        盛明巴蜀文人楊慎對孔明的批評

        Yang Shen had made many comments on Kong Ming, which can be seen in his works, poems, poetic criticisms and “the Whole Shu Yiwenzhi”. Yang Shen mainly focused on the analysis of hot issues such as the moral and utilitarian achievements, the academic and governance abilities of Kong Ming. Yang Shen’s purpose in comments on Kong Ming aimed was to admire for former worthies and educate the future generation, not pedantically to look for quotes in order to embellish his writings.

        MLL4 Regulates the Progression of Non-Small-Cell Lung Cancer by Regulating the PI3K/AKT/SOX2 Axis

        Purpose Mixed-lineage leukemia protein 4 (MLL4/KMT2D) is a histone methyltransferase, and its mutation has been reported to be associated with a poor prognosis in many cancers, including lung cancer. We investigated the function of MLL4 in lung carcinogenesis. Materials and Methods RNA sequencing (RNA-seq) in A549 cells transfected with control siRNA or MLL4 siRNA was performed. Also, we used EdU incorporation assay, colony formation assays, growth curve analysis, transwell invasion assays, immunohistochemical staining, and in vivo bioluminescence assay to investigate the function of MLL4 in lung carcinogenesis. Results We found that MLL4 expression was downregulated in non–small cell lung cancer (NSCLC) tissues compared to adjacent normal tissues and tended to decrease with disease stage progression. We analyzed the transcriptomes in control and MLL4- deficient cells using high-throughput RNA deep sequencing (RNA-seq) and identified a cohort of target genes, such as SOX2, ATF1, FOXP4, PIK3IP1, SIRT4, TENT5B, and LFNG, some of which are related to proliferation and metastasis. Our results showed that low expression of MLL4 promotes NSCLC cell proliferation and metastasis and is required for the maintenance of NSCLC stem cell properties. Conclusion Our findings identify an important role of MLL4 in lung carcinogenesis through transcriptional regulation of PIK3IP1, affecting the PI3K/AKT/SOX2 axis, and suggest that MLL4 could be a potential prognostic indicator and target for NSCLC therapy.

        楊愼 《全蜀藝文志》 編輯的文化背景考論

        Yang Shen, in the introduction to “Arts and Literatures History of the Whole Shu nation”, provided that the cultural background of editting this book was to “carry forward the fragrance of the Si Zi, and write the Su nation’s history”. Thus the selected works in this book were not only the literatural ones but also those that reflected the Bashu’s history and academic thoughts. Meanwhile, he had self-consciously assumed the responsibilities of the inheritance of the rural culture and encouraging the backwards through editting the book. Yang Shen and his son had carried forward the local culture through the following three aspects, that is, to devote much attention to the nation’ history, to praise the local loyal bachelors and to place emphasize on the moral education.Besides, Yang Shen and his son had taken great pains to discuss these issues and carry out the social pracitice.

        論錢穆對漢代『春秋』學的硏究

        錢穆對秦漢史、漢代經學研究飮譽學林, 影響很大。他這兩方面的論著大多已逾半世紀, 在出土文獻日益增多、研究方法多元化等學風下, 他的論著有哪些價值?對當代相關研究有什麼影響?筆者結合他這兩方面的研究, 聚焦他的漢代『春秋』學, 以見他治學、見解之一斑。 要探討他對漢代『春秋』學的看法, 則先簡介他對『春秋』及三傳的看法。他與傳統主張孔子作『春秋』的看法相同, 並從史學肯定『春秋』、『左傳』的史學價值。他認為漢代經學今古文之別是王官學與百家言之殊, 而非文字古今之異。這是他論漢代經今古文最具特色的部分。他撰『孔子與春秋』一文, 著重闡釋『公羊』學的精神, 強調孔子『春秋』以百家言而成為新的王官學。他在不同時期對『公羊』學與董仲舒的評價不同。他認為漢儒多通古今, 因此治『春秋』者多能兼採三傳或兩傳之義、說。他又指出, 『春秋』學與漢代思想尤其是災異說、禪讓說、禮制說有密切的關係。 Prof. Qian Mu has a great influence on the honorary scholars of Qin and Han history and Han dynasty studies. Most of his works in these two areas have been over half a century. With the increasing number of unearthed documents and diversified research methods, what is the value of his works? And what is the impact on contemporary related research? This essay combines his research in these two areas and focuses on his study of Chun Qiu in the Han Dynasty, so as to see his academic research methods and insights. To explore his views on the study of Chun Qiu in the Han Dynasty, the first part of the essay introduces his views on Chun Qiu and the three ‘Zhuan’. He is the same as the traditional view that Confucius wrote Chun Qiu, and affirmed the historical value of Chun Qiu and Zuo Zhuan from his historiography. He believes that the difference between modern and ancient literatures in Han dynasty is the difference between Ancient Official Document and Philosophers, rather than the difference between ancient and modern writing. He wrote the article Confucius and Chun Qiu, focusing on explaining the spirit of school of Gong Yang, emphasizing that Confucius' Chun Qiu became a new official ideology with the philosophical thought in Han China. He had different evaluations of Gong Yang and Dong Zhongshu in different periods. He believes that the Han scholars are mostly integrated with the ancient and modern literatures, so most of them who studied Chun Qiu can adopt the meaning and theories of the three or two ‘Zhuan’ at the same time. He also pointed out that the study of Chun Qiu has a close relationship with the thoughts of the Han Dynasty, especially the theory of catastrophe, concession, and etiquette.

        TSHR Variant Screening and Phenotype Analysis in 367 Chinese Patients With Congenital Hypothyroidism

        Background: Genetic defects in the human thyroid-stimulating hormone (TSH) receptor (TSHR) gene can cause congenital hypothyroidism (CH). However, the biological functions and comprehensive genotype–phenotype relationships for most TSHR variants associated with CH remain unexplored. We aimed to identify TSHR variants in Chinese patients with CH, analyze the functions of the variants, and explore the relationships between TSHR genotypes and clinical phenotypes. Methods: In total, 367 patients with CH were recruited for TSHR variant screening using whole-exome sequencing. The effects of the variants were evaluated by in-silico programs such as SIFT and polyphen2. Furthermore, these variants were transfected into 293T cells to detect their Gs/cyclic AMP and Gq/11 signaling activity. Results: Among the 367 patients with CH, 17 TSHR variants, including three novel variants, were identified in 45 patients, and 18 patients carried biallelic TSHR variants. In vitro experiments showed that 10 variants were associated with Gs/cyclic AMP and Gq/11 signaling pathway impairment to varying degrees. Patients with TSHR biallelic variants had lower serum TSH levels and higher free triiodothyronine and thyroxine levels at diagnosis than those with DUOX2 biallelic variants. Conclusions: We found a high frequency of TSHR variants in Chinese patients with CH (12.3%), and 4.9% of cases were caused by TSHR biallelic variants. Ten variants were identified as loss-of-function variants. The data suggest that the clinical phenotype of CH patients caused by TSHR biallelic variants is relatively mild. Our study expands the TSHR variant spectrum and provides further evidence for the elucidation of the genetic etiology of CH.

        Ablation resistance under different heat fluxes of HfC‑ZrC‑SiC multiphase coating prepared by supersonic atmospheric plasma spraying for C/C composites

        To prevent the ablation of C/C composites, an HfC-ZrC-SiC multiphase coating was prepared by supersonic atmospheric plasma spraying. The morphology and microstructure of the coating were examined by X-ray diffraction and scanning electron microscopy, respectively. The as-prepared coating showed a compact structure with a few defects. The ablation resistance test of the coatings was carried out using an oxyacetylene torch under different heat fluxes. The samples ablated under different heat fluxes showed different ablation properties and ablation morphologies. During the ablation, the thickness and weight of the coating decreased with an increase in the heat flux from 2.38 to 4.18 MW/m2. Under the heat flux of 2.38 MW/m2, the coating remained intact because of its low surface temperature and the pinning effect of the newly formed HfSiO4 and ZrSiO4 phases. With an increase in the heat flux, the surface temperature increased beyond the melting point of HfO2 and ZrO2 and the coating suffered a violent flush of the oxyacetylene torch, which resulted in severe evaporation of the oxides and the weakening of the pinning effect.

        TORQUE CONTROL ALLOCATION BASED ON CONSTRAINED OPTIMIZATION WITH REGENERATIVE BRAKING FOR ELECTRIC VEHICLES

        This paper proposes a constrained optimization-based torque control allocation method aimed to improve energy efficiency, and thus, driving range for electric vehicles. In the proposed method, the cost function is defined not only to achieve desired yaw moment for vehicle handling and stability, but also to minimize power losses for energy efficiency. The particular attention is paid to the power losses due to tire slips both longitudinally and laterally. The constraints are also set based on thorough investigation on various causes of power disppation such that the torque is allocated with restraint to use regenerative braking in its maximum capacity. The proposed control allocation method has been tested and verified to be effective on energy efficiency improvement through both simulation and experiment under various driving maneuvers.

        Rhizosphere Microbial Community and Metabolites of Susceptible and Resistant Tobacco Cultivars to Bacterial Wilt

        Soil-borne diseases are closely related to rhizosphere microecosystem. While, plant species and genotypes are important factors affected rhizosphere microecosystem. In this study, the rhizosphere soil microbial community and metabolites of susceptible and resistant tobacco cultivars were investigated. The results showed that there were significant differences in the rhizosphere microbial community and metabolites between susceptible cultivar Yunyan87 and resistant cultivar Fandi3. Furthermore, the rhizosphere soil of Fandi3 showed a higher microbial diversity than that of Yunyan87. The abundance of R. solanacearum was much higher in the rhizosphere soil of Yunyan87 than in the rhizosphere soil of Fandi3, resulting in a higher disease incidence and index. While the abundance of beneficial bacteria in the rhizosphere soil of Fandi3 were higher than that of Yunyan87. Additionally, there were significant differences in metabolites between Yunyan87 and Fandi3 cultivars, and 4-hydroxybenzaldehyde, 3-hydroxy-4-methoxybenzoic acid, vamillic aldehyde, benzoic acid, 4-hydroxybenzyl alcohol, p-hydroxybenzoic acid and phthalic acid were notably high in Yunyan87. Redundancy analysis (RDA) indicated that the rhizosphere microbial community of Fandi3 and Yunyan87 were highly correlated with various environmental factors and metabolites. Overall, susceptible and resistant tobacco cultivars had different impact on rhizosphere microbial community and metabolites. The results expand our understanding of the roles of tobacco cultivars in plant-micro-ecosystem interactions, and provide a basis for the control of tobacco bacterial wilt.

        Transcriptome analysis reveals critical genes and key pathways involved in early phyllotaxy development in maize

        Integrated networks of gene expression, hormonal signaling and metabolite sensing regulate phyllotaxy pattern development. In this study, we characterized differentially expressed genes (DEGs) between maize plants with alternate and opposite phyllotaxies. Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses showed that 2432 DEGs were involved in biological processes, molecular functions, cellular components and many pathways. Importantly, we identified 19 DEGs related to plant hormone signal transduction. Additionally, we identified four main alternative splicing types: skipped exons, retained introns, alternative 50-splice sites, and alternative 30-splice sites, which exhibited different characteristics in the alternate and opposite phyllotaxy libraries. The reliability of the sequencing data was verified through using quantitative real-time reverse transcription PCR analysis of the 19 genes: 15 were validated to play a role in phytohormone signal transduction pathways. Taken together, our data provide new insight into the mechanisms of phyllotaxy pattern development, and will increase our understanding of how relative changes in gene expression determine alternate/opposite phyllotaxy in maize.
