular alteration.</P>

      • 유방의 침윤성 관암종에서 TGF-β isoforms의 발현에 관한 면역조직화학적 연구

        To study the roles of TGF-β related with the aggressiveness of the cancer behavior, the expression of TGF-β isoforms were examined by immunohistochemically in the thirty five cases of infiltrating duct carcinoma of the breast. Analyzed prognostic parameters of the breast cancer along with TGF-β immunohistochemistry were tumor size, axillary lymph node metastasis, clinical stage, histological grade, nuclear grade, estrogen receptor and progesterone receptor status. The results were noted as follows : 1) Of 35 cases of infiltrating duct carcinoma of breast, 34 cases (97.1%) expressed pan TGF-β, 33 cases (94.3%) expressed TGF-β1, 34 cases(97.1%) expressed TGF-β2, and 34 cases(97.1%) expressed TGF-β3. 2) The staining indices (extent X intensity) of TGF-β isoforms were 4.89±2.4 in panTGF-β, 4.69±2.35 in TGF-β, 5.91±2.54 in TGF-β2, and 3.97±2.27 in TGF-β3. 3) The expression of panTGF-β, TGF-β1, and TGF-β2 were closely related to clinical stage and presence of axillary lymph node metastasis(P,0.05). 4) The TGF-β expression was not related to tumor size, histologic grade, nuclear grade, estrogen receptor and progesterone receptor statue(P>0.05). Therefore, the above results suggested that TGF-β expression in infiltrating duct carcinoma of breast may closely related with important prognostic factors, such as lymph node metastasis and clinical stage by increasing the invasiveness of cancer cells.
