      • Novel Methylation Biomarker for Non-invasive Diagnostics in Lung Cancer

        To identify aberrantly hypermethylated DNA in lung cancer cells we established a genome-wide analysis for hypermethylation sites, namely Methyl DNA Isolation and Amplification (MeDIA) coupled-CpG microarray analysis. In the comprehensive methyaltion profiling analysis between human lung cancer, A549 cells and normal NHBE cells, we observed that several clusters of genes show a significant level of aberrancy in CpG island methylation pattern in cancer cells compared to normal cells. We further identified PCDHGA12 gene as a new marker of non-invasive diagnostics for lung cancer based on followings. 1) Transcription of PCDHGA12 gene is reactivated after treatment of A549 cells with demethylating agent. 2) Bisulfide clonal-sequencing reveals that CpG island of PCDHGA12 shows a distinctive differential methylation between two cell lines. 3) Pyrosequencing-based quantitative methylation assay for such region in tumor and non-tumorous tissues from lung cancer patients shows aberrant hypermethylation in 37 (92%) of the 40 tumor tissues. In clinical validation by pyrosequencingin induced-sputum of lung cancer patients (n=87) and healthy controls (n=51), we observed aberrant hypermethylation incident at significantly elevated level in samples derived from lung cancer patients. According to the optimal threshold calculated by ROC curve analysis, sensitivity and specificity of PCDHGA12 was 86.2% and 82.4%, respectively. PCDHGA12 methylation status could be a potential methylation biomarker alone or combined with others for the screen and the detection of relapse of lung cancer.

        건물구조체의 통기시스템을 이용한 냉ㆍ난방부하 저감효과 및 경제성 평가

        To evaluate the energy efficiency of the building envelopes with ventilated cavity, the reduction on cooling load and heating load were analyzed with ESP-r simulation program which is possible to interpret the energy consumption on various types of building envelopes. For the economic assessment of the ventilated cavity in the internal part of the office building envelope, the column & spandrel type was compared with the conventional curtain wall. The result of the computer simulation shows that 11.2~13.7% of the cooling & heating energy can be reduced in building envelope with ventilated cavity.

        실물실험에 의한 통기구조를 가진 외벽체의 에너지 성능평가 및 예측모델 개발

        This field-based study focused on evaluating the thermal performance in building envelope with ventilated cavity, which is set in the internal part of the conventional curtain wall. The fire resistance, the airtightness and the insulation efficiency were evaluated through mock-up tests. Temperature and air velocity were measured and analyzed by CFD according to the mechanical or natural control of the ventilated cavity. We made a regression model based on the results of this study to predict the rate of temperature drop on the curtain wall with ventilated cavity in summer.

        지하저장시설 통기순환형 결로발생유도시스템에 관한 실험적 연구 : 군 이글루 탄약고를 중심으로 Focused on Igloo Type Ammunition Storage Facility

        The objective of this study is to estimate applicability and performance of the Condensation Induction System by the experiment. This system consisted of the dehumidification system, thermal exchange system and air rotation apparatus. The concept of this system is to remove water vapor in room air of the powder magazine taking advantage of the existing wall and the condensation production apparatus(dehumidification system), and to make dry stored goods and room by circulating high dry air through the thermal exchange system. The results show that the Condensation Induction System dehumidified sufficiently the room and prevented an corrosion of stored ammunition. The dehumidification effect per one power[W] of this system is three times as much as that of the existing ready-made dehumidifier.

      • 활성화된 석탄회분 칼럼을 이용한 염료의 흡착특성

        유연탄의 연소시에 발생되는 산업폐기물인 coal ash를 산처리하여 activated coal ash로 개량하여 흡착제로 사용하고 반응성 염료인 Sumifix Red와 산성염료인 Red EB-L을 시료수로 사용하여 흡착실험을 한 결과 pH에 따른 흡착제거율은 시료수 모두 pH 3~9범위에서는 거의 변동이 없었으나, pH 11에서는 급격하게 감소하였고, 반응시간에 따른 흡착제거율은 activated coal ash를 10g 주입했을 경우 0.5시간 경과후에 Sumifix Red는 54.8%, Red EB-L은 88.7%로 나타났으며, 5시간 경과 후엔 변화가 거의 없었고, 온도에 따른 흡착제거율은 Red EB-L에 있어서는 거의 변동이 없었고, Sumifix Red에서는 15℃에서 가장 높았고 온도가 증가함에 따라 제거율은 감소하는 경향을 나타내었다. 또한, Freundlich 흡착등온식을 이용하여 흡착이 평형에 도달했을 때의 평형농도와 흡착량과의 관계에서 1/n과 k값은 반응성 염료인 Sumifix Red가 각각 0.604, 0.016이었고, 산성 연료인 Red EB-L은 0.921, 0.058로 나타났다. 칼럼을 이용한 activated coal ash의 흡착제거율을 살펴본 결과는 회분식에서와 마찬가지로 Red EB-L이 Sumifix Red보다 높았고, Bohart와 Adams의 식을 이용하여 흡착칼럼을 설계 해 본 결과 흡착용량은 Red EB-L이 74.96kg/m³,17.22kg/m³ 이었으며, activated coal ash이 이론적 충진깊이는 Red EB-L과 Sumifix Red 모두 0.1m 정도로 나타났다. This study was conducted to investigate the characteristics of absorption in continuous waste water treatment system containing activated coal ash. Activated coal ash was produced from bituminous coal and treated by direct acid leaching(DAL) in order to get better activity as porosity absorbent. On this experiment performed by batch and continuous methods, acid and reactive type dyes were selected to find out the feasibility of activated coal ash as absorbent. The absorption removal rate of activated coal ash were nearly constant at the pH range of 3 to 9 on Red EB-L and Sumifix Red. But reduced rapidly at pH 11. The adsorption removal rate of absorbent was not changed at the temperature range of 15℃ ~ 45℃ on acid dye Red EB-L. But in case of reactive dye Sumifix Red, absorption removal rate was reduced at 15℃. When the absorption equilibrium was calculated based on the Freundlich isotherm equation, constant k was 0.016, 0.058 and 1/n was 0.604, 0.921 for Sumifix Red, Red EB-L, respectivey. Removal rate of acid dye Red EB-L was higher than that of reactive dye Sumifix Red by activated coal ash at the continuous column system. Batch system illustrated same tendency. The absorption capacities of absorption column, designed by Bohart and Adams equation, were 74.96 kg.m³17.22 kg/m³ for Red EB-L, Sumifix Red, respectively, and theoretical packing depth of activated coal ash was 0.1 m for all.

        건축물 건설단계에서의 에너지소비량 및 CO₂배출량 원단위 산출

        The purpose of this study is to establish the database that could be applied to the building LCA Firstly, in this paper, the methodologies of building LCA and energy based I/O model for estimating embodied energy of building materials were examined Then, the intensity of energy consumption and CO₂ emission for building materials and construction stage were estimated The intensity database for building materials calculated by the I/O table published in 2000 was compared with the database by the I/O table published in 1995 According to the result of comparison, there were a little differences between the results calculated by the new and the previous data, and two results showed nearly the same patterns So, it could be said that the industrial activity change has little effect on the energy intensity of building materials during the past 5 years For the calculation of the energy intensity in the construction stages, the payment documents of an ad and electricity consumptions during construction for the sample buildings were surveyed and the intensity per a building gross area were calculated The database presented in this paper could be applied to the building LCA process as the basic data for analysing global worming effect caused by building construction

        Systemic immunity of obese-diabetes model (db/db) mice

        Obesity has recently been defined as a chronic inflammatory disease and is considered as a major cause of adult health problems including incurable diseases like diabetes and cancer. In this study, the systemic immunity, including the innate and adaptive immunity parameters, of naturally occurring (leptin receptor mutation) obesity-diabetes mice (db/db) was examined to increase our knowledge of these mice for obesity-related studies. Severe fatty liver with blood engorgement was observed in the db/db mice. Compared to background C57BL/6J mice, the adaptive immunity, as measured by mitogen-induced T and B cell proliferation and cytokine release, was significantly suppressed in db/db mice. However, significant upregulation of innate immune-inflammatory parameters including macrophage function and NK cell activity was observed in db/db mice without external stimulation. These data conclusively confirm the systemic immune-inflammatory micro-environment with suppressed adaptive immunity of db/db mice, which may cause the secondary obesity-related life-threatening diseases like diabetes or cancer.
