Tacrolimus Differentially Regulates the Proliferation of Conventional and Regulatory CD4+ T Cells

Tacrolimus is a widely used T cell targeted immunosuppressive drug, known as a calcineurin inhibitor. However, the exact pharmacological effects of tacrolimus on CD4+ T cells have yet to be elucidated. This study investigated the effects of tacrolimus on CD4+ T cell subsets. Mouse or human CD4+ T cells were cultured with immobilized anti-CD3/CD28 antibodies in the presence of tacrolimus. The cell division of CD4+ T cells was analyzed using a flow cytometer according to the expression of Foxp3. The gene expression patterns of tacrolimus-exposed T cells were examined by quantitative PCR. In the case of conventional CD4+ T cells (Tconv cells), tacrolimus inhibited T cell receptor stimulation-induced cell division. In contrast, the cell division of regulatory CD4+ T cells (Treg cells) was even promoted in the presence of tacrolimus, especially in humans. Tacrolimus did not promote conversion of Tconv to Treg cells in mice. Furthermore, tacrolimus modified the expression levels of Foxp3-regulated T cell receptor signal related-genes, PTPN22 and Itk, in human Treg cells. Immunosuppressive effect of tacrolimus may be attributed to the relatively enhanced proliferation of Treg cells in association with altered gene expression levels of TCR signaling molecules.

Aim(s): To examine the factors related to high expressed emotion in family members of people with dementia. Method(s): A cross-sectional research design was used. Interviews and questionnaires were employed, and the key family members of people with dementia were the participants. The first survey was administered to 24 individuals. Data collected include demographic details, the Family Attitude Scale (FAS), which assesses expressed emotion, and measures of caregiving burden and depression in family members. This study was approved by the Institutional Review Board of the institution we are affiliated with. And it was also supported by Nissei Foundation. Result(s): We show the preliminary results in this abstract although the number of participants is still increasing. Currently, 24 family members (8 male, 17 female) have participated in this study. The preliminary results indicate that nine had FAS scores above 40, which is the threshold for high Expressed Emotion (EE) in family members caring for persons with dementia, of potentially demented individuals. The nine participants with high EE were female, 66.7% were children of patient with dementia, 55.6% had depressive tendencies, 22.2% were identified as depressed, and 77.8% had a moderate to severe sense of burden. Conclusion(s): Factors influencing the high EE of family members of people with dementia could be related to their role in the family and their mental state. Early identification of risk factors might help in identifying and developing relevant interventions to prevent high EE in caregivers. The validity of this hypothesis will be further tested.
Wireless Sensor Networks for Monitoring the Daily Interactions between User and Objects

This article presents a hybrid system of wireless sensor networks and active tags in order to monitor user"sbehaviors of handling objects. The sensor networks consist of an acceleration sensor node that the user carries and occupancy sensor nodes that are deployed in a workplace. The active tags, which are embedded with switch sensors, vibration sensors and occupancy sensors, are attached to objects. The sensor networks capture the user"s movements andthe active tags detect the objects" movements. The sensor nodes have the parent selection function that can allocate resources to the nodes. We demonstrate monitoring the user"s behaviors of handling the objects using the hybrid system.We also show that the proposed system provides the useful information for behavior classification.

Objective: The antitumor effects of anti-PD-1 antibody against mismatch repair deficiency (MMR-D)-associated cancers have been reported. MMR-D is found in approximately20%–30% of endometrial carcinomas (ECs) and frequently occurs due to promoter hypermethylation ( -PHM). ECs with -PHM are classified according to the molecular screening of Lynch syndrome (LS), but few detailed reports are available. The purpose of this study was to clarify the clinical features of EC with -PHM. Methods: Immunohistochemistry of MMR proteins (MLH1, MSH2, MSH6, and PMS2) was performed on specimens from 527 ECs treated at our university hospital from 2003 to 2018. methylation analysis was added to cases with MLH1/PMS2 loss. ECs were classified as follows: cases that retained MMR proteins as “MMR-proficient;” cases with MLH1/PMS2 loss and -PHM as “met-EC;” and cases with other MMR protein loss and MLH1/PMS2 loss without -PHM as “suspected-LS.” The clinical features, including long-term prognosis, of each group, were analyzed. Results: Accordingly, 419 (79.5%), 65 (12.3%), and 43 (8.2%) cases were categorized as “MMR-proficient,” “suspected-LS,” and “met-EC,” respectively. Significantly, “met-EC” had a lower proportion of grade 1 tumors (37.5%) and a higher proportion of stage III/IV tumors (37.2%) than the other groups. The overall and progression-free survival of “met-EC” were significantly worse than those of “suspected-LS” in all cases. Conclusion: In ECs with MMR-D, “met-ECs” were a subgroup with a poorer prognosis than “suspected-LS.” “Met-ECs” would be the main target for anti-PD-1 antibody treatment, and its clinical susceptibility should be verified individually.

<P>Wall teichoic acid (WTA) of Staphylococcus aureus is a major cell envelope-associated glycopolymer that is a key molecule in promoting colonization during S. aureus infection. The complement system plays a key role in the opsonization and clearance of pathogens. We recently reported that S. aureus WTA functions as a ligand of human serum mannose-binding lectin (MBL), a recognition molecule of the lectin complement pathway. Intriguingly, serum MBL in adults does not bind to WTA because of an inhibitory effect of serum anti-WTA-IgG. In this study, serum anti-WTA-IgG was purified to homogeneity using a purified S. aureus WTA-coupled affinity column to examine the biological function of human anti-WTA-IgG. The purified anti-WTA-IgG contained the IgG2 subclass as a major component and specifically induced C4 and C3 deposition on the S. aureus surface in the anti-WTA-IgG-depleted serum, but not in C1q-deficient serum. Furthermore, the anti-WTA-IgG-dependent C3 deposition induced phagocytosis of S. aureus cells by human polymorphonuclear leukocytes. These results demonstrate that serum anti-WTA-IgG is a real trigger for the induction of classical complement-dependent opsonophagocytosis against S. aureus. Our results also support the fact that a lack of the lectin complement pathway in MBL-deficient adults is compensated by Ag-specific, Ab-mediated adaptive immunity.</P>
Relation between Earthward magnetotail flow and low-latitude Pi2 pulsations

It has been suggested that oscillatory earthward bursty bulk flows (BBFs) in the magnetotail are driving Pi2 oscillations on the ground. However, only a few studies examined the BBF-driven Pi2 model. The goal of this study is to establish the relation between BBFs and Pi2s. In order to examine BBF-drive model, we will conduct the timing analysis between BBFs and Pi2s for the intervals during which both Pi2 and BBF are present. In our study BBFs are identified with earthward flow speeds up to 200 km/s from THEMIS probes with a geocentric distance between 7 and 12 RE in the nightside magnetosphere for the interval from January 2008 to March 2008. Pi2 pulsations are identified at three low/mid-latitude ground stations having different local times; Bohyun (L = 1.35, LT ~ UT +9 hr), Hermanus (L = 1.83, LT ~ UT +2 hr) and Carson city (L = 2.00, LT ~ UT -2 hr). It is found that ~28% of BBFs have onset timing nearly identical to Pi2 onset. To investigate whether there is high similarity in the waveform between BBF and Pi2 oscillations, the coherence analysis is applied to the time series of BBF and Pi2 events. We found that there is no high coherence between BBF and Pi2 oscillations. Thus our observations suggest that low-latitude Pi2 oscillations are not directly driven by earthward flow bursts.
