Aims: Daclatasvir (DCV) plus asunaprevir (ASV) has demonstrated highsustained virologic response (SVR) in HCV genotype (GT-)1b infection.NS5A-Y93H and NS5A-L31 resistance-associated polymorphisms(RAPs) to DCV are known to impact DCV+ASV response in GT-1b-infectedJapanese. The effect of RAPs on SVR at posttreatment week12 (SVR12) to DCV+ASV was explored in mainland Chinese, Korean,and Taiwanese.Methods: Pooled data from 2 studies of DCV (60 mg daily) + ASV(100 mg capsule, twice-daily) for 24 weeks in GT-1b-infected interferon/ribavirin-naive and -experienced patients from mainland China,Korea, and Taiwan. Similar Japanese data (4 studies; n=445) werepooled for comparison. SVR12 with versus without baseline Y93Hand/or L31 RAPs was compared by age (<65 vs ≥65 years), cirrhosisstatus, and baseline HCV-RNA.Results: SVR12 and baseline NS5A sequences were available for 282patients (126 mainland Chinese [45%〕, 80 Koreans [28%〕, 76Taiwanese [27%〕). NS5A-Y93H and/or -L31 RAPs were observed pretreatmentin 8% mainland Chinese, 14% Korean, and 18%Taiwanese patients, compared with 19% in Japanese. SVR12 in allnon-Japanese patients is shown (Figure); rates were broadly similarbetween countries and with Japanese data (Japanese: 96% overallwithout RAPs, 41% with RAPs). Responses were lower among patientswith baseline RAPs. By contrast, SVR12 in patients without RAPs washigh (92-100%), irrespective of cirrhosis, age, or baseline HCV-RNA.Conclusions: At least 95% of HCV GT-1b-infected patients from mainlandChina, Korea or Taiwan without baseline NS5A-Y93H or -L31polymorphisms who had HCV-RNA ≤7 log10 IU/mL achieved SVR12on DCV+ASV, regardless of cirrhosis status and age.

A laser pistonphone system capable of measuring the sensitivity modulus and phase of microphones is proposed. For the primary vibration calibration, the sine-approximation method has been applied which is used to calculate the complex sensitivity of accelerometers. This method is based on a laser interferometer with quadrature output instead of the fringe-counting method that is used in conventional laser pistonphones. Here, the laser pistonphone system based on the sine-approximation method with the quadrature signal is proposed and the measured results are compared to those obtained by the reciprocity method. The measured sensitivity with the proposed method agrees well with the result of the reciprocity method for the infrasound range. However, in case of the phase sensitivity, the discrepancy is relatively high in comparison to the sensitivity modulus. The results show that the proposed system is capable of calibrating the complex sensitivity of microphones. The improvement of the uncertainty and extension of the frequency range can be achieved by modification of the excitation system for obtaining the required stable excitation with long stroke.

The γ-secretase, composed of presenilin-1 (PS1) or presenilin-2 (PS2), nicastrin (NCT), anterior pharynx-defective phenotype 1 (APH-1), and PEN-2, is critical for the development of Alzheimer's disease (AD). PSs are autoproteolytically cleaved, producing an N-terminal fragment (NTF) and a hydrophilic loop domain-containing C-terminal fragment. However, the role of the loop domain in the γ-secretase complex assembly remains unknown. Here, we report a novel PS2 isoform generated by alternative splicing, named PS2β, which is composed of an NTF with a hydrophilic loop domain. PS2β disturbed the interaction between NCT and APH-1, resulting in the inhibition of amyloid-β production. We concluded that PS2β may inhibit γ-secretase activity by affecting the γ-secretase complex assembly. The NCT Program, in which the present JT-60U is being modified into a superconducting coil machine, is discussed under nationwide collaboration in Japan. Its mission is to establish high-beta steady-state operation for DEMO and to contribute to ITER. NCT is designed to have potential to investigate such research.

Temperature and diameter effect on hydrodynamic characteristic of surfactant drag-reducing flows

Hydrodynamic characteristic of surfactant drag-reducing flows is still not fully understood. This work investigated the temperature and diameter effect on hydrodynamic characteristic of cationic surfactant drag reducing flows in pipes. Solution of oleyl bishydroxyethyl methyl ammonium chloride (Ethoquad O/12), 900 ppm, as a cationic surfactant and sodium salicylate (NaSal), 540 ppm, as a counter-ion was tested at 12, 25, 40, and 50oC in pipes with diameter of 13, 25, and 40 mm. Drag reduction effectiveness of this surfactant solution was evaluated in 25 mm pipe from 6 to 75oC. Rheological characteristic of this solution was measured by stress control type rheometer with cone-and-plate geometry. Scale-up laws proposed by previous investigators were used to evaluate the flow characteristic of the solution. It was found that this surfactant solution has clear DR capability until 70oC. Result of this work suggested that temperature has a significant influence in changing the hydrodynamic entrance length of surfactant drag reducing flows. From rheological measurement, it was found that the solution exhibits Shear Induced Structure at all temperatures with different degree of peak viscosity and critical shear rate.

Adipokines, adipocyte-derived protein, have important roles in various kinds of physiology including energy homeostasis. Chemerin, one of adipocyte-derived adipokines, is highly expressed in differentiated adipocytes and is known to induce macrophage chemotaxis and glucose intolerance. The objective of the present study was to investigate the changes of chemerin and the chemokine-like-receptor 1 (CMKLR1) gene expression levels during differentiation of the bovine adipocyte and in differentiated adipocytes treated with tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), adiponectin, leptin, and chemerin (peptide analog). The expression levels of the chemerin gene increased at d 6 and 12 of the differentiation period accompanied by increased cytoplasm lipid droplets. From d 6 onward, peroxisome proliferator-activated receptor-${\gamma}2$ (PPAR-${\gamma}2$) gene expression levels were significantly higher than that of d 0 and 3. In contrast, CMKLR1 expression levels decreased at the end of the differentiation period. In fully differentiated adipocytes (i.e. at d 12), the treatment of TNF-${\alpha}$ and adiponectin upregulated both chemerin and CMKLR1 gene expression levels, although leptin did not show such effects. Moreover, chemerin analog treatment was shown to upregulate chemerin gene expression levels regardless of doses. These results suggest that the expression of chemerin in bovine adipocyte might be regulated by chemerin itself and other adipokines, which indicates its possible role in modulating the adipokine secretions in adipose tissues.
