        Comparison of Six Automated Immunoassays With Isotope-Diluted Liquid Chromatography–Tandem Mass Spectrometry for Total Thyroxine Measurement

        Background: Accurate serum total thyroxine (TT4) measurement is important for thyroid disorder diagnosis and management. We compared the performance of six automated immunoassays with that of isotope-diluted liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) as the reference method. We also evaluated the correlation of thyroid stimulating hormone (TSH) with TT4 measured by ID-LC-MS/MS and immunoassays. Methods: Serum was collected from 156 patients between October 2015 and January 2016. TT4 was measured by immunoassays from Abbott (Architect), Siemens (ADVIA Centaur XP), Roche (E601), Beckman-Coulter (Dxi800), Autobio (Autolumo A2000), and Mindray (CL-1000i), and by ID-LC-MS/MS. Results were analyzed using Passing–Bablok regression and Bland–Altman plots. Minimum requirements based on biological variation were as follows: a mean bias of ≤4.5% and total imprecision (CV) of ≤3.7%. Results: All immunoassays showed a correlation >0.945 with ID-LC-MS/MS; however, the slope of the Passing–Bablok regression line varied from 0.886 (Mindray) to 1.23 (Siemens) and the intercept from -12.8 (Siemens) to 4.61 (Mindray). Only Autobio, Beckman-Coulter, and Roche included the value of one in the 95% confidence interval for slope. The mean bias ranged from -10.8% (Abbott) to 9.0% (Siemens), with the lowest value noted for Roche (3.5%) and the highest for Abbott (-10.8%). Only Abbott and Roche showed within-run and total CV ≤3.7%. Conclusions: Though all immunoassays correlated strongly with ID-LC-MS/MS, most did not meet the minimum clinical requirement. Laboratories and immunoassay manufacturers must be aware of these limitations.

        Effects of high moisture ear corn on production performance, milk fatty acid composition, serum antioxidant status, and immunity in primiparous dairy cows

        Objective: This study evaluated the effects of high moisture ear corn (HMEC) on production performance, milk fatty acid composition, serum antioxidant status, and immunity in primiparous dairy cows.Methods: A total of 45 healthy primiparous Holstein cows (36.50±4.30 kg of milk/d, 201±9.00 lactating days in milk) were sorted into 3 groups: control group (CG, n = 15); 50% HMEC (replacing 50% steam-flaked corn with HMEC, n = 15); and 100% HMEC (replacing steam-flaked corn with HMEC, n = 15) on an equal dry matter (DM) basis. The study consisted of adaptation period of 14 days, followed by a formal period of 60 days. Feed intake and milk yield were recorded daily. Milk and blood samples were collected on 1, 30, and 60 d of the experimental period.Results: The 50% HMEC group and 100% HMEC group significantly increased (p<0.05) milk yield and DM intake in dairy cows compared to the control group (CG). The 100% HMEC group showed an increase (p<0.05) in 4% fat-corrected milk (4% FCM). Both the 50% HMEC group and 100% HMEC group exhibited significant decreases (p<0.05) in the content of C10:0, C12:0, and C14:0 fatty acids, along with a significant increase (p<0.05) in cis-9C18:1 content. The saturated fatty acid content was significantly lower (p<0.05) in the 50% HMEC and 100% HMEC groups than that of CG. Conversely, the monounsaturated fatty acid content was higher (p<0.05) in the 50% HMEC and 100% HMEC groups than that in CG. Notably, the 100% HMEC group significantly increased (p<0.05) the serum superoxide dismutase and glutathione peroxidase content, while also decreasing the serum malondialdehyde content (p<0.05). Moreover, the 100% HMEC group significantly increased (p<0.05) the content of immunoglobulin G (IgG) and IgM.Conclusion: High moisture ear corn could improve production performance and milk fatty acid levels and enhance immunity and antioxidant capacity in dairy cows. These results lay the foundation for the wider application of HMEC in ruminant animal diets.         한국 및 중국 기업의 기술개발투자가 기업가치에 미치는 영향: 외국인 투자자를 중심으로

        In this study, when technological innovation has become a key driver of corporate competitiveness and national economic growth, Korea and China have made remarkable progress in the technology field in recent decades. This study investigates the impact of technology development and foreign investments on corporate value in listed companies in South Korea and China. Over recent decades, both nations have rapidly advanced technologically, with increased investments driving economic growth and enhancing corporate profitability. Analyzing data from 2010 to 2023, the research finds that substantial technology investments lead to notable improvements in financial performance and corporate value. Foreign investments also positively influence corporate value. Notably, technology investments have a greater effect on financial performance in South Korea, while they more significantly impact corporate value in China. Foreign investments further enhance the positive effects of technology development on corporate value in both countries. The study offers insights into how South Korean and Chinese companies can boost management performance through technology investments and highlights the role of foreign investments in moderating these effects, considering the distinct economic and policy contexts of each country.

        Development of an immunochromatographic strip for detection of antibodies against porcine reproductive and respiratory syndrome virus

        A simple and rapid immunochromatographic test strip incorporating a colloidal gold-labeled recombinant Nsp7 antigen probe was successfully developed for the detection of anti-porcine reproductive and respiratory syndrome virus (PRRSV) antibodies in swine. Recombinant Nsp7 protein of PRRSV labeled with colloidal gold was dispensed on a conjugate pad for use as the detector. Staphylococcal protein A and purified porcine anti-Nsp7 antibodies were blotted on a nitrocellulose membrane to form test and control lines, respectively. A comparison of the strip with standard diagnostic tests, enzyme-linked immunosorbent assays and immunoperoxidase monolayer assay, was also performed. The immunochromatographic test strip was shown to be of high specificity and sensitivity. Furthermore, the strip assay is rapid and easy to perform with no requirement for professional-level skills or equipment. It is suggested that the immunochromatographic test strip can be used to quickly and accurately detect PRRSV antibody and to be suitable for diagnostic purposes in the field.

      • Tests on explosion-resisting properties of high-performance equal-sized-aggregate concrete composite sandwich plates

        Targeted introduction of explosion-resisting and energy-absorbing materials and optimization of explosion-resisting composite structural styles in underground engineering are the most important measures for modern engineering protection. They could also improve the survivability of underground engineering in wartime. In order to test explosion-resisting and energy-absorbing effects of high-performance equal-sized-aggregate (HPESA) concrete, the explosive loading tests were conducted on HPESA concrete composite plates by field simple explosion craters. Time-history curves of the explosion pressure at the interfaces were obtained under six conditions with different explosion ranges and different thicknesses of the HPESA concrete plate. Test results show that under the same explosion range, composite plate structures with different thicknesses of the HPESA concrete plate differ significantly in terms of the wave-absorbing ability. Under the three thicknesses in the tests, the wave-absorbing ability is enhanced with the growing thickness and the maximum pressure attenuation index reaches 83.4%. The energy attenuation coefficient of the HPESA concrete plate under different conditions was regressively fitted. The natural logarithm relations between the interlayer plate thickness and the energy attenuation coefficient under the two explosion ranges were attained.

      • Seismic performance of a novel bolt-and-welded connection of box-section beam and box-section column

        The H-shaped steel beam is popular due to its ease of manufacturing and connection to the column. This profile, which is used as a shallow beam, needs the high weak-axis bending stiffness and torsional stiffness to meet the overall stability. Achieving the local beam flange stability, bearing capacity, bending stiffness, and torsional requirements need a great thickness and width of the beam flange, which causes, which will cause more uneconomical structural design. So, the box-section beam is the ideal alternative. However, the current design specifications do not have design rules for the bolt-and-welded connection of the box-section beam and box-section column. The paper proposes a novel bolt-and-welded connection of the box-section beams and box-section columns based on a high-rise structural design scheme. Three connection models, BASE, WBF, and RBS, are analyzed under cyclic loading in ABAQUS software. The failure modes, hysteresis response, bearing capacity, ductility, plastic rotation angle, energy dissipation, and stiffness degradation of all models are determined and compared. Compared with the other two models, the model WBF exhibited excellent seismic performance, ductility, and plastic rotation ability. Finally, model WBF was chosen as the connection scheme used in the project design.

        Novel multiple-acidic ionic liquids: Green and efficient catalysts for the synthesis of bis-indolylmethanes under solvent-free conditions

        Four novel multiple-acidic ionic liquids based on triethanolamine (TEA) were prepared and used as efficient catalysts to synthesize bis-indolylmethanes at room temperature without any organic solvent. [TEOA][HSO4] showed the best catalytic performance. The optimal amount of catalyst was 10 mol%. Various aldehydes/ketones reacted with indole/substituted indole smoothly and afforded to corresponding products in 70–99% yields within minutes. Additionally, the ionic liquid could be reused up to five times with only a slight decrease in catalytic activity. Finally, a possible reaction mechanism was given. Techniques of acidity test and NMR were introduced to verify the proposed mechanism.

        Anti-tumor activities of Panax quinquefolius saponins and potential biomarkers in prostate cancer

        Background: Prostate carcinoma is the second most common cancer among men worldwide. Developing new therapeutic approaches and diagnostic biomarkers for prostate cancer (PC) is a significant need. The Chinese herbal medicine Panax quinquefolius saponins (PQS) have been reported to show anti-tumor effects. We hypothesized that PQS exhibits anti-cancer activity in human PC cells and we aimed to search for novel biomarkers allowing early diagnosis of PC. Methods: We used the human PC cell line DU145 and the prostate epithelial cell line PNT2 to perform cell viability assays, flow cytometric analysis of the cell cycle, and FACS-based apoptosis assays. Microarray-based gene expression analysis was used to display specific gene expression patterns and to search for novel biomarkers. Western blot and quantitative real-time PCR were performed to demonstrate the expression levels of multiple cancer-related genes. Results: Our data showed that PQS inhibited the viability of DU145 cells and induced cell cycle arrest at the G1 phase. A significant decrease in DU145 cell invasion and migration were observed after 24 h treatment by PQS. PQS up-regulated the expression levels of p21, p53, TMEM79, ACOXL, ETV5, and SPINT1 while it down-regulated the expression levels of bcl2, STAT3, FANCD2, DRD2, and TMPRSS2. Conclusion: PQS promoted cells apoptosis and inhibited the proliferation of DU145 cells, which suggests that PQS may be effective for treating PC. TMEM79 and ACOXL were expressed significantly higher in PNT2 than in DU145 cells and could be novel biomarker candidates for PC diagnosis.

        Development of a dual-mode energy-resolved neutron imaging detector: High spatial resolution and large field of view

        Energy-resolved neutron imaging is an effective way to investigate the internal structure and residual stress of materials. Different sample sizes have varying requirements for the detector’s imaging field of view (FOV) and spatial resolution. Therefore, a dual-mode energy-resolved neutron imaging detector was developed, which mainly consisted of a neutron scintillator screen, a mirror, imaging lenses, and a time-stamping optical fast camera. This detector could operate in a large FOV mode or a high spatial resolution mode. To evaluate the performance of the detector, the neutron wavelength spectra and the multiple spatial resolution tests were conducted at CSNS. The results demonstrated that the detector accurately measured the neutron wavelength spectra selected by a bandwidth chopper. The best spatial resolution was about 20 μm in high spatial resolution mode after event reconstruction, and a FOV of 45.0 mm × 45.0 mm was obtained in large FOV mode. The feasibility was validated to change the spatial resolution and FOV by replacing the scintillator screen and adjusting the lens magnification.
