국화(C. morifolium)의 꽃을 80 % MeOH 수용액으로 추출한뒤, 감압농축한 추출물을 EtOAc, n-butyl alcohol 및 물분획으로 나누었다. EtOAc 분획과 n-butyl alcohol 분획에 대하여SiO2 및 ODS column chromatography를 반복 실시하여 4종의 flavonoid를 분리, 정제하였다. Nuclear magnetic resonance, infrared spectrometry 및 FAB-MS data를 해석하여, 화합물 1-4를 각각 apigenin, acacetin, cosmetin (apigenin-7-O-β-Dglucopyranoside), acacetin-7-O-β-D-glucoside으로 구조 동정 하였다. Chrysanthemum morifolium flowers were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned into EtOAc, n-butyl alcohol (n-BuOH), and water fractions. The repeated silica gel and octadecyl silica gel column chromatographies for the EtOAc and n-BuOH fractions led to isolation of four flavonoids. The chemical structures of the compounds were determined as acacetin (1), apigenin (2), apigenin- 7-O-β-D-glucopyanoside (3), acacetin-7-O-β-D-glucopyranoside (4) based on spectroscopic data analyses including nuclear magnetic resonance, mass spectrometry, and infrared spectrometry.
투과선량을 이용한 생체내 (in vivo) 선량측정을 위한 알고리즘

연구 목적 : 각종 암 환자의 방사선치료시 환자에 조사되는 방사선량을 매 치료시마다 간편하게 확인하기 위한 생체내(in vivo) 선량측정의 한 방법으로 투과선량을 이용하는 새로운 시스템에 필요한 알고리즘을 개발하고자 하였다. 이를 위하여 본 교실에서 이미 개발한 바 있는 알고리즘을 개선하고자 하였다. 연구 재료 및 방법 : 알고리즘은 투과선량의 기본 측정치를 A/P (area-perimeter ratio)의 4차 함수로 회귀한 후에 각 계수들을 PCD (phantom-chamber distance)의 3차 함수로 회귀하여 구성하였다. 또한 기본 측정조건들의 감소 가능성을 확인하기 위하여 각각의 PCD 및 Tp(phantom thickness)당 조사야를 8개만 사용한 경우와 15개를 사용한 경우에서 측정치와 계산치를 비교하였다. 임의의 방사선조사조건에서의 알고리즘의 정확도를 확인하기 위하여 기본 측정 조건에 포함되지 않은 장방형의 방사선조사야, 기본 측정에 포함되지 않은 임의의 팬톰 두께, 임의의 PCD 조건에서 측정을 시행하고 알고리즘을 이용한 계산치와 비교하였다. 연구 결과 : 기본 측정치와 알고리즘을 이용한 계산치 간의 오차를 분석한 결과 기존의 알고리즘에 비하여 정확도가 크게 향상되었으며 정방형 개방 조사야의 경우 오차의 범위를 ±0.5% 이내로 제안할 수 있었다. 또한 기본 측정조건을 약 2분의 1로 감소시킬 수 있었다. 임의의 개방조사야의 방사선조사조건에서 측정을 시행하고 알고리즘을 이용한 계산치와 비교한 경우도 대부분의 경우 ±1.0% 이하의 오차를 나타내었다. 결론 : 악성종양환자의 방사선치료시 투과선량을 이용하여 환자에 조사되는 방사선량을 실시간으로 측정하기 위한 생체내 선량측정시스템에 필요한 개선된 알고리즘은 기본 측정자료를 약 2분의 1로 축소한 경우에도 정방형 개방 조사야의 경우 ±0.5% 이하의 오차 범위, 장방형의 개방 조사야에서는 ±1.0% 이하의 오차 범위로 정확히 투과선량을 계산할 수 있음을 확인하였다. The objective of this study was to evaluate effects of Gaeddongssuk powder (GP) on quality characteristics and shelf stability of emulsion sausages during storage. Proximate composition properties showed no significant differences in all treatment (p>0.05). Control showed the highest cooking loss while the treatment with GP showed decreased cooking loss depending on increasing GP content (p< 0.05). Apparent viscosity of batter was increased as the amount of GP increased, whereas hardness of emulsion sausages was decreased with increasing GP level. In sensory evaluation, emulsion sausage with 0.1% GP resulted in the highest score in overall acceptability. The pH values of all treatments decreased at the early storage stage, followed by gradual increase. The lightness and redness of treatments were decreased when the level of GP was increased. However, the yellowness of sausages with GP were higher than that of control (p0.05). Therefore, Gaeddongssuk powder up to 0.1% has a potential as a natural antioxidant for meat products because it can inhibit lipid oxidation of sausages without decreasing their sensory properties.

<P>N-glycan structures released from miniature pig endothelial and islet cells were determined by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF), negative ion electrospray ionization (ESI) MS/MS and normal-phase high performance liquid chromatography (NP-HPLC) combined with exoglycosidase digestion. Totally, the identified structures were 181 N-glycans including 129 sialylated and 18 α-galactosylated glycans from pig endothelial cells and 80 N-glycans including 41 sialylated and one α-galactosylated glycans from pig islet cells. The quantity of the α-galactosylated glycans from pig islet cells was certainly neglectable compared to pig endothelial cells. A number of NeuGc-terminated N-glycans (80 from pig endothelial cells and 13 from pig islet cells) are newly detected by our mass spectrometric strategies. The detailed structural information will be a matter of great interest in organ or cell xenotransplantation using α 1,3-galactosyltransferase gene-knockout (GalT-KO) pig.
