Anti-inflammatory and Anti-oxidant Effects of PHBV/Collagen (PHCP) on Lipopolysaccharide-Induced Skin Inflammation

The anti-inflammatory effect of PHBV/Collagen (PHCP) was examined in a mouse model of lipopolysaccharide (LPS)-induced skin inflammation. Vascular permeability on the back skin was measured by the local accumulation of Evan’s blue dye after subcutaneous injection of LPS (30 μg site-1). Dye leakage in the skin showed a significant increase at 2 h after injection of LPS. This LPS-induced dye leakage was also completely inhibited by HO-1 inhibitor, ZnPP, and antioxidants, including methyl gallate, trolox, and mannitol. To study the possible mechanisms underlying the in vivo anti-inflammatory effect of PHCP against LPS-induced inflammation, we also examined the effects of PHCP on malondialdehyde (MDA) and glutathione levels in skin tissues and found that pretreatment with PHCP resulted in inhibited MDA elevation and a remarkable reduction of glutathione level. In addition, similar results were obtained after pretreatment with antioxidants, including trolox and mannitol, and HO-1 inhibitor, ZnPP. Histopathologically, an influx of neutrophils into the skin dermis was detected between 24 h and 72 h after LPS injection (30, 100 μg site-1), compared to control animals after injection of saline. This increase was greater in mice treated with 100 μg of LPS than in those treated with 30 μg of LPS and was significantly suppressed by pretreatment with PHCP, antioxidants, and HO-1 inhibitor. These results collectively suggest that PHCP has an anti-inflammatory effect against LPS-induced inflammation model in vivo and may be a good candidate for the skin tissue engineering biomedical application primarily through manipulation of the redox state.

The Gene Polymorphism of VMAT2 Is Associated with Risk of Schizophrenia in Male Han Chinese

Objective To investigate the association between gene polymorphism of vesicular monoamine transporter type 2(VMAT2) and schizophrenia in Han Chinese population.Methods 430 patients with schizophrenia and 470 age-sex matched controls were recruited from four mental health centers. All patients were diagnosed by two psychiatrists based on the Structured Clinical Interview for DSM Disorders (SCID). The ligase detection reactions (LDR) method was used to assess the polymorphism of the two SNPs (rs363371 and rs363324) of VMAT2.Results No associations of two SNPs with schizophrenia was found. When we stratified males and females for the analysis, we found that that in the recessive model of rs363371, there was an obvious significant association between rs363371 and schizophrenia in males (OR=0.564, 95% CI=0.357–0.892, p=0.014) but not females. For the association between rs363324 and schizophrenia, no association was found in either males or females. No association was found when stratifying early-onset schizophrenia and late-onset schizophrenia.Conclusion Our findings indicate that both rs363371 and rs363324 were not associated with schizophrenia, while it seemed that the AA genotype of rs363371 plays a protective effect in male Chinese in developing schizophrenia.

RNAi suppression of nuclear receptor genes results in increased susceptibility to sulfoxaflor in brown planthopper, Nilaparvata lugens

Nuclear receptors (NRs), which belong to a superfamily of ligand-dependent transcription factors, play essential roles in gene regulation to affect numerous physiological pathways. Twenty NR genes were identified in Nilaparvata lugens by using genomic, transcriptomic and GenBank databases, and categorized into NR0-NR6 subfamilies according to standard nomenclature. Among them, three NR geneswere classed into NR0, four into NR1, eight into NR2, one into NR3, one into NR4, two into NR5, and one into NR6, respectively. A phylogenetic tree of NRs from N. lugens and other representative species was constructed, which provided evolutionary insight into genetic distance. In order to investigate the NRs,whichwere induced by sulfoxaflor, time- and tissue-specific expression profiles of NR genes in fourth-instar nymphs were determined following LD50 sulfoxaflor treatment (0.28 ng/insect) and compared with control samples. NlUSP, NlE78, NlTLL, NlHR51, NlHR83, NlPNR and NlFTZ-F1 were highly expressed following 12–48 h of sulfoxaflor treatment. NlHR39 and NlDSF transcripts were detected in the head and the thorax, respectively. High-level and constitutive expression of NlHR3, NlTLL, NlHR83, NlFTZF1 and NlHR4 were found in the abdomen. Feeding of dsRNAs reduced the expression of NlHR3, NlUSP, NlTLL, NlHR83, NlPNR, NlFTZ-F1 and NlHR4 (35.48–49.77%) and caused significant nymph mortality (69.21–81.45%). These NRs, considered as insecticide targets, may play important function in sulfoxaflor detoxification.

Systematic Review on Epstein-Barr Virus (EBV) DNA in Diagnosis of Nasopharyngeal Carcinoma in Asian Populations

Objective: To conduct a meta-analysis to investigate the value of EBV DNA in diagnosis of nasopharyngeal cancer (NPC) in Asian populations, and provide important evidence for screening. Methods: Prospective or respective case-control or cohort studies regarding the detection role of EBV DNA for NPC were included in our study. We conducted a comprehensive literature search in PubMed, EMBASE, and the Chinese Biomedical Database (CBM database between January 1980 and March 2012. Results: A total of 18 studies with 1492 NPC cases and 2641 health controls were included. Almost of the included studies were conducted in China, and only one other conducted in Thailand. The overall results demonstrated that the pooled sensitivity, specificity, positive likelihood (+LR) and negative likelihood (-LR) were 0.73 (0.71-0.75), 0.89 (0.88-0.90), 8.84 (5.65-13.84) and 0.19(0.11-0.32), respectively. The overall EBV DNA detection showed the largest area of 0.932 under the summary receiver operator curve (SROC). The accuracy of detection by plasma for NPC (0.86) was higher than in serum (0.81), with largest areas under the SROC of 0.97 and 0.91, respectively. Conclusion: Our results demonstrated the EBV DNA detection in plasma or serum has high sensitivity and specificity in diagnosis of NPC, especially in Chinese populations with a high risk of cancer.

Numerical Simulation of Seismic Waves with Peak Arrival Time and Amplitude-Frequency Correlation

To improve the accuracy with which the non-stationary intensity characteristics of artificial acceleration-time-history curves are determined, a method of adjusting the frequency correlation between the artificial peak arrival time and amplitude spectrum is proposed. A calculation method for determining the peak arrival time to reflect the physical meaning of the phase-differenc espectrum was first deduced. Subsequently, the amplitude-distribution characteristics corresponding to the peak arrival times for different durations of natural earthquakes were analyzed. In accordance with the distribution characteristics of the reference amplitude, the frequency distribution of the artificial peak arrival time was divided into four parts: the beginning, strong-earthquake, attenuation, and tail sections. By adjusting the arrival frequency of peak points for the different earthquake durations considered, the corresponding relationship between the artificial peak arrival time and amplitude spectrum was determined to be more consistent with the general law of natural earthquakes. The sample calculation described in this paper demonstrates that adjusting the frequency correspondence between the artificial peak arrival time and amplitude spectrum greatly improves the accuracy of the non-stationary intensity characteristics of artificial acceleration-time-history curves.

Targeting EGFL7 Expression through RNA Interference Suppresses Renal Cell Carcinoma Growth by Inhibiting Angiogenesis

Renal cell carcinoma (RCC) is the most lethal of all urological cancers and tumor angiogenesis is closely related with its growth, invasion, and metastasis. Recent studies have suggested that epidermal growth factor-like domain multiple 7 (EGFL7) is overexpressed by many tumors, such as colorectal cancer and hepatocellular carcinoma; it is also correlated with progression, metastasis, and a poor prognosis. However, the role of EGFL7 in RCC is not clear. In this study, we examined how EGFL7 contributes to the growth of RCC using a co-culture system in vitro and a xenograft model in vivo. Downregulated EGFL7 expression in RCC cells affected the migration and tubule formation of HMEC-1 cells, but not their growth and apoptosis in vitro. The level of focal adhesion kinase (FAK) phosphorylation in HMEC-1 cells decreased significantly when co-cultured with 786-0/iEGFL7 cells compared with 786-0 cells. After adding rhEGFL7, the level of FAK phosphorylation in HMEC-1 cells was significantly elevated compared with phosphate-buffered saline (PBS) control. However, FAK phosphorylation was abrogated by EGFR inhibition. The average size of RCC local tumors in the 786-0/iEGFL7 group was noticeably smaller than those in the 786-0 cell group and their vascular density was also significantly decreased. These data suggest that EGFL7 has an important function in the growth of RCC by facilitating angiogenesis.

Experimental Study and Numerical Analysis on Mechanical Behavior of T-shape Stiffened Orthotropic Steel-concrete Composite Bridge Decks

A new-type of orthotropic steel-concrete composite bridge deck system was developed, by casting the concrete overlay on the top of the orthotropic steel deck ribbed with T-shape steel members. To study its mechanical behavior (in terms of failure mode, load-deflection relationship, concrete crack initiation and propagation, strength, stiffness and so on), two new-type orthotropic steel-concrete composite bridge decks with different section dimensions were experimentally investigated and two reference decks (reinforced concrete deck and orthotropic steel deck) were also involved in the research for comparison. For the two new-type orthotropic steel-concrete composite decks, the average value of ultimate loads per width is 885.7kN, which is 2.35 and 1.61 times of that of the concrete and steel reference decks with almost the same section height. Experimental results proved that the composite deck can effectively control the crack initiation and propagation in the concrete and postpone the yielding of the steel bars and steel plates, due to the composite action between the concrete overlay and the underlying steel plate. Furthermore, the Finite Element (FE) model of the orthotropic steel-concrete composite deck was developed and validated by test results. A parametric study is conducted regarding to the stiffness of shear studs. With the validated FE model, stress distribution in the underlying steel plate and T-shape stiffeners and development of concrete cracking in the concrete overlay were characterized at different load levels.

Amperometric Determination of Nitrite at Poly(Methylene Blue)-Modified Glassy Carbon Electrode

Electrochemical characteristics of nitrite ion were investigated at a poly(methylene blue)-modified glassy carbon electrode by cyclic voltammetry and differential pulse voltammetry. The poly(methylene blue)-modified glassy carbon electrode exhibited enhanced anodic signals for nitrite. The effects of key parameters on the detection of nitrite were evaluated at the modified electrode, such as pH, accumulation time, and scan rate. Under optimum condition, the chemically modified electrode can detect nitrite in the concentration range $2.0{\times}10^{-6}$ to $5.0{\times}10^{-4}$ M with the detection limit of $2.0{\times}10^{-6}$ M and a correlation coefficient of 0.999. The detection of nitrite using the chemically modified electrode was not affected by common ions such as $Na^+$, $K^+$, $Ca^{2+}$, $Cl^-$, $HPO_4^{2-}$ and $H_2PO_4^- $. The modified electrode showed good stability and reproducibility. The practical application of the present method was successfully applied to the determination of nitrite ion in cabbage samples.

Through the analysis of such curriculum policies, we can found that movements and intrinsic curriculum values of the curriculum policy of vocational education in our country since the new century. This paper tries to make people think about the curriculum policy of vocational education deeply and provide directional guidance for the curriculum reform of vocational education in our country.

On the Application of Game Theory to the Competition among Major Ports in NE Asia for T/S Cargos

동북아시아 경제의 급성장으로 인한 지역 물동량 및 해운시장 환경 변화에 따라서 부산항과 상해항은 동북아 물류 중심 허브가 되기 위하여 항만개발 및 항만 경쟁력을 강화해 왔으며, 최근에는 환적 컨테이너 화물 유치경쟁이 더욱 심화되고 있다. 따라서 본 연구에서는 동북아 주요 컨테이너 항만중 경쟁관계에 있는 부산항과 상해항을 대상으로 이 두 항만이 피더항의 환적화물을 유치하기 위한 경쟁의 게임모형을 수립하여 경쟁상황을 분석하였다. Due to rapid growth of East Asian economy, aome changes are being made in trade volume and shipping market of this area. Busan port in Korea and Shanghai port in China are continually increasing their investment in port development and competitiveness in order to become the logistics hub of Northeast Asia. Especially the competition for transshipment containers becomes more and more fierce. So in this study, we set a goal of increasing the port competitiveness on transshipment cargo by an analysis of competition strategies. We choose the Busan port and the Shanghai port as the research objects, games competition model and real data is used to analyze the two ports" price strategies and merket share. According to the results, some sdvices will be put forward to enhance competitiveness of the two ports.
