      • Development of a parallel microbore hollow fiber enzyme reactor platform for online <sup>18</sup>O-labeling: Application to lectin-specific lung cancer <i>N</i>-glycoproteome

        <P><B>Abstract</B></P> <P>We introduce a simple online <SUP>18</SUP>O-labeling protocol for protein samples that uses a parallelizing microbore hollow fiber enzyme reactor (mHFER) as an alternative tool for online proteolytic digestion. Online <SUP>18</SUP>O-labeling is performed by separately attaching two mHFERs in parallel to a 10-port switching valve with a high-pressure syringe pump and two syringes containing <SUP>16</SUP>O- or <SUP>18</SUP>O-water. <SUP>16</SUP>O-/<SUP>18</SUP>O-labeled peptides are formed in this manner and simultaneously analyzed online using nanoflow liquid chromatography-tandem mass spectrometry (nLC-MS/MS) without any residual trypsin activity. The usefulness of a parallel mHFER platform (P-mHFER) in <SUP>18</SUP>O-labeling was tested using both cytochrome C and alpha-1-acid-glycoprotein to verify the incorporation level of two <SUP>18</SUP>O atoms into tryptic peptides and to provide a quantitative assessment with varied mixing ratios. Additionally, our <SUP>18</SUP>O-labeling approach was used to study the serum <I>N</I>-glycoproteome from lung cancer patients and controls to evaluate the applicability of lectin-based quantitative <I>N</I>-glycoproteomics. We successfully quantified 76 peptides (from 62 <I>N</I>-glycoproteins). Nineteen of these peptides from lung cancer serum were up-/down-regulated at least 2.5-fold compared to controls. As a result, the P-mHFER-based online <SUP>18</SUP>O-labeling platform presented here can be a simple and reproducible way to allow quantitative proteomic analysis of diverse proteome samples.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Developing MW sorting-based parallel mHFER (P-mHFER) for <SUP>18</SUP>O-labeling of protein(s) </LI> <LI> P-mHFER allows to form <SUP>18</SUP>O<SUB>2</SUB>-labeled peptides online in a short time (~30 min). </LI> <LI> MW sorting-based P-mHFER lowers the back-exchange caused by trypsin activity. </LI> <LI> P-mHFER has the potential in proteolytic <SUP>18</SUP>O-labeling for quantitative proteomics. </LI> </UL> </P>

        How COVID-19 Affected Healthcare Workers in the Hospital Locked Down due to Early COVID-19 Cases in Korea

        Background: The healthcare workers (HCWs) were exposed to never-experienced psychological distress during the early stage of the coronavirus disease 2019 (COVID-19) pandemic. The aim of this study was to investigate how the COVID-19 pandemic affected the mental health of HCWs during the hospital lockdown period due to mass healthcareassociated infection during the early spread of COVID-19. Methods: A real-time online survey was conducted between April 14–18, 2020 among HCWs who worked at the university hospital where COVID-19 was confirmed in a patient, and the hospital was shut down for 3 weeks. Along with demographic variables and work-related information, psychological distress was measured using the Generalized Anxiety Disorder-7 (GAD-7), the Patient Health Questionnaire-9 (PHQ-9), the Maslach Burnout InventoryGeneral Survey scale, and the Stress and Anxiety to Viral Epidemics-9. Results: The HCWs working in the cohort ward and those who have experienced social discrimination had significantly higher level of depression (PHQ-9 score; 5.24 ± 4.48 vs. 4.15 ± 4.38; P < 0.01 and 5.89 ± 4.78 vs. 3.25 ± 3.77; P < 0.001, respectively) and anxiety (GAD-7 score; 3.69 ± 3.68 vs. 2.87 ± 3.73; P < 0.05 and 4.20 ± 4.22 vs. 2.17 ± 3.06; P < 0.001, respectively) compared to other HCWs. Worries regarding the peer relationship and the skepticism about job were associated with depression (odds ratio [OR], 1.39; 95% confidence interval [CI], 1.07–1.79; P < 0.05 and OR, 1.69; 95% CI, 1.31–2.17; P < 0.001, respectively) and anxiety (OR, 1.73; 95% CI, 1.21–2.49; P < 0.01 and OR, 1.54; 95% CI, 1.09–2.17; P < 0.05, respectively), while fear of infection or worsening of health was not. Path analysis showed that work-related stress associated with the viral epidemic rather than anxiety about the viral epidemic mainly contributed to depression. Conclusion: The present observational study indicates that mental health problems of HCWs exposed to COVID-19 are associated with distress in work and social relationship. Early intervention programs focusing on these factors are necessary.

      • An Optimal Estimation of Paramter Functions in ODEs

        A mathematical model which consists of a set of differential equations is widely used in many scientific fields. However, the inverse problem, the problem of parameter estimation based on the measurements of output variables, has not been well explored, although some least squares based approaches have been proposed and studied. It happens frequently that the model equations contain a number of unknown parameters. The aim of parameter estimation problem is to determine these unknown parameters. To do that, one formulates an appropriate constrained optimization problem to minimize the error between the measurements from the laboratory or the plant and the theoretical results obtained by solving the set of differential equations. In this talk, we will concerned with the problem of fitting a mathematical model involving a parameter function to given data. We focus on the case where the mathematical model is a linear differential equation and the parameter function is a piecewise constant function. We will propose the sufficient and necessary conditions for the given data set to derive the parameter function which fits the corresponding solution to the data set. Moreover, we suggest an algorithm for parameter function estimation in the case where the subintervals for piecewise constant function are yet to be determined.

      • ANTHROPOMORPHISM IN LUXURY GOODS AND LUXURY SERVICE BRAND EVALAUTIONS

        Even though luxury brands embraced social media (Godey et al., 2016; Kim & Ko, 2012), research on the use of social media for luxury brand marketing is limited. Given the increasing popularity of social media, it is imperative to examine social media-based communication strategies for luxury brands to achieve desirable consumer evaluations of brands and advertising. This research explored the role of brand anthropomorphism strategies in social media in enhancing consumers’ advertising and brand evaluations. Specifically, this research investigated (1) the effects of verbal-based brand anthropomorphism in the luxury goods brand context (Study 1), (2) the effects of verbalbased brand anthropomorphism in the luxury service brand context (Study 2), and (3) the effects of verbal-based brand anthropomorphism cues (Study 3) on consumers’ luxury brand and advertising evaluations. Studies 1 and 2 both collectively examined the role of self-focused anthropomorphized brand effects in two different contexts. Consumers easily connect themselves with an anthropomorphized advertising, compared to an objectified advertising for luxury brands. Study 3 identified the fit between luxury brand type and an anthropomorphized brand cue to explore the effectiveness of brand anthropomorphism. Depending on the type of luxury brands, consumers processed anthropomorphized brand cues differently. Consumers favorably perceived the fit between luxury good brands and partner cues; on the other hand, consumers favorably perceived the fit between luxury service brands and servant cues. Results of this research contribute to the social media advertising and luxury brand marketing literature by clarifying the role of brand anthropomorphism in consumer-brand relationships as well as in brand-advertising relationships. Results also provide industry suggestions for delivering personified brand messages to have desirable consumer brand and advertising evaluations.

        Enhancing customers’ positive responses: Applying sensory marketing to the hotel website

        Sensory marketing enables visualization of intangible services, helping to promote customers’ future hotel experiences. This study investigates how sensory information, brand relationships, and review types affect customers’responses in the context of the hotel industry. Customers’response types include narrative persuasion, experiential, emotional, and cognitive responses. Conducting two 2 × 2 scenario- based between-subjects experiments with previous hotel customers in the US, Study 1 confirms the effect of brand relationships on story- format sensory information that promote customers’ transportation experience, brand experience, and emotional responses. Study 2 also identifies effects of review types on story-format sensory information that enhance customers’transportation experience, brand experience, and trust. Results inform theoretical contributions and practical implications to promote customers’ future experiences.

      • P-32 Prognostic Factors for Lung Cancer in Idiopathic Pulmonary Fibrosis

        Background: Lung cancer in idiopathic pulmonary fibrosis (IPF) shows poorer prognosis than lung cancer without. However, prognostic factors for lung cancer in IPF are not well defined. Methods: Clinical data were retrospectively analyzed in 610 patients with lung cancer (IPF=122) diagnosed from 2007 to 2015 at Asan Medical Center. Lung cancer subjects without IPF (n=488) were randomly selected as controls from registry, matched by age, sex, histologic subtype, stage and date of diagnosis. Results: Median follow-up period was 16.0 months. IPF group showed more frequent cough (68.6 vs. 49.9%), dyspnea (70.1 vs. 31.5%) and location at lower lobe (61.1 vs. 45.2%) and lower FVC (72.9 vs. 80.4% predicted), DLco (53.3 v.s 72.7% predicted) and TLC (73.3 vs. 90.3% predicted) compared to non-IPF group (all p<0.05). IPF group had poorer prognosis (median survival: 12.0 vs. 22.0 months, p<0.001) compared to non-IPF group, however, in the case of stage 4 or small cell lung cancer, prognosis was not different. IPF was an independent prognostic factor (HR 1.578) for overall lung cancer, with ever-smoker (HR, 2.747), FVC (HR 0.987), DLco (HR 0.992) and advanced stage (HR, 3.539). Body mass index (HR,0.860) and walking distance of 6 minute walk test (HR 0.996) were independent prognostic factors for IPF group with advanced stage (HR 5.596). Conclusions: IPF group had more respiratory symptoms, lower pulmonary function and poorer prognosis compared to non-IPF group. Low BMI and short distance of 6MWT may imply poor prognosis in IPF group.

        This study explores the role of the digital communication in determining customers’ e-service agent use behavior and examines whether perceived communicator authority interacts with communication style toward strengthening or weakening future e-service agent use intention. The results of this study provide pioneering evidence on the effects of digital service communication strategies. This study contributes to digital service research by exploring the communication strategies of e-service agents. Results also provide implications for practitioners on how to achieve most out of deploying digital communications in the digital customer service platform.

        Gait Variability Can Predict the Risk of Cognitive Decline in Cognitively Normal Older People

        <P><B><I>Background:</I></B> The aim of this study was to investigate the association of gait speed and gait variability, an index of how much gait parameters, such as step time, fluctuate step-to-step, with risk of cognitive decline in cognitively normal elderly individuals. While high gait variability is emerging as an early indicator of dementing illnesses, there is little research on whether high gait variability predicts cognitive decline in cognitively normal elderly who have no evidence of cognitive impairment. <B><I>Methods:</I></B> In this 4-year prospective cohort study on 91 community-dwelling cognitively normal elderly individuals without cerebral ischemic burden or Parkinsonism, we evaluated gait speed and step time variability using a tri-axial accelerometer placed on the center of body mass, and diagnosed mild cognitive impairment (MCI) according to the International Working Group on MCI. We performed Kaplan-Meier analysis with consecutive log-rank testing for MCI-free survival by cohort-specific tertiles of gait speed; hazard ratios (HR) of incident MCI were estimated using Cox proportional hazards regression analysis adjusted for age, sex, education level, Cumulative Illness Rating Scale score, GDS score, and presence of the apolipoprotein E ε4 allele. <B><I>Results:</I></B> Out of the 91 participants in the baseline assessment, 87 completed one or more 2-year follow-up assessments, and the median duration of follow-up was 47.1 months. Kaplan-Meier curves of incident MCI show evident differences in risk by gait variability group (χ<SUP>2</SUP> = 9.64, <I>p</I> = 0.002, log-rank test). Mean MCI-free survival in the high variability group was 12% shorter than in the mid-to-low tertile group (47.4 ± 1.74 [SD] vs. 54.04 ± 0.52 months), while it was comparable between gait speed groups (51.59 ± 0.70 vs. 50.64 ± 1.77 months; χ<SUP>2</SUP> = 1.16, <I>p</I> = 0.281). In multivariate analysis, subjects with high gait variability showed about 12-fold higher risk of MCI (HR = 11.97, 95% CI = 1.29–111.37) than those with mid-to-low variability. However, those with slow gait speed showed comparable MCI risk to those with mid-to-high speed (HR = 5.04, 95% CI = 0.53–48.18). <B><I>Conclusions:</I></B> Gait variability may be a better predictor of cognitive decline than gait speed in cognitively normal elderly individuals without cerebral ischemic burden or Parkinsonism.</P>

        Anhedonia and Dysphoria Are Differentially Associated with the Risk of Dementia in the Cognitively Normal Elderly Individuals: A Prospective Cohort Study

        Objective We investigated the impact of depressed mood (dysphoria) and loss of interest or pleasure (anhedonia)on the risk of dementia in cognitively-normal elderly individuals. Methods This study included 2,685 cognitively-normal elderly individuals who completed the baseline and 4-year follow-up assessments of the Korean Longitudinal Study on Cognitive Aging and Dementia. We ascertained the presence of dysphoria and anhedonia using the Mini International Neuropsychiatric Inventory. We defined subjective cognitive decline as the presence of subjective cognitive complaints without objective cognitive impairments. We analyzed the association of dysphoria and anhedonia with the risk of cognitive disorders using multinomial logistic regression analysis adjusted for age, sex, education, Cumulative Illness Rating Scale score, Apolipoprotein E genotype, and neuropsychological test performance. Results During the 4-year follow-up period, anhedonia was associated with an approximately twofold higher risk of mild cognitive impairment (OR=2.09, 95% CI=1.20–3.64, p=0.008) and fivefold higher risk of dementia (OR=5.07, 95% CI=1.44–17.92, p=0.012) but was not associated with the risk of subjective cognitive decline. In contrast, dysphoria was associated with an approximately twofold higher risk of subjective cognitive decline (OR=2.06, 95% CI=1.33–3.19, p=0.001) and 1.7-fold higher risk of mild cognitive impairment (OR=1.75, 95% CI=1.00–3.05, p=0.048) but was not associated with the risk of dementia. Conclusion Anhedonia, but not dysphoria, is a risk factor of dementia in cognitively-normal elderly individuals.
