Although mammalian sperms are cryopreserved for in vitro fertilization a process of cryopreservation decreases the fertility. Acrosome reaction requires depolarization-induced ca^(2+) influx and ca^(2+) releases from the ca^(2+) stores. To examine whether the cellular ca^(2+) mobilization is altered by a sperm cryopreservation we compared cytosolic ca^(2+) signals between fresh and cryopreserved pig sperms using confocal ca^(2+) imaging. The magnitudes of depolarization-induced ca^(2+) increases were significantly smaller in cryopreserved sperms. Exposures to 10 mM caffeine or 5 μM thapsigargin elicited less ca^(2+) increases in the cryopreserved sperms compared to fresh sperms. In addition, progesterone-triggered ca^(2+) rises, that are thought to enhance acrosome reaction, were completely abolished in the cryopreserved sperms. These results suggest that storage and(/or) release of ca^(2+) from the intracellular ca^(2+) stores in pig sperms are significantly impaired by the process of cryopreservation.

It is necessary to add protective agents for protecting the probiotic viability in the preparation process of probiotics starter. In this study, we used whey protein concentrate (WPC), pullulan, trehalose, and sodium glutamate as the protective agent and optimized the proportion of protective agent and spray-drying parameters to achieve the best protective effect on Lactobacillus plantarum. Moreover, the viable counts of L. plantarum in starter stored at different temperatures (–20℃, 4℃, and 25℃) for 360 days were determined. According to response surface method (RSM), the optimal proportion of protective agent was 24.6 g/L WPC, 18.8 g/L pullulan, 16.7 g/L trehalose and 39.3 g/L sodium glutamate. The optimum spray-drying parameters were the ratio of bacteria to protective agents 3:1 (v: v), the feed flow rate 240 mL/h, and the inlet air temperature 115℃ through orthogonal test. Based on the above results, the viable counts of L. plantarum was 12.22±0.27 Log CFU/g and the survival rate arrived at 85.12%. The viable counts of L. plantarum stored at –20℃ was more than 1010 CFU/g after 200 days.
Effects of Vietnamese Plant Extracts on L-type Ca2+ Channel in Rat Ventricular Myocytes

In mammalian cardiac myocytes, influx of Ca2+ through the L-type Ca2+ channel triggers Ca2+ release from the sarcoplasmic reticulum during action potential, which induces contraction. Blockers of L-type Ca2+ channel have been used to treat several cardiac diseases including hypertension and heart failure. In the present study, we screened the effects of herbal extracts, originated from Vietnam, on L-type Ca2+ current (ICa) using whole-cell patch-clamp technique in isolated adult rat ventricular myocytes. Among seventeen extracts tested, three showed significant suppressive effects on ICa. Butanol extract of Celastrus sp. leaves, hexane extract of the whole plant of Bousingonia mekongense, and methanol extract of the stem of Phyllanthus reticulatus significantly decreased the magnitude of ICa by 9.5 ± 2.1%, 13 ± 2.5%, and 68 ± 9.2%, respectively, at the concentrations of 3 μg/ml. These results suggest a possible use of these substances as drug-candidates for cardiac diseases involved.

This paper investigates the traffic offloading over unlicensed bands for two-tier multi-mode small cell networks. We formulate this problem as a Stackelberg game and apply a hierarchical learning framework to jointly maximize the utilities of both macro base station (MBS) and small base stations (SBSs). During the learning process, the MBS behaves as a leader and the SBSs are followers. A pricing mechanism is adopt by MBS and the price information is broadcasted to all SBSs by MBS firstly, then each SBS competes with other SBSs and takes its best response strategies to appropriately allocate the traffic load in licensed and unlicensed band in the sequel, taking the traffic flow payment charged by MBS into consideration. Then, we present a hierarchical Q-learning algorithm (HQL) to discover the Stackelberg equilibrium. Additionally, if some extra information can be obtained via feedback, we propose an improved hierarchical Q-learning algorithm (IHQL) to speed up the SBSs` learning process. Last but not the least, the convergence performance of the proposed two algorithms is analyzed. Numerical experiments are presented to validate the proposed schemes and show the effectiveness.
Determining Osteogenic Differentiation Efficacy of Pluripotent Stem Cells by Telomerase Activity

BACKGROUND: Bone tissue engineering based on pluripotent stem cells (PSCs) is a new approach to deal with bone defects. Protocols have been developed to generate osteoblasts from PSCs. However, the low efficiency of this process is still an important issue that needs to be resolved. Many studies have aimed to improve efficiency, but developing accurate methods to determine efficacy is also critical. Studies using pluripotency to estimate efficacy are rare. Telomerase is highly associated with pluripotency. METHODS: We have described a quantitative method to measure telomerase activity, telomeric repeat elongation assay based on quartz crystal microbalance (QCM). To investigate whether this method could be used to determine the efficiency of in vitro osteogenic differentiation based on pluripotency, we measured the pluripotency pattern of cultures through stemness gene expression, proliferation ability and telomerase activity, measured by QCM. RESULTS: We showed that the pluripotency pattern determined by QCM was similar to the patterns of proliferation ability and gene expression, which showed a slight upregulation at the late stages, within the context of the general downregulation tendency during differentiation. Additionally, a comprehensive gene expression pattern covering nearly every stage of differentiation was identified. CONCLUSION: Therefore, this assay may be powerful tools for determining the efficiency of differentiation systems based on pluripotency. In this study, we not only introduce a new method for determining efficiency based on pluripotency, but also provide more information about the characteristics of osteogenic differentiation which help facilitate future development of more efficient protocols.

Proton-conducting solid oxide fuel cell (H-SOFC) based on layered perovskite type GdBaCuCoO5+x (GBCC) cathodewas fabricated with in situ drop-coating BaZr0.1Ce0.7Y0.2O3−δ (BZCY) electrolyte membrane. The influences of Cu dopinginto Co sites of GdBaCo2O5+x on the electrical conductivity and conduction mechanism, thermal expansion propertyand electrochemical performance of cathode materials and corresponding single cell were investigated. Results show thatthe electrical conductivity decreased and the conduction mechanism would gradually transform to the semiconductor-likebehavior. A high maximum power density of 480 mW cm−2 was obtained for the anode supported NiO–BZCY/NiO–BZCY/BZCY/GBCC single cells with wet H2fuel at 700 °C. The corresponding polarization resistance was as low as 0.17 Ω cm2. The excellent electrochemical performance of as-prepared single cell indicates that GBCC is a good candidate of cathodematerials for H-SOFCs.
