      • In vitro Mammalian Chromosomal Aberration Test of Fullerene-C60

        Fullerene의 유전독성을 평가하기 위하여 Chinese hamster유래의 난소유아세포(CHO-K1 cell)를 이용하여 직접법(-S9)과 대사활성화법(+S9 mix)의 염색체이상시험을 실시하였다. 시험물질은 1% CMC 나트륨염의 현탁액(l% CMC 용액)에 희석하여 조제하였다. 대사활성화를 시키지 않은 직접법의 염색체이상시험에서 24시간 투여군은 8단계의 농도(0.078, 0.156, 0.313, 0.625, 1.25, 2.5, 5, 10mM)로 투여하여 실시하였다. 투여 농도 증가에 따른 염색체이상의 빈도가 증가하는 양상이 나타나지 않았다. 48시간의 투여군에서는 8단계의 농도(0.078, 0.156, 0.313, 0.625, 1.25, 2.5, 5, 10mM)로 투여하여 실시하였는데 투여 농도 증가에 따른 염색체이상의 빈도가 증가하는 양상이 나타나지 않았다. 배수체의 염색체이상은 직접법에서 관찰되지 않았다. 대사활성화법을 이용하여 6시간 시험물질을 투여한 시험에 있어서는 8단계의 용량단계 (0.078, 0.156, 0.313, 0.625, 1.25, 2.5, 5, 10mM)를 설정하였는데 투여 농도가 증가함에 따른 염색체이상빈도의 증가양상이 관찰되지 않았다. 이상의 결과를 종합할 때 본 시험물질은 본 시험 조건하에서 CHO-K1세포에서 대사활성화를 시켰을 때 염색체 이상을 유발하지 않는 것으로 판단된다.

        과실이 크고 품질이 우수한 조생종 배 ‘금촌조생’ 육성

        ‘금촌조생’ 품종은 농촌진흥청 원예연구소 나주배연구소에서 1971년 ‘금촌추’의 품질을 개선하기 위해 당도가 높고 육질이 유연한 ‘단배’를 교배하여 1982년 1차 선발되었고, 1997년부터 5년 간 지역적응시험을 거쳐 2001년 최종 선발 및 품종보호출원 되었다. 수세는 비교적 강하고, 수자는 반개장성이다. 꽃가루가 풍부하며, 주요 재배품종과 교배친화성을 보인다. 또한 흑반병에 대하여 강한 포장저항성을 나타내었다. 숙기는 재배중인 ‘원황’ 품종보다 약 5일 정도 늦은 9월 3일로 추석에 출하하기 알맞은 품종이다. 과형은 ‘금촌추’와 같은 도원추형이고, 과피색은 황갈색이다. 과중은 593g이고, 당도는 13.2Bx이다. 육질은 유연하며 과즙이 풍부하고, 석세포가 없어 식미가 매우 우수하다. 그러나 과육선숙형 품종으로 원예적 수확기에도 과피에 녹색이 남지만, 완숙과가 되면 황갈색으로 착색된다. 상온저장 기간은 20일 정도로 다른 조생종에 비해 저장력은 좋은 편이다. ‘Geumchonjosaeng’ pear cultivar (Pyrus pyrifolia var. culta Nakai) originated from the cross between ‘Imamuraaki’ and ‘Danbae’ to improve the fruit quality of ‘Imamuraaki’ cultivar in 1971 at Naju Pear Research Institute of National Horticultural Research Institute (NHRI), Rural Development Administration (RDA). It was preliminarily selected in 1982 and named in 2001. The tree habit shows vigorous growth and upright spreading. It carries abundant pollen grains and shows cross compatibility with leading cultivars. Also it shows high resistance to black leaf spot (Alternaria kikuchiana) in the field condition. Optimum harvest time is around Sep. 3, which is 5 days later than that of ‘Wonwhang’, and it could satisfy consumer’s demand for high quality pear fruit in “Chuseok”. The obovate fruit shape resembles maternal parent, ‘Imamuraaki’ and skin color is green yellowish brown at normal harvesting time and gradually turned to yellowish brown at full ripennig state. Fruit weighs about 593 g, and the soluble solid content is 13.5 ˚Bx. The flesh is very soft and juicy and, rendering good eating quality. Shelf life is about 20 days under the ambient temperature, and it is a relatively long period as compare with other early season cultivars.

        가스크로마토그래피를 이용한 요중 마뇨산 측정시 새로운 시료 전처리 방법의 검증

        Urinary hippuric acid was analyzed by gas chromatography/mass spectrometry method. A simple and economical derivatization procedure using excessive methanol in acidic condition was adopted to esterify hippuric acid. Quantitative analysis was performed with two internal standards, heptadecanoic acid and hydrocinnamic acid, to evaluate the derivatization procedure as an analytical method. Using the standard addition method, linear and reproducible calibration curves were obtained for both internal standards. Calibration experiment showed that good quantitative result could also be obtained with the calibration curve obtained without urine matrix. Recovery rates for the urine samples were in the range of 95.6∼111.4%. Using hydrocinnamic acid as internal standard, analysis time could be reduced.

        전환교육 관점에서 본 정신지체아 여가 교육 활동의 이론적 탐색

        전환교육의 영역이 최근 협의의 영역에서 포괄적인 영역으로 이동함에 따라 여가 및 레크리에이션에 관한 중요성이 부각되고 있다. 이에 본 연구는 정신지체 학생에게 여가교육이 차지하는 중요성에 대해 살펴봄으로써 우리나라 정신지체 학교교육 현장에서 여가교육 프로그램을 계획ㆍ구성하기 위한 기초연구이다. 연구방법으로 관련 문헌을 중심으로 고찰했다. 선행연구들(Schleien & Ray, 1998; Hughes & Fullwood, 1985; Moon & Bunker, 1987; Meyer & Evans, 1989; Heyne, Schlein & Rynders, 1997)은 정신지체 학생들이 일반 학생들에 비해 실패감을 많이 경험하고 자아 개념과 동기수준 결여 및 사회적 상호작용 결핍을 보이기 때문에, 인지적 요구가 적은 여가활동을 통해 자아개념과 자신감, 정상화 촉진, 부적절한 행동 감소 등의 교육적 가치를 강조하여 여가교육을 중요한 영역으로 다루고 있다. 연구를 통해 얻은 결론은 학교 현장에서 여가교육을 실시할 때 학교 교육과정 내에서 실시할 수 있는 방법과 여가프로그램을 구성할 시 정신지체 학생에게 적합한 여가 활동 프로그램을 고려하여 시행해야 한다. Recently, as the domains of transition education move it from a narrow sense to comprehensive sense, importance of leisure and recreation in domains of transition education be highlighted. Therefore, this study is a basic search that may use for make a planning and constitution of leisure education programs at school for mental retardation in korea, as we look into importance of leisure education for children with mental retardation described in literature. As preceding literature(Schleien & Ray, 1998; Hughes & Fullwood, 1985; Moon & Bunker, 1987; Meyer & Evans, 1989; Heyne, Schlein & Rynders, 1997) showed that children with mental retardation have a more failure experience, deficient in self concept, motive and social interrelation than normal children stress value of education of leisure activities, because leisure activities demanded less intellectual accelerate for children with mental retardation to self concept, confidence, normalization, reduction of inadequacy behavior. In conclusion, when implementation to leisure education in the field of school, we should be several considerations that how to act in school curriculum and leisure activities programs fitted to children with mental retardation.

      • 분산 네트워크 관리 시스템 지원을 위한 멀티캐스트 프로토콜의 설계 및 구현

        This paper presents a new model for group communication facility which is gradually magnified. This model has been implemented in LAN environment and can get the enhancement in performance because a new manager can be dynamically created determined depending on the type of receiving message. As an object oriented approach has been introduced in designing the system, it provides an application programmer with the means to develop a new application facility without having to build the detail functions. As the members in a group are only identified by the index in a table, the overhead for the address referencing can be minimized. In this paper, some requirements for multicast communication, the structure of the system, and service primitives will be describe in detail. As the number of manager can be dynamically created and destroyed relying on the requirement, the system can efficeintly operated. Multicast facility designed in this paper is for supporting a distributed network management system.

        Effect of Ultrasonic Melt Treatment on Solidification Microstructure of Al–5Ti–1B Alloy Containing Numerous Inoculant Particles

        The effect of ultrasonic melt treatment (UST) on the solidification microstructure of an Al–5Ti–1B alloy containing highvolumefractions of Al3Tiand TiB2particles is investigated for various UST times with different melt holding times. Theas-cast Al–5Ti–1B alloy is composed of TiB2and polygonal Al3Tiparticles (present prior to UST), plate-like Al3Tiparticles,and Al grains (formed during UST and/or solidification). The UST causes a size reduction and homogeneous distributionof the TiB2-agglomerated region containing many submicron-sized TiB2particles pushed to the grain boundaries. The USTslightly decreases the size and improves the distribution of polygonal Al3Tiparticles enriched in the TiB2-agglomeratedregion. Unlike the TiB2and polygonal Al3Tiparticles, which exhibit a minor refining effect, the plate-like Al3Tiparticlesshow a significant refinement with UST application. The UST has a significant effect on the size distribution of Al grains byinducing the formation of medium-sized grains at the expense of small and large grains; however, it only has a slight effecton grain refinement. The degree of microstructure modification increases with increasing UST time but decreases with meltholding time after UST. The mechanisms for the refinement and dispersion of the TiB2and Al3Tiparticles and Al grains arediscussed considering fragmentation, nucleation, and growth behaviors induced by the UST and subsequent solidification.

      • 생쥐 뇌조직 체외배양법을 활용한 향산화제의 향독성효과에 관한 연구

        In an effort to establish an in vitro model of Parkinson's disease (PD), we prepared slice cultures of midbrain from newborn mouse. The slice section containing substantia nigral region was cultured for 10days. The striatal culture was grown to preserve the distribution of interneurons with minimal contamination of any other brain tissue. Using this explant culture containing both glial cells and nitro-striatal dopamine neurons, the protective effect of antioxidant pineal hormone and melatonin on 1-methyl-4-phenyl-1, 2,3,6-tetrahydropyridine (MPTP) -induced loss of the dopamine neurons was investigated. MPTP selectively damaged dopaminergic neurons, as indicated by immunoblot and immunohistochemistry for glial fibrillary acidic protein (GFAP) and tyrosine hydroxylase, the marker enzymes for glial and dopaminergic neurons, respectively. Twenty-four-hour treatment of MPTP (50 μM) in the explant culture induced 37% loss of dopaminergic neurons while no apparent damage was observed in glial region. Type B monoamine oxidase (MAO-B) that resides in glial cells, produces free radicals during catecholamine metabolism and also oxidizes MPTP to its active form, MPP+, was activated by 2.2 times after MPTP treatment. Melatonin was able to recover both the dopaminergic neuronal loss and the elevated MAO-B activity level fully to the untreated control level. Similar observation was made bur the co-treatment of vitamin E with MPTP, suggesting the generation of oxygen radical species by MPTP. On behalf of pursuing molecular mechanisms of Parkinsonism, the data illustrate the proposed mechanism of MPTP-induced dopaminergic cell death ; namely, MPP+ that is processed by MAO-B in glial cells is taken up avidly by dopaminergic neurons and causes a toxic effect at least partly, if not mostly, through oxidative stress. The culture system, therefore, can serve as an experimental model of PD in which the putative neuroprotective effects of antioxidants can be investigated. This model is also useful for screening novel drugs and growth factors without a massive sacrifice of animals, and ultimately for developing neuroprotective therapies.

        Antimutagenic Effects of Ginsenoside Rb$_1$, Rg$_1$ in the CHO-K1 Cells by Benzo[a]pyrene with Chromosomal Aberration Test and Comet Assay

        The usage and types of chemicals are advancing, specializing, large-scaled increasing, and new chemical exposed workers are concerning to occupational disease. The generation of reactive oxygen in the body from carcinogen, mutation and DNA damage in cancer is protected by natural antioxidants (phytochemicals) with antimutagenic effect. There were many reports of ginsenoside Rb$_1$, Rg$_1$ grievances of the genetic mutation to suppress the effect confirm the genetic toxicity test with chromosomal aberration test and the Comet (SCGE) assay confirmed the suppression effect occurring chromosomal DNA damage. We had wanted to evaluate the compatibility and sensitivity between the chromosomal aberration (CA) test and the Comet assay. We used the CA test and Comet assay to evaluate the anti-genotoxicity of ginsenoside Rb$_1$ and Rg$_1$, in CHO-K1 (Chinese hamster ovary fibroblast) cell in vitro, composed negative control (solvent), positive control (benzo[a]pyrene), test group (carcinogen+variety concentration of ginsenoside) group. The positive control was benzo[a]pyrene (50 $\mu$M), well-known carcinogen, and the negative control was the 1 % DMSO solvent. The test group was a variety concentration of ginsenoside Rb$_1$, Rg$_1$ with 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1%, 10%. In chromo-somal aberration test, we measured the number of cells with abnormally structured chromosome. In Comet assay, the Olive tail moment (OTM) and Tail length (TL) values were measured. The ratio of cell proliferation was increased 8.3% in 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1%, 10% Rb$_1$ treated groups, and increased 10.4% in 10$^{-10}$%, 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1% Rg$_1$ treated groups. In the CA test, the number of chromosomal aberration was decreased all the Rb$_1$ and Rg$_1$ treated groups. In the Comet assay, the OTM values were decreased in all the Rb$_1$ and Rg$_1$ treated groups. To evaluate the compatibility between CA and Comet assay, we compared the reducing ratio of chromosomal abnormalities with its OTM values, it was identified the antimutagenicity of ginsenoside, but it was more sensitive the CA test than the Comet assay. Ginsenoside Rb$_1$ and Rg$_1$ significantly decrease the number of cells with chromosomal aberration, and decrease the extent of DNA migration. Therefore, ginsenoside Rb$_1$, Rg$_1$ are thought as an antioxidant phytochemicals to protect mutagenicity. The in vitro Comet assay seems to be less sensitive than the in vitro chromosomal aberration test.
