• Bioengineered mussel glue incorporated with a cell recognition motif as an osteostimulating bone adhesive for titanium implants

Jo, Yun Kee,Choi, Bong-Hyuk,Zhou, Cong,Ahn, Jin-Soo,Jun, Sang Ho,Cha, Hyung Joon The Royal Society of Chemistry 2015 Journal of Materials Chemistry B Vol.3 No.41

Successful titanium implantation strongly depends on early fixation through an osseointegration between the titanium fixture and adjacent bone tissue. From a clinical perspective, rapid recruitment of functional biomolecules from the blood and osteogenic cell binding is critical for osseointegration immediately after implant insertion. Thus, surface modifications aiming to improve the interactions between the blood and implant and to enhance the binding of osteogenic cells onto the implant surface can contribute to successful osseointegration. Mussel adhesive proteins (MAPs) derived from marine mussels have been considered as promising bioadhesives that have strong adhesion and coating abilities onto organic and inorganic surfaces, even in wet environments. Here, we investigated thein vitroandin vivoosteostimulating ability of the bioengineered mussel glue MAP-RGD, which is a recombinant MAP fused with an Arg-Gly-Asp (RGD) peptide, an effective cell recognition motif for activating intracellular signaling pathways, using a titanium mesh (Ti-mesh) as a model titanium implant. We found that thein vitrocell behaviors of pre-osteoblast cells, such as attachment, proliferation, spreading, and osteogenic differentiation, increased significantly on the MAP-RGD-coated Ti-mesh surface. In vitroblood responses including blood wetting, blood clotting, and platelet adhesion were also highly enhanced on the MAP-RGD-coated surface. Importantly, implantation of the MAP-RGD-coated Ti-mesh resulted in a remarkable acceleration ofin vivobone regeneration and maturation of a new bone in a rat calvarial defect. Consequently, the bioengineered mussel glue can be successfully utilized as an osteostimulating bone bioadhesive for titanium implant applications with further expansion to general bone tissue engineering.

Differences in Insulin Sensitivity and Secretory Capacity Based on OGTT in Subjects with Impaired Glucose Regulation

( Sang Youl Rhee ),( Mi Kwang Kwon ),( Byong Jo Park ),( Suk Chon ),( In Kyung Jeong ),( Seung Joon Oh ),( Kyu Jeung Ahn ),( Ho Yeon Chung ),( Sung Woon Kim ),( Jin Woo Kim ),( Young Seol Kim ),( Jeon 대한내과학회 2007 The Korean Journal of Internal Medicine Vol.22 No.4

Background: This study examined whether defects in insulin secretion contribute to the development and progression of type 2 diabetes mellitus (T2DM). Methods: Plasma insulin and glucose were measured after a glucose tolerance test to calculate the insulinogenic index (IGI) and the HOMA-IR Homeostasis model assessment of insulin resistance in subjects with normal glucose tolerance (NGT), prediabetes (preDM, n=49), and T2DM patients with disease duration <1 year (n=84), 1~5 years (n=45), or >5 years (n=37). Plasma proinsulin and adiponectin levels were also measured as a parameter of insulin secretion and resistance. Results: The mean HOMA-IR increased and the adiponectin levels decreased relative to the deterioration of glucose tolerance in NGT and preDM subjects. However, differences in the HOMA-IR were not related to disease duration in T2DM subjects. The mean IGI was similar in NGT and preDM subjects, but there were significant deteriorations in IGI relative to the duration of diabetes. Conclusions: Defects in both insulin sensitivity and insulin secretion contribute to T2DM, but decreased insulin secretion may be more important in the development and progression of T2DM.

Quality of Life in Newly Diagnosed Moderate-to-Severe Ulcerative Colitis: Changes in the MOSAIK Cohort Over 1 Year

        Jo Ik Hyun,Lee Kang-Moon,Kim Dae Bum,Kim Ji Won,Lee Jun,Jeen Yoon Tae,Kim Tae-Oh,Kim Joo Sung,Park Jae Jun,Hong Sung Noh,Park Dong Il,김현수,Lee Yoo Jin,Kim Youngdoe 거트앤리버 소화기연관학회협의회 2022 Gut and Liver Vol.16 No.3

Background/Aims: Improving quality of life has been gaining importance in ulcerative colitis (UC) management. The aim of this study was to investigate changes in health-related quality of life (HRQL) and related factors in patients with moderate-to-severe UC. Methods: A multicenter, hospital-based, prospective study was performed using a Moderateto- Severe Ulcerative Colitis Cohort in Korea (the MOSAIK). Changes in HRQL, evaluated using the 12-Item Short Form Health Survey (SF-12) and Inflammatory Bowel Disease Questionnaire (IBDQ), were analyzed at the time of diagnosis and 1 year later. Results: In a sample of 276 patients, the mean age was 38.4 years, and the majority of patients were male (59.8%). HRQL tended to increase in both the IBDQ and SF-12 1 year after diagnosis. A higher partial Mayo score was significantly related to poorer HRQL on the IBDQ and SF-12 in a linear mixed model (p<0.01). Inflammatory markers such as C-reactive protein (CRP) or erythrocyte sedimentation rate also showed a negative correlation on HRQL (p<0.05). Patients whose IBDQ score improved by 16 or more (71.2%) in 1 year were younger, tended to be nonsmokers, and had a lower partial Mayo score and CRP than those whose IBDQ score did not. There was no significant association between HRQL and disease extent, treatments at diagnosis, or the highest treatment step during the 1-year period. Conclusions: Optimally controlled disease status improves HRQL in patients with moderate-tosevere UC. The partial Mayo score and inflammatory markers may be potential indicators reflecting the influence of UC on patient`s daily lives.

Chrotective Effects of Wogonin in Experimental Models of Osteoarthritis in vitro and in vivo

( Jin Sung Park ),( Hyun Jae Lee ),( Dong Yeong Lee ),( Ho Seung Jo ),( Jin Hoon Jeong ),( Dong Hee Kim ),( Dae Cheol Nam ),( Choong Jae Lee ),( Sun-chul Hwang ) 한국응용약물학회 2015 Biomolecules & Therapeutics(구 응용약물학회지) Vol.23 No.5

We evaluated the chondroprotective effects of wogonin by investigating its effects on the gene expression and production of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well as on production of MMP-3 in the rat knee. Rabbit articular chondrocytes were cultured in a monolayer, and RT-PCR was used to measure interleukin-1β (IL-1β)-induced expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), and type II collagen. In rabbit articular chondrocytes, the effects of wogonin on IL-1β-induced production and proteolytic activity of MMP-3 were investigated using western blot analysis and casein zymography, respectively. The effect of wogonin on MMP-3 protein production was also examined in vivo. In rabbit articular chondrocytes, wogonin inhibited the expression of MMP-3, MMP-1, MMP-13, and ADAMTS-4, but increased expression of type II collagen. Furthermore, wogonin inhibited the production and proteolytic activity of MMP-3 in vitro, and inhibited production of MMP-3 protein in vivo. These results suggest that wogonin can regulate the gene expression and production of MMP-3, by directly acting on articular chondrocytes.

      • Surface-diffusion-limited growth of atomically thin WS2 crystals from core-shell nuclei

        Jo, Sunghwan,Jung, Jin-Woo,Baik, Jaeyoung,Kang, Jang-Won,Park, Il-Kyu,Bae, Tae-Sung,Chung, Hee-Suk,Cho, Chang-Hee The Royal Society of Chemistry 2019 Nanoscale Vol.11 No.18

Atomically thin transition metal dichalcogenides (TMDs) have recently attracted great attention since the unique and fascinating physical properties have been found in various TMDs, implying potential applications in next-generation devices. The progress towards developing new functional and high-performance devices based on TMDs, however, is limited by the difficulty in producing large-area monolayer TMDs due to a lack of knowledge of the growth processes of monolayer TMDs. In this work, we have investigated the growth processes of monolayer WS2 crystals using a thermal chemical vapor deposition method, in which the growth conditions were adjusted in a systematic manner. It was found that, after forming WO3-WS2 core-shell nanoparticles as nucleation sites on a substrate, the growth of three-dimensional WS2 islands proceeds by ripening and crystallization processes. Lateral growth of monolayer WS2 crystals subsequently occurs by the surface diffusion process of adatoms toward the step edge of the three-dimensional WS2 islands. Our results provide understanding of the growth processes of monolayer WS2 by using chemical vapor deposition methods.

( Sang Woong Youn ),( Tae Yoon Kim ),( Byung Soo Kim ),( Seung Chul Lee ),( Jeung Hoon Lee ),( Yong-beom Choe ),( Joo-heung Lee ),( Jee-ho Choi ),( Joo Young Roh ),( Seong Jin Jo ),( Eun-so Lee ),( Mi 대한피부과학회 2018 대한피부과학회 학술발표대회집 Vol.70 No.2

Background: The Phase Ⅲ studies, VOYAGE 1 and 2, were conducted to assess guselkumab for the treatment of patients with moderate to severe psoriasis. However, these studies included a worldwide study population. Objectives: To determine the efficacy and safety of guselkumab in Korean patients. Methods: This analysis included 28 patients from VOYAGE 1 and 98 patients from VOYAGE 2. In total, 126 patients were analyzed. Outcomes through week 28 were analyzed due to differences in design after week28. Results: Of 126 Korean patients randomized, 30 received placebo, 63 were assigned to guselkumab, and 33 received adalimumab. Guselkumab was superior to adalimumab in achieving an IGA 0 score (clear skin) at week 12 (31.7% vs 0.0%, p<0.001) and at week 24 (52.4% vs 21.2%, p=0.004), respectively. Among patients treated with guselkumab, 40 of 63 (63.5%) achieved a PASI75 response at week 8; at week 24, a significantly higher proportion of guselkumab patients achieved a PASI 75 compared to adalimumab patients (93.7% vs 66.7%, respectively, p<0.001). In addition, higher proportions of guselkumab patients achieved PASI 90 and PASI 100 than adalimumab patients at week 24 (PASI90; 73.0% vs 57.6%, p=0.168 / PASI100; 20.6% vs 15.2%, p=0.591, respectively). Through week 28, guselkumab and adalimumab showed comparable safety profiles. Conclusion: The efficacy and safety of guselkumab in Korean psoriasis patients through 28 weeks was consistent with findings reported for the VOYAGE1 and 2 trials.
