Comparative analysis of yeast cell viability at exponential and stationary growth phases

This paper describes a comparative analysis of yeast cell viability at exponential and stationary growth phases using multiple conventional techniques and statistical tools. Overall, cellular responses to various viability assays were asynchronous. Results of optical density measurement and direct cell counting were asynchronous both at exponential and stationary phases. Proliferative capacity measurement using SP-SDS indicated that cells at the end of the stationary phase were proliferative as much as exponentially growing cells. Metabolic activity assays using two different dyes concluded that the inside of cells at stationary phase is slightly less reducing compared to that of exponentially growing cells, implying that the metabolic activity imperceptibly declined as cells were aged. These results will be helpful to understand the details of yeast cell viability at exponential and stationary growth phases.

최근 페이스북, 트위터 그리고 카카오톡과 같은 SNS (Social Network Service)는 많으 사람들에게 인기를 얻고 있다. 사용자들은 SNS를 통해 자신의 일상을 다른 사람들에게 가시화하고, 오프라인에서 서로 모르는 사람과 친구 관계를 맺으며 서로의 생각이나 정보 등을 실시간으로 공유한다. 또한 SNS을 통해 연구 설문조사가 이루어지기도 하고, 기업은 마케팅, PR활동 그리고 고객 관리까지 SNS을 이용하고 있다. 본 논문에서는 SNS가 가진 장단점을 면밀히 분석하여 구글의 개방형 모바일 플랫폼인 안드로이드기반의 소셜 네트워크 애플리케이션, '슈게더'를 제작하였다. '슈게더'애플리케이션을 이용해 서울여대 학생들은 모르고 지냈던 사람들과의 관계 형성을 할 수 있으며, 교내에서 본인의 관심사를 타인과 함께 나눌 수 있는 효과를 기대할 수 있다.

<P><B>Abstract</B></P> <P>Bilirubin (BR) is generated by the reduction of biliverdin (BV), a metabolite that results from the catalytic degradation of heme by the isoforms of heme oxygenase (HO). BV is nontoxic and water-soluble but BR is potentially toxic and lipophilic. Therefore, a further metabolic step is required for BR before excretion is possible. The reductive conversion of BV to BR costs energy and is evolutionarily conserved in human physiology. There must be a compelling reason for this apparently nonsensical evolutionary conservation. In addition to the differences between BR and BV—such as water solubility, antioxidant activity, and participation as a receptor ligand—in the present study, we focused on the chemistry of the two metabolites with regard to an electrophilic functional group called a Michael reaction acceptor (MRA). Our data reveal that the BR reacts with thiol compounds forming adducts, whereas no reaction occurs with BV. Furthermore, the binding of biotin-tagged BR to Kelch-like ECH-associated protein 1 (KEAP1)—a biological electrophile sensor—was prevented by pretreatment with BR or a thiol compound, but was not by pretreatment with BV. In cells, BR could bind to KEAP1 to release and activate nuclear factor-erythroid 2 (NF-E2) p45-related factor 2, a cytoprotective transcription factor, leading to the induction of HO-1. 울금 주정 추출물이 자연살해세포와 T 면역세포에 미치는 영향

본 연구에서는 국내에서 재배되는 울금의 면역조절 효과에 대해 평가하고자 20% 주정 추출물을 이용하여 자연살해세포와 T 세포에 미치는 영향을 관찰하였다. 마우스의 비장세포에서 분리한 자연살해세포를 종양세포 YAC-1 세포와 함께 배양시켜 울금 20% 주정 추출물의 처리에 따른 변화를 관찰하였다. 그 결과 울금 20% 주정 추출물의 처리는 자연살해세포의 CD69 발현과 IFN-γ의 발현을 증가시켰고 결과적으로 활성이 증가되어 YAC-1 세포의 제거를 증가시켰음을 확인하였다. 또한 마우스의 비장세포에서 울금 20% 주정 추출물의 처리에 따른 T 세포의 변화를 관찰한 결과에서는 CD4+ T 세포보다는 CD8+ T 세포를 증가시켰음을 확인하여 감염된 세포나 종양세포 제거를 효과적으로 할 수 있을 것이라고 예상할 수 있었다. 결론적으로 울금 20% 주정 추출물은 내재면역과 적응면역에 영향을 미쳐 면역조절에 긍정적인 변화를 보였음을 확인하였다. The present study investigated the immunomodulatory effects of Curcuma longa L. ethanol extracts on natural killer (NK) cells and T cells. We treated Curcuma longa L. ethanol extracts at concentrations of 20, 50, 100, and 150 μg/mL to murine NK cells co-incubated with YAC-1 cells. Curcuma longa L. ethanol extracts resulted in increased NK cell activity compared to the control group at all concentrations. In the groups treated with Curcuma longa L. ethanol extracts, CD69 and IFN-γ expression levels significantly increased compared to the control group at 100 and 150 μg/mL. In addition, Curcuma longa L. ethanol extracts induced significant elevation of CD8+ T cell numbers in a dose-dependent manner. However, Curcuma longa L. ethanol extracts also led to reduction of CD4+ T cell and MHCⅡ numbers. The findings of this study suggest that Curcuma longa L. ethanol extracts could enhance the immune response through activation of NK and cytotoxic T cells due to a proliferative shift of antigen presentation from MHCⅡ to MHCⅠ, presumably.
