      • A novel copper-chelating strategy for fluorescent proteins to image dynamic copper fluctuations on live cell surfaces

        <▼1><P>A strong but selective copper-binding tripeptide was employed to develop a highly sensitive and selective copper(<SMALL>II</SMALL>) protein reporter.</P></▼1><▼2><P>Copper is indispensable in most aerobic organisms although it is toxic if unregulated as illustrated in many neurodegenerative diseases. To elucidate the mechanisms underlying copper release from cells, a membrane-targeting reporter which can compete with extracellular copper-binding molecules is highly desirable. However, engineering a reporter protein to provide both high sensitivity and selectivity for copper(<SMALL>II</SMALL>) has been challenging, likely due to a lack of proper copper(<SMALL>II</SMALL>)-chelating strategies within proteins. Here, we report a new genetically encoded fluorescent copper(<SMALL>II</SMALL>) reporter by employing a copper-binding tripeptide derived from human serum albumin (HSA), which is one of the major copper-binding proteins in extracellular environments. Optimized insertion of the tripeptide into the green fluorescent protein leads to rapid fluorescence quenching (up to >85% change) upon copper-binding, while other metal ions have no effect. Furthermore, the high binding affinity of the reporter enables reliable copper detection even in the presence of competing biomolecules such as HSA and amyloid beta peptides. We also demonstrate that our reporter proteins can be used to visualize dynamic copper fluctuations on living HeLa cell surfaces.</P></▼2>

        웨이트트레이닝이 남자 펜싱선수의 연속동작 수행시간에 미치는 영향

        옥정석, 최태석, 전태원, 엄우성. 웨이트트레이닝이 펜싱선수의 연속 동작수행시간에 미치는 영향. 운동과학, 제11권 제1호, 221-231, 2002. 본 연구의 목적은 8주간의 웨이트트레이닝이 펜싱선수의 연속 동작수행시간에 미치는 효과를 분석하기 위함이었다. 이러한 목적을 달성하기 위해 신체적·정신적으로 건강하고 의학적으로 특별한 질환이 없는 남자 대학생 펜싱선수 30명을 무작위 추출하여 실험군과 대조군을 각 15명씩 구성하였다. 근지구력은 웨이트트레이닝 종목별로 20RM에 해당하는 무게를 1㎏ 단위로 측정하였으며, 웨이트트레이닝 프로그램 처치 전·후를 비교하였다. 또한, 펜싱 전진 이동-찌르기 두 가지 기본동작의 연속 동작수행시간, 반응시간, 그리고 동작시간은 자체 제작한 동작수행시간 측정기로 측정하였으며, 전진 이동-찌르기 동작시 웨이트트레이닝 프로그램 처치전후의 연속 동작수행시간, 반응시간, 동작시간을 비교·검토하여 다음과 같은 결과를 얻었다. 첫째, 펜싱선수들의 근지구력과 연속 동작수행시간 사이에 유의한 상관관계가 있었다. 둘째, 8주간의 웨이트트레이닝 참여에 따른 처치전·후 및 집단간의 연속 동작수행시간, 반응시간, 그리고 동작시간은 유의한 차이가 있었다. The purpose of this study was to investigate the effect of 8-week weight training on continuous response time. To achieve this purpose subjects were 30 colleges male fencing players who have not medical diseases, were divided into experiment group and control group. The muscle fitness was measured by 1㎏ unit of weight equivalent to 20RM of weight training items respectively, muscle fitness of pre weight training program treatment was compared with those of post weight training program treatment. Also response time of two basic motions of fencing forward movement-fente were measured by a self-produced response time measuring instrument, and continuous response time, reaction time, movement time of pre and post weight training program treatment during fencing forward movement-fente motion were inspected comparatively. The results of this study were as follows. First, muscle endurance and continuous response time of fencing players were correlated in significantly. Two, after 8 weeks weight training program, continuous response times, reaction time, and movement time between experiment group and control group were showed significant difference. As this results, muscle endurance was closely related to continuous response time, weight training program developed on the basis of this had an affirmative effect on continuous response time, reaction time, and movement time of the basic movement of a fencing.

        Ketamine의 Guinea Pig 기관 평활근 세포내 Ca2 + 축적과 방출에 대한 작용

        Background: The potent bronchodilatory effects of ketamine on airway smooth muscle tone are important in the management of patients with asthma, but its mode of action is unclear. In the present study we evaluated that effects on isolated guinea pig tracheal smooth muscle. Methods : Changes of isometric contraction of strip were measured. (1) Serial stimulation with acetylcholine(ACh) in Krebs solution or with A23187, nifedipine, ketamine were evaluated. After that, ACh stimulation was induced in Ca2+ free solution. (2) In Ca2+ free solution, ACh contraction was obtained(L1) and emptied by repetitive ACh stimulation. Internal stores were refilled by Ca2+ with ACh stimulation. During the incubation period, A23187, nifedipine, ketamine, cyclopiazonic acid + ketamine was added and tested for their ability to inhibit refilling. Refilling was evaluated by ACh produced contraction (L2) with ratio (L2/L1). (3) Effects of ketamine on the contraction induced by caffeine were also checked. Results : Ketamine inhibited amplitude dose-dependently by successive application of ACh in modified Krebs solution and Ca2+ free solution. Ca2+ influx through voltage gated channels were inhibited with nifedipine but not with A23187. ACh sensitive internal store were different when A23187, nifedipine and ketamine were applied in Ca2+ free solution. Refilling of internal store were potentiated by A23187, but decreased by nifedipine and ketamine. Caffeine produced contractions in the presence of ketamine were not significantly different from control. Conclusion : We concluded that the inhibitory effects of ketamine in guinea pig trachea were by acting through voltage and receptor gated channels in dose-depedent manner and these effects may be interferences of intracellular second messengers system.         Humphrey Circuit System 의 임상적 관찰

        The Humphrey circuit system functions with the efficiency of the Lack system for spontaneous respiration and of the Bain system for controlled ventilation. It is designed to incorporate into a single system the advantages of the Mapleson A, D and E type systems and to be available in coaxial and non-coaxial forms. This study was done to sudy the functions of a new anesthetic breathing system and to evaluate the adequacy of the recommended fresh gas flow. For spontaneous and controlled ventilation the system required a low fresh gas flow to achieve normocarbia(In the Lack circuit system, 60 ml/min., is useded and in the Bain circuit system, 70 ml/mn.). The advantage of this system was that it was simple to poerate and maintain, more easily sterilized and applicable in academic use. Further more the non-coaxial from of the circuit overcame the hazard of the inner tube complication. But this form was thought to have the provlem of humidifying the inspired gas which still has to be studied. This multicircuit system was used satisfactorily with low fresh gas flow under spontaneous and controlled ventilation maintaining normal range of arterial carbon dioxide. It may become one of the preferred semiclosed breathing systems because of simplicity, multipurpose usefulness and other advantages.

        중환자실 환자의 임상통계 고찰

        The intensive care unit (ICU) of our hospital is a 12 bed multidisciplinary ICU which is under the auspices of the Department of Anesthesiology. The ICU is essentially a respiratory ICU in which all hospital patients requiring ventilatory support are cared for. The patient population encompasses all causes of respiratory failure, medical and surgical. Another source of referral (surgical and medical) is a large group of critically ill patients with multiorgan failure, especially due to trauma. Our experience of intensive care therapy began in the recovery room starting in 1959. Since that time the size and facilities of the ICU have been enlarged. We analyzed 4,514 ICU patients admitted between Mar. 1980 and Feb. 1984, to obtain better guidance and management in the ICU. The results of analysis are as follows: 1) Total number of patients was 4,514; 874 patients in 1980, 832 patients in 1981, 895 patients in 1982, 862 patients in 1983 and 1,051 patients in 1984. The ratio of male to female patients was nearly 2:1. 2) Total admission time in the ICU was 17,473 days. The average patient stay was 3.9 days. The number of patients staying in the ICU more than 9 days increased from 54 cases(6%) in 1980 to 101 cases(10%) in 1984. 3) The forty year age group outrumbered the order age group and was 20.5%. The mortality rate of the pre-teen age group was the highest(19.7%) and the overall mortality was 14.2%(640 cases) during these 5 years. 4) The number of patients admitted to the ICU from the Department of Neurosurgery was 1,218 (27%)being the highest among all departments. Out of 232 patients who died, the Department of Neurosurgery had the highest mortality rate, 19.1%. Form the above results, it can be seen that the number of patients admitted to the ICU., the mortality rate and the patients staying more than 9 days in the ICU has increased annually It is claimed that more facilities allowing for monitoring and support, special qualified staffs and a co-ordinated organization will reduce the mortality rate, particularly in the pre-teen age group.

        Rhabdomyolysis is a syndrome characterized by muscle necrosis and release of intracellular muscle constituents into the circulation. It can occur in various diseases and conditions involving destruction of muscle membranes, including muscle strain, persistent coma, drug or alcohol abuse, connective tissue disease, excessive exercise, and surgery. Many factors have been implicated in the development of rhabdomyolysis during surgery; these include patient positioning with obstructed blood flow of either a femoral vessel at the hip or a popliteal vessel at the knee. Rhabdomyolysis can also be caused by muscle compression due to operative positioning during prolonged surgery. Surgery performed in the lithotomy position sometimes results in serious complications of rhabdomyolysis and acute renal failure. We report a case of rhabdomyolysis of both calves associated with the lithotomy position for laparoscopic myomectomy. (Korean J Med 2012;83:802-806)
