        무청 추출물의 생리활성

        건조 무청을 물, 50% 에탄올 및 50% 메탄올로 추출한 물질의 생리활성 효과를 조사하였다. 건조 무청의 추출물 수율은 브랜칭하지 않은 건조 무청인 관동 무청(DRLK)은 26.56∼32.56%이고, 미농단백 무청(DRLM)은 34.34∼36.90%였다. 반면에 브랜칭한 무청군 수율은 18.70∼23.56%로 브랜칭하지 않은 시료보다 수율이 낮았다. 총 폴리페놀 함량의 경우 브랜칭한 무청 시료는 49.06∼59.37 mg/g 범위였고, 브랜칭을 하지 않은 시료는 53.41∼62.08 mg/g이었다. 시료에 따라서는 관동 무청시료(DRLK, BDRLK)보다 미농단백무 시료(DRLM, BDRLM)가 약간 더 높은 폴리페놀 함량을 나타내었다. 전자공여능은 브랜칭하지 않은 시료는 80% 이상의 활성을 보였고, 아질산염 소거작용은 36.63~51.17% 범위의 활성을 보였으며, SOD 유사활성은 38.53~45.38%의 범위의 활성을 보인 반면 브랜칭한 시료는 각각의 생리활성 측정에서 브랜칭하지 않은 시료보다 낮은 활성을 보였다. 전반적으로 관동 무청군이 미농단백 무청군보다 전자공여능, 아질산 소거능 및 SOD 유사활성에 있어서 약간 높은 활성을 보였고, 브랜칭하지 않은 무청이 높은 활성을 나타내었다. This study was conducted to investigate yield of extract, total phenolic compounds content, electron donating activity, nitrite scavenging activity, superoxide dismutase like activity of the radish leaves (Raphanus sativus L.) extracts. Radish leaves extracts were prepared using water extraction, 50% ethanol extraction and 50% methanol extraction. The yield of Kwandong radish leaves extracts without blanching treatment (DRLK) was 26.56∼32.56% and the extract yield of Minongdanbaek radish leaves without blanching treatment (DRLM) was 34.34∼36.90%. On the other hand, the yield of samples with blanching treatment was a lower range value of 18.70∼23.56% than samples without blanching treatment. In the total contents of phenolic compound, samples with and without blanching treatment were 49.1∼59.4 ㎎/g and 53.41∼62.08 ㎎/g, respectively. Minongdanbaek radish leaves extracts (DRLM, BDRLM) showed higher contents value than Kwandong radish leaves extracts (DRLK, BDRLK) in the total phenolic compounds. Samples without blanching treatment showed electron donating activity above 80%, nitrite scavenging activity of 36.63~51.17% and superoxide dismutase like activity of 38.53~45.38%. Generally, Kwandong samples showed a little higher activity more than Minongdanbaek samples in the electron donating activity, nitrite scavenging activity and superoxide dismutase like activity. Also, radish leaves extracts without blanching treatment showed high physiological activities.

      • 뽕나무 부위별 폴리페놀화합물 및 생리활성

        Polyphenolic compounds and biological activity of four different parts (fruit, twig, leaf, root) of two mulberry trees, Morus alba (MA) and Cudrania tricuspidata (CT), were investigated. Yield of the EtOH extracts of four different parts of MA and CT were in that order; fruits > roots > twigs > leaves. The fruits, twigs, leaves, and roots of CT had higher contents of total polyphenol than those of MA, whereas those of MA had higher contents of total flavonoid than those of CT. Among 80% EtOH extracts of MA and CT examined, the EtOH extracts of CT root showed the strongest DPPH radical scavenging activity and α-glucosidase inhibitory activity, meanwhile that of MA root exhibited the strongest tyrosinase inhibitory activity. HPLC analysis revealed that several prenylflavonoids, resveratrol, and mulberrofuran derivatives were present in the root extract of MA, while several flavonoid, prenylflavonoids, and prenylxanthones were in that of CT. These results suggest that roots of CT and MA may be useful as potential sources of dietary supplement with antioxidant, anti-diabetic and anti-melanogenic activity.

        고추씨의 물과 에탄올 추출물의 생리활성

        품종별 고추씨의 추출 수율은 물 추출물 11.30~18.93%, 에탄올 추출물 3.00~5.25%로 에탄올 추출보다는 물 추출물이 더 높은 수율을 보였다. 품종별 고추씨의 총 폴리페놀 함량을 조사한 결과, 물 추출물은 10.22±1.29~25.98±0.55㎎/g의 범위인 반면 에탄올 추출물은 8.00±0.57~33.99±0.09 ㎎/g이었다. 또 품종별 고추씨 용매 추출물의 아질산염 소거 작용을 측정한 결과 물 추출물은 70.11±0.71~94.07±0.86%, 에탄올 추출물은 81.93±1.77~99.90±0.70%의 높은 소거능을 보였고, 전자공여능의 경우 물 추출물은 0.04±0.07~14.31±0.06%로 전자공여능이 낮았으나, 에탄올 추출물은 35.48±0.23~73.83±0.04%의 전자공여능을 보였다. This study was carried out to investigate the extract yield, total polyphenolic compounds content, electron donating activity (EDA) and nitrite scavenging activity (NSA) of various red pepper (Capsicum annuum L.) seeds. The water and ethanol extracts showed yields in the ranges of 11.30~18.93% and 3.00~5.25%, respectively. Ethanol extract yield was higher than water. In the total polyphenol content, water and ethanol extracts were in the range of 10.22±1.29~25.98±0.55 ㎎/ and 8.00±0.57~33.99±0.09 ㎎/g, respectively. Also, nitrite scavenging activities were 70.11±0.71~94.07±0.86% and 81.93±1.77~99.90±0.70% for water and ethanol extracts, respectively. In the electron donating activity, water extracts showed lower activity than ethanol extracts. The electron donating activity of water extracts was in the range of 0.04±0.07~14.31±0.06% while that of NSA of ethanol extracts was 35.48±0.23~73.83±0.04%.

        복숭아 유과 씨 추출물을 이용한 화장품 소재 개발에 관한 연구

        본 연구에서는 복숭아 유과 씨의 품종에 따른 총 폴리페놀 함량과 항산화능, tyrosinase 저해활성 및 elastase 저해활성을 알아보고자 하였으며 폴리페놀 화합물과 총 항산화력 과의 상관관계를 비교?분석하고자 하였다. 품종별 복숭아 유과씨의 추출수율을 관찰한 결과, 몽부사에서 가장 높게 나타났으며, 5.20~6.16%의 수율을 갖는 것으로 관찰되었다. 총 폴리페놀 함량은 18.33~27.08 mg/g의 함량을 나타내었으며, DPPH 및 ABTS radical 소거능에서 천중도의 radical 소거능이 가장 높게 나타났다. Tyrosinase 저해활성에서는 용택골드, 천중도백도에서 높은 tyrosinase 저해능을 보였으나, elastase 저해활성에서는 마도카에서 다른 품종에 비해 유의적으로 높은 elastase 저해능을 보였다. 총 폴리페놀 함량과 항산화활성과 높은 상관관계를 가졌으며 항산화능과 tyrosinase 저해활성 또한 비교적 높은 상관관계를 나타내었다. 따라서 본 연구 결과에 따라 복숭아 유과 씨는 항노화, 미백, 주름방지로 화장품 산업에서 천연 물질로 잠재적인 기능성을 가지고 있을 것으로 사료된다. Various unripe peach (Prunus persica L. Batsch) seeds, which were picked on late May, were subjected to 80% methanol extracts to investigate the antioxidant activities, whitening effects, and anti-wrinkle activities. The yield of 6 cultivars (Takinosawa Gold, Kawanakawase Hakuto, Madoka, Yumefuji, Nagasawa Hakuho, and Hong Bak) from unripe peaches were the highest on Yumefuji, followed by Madoka, Kawanakawase Hakuto, Takinosawa Gold, Hong Bak, and Nagasawa Hakuho (6.16%, 6.08%, 5.65%, 5.25%, 5.23%, and 5.20%). The content of the total polyphenol of seed extracts ranged from 18.33 to 27.08 mg/g. The antioxidant in vitro assays including the DPPH and ABTS radical scavenging activities were the highest with Hong Bak and Kawanakawase Hakuto. The tyrosinase inhibition activity greatly affected the Takinosawa Gold and Kawanakawase Hakuto. Compared to the elastase inhibition activity of 6 cultivars, Madoka was higher than others, while the lowest was Hong Bak. The significant correlation coefficient values were determined (more than p=0.05) between the yield, total polyphenol content, antioxidant activities, tyrosinase inhibition, and elastase inhibition activities. The results suggested that the total polyphenol content has a high positive value on the DPPH, ABTS radical scavenging activity, and tyrosinase inhibition activity. Also, the tyrosinase inhibition activity and elastase inhibition activity showed a significant correlation (r=+0.594). The ABTS inhibition activity and tyrosinase inhibition activity that is the most suitable character represented significant positive correlation (r=+0.846). This data suggested that the methanol extracts from these unripe peaches could be potential candidates for natural cosmetics.

        들깻잎의 caffeic acid와 rosmarinic acid 함량과 항산화 활성 비교를 통한 우수 유전자원 선발

        Perilla is an annual plant in the family Lamiaceae and are widely cultivated in Asian countries. Perilla leaves are important sources of bioactive compounds and are reported to exhibit anti-inflammatory, antibacterial, anti-cancer and antioxidant effects, drawing attention as functional food materials. We examined caffeic acid, rosmarinic acid, total phenol content, and antioxidant activity in the leaves of 18 perilla accessions obtained from the gene bank of the National Agrobiodiversity Center, Jeonju, Korea. The caffeic acid content ranged between 9.86-27.52 ㎎/g with an average content of 17.75 ㎎/g while the level of. rosmarinic acid was in the range between 49.14 and 90.30 ㎎/g with an average content of 61.88 ㎎/g. The total polyphenol content ranged between 138.39 ㎍ GAE/㎎ dried extract (DE) and 378.19 ㎍ GAE/㎎ DE with an average content of 225.93 ㎍ GAE/㎎ DE. Cluster analysis based on the content of caffeic acid, rosmarinic acid. and antioxidant activity showed that the accessions collections were grouped in two distinct classes. The first group contained six genetic resources with high content of rosmarinic acid, and antioxidant activities respectively. The second group contained 12 genetic resources with high content of caffeic acid. These results could help develop new varieties of nutrient dense perilla resources.

        대추 추출물이 장내 미생물의 생육에 미치는 영향 및 항산화 활성

        본 연구에서는 대추 추출물과 장내 유익균의 생육을 조절하여 장내 부패를 방지하기 위한 probiotics로서 유산균을 조합하기 위하여 대추 추출물이 장내 유익균 중에서도 유산균의 생육에 미치는 영향을 in vitro 상에서 알아보고자 하였다. 또한 증식인자로서 대추 추출물에 존재하는 당이 유산균의 생육에 어떠한 영향을 미치는가를 알아보기 위하여 추출방법을 달리하여 당 함량이 다른 대추 1차 및 2차 열수, 에탄올 추출물을 각각 제조하였다. 이와 더불어 지역 특산물인 보은대추를 기능성식품 소재로 활용하기 위하여 페놀성 화합물과 항산화성을 검색하여 기능성식품으로 상품화하기 위한 기본 자료를 확보하고자 하였다. TLC와 HPLC 분석을 통해 장내 유익균의 증식인자로 작용할 수 있는 대추 추출물의 당 조성은 fructose, glucose, sucrose인 것을 확인하였고, 1차 추출물에 비하여 2차 열수 추출물은 fructose, glucose, sucrose가 각각 69, 74, 89% 감소하였으며 2차 에탄올 추출물은 63, 62, 83% 감소한 것을 확인할 수 있었다. 인체 장내세균에 대하여 대추 추출물을 50 mg/mL 농도로 하여 생육증진 효과를 분석한 결과, 대추 1차 열수 추출물에서 B. bifidum과 B. adolescentis가 다른 시험균주들에 비하여 대조군과 대수기에 도달하는 시간은 비슷하였지만, 대조군보다 높은 흡광도 값을 나타내어 생육을 증진시키는 결과를 보였다. 이는 대추 추출물에 존재하는 당이 prebiotics로 작용하여 Bifidobacterium 속의 생육을 증진시킨 것으로 사료되며 인체 장내 균총 조성을 바람직한 방향으로 개선시켜 장내 기능을 향상시킬 것으로 기대된다. 또한 당이 아닌 다른 기능성 성분에 의해 생육이 증진되는지를 알아보기 위해 기능성 물질인 페놀류의 함량과 항산화성을 분석하였다. 총 폴리페놀과 총 플라보노이드 함량을 측정한 결과, 대추 1차 열수 추출물에서 각각 11.63 mg/g, 247.45 μg/g으로 다른 추출물에 비해 높은 함량을 보여 주었다. DPPH에 의한 라디칼 소거능을 살펴본 결과, 대추 추출물의 농도가 높을수록 우수한 전자공여능을 나타내었으며 대추 1차 열수 추출물의 경우 다른 추출물에 비해 가장 높은 85.60%의 전자공여능을 보여 천연 항산화제인 ascorbic acid에 견줄 수 있는 기능성이 있음을 확인하였다. ABTS free 라디칼 소거활성을 통해서도 대추 1차 열수 추출물은 679.91 mg AA eq/g의 항산화력을 지니고 있어 다른 추출물에 비해 가장 좋은 항산화 활성을 나타내었다. 따라서 대추 1차 열수 추출물에는 당뿐만 아니라 항산화능, 폴리페놀, 플라보노이드 함량도 높아 B. bifidum과 B. adolescentis의 생육이 증진된 것으로 사료되며, 기능성 식품소재로 생리활성 성분을 함유하고 있는 대추추출물에 Bifidobacterium 속을 이용하여 유산발효를 진행함으로써 기능성 발효 음료 개발 및 이유식과 같은 다양한 식품 개발에 응용될 수 있을 것으로 판단된다. This study was carried out to investigate the effects of jujube extracts on intestinal microflora, along with their antioxidant activities, according to extraction method. The antimicrobial activities of the extracts were measured using the agar diffusion method with a jujube extract concentration of 50 mg/mL. Neither the first nor second jujube extracts were inhibitory against the tested intestinal bacteria. However, water extracts of jujube significantly enhanced the growth of lactic acid bacteria, especially Bifidobacterium bifidum and Bifidobacterium adolescentis. Total phenol compounds and flavonoid compounds were higher in the 1st than in the 2nd water extracts. The EDA values of both water and ethanol extracts increased in proportion to the extract concentration. The 1st water extract showed the highest value among all the others, which was 85.60% at the concentration of 0.05 mg/mL. Furthermore, the 1st water extract showed stronger antioxidant activity than the other samples with an activity of 679.91 mg AA eq/g. These results support the potential use of jujube water extracts as a functional food component and a valuable resource for the development of nutraceutical foods, to increase the growth of Bifidobacterium spp. in the human intestine.

        함초와 칠면초의 이화학적 특성 조사연구

        This study was conducted to investigate for mineral contents, total polyphenol compounds,betaine, choline and DPPH free radical scavenging activity of halophyte. The mineral concentrations of Salicornia herbacea (top part) were Na 100,006 mg/kg, K 1,385 mg/kg, Mg 6,263 mg/kg, Ca 2,750 mg/kg, Fe 90.4 mg/kg, Mn 98.9 mg/kg, Zn 33.3 mg/kg, Cu 3.4 mg/kg respectively. And Suaeda Japonica (top part) were Na 85,332 mg/kg, K 710 mg/kg, Mg 7,005 mg/kg, Ca 4,344 mg/kg, Fe 1,434.9 mg/kg, Mn 119.1 mg/kg, Zn 19.2 mg/kg, Cu 2.7 mg/kg respectively. The betaine contents of Salicornia herbacea (top part) were 15.09 mg/g and Suaeda Japonica (top part)were 14.64 mg/g. The choline contents estimated by the DBAP-choline derivatives of Salicornia herbacea (top part)were 20.9 mg/100 g, Salicornia herbacea (root) were 23.4 mg/100 g, Suaeda Japonica (top part) were 23.1 mg/100g and Suaeda Japonica (root) were 23.8 mg/100 g. Total polyphenol compounds of Salicornia herbacea (top part) were high 36.0 mg/g in growth phase. The DPPH radical scavenging activities of methanol extract Salicornia herbacea (top part) were high 90.1% in growth phase. The frozen dried powder of Salicornia herbacea (top part) 1 g was equal to Quercetin 30.26 mg, Rutin 42.65 mg, TBHQ 20.32 mg, BHA 25.86 mg, BHT 40.75 mg, Ascorbic acid 22.86 mg in DPPH radical scavenging activities.

        슈퍼홍미 벼품종의 품질특성

        ‘Superhongmi’ is a new rice variety with reddish brown color developed from a crossing a high cyanidin 3-glucoside (C3G) content beeeds including Heugjinju and Suwon 425 and a big grain size breed such as Daeribbyeo. This variety heads on Sep. 5 and has 86.7 cm culm length. The fertility ratio of Superhongmi was about 81.7% and weight of 1,000-grain was about 26.8 g. The 1,000 grain weight of Superhongmi was heavier than that of Superjami. The total essential amino acid content of Superhongmi was lower than those of others. However, Superhongmi was high in tyrosine and arginine. Total polyphenol content of Superhongmi was 2 times higher than that of Heugjinju and flavonoid content was 8 times higher than that of Dongjinbyeo. In addition, hydroxy radical scavenging activity of Superhongmi was significantly higher than that of Dongjinbyeo. However, significant differences were not found in the newly breed colored rice. These results suggest that Superhongmi variety has very high value as a source of various functional food as well as stable food.

        Antibacterial Activity of Puer Tea Extract on Pathogenic Microorganisms

        We have analyzed 11 different commercial brands provided by Daboo Culture and Art Center. This study was carried out to determine the antibacterial activity of water extract from puer tea ingredients against Staphylococcus aureus (S. aureus) and Listeria monocytogenes (L. monocytogenes). Antibacterial activity of the water extracts was shown against S. aureus and L. monocytogenes by paper disc method. In case of S. aureus, antibacterial activity of the extracts appeared as clear inhibitory zone. This antibacterial activity was greater along with pH and showed the highest effect at pH 10. In case of L. monocytogenes, antibacterial activity of the extract appeared as clear inhibitory zone. Some Puer tea appeared to be more powerful in antibacterial activity against S. aureus and L. monocytogenes. The polyphenol compounds were 3.46~8.68%, depending upon samples. In conclusion, puer tea is considered as a good choice for well-being food.
