        This study was performed to investigate the effect of Bunsimgieum on antitumor effect after sarcoma-180 cells transplantation into peritoneal cavity or left groin and immune responses on the depressed immunity induced by methotrexate in mice. The Bunsimgieum extract of 10㎎/㎏ was orally administered 14 days for antitumor effects and 21 days for immune responses. 50% inhibitory concentration(IC_(50)) of SUN-1, SUN-C4. and SUN-396 cancer cell, mean sunvival days and body weight of tumor bearing mice, and growth of tumor mass for antitumor effect; delayed type hypersentivity, hemagglutinin titer, hemolysis titer, rosette forming cells, natured killer cell activity, lymphocyte transformation. productivity of interleukin-2 and phagocytic activity for their immune responses were measured in ICR mice. Significance in antitumor effect is noted in the enlongation of mean life days and inhibition of tumor growth(p<0.01 respectively). Significance of immune responses is also noted in hemolysis titer, lymphocyte transfumotion IL-2 productivity, phagocytic activity, and natural killer cell activity at E/T ratio 100 : 1(p<0.01, respectively). Significant in rosette cell formation was seen dosage of 20㎎/㎏(p<0.01) However, Difference of body weight as antitumor effect, delayed type hypersensitivity, and hemagglutinin titer were not shown significantly. According to the above results, it could be suggested that Bunsimgieum has prominent antitumor and immunity enhancing effect.

        분심기음의 항암작용 및 면역기능에 관한 연구

        Cholecystokinin 길항제인 L-364 , 718이 시험관내 췌장암세포 성장에 미치는 영향

        N/A Background/Aims: It has been reported that some gastrointestinal regulatory peptides including cholecystokinin(CCK) may ac: as growth modulators of pancreatic cancer and CCK antagonists have antitumor effect. However, there have been still debates on the role of CCK antagonists. This study was done to answer the guestions: Does L-364,7 l8 inhibit the growth of cultured pancreatic cancer cells?, What is the growth inhibitory mechanism and is it cell-cycle specific?, and finally, Does the combinatior with L-364,718 and 5-fluorouracil(5-FU) have a synergistic effect on the growth inhibition? Methods: Using Panc-1 and Capan-2 cells, the effects of caerulein, I -364,718 and 5-FU were evaluated, respectively and combined effect of L-364,718 and 5-FU measured. To evaluate the effect of L-364,718 on DNA synthesis and cell cycles, [3H]-thymidine uptake and flow cytometric analysis were performed. The in vitro cytotoxicity was measured by MTT assay and an isobologram model used to analyze the synergism. Results: The both cells, especially Capan-2 cells, grev faster in a medium of CCK supplementation. Dose-dependent powth inhibition o, both cell.', was noted by L-364,718. The IC50 of Panc-1 and Capan-2 cells were 2.43x10-5 M/L and 3.25:cl0-5 M/L, respectively. The IC50 of 5-FU on Panc-I and Capan-2 cells were 13.83 pg/ml and 2.19 pg/ml, respectively. ['H]-thymidine uptake of both cells decreased dose-dependently by L-364,715. The proportion of S phase cells decreased and that of G0/G1 phase cells increased dose-dependently by L-364,718. When L-364,718 was combined with 5-FU, a synergistic effect was observed in 62.5% in Capan-2 cells, however 14.3% in Panc-1 cells. Conclusions: L-364,718 has a antitumor effect on pancreatic cancer cells cultured in vitro and its synergistic effect with certain anticancer agent can be expected. The antiproliferative effect of L-364,718 is considered as the inhibition of DNA synthesis, which cause the blockage of GO/Gl phase of cell cycle.(Korean J Gastroenterol 1996; 28:861-873)

        고삼(苦參)이 항암제(抗癌劑) cisplatin의 간(肝),신장(腎臟) 부작용(副作用) 감소(減少)에 미치는 영향(影響)

        Objective: The objective of this study is to investigate the inhibitor effects of an traditional oriental herb, Sophora flavescens on the hepatic and renal side effects of chemotherapy by using B16-BL6 melanoma-injected C57BL6 mouse tumor model. Methods: In this study, the effects of an traditional oriental herb, Sophora flavescens, on the side effects of chemotherapy were studied using B16 melanoma-injected C57BL6 mouse tumor model. Results: Sophora flavescen has significant effect on the reduction of the side effects of chemotherapy. Sophora flavescen recovered the reduction of WBC and RBC during cisplatin chemotherapy. Water extract of Sophora flavescens significantly inhibited cisplatin-induced increase of serum blood urea nitrogen (BUN) which is a good indicator of renal toxicity. Sophora flavescens extract does not decrease the anti-tumor activity of cisplatin showing that it can selectively inhibit side effects of anticancer drugs preserving beneficial effect. Conclusion: Theses results suggest a possibility that Sophora flavescens extract can be used for cancer patients for the reduction of the side effects and improving the quality of Life during chemotherapy of cancer patients.

        군리탕가감방(君理湯加減方)이 항종양(抗腫瘍) 면역반응(免疫反應)과 항암제로 유발(誘發)한 부작용(副作用)에 미치는 영향(影響)

        Even though appropriate immune response is necessary for the survival of the individual, excessive or insufficient immune Response might cause autoimmune or allergic disease. So the immune response must be controlled to the degree that is beneficial for the well being of the individual. This study was undertaken to know the effects of Gunleetang Gagambang on the immune system of the mouse. Gunleetang Gagambang has been used for cure of tumor as a traditional medicine without any experimental evidence to support the rational basis for its clinical use. This study was carried out to evaluate the possible therapeutic or antitumoral effects of Gunleetang Gagambang extract against tumor, and to carry out some mechanisms responsible for its effect. Some kinds of tumor were induced by the typical application of 3-methylcholanthrene(MCA) or by the implantation(s.c) of malignant tumor cells such as leukemia cells(3LL cells) or sarcoma cells(S180 cells). Treatment of the Gunleetang Gagambang on water-extract(dailly 1mg/mouse, i. p.) was continued for 7 days prior to tumor induction and after that the treatment was lasted for 20 days. Against squamous cell carcinoma induced by MCA, Gunleetang Gagambang decreased not only the frequency of tumor production but also the number and the weight of tumors per tumor bearing mice(TBM). Gunleetang Gagambang on also significantly suppressed the development of 3LL cell and S180 cell-implanted tumors in occurrence-frequency and their size. and some developed tumors were regressed by the continuous treatment of Gunleetang Gagambang extract into TBM. In vitro, treatment of Gunleetang Gagambang extract had no effect on the growth of some kinds of cell line such as FsaII, A431 strain but significantly inhibited the proliferation of 3LL, S180 cells and augmented the DNA synthesis of mitogen-activated lymphocytes. Gunleetang Gagambang also stimulated the migrative ability of leukocyte, the MIF and IL-2 production of T lymphocytes, but not IL 6 production of B cells. Gunleetang Gagambang administration to mice enhanced NK cells activities. These results demonstrated that Gunleetang Gagambang extract exhibited a significant prophylactic benefits against tumors and its antitumor activity was manifested depending on the type of tumor cells. And these results also suggested that effect of Gunleetang Gagambang might be chiefly due to nonspecitie enhancement of NK cell activities and cell-mediated immune responses.

        과학기술(科學技術)의 발전(發展)에 따른 생활(生活) 및 의료(醫療) 수준의 향상(向上)에도 불구하고 산업발전(産業發展)에 따른 환경오염(環境汚染), 스트레스 증가(增加) 등 그 역기능(逆機能) 때문에 발암물질(發癌物質) 또는 자극(刺戟)에의 노출(露出) 등으로 인해서 전세계적(全世界的)으로 암(癌)으로 인(因)한 사망율(死亡率)이 증가(增加)되고 있다. 암(癌)은 현재(現在) 인류(人類)의 건강(健康)을 위협하는 주요(主要)한 질환(疾患)의 하나로, 오늘날 우리 나라에서 질환(疾患)으로 인(因)한 사망원인중(死亡原因中) 수위(首位)를 차지하고 있다. 이러한 암(癌)의 발생(發生)은 80%-90%가 환경적(環境的) 인자(因子)에 기인(起因)한다고 보고(報告)되고 있는데 앞으로 환경성(環境性) 발암물질(發癌物質)이 점차 증가(增加)됨에 따라 암환자(癌患者)의 발생(發生)도 더욱 증가(增加)될 것으로 예상(豫想)된다. 암(癌)의 서의학적(西醫學的) 치료(治療)는 주로 수술요법(手術療法), 방사선요법(放射線療法), 화학요법(化學療法) 및 면역요법(免疫療法) 등이 활용(活用)되고 있다. 이중 수술요법(手術療法)과 방사선요법(放射線療法)은 암(癌)이 전신(全身)에 전이(轉移) 증식(增殖)되기 전에 실시(實施)함으로써 효과(效果)를 거둘 수는 있으나, 암(癌)이 원발병소(原發病巢) 이외(以外)에까지 전이(轉移) 증식(增殖)되었을 경우에는 이 두가지 방법(方法)의 국소요법(局所療法)만으로서는 좋은 치료효과(治療效果)를 거두기 어렵다. 따라서 최근 항암제(抗癌劑)에 의한 화학요법(化學療法)이 다른 방법(方法)보다 유효(有效)하다고 인정(認定)이 되어 가장 많이 활용(活用)되고 있으나, 암종(癌種)에 대한 감수성차이(感受性差異), 치료후(治療後)의 부작용(副作用), 재발(再發) 및 합병증(合倂症) 등의 문제점(問題點)때문에 부작용(副作用)을 감소(減少)시키면서 항암효과(抗癌效果)를 높이는 치료법(治療法)이 계속 연구(硏究)되고 있다. 이에 1972년(年) 미국(美國) 국립암연구소(國立癌硏究所)에서 실시(實施)한 항암제(抗癌劑) 개발연구(開發硏究)에 있어서 항종양성(抗腫瘍性) Screening test에 이용(利用)된 물질중(物質中) 약 47%가 천연물질(天然物質)이었다는 사실은 주목(注目)할만 하다. 한의학적(韓醫學的) 치료법(治療法)은 최근(最近)에 변증시치(辨證施治)를 근간(根幹)으로 하여 건비익기(健脾益氣)를 위주(爲主)로한 부정거사법(扶正祛邪法)과 부정배본법(扶正培本法)이 많이 활용(活用)되고 있으며 부정법(扶正法)으로는 익기건비(益氣健脾), 자음보혈(滋陰補血), 온보비신(溫補裨腎) 등이, 거사법(祛邪法)으로는 활혈거어법(活血祛瘀法), 청열해독법(淸熱解毒法), 연견화담법(軟堅化痰法) 등이 활용(活用)되고 있는데, 이를 근거(根據)로 하여 한약(韓藥)의 복방(複方).단미(單味).단미추출물(單味抽出物) 등의 투여(投與)와 침(鍼) 구(灸) 수침(水鍼) 전침(電鍼) laser침(鍼)등의 치료방법(治療方法)을 수술(手術) 후(後) 혹(或)은 화학요법(化學療法)이나 방사선요법(放射線療法)와 병행(幷行)하여 치료효과(治療效果)가 향상(向上)되었다고 보고(報告)하고 있다. 항암제(抗癌劑)의 항암효과(抗癌效果) 정도(程度)를 검사(檢査)하는 방법(方法)은 크게 5가지로 분류(分類)되는데, 하나는 세포(細胞)를 약제(藥劑)에 노출(露出)시킨 후 현미경(顯微鏡)을 통하여 나타난 형태학적(形態學的) 변화(變化)를 가지고 약제(藥劑)의 효과정도(效果 The sprig of Jinryungtang Gagambang has been used for curing as a traditional medicine without any experimental evidence to support the rational basis for their clinical use. This experiment was carried out to evaluate the possible therapeutic or antitumoral effects of Jinryungtang Gagambang extract against cancer, and to study some mechanisms responsible for its effect. The cytotoxic and antitumor effects were evaluated on human cell liens (A549, hep3B, Caki-1, Sarcoma 180) after exposure to Jinryungtang Gagambang extract using in ILS, colony forming efficency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. As a result of exposure to Jinryungtang Gagambang extract, the proliferation of A549, hep3B, Caki-1, good correlations were shown from the results of SRB assay and those of clogenetic assay. 2. The oral administration of Jinryungtang Gagambang extract showed significant effects of increase of MST(mean survival time) and ILS(increased life span) depending on the increasing concentration. 3. Against squamous cell carcinoma induced by MCA, Jinryungtang Gagambang decreased not only the frequency of tumor production but also the number and weight of tumors per tumor bearing mice(TBM). Jinryungtang Gagambang also significantly suppressed the development of 3LL cell-implanted tumors by frequency and their size, and some developed tumors were regressed by the continuous treatment of Jinryungtang Gagambang extract into TBM. 4. Jinryungtang Gagambang extract also increased NK cell activities. According to the above results, it could be suggested that Jinryungtang Gagambang extract has prominent antiutmor effect.

      • Antitumor effect of dendritic cell loaded <i>ex vivo</i> and <i>in vivo</i> with tumor-associated antigens in lung cancer model

        <P>Various <I>ex vivo</I> or <I>in vivo</I> loading protocols have been developed or evaluated for the delivery of tumor antigens to dendritic cells (DCs). We compared the antitumor effect of mature DCs electroporation-pulsed (EP/mDC) <I>ex vivo</I> with tumor cell lysate and immature DCs (iDCs) injected into the tumor apoptosed by ionizing radiation (IR/iDC) in lung cancer model. DCs were generated from bone marrow of C57BL/6 mice. Ionizing radiation (IR) was applied at a dose of 10 Gy to the tumor on the right thigh. iDCs were intratumorally injected into the irradiated tumor and EP/mDC was injected subcutaneously in the right flank. DC injection induced strong tumor-specific immunity against Lewis lung carcinoma, as compared with the tumor-bearing control and IR only treated mice. The growth of a distant tumor on the right and left flank was inhibited by IR/iDC and EP/mDC. Particularly, IR/iDC resulted in a more significant inhibition of tumor growth and prolonged survival time. It was related to increase of tumor-specific interferon-gamma, cytotoxicity, and decrease of regulatory T-cells. The results indicate that DCs electroporation-pulsed with tumor cell lysate induce a potent antitumor effect, but that iDCs intratumoral injected into the irradiated tumor induce a more potent antitumor effect.</P>

        청열해독복합방의 항종양작용에 대한 실험적 연구

        N/A Objectives : To evaluate the antitumor activity and antimetastatic effects of Cheongyoelhaedokbang(CHB), studies were done experimentally. Method : The study was carried out to evaluate antitumor activity, cytotoxicity, topoisomerase 1 activity, apoptosis and T/C% were measured, and also pulmonary colonization and histological changes of lung in B16-F10 injceted mice, levels of blood vessel formation in the CAM assay and platelet coagulation were measured to prove antimetastatic effects. Results : The results were obtained as follows 1. Treatment of CHB showed cytotoxic effect on A549, SK-OV-3, NIH: OVCAR-3, MCF-7 and MDA-MB-231 cancer cell line by 40% or higher than non-treated control cells at conc. of 100 ㎍/ml. 2. IC_50 value for the inhibition of topoisomerase 1 enzyme activity by CHB was 50-100 ㎍/ml. 3. Treatment of Jurkat cells with CHB and FACS analysis didn`t showed significant changes compared to non-treated control cells. 4. The T/C% was 127.65% in treatment of CHB in S-180 hearing ICR mice. 5. CHB treatment inhibited the adhesion of A549, SK-OV-3 cancer cell line to the complex substrates of culture dish surface. Cell detachment with CHB treatment (100 ㎍/ml) was higher than 40% compared to non-treated control cells. 6. In pulmonary colonization assay, a number of colonies in the lungs were decreased significantly in treatment of CHB compared to non-treated control cells. 7. In the histological changes of lung in B16-F10 injected mice, infiltration area of cancer cells were inhibited effectively in CHB treated group and also infiltration of lymphocytes were shown in pulmonary vein of CHB treated group. 8. In the CAM assay by which the levels of blood vessel formation can be determined quantitatively, CHB treatment inhibited vessel formation by 50%. 9. CHB treatment (10 ㎍/ml or higher concentrations) completely inhibited platelet coagulation by ADP. And also inhibited platelet coagulation by epinephrine in dose dependent manner. Conclusion : From above results it was concluded that CHB could be usefully applied for the prevention and treatment of cancer. and it is still necessary to research on synergic effect with key prescreptions in relation to cancer therapy in oriental medicine
