In our recent work (Ma et al., in Planta229(3):457–469, 2009a and 229(4):1077–1086, 2009b),two three-intron type III PKS genes, PcPKS1 and PcPKS2,were isolated from Polygonum cuspidatum Sieb. et Zucc. Phylogenetic and functional analyses revealed PcPKS1 is athree-intron chalcone synthase (CHS) gene, and PcPKS2 isfound to be a three-intron benzalacetone synthase (BAS)gene. The regular CHS encoded by a single intron genehave not been isolated and characterized from P. cuspidatum. In this work a further CHS with one intron (PcPKS3)and a stilbene synthase (STS) gene with three-intron(PcPKS5) were isolated and characterized by functionaland phylogenetic analyses. In comparison with PcPKS1, abifunctional enzyme with both CHS and BAS activity, theenzymatic product of recombinant PcPKS3 was naringenin,bis-noryangonin (BNY) and 4-coumaroyltriacetic acidlactone (CTAL) occurred as side products. The PcPKS5synthesized resveratrol and a trace amount of naringeninfrom p-coumaroyl-CoA. To our knowledge, PcPKS5 is thefirst reported three-intron STS gene in flowering plants. Inthis work, we speculated that this involved a possibleevolutionary route of plant-specific type III PKS superfamilyin P. cuspidatum.

Background: To investigate the expression of CD44v3 and vascular endothelial growth factor-C (VEGF-C) and their relationship with lymph node metastasis in squamous cell carcinomas (SCC) of the uterine cervix. Materials and Methods: Expression of CD44v3 and VEGF-C was analyzed in 109 cases of cervical SCC by immunohistochemistry (IHC). The relationship was analyzed between expression and the patient age, histological differentiation, formation of tumor emboli in lymphoid vessels, lymph node metastasis, FIGO staging, and TNM classification. Results: Expression rates for both CD44v3 and VEGF-C were 43.1% in cervical SCC. The cells with positive immunohistochemical staining of CD44v3 were distributed mainly around the keratin pearls in well differentiated carcinomas, but distributed diffusely in the moderately and poorly differentiated lesions. VEGF-C was found stained positively in most of the tumor cells. There were differences in expression between normal epithelium and atypical hyperplasia as well as carcinoma. Both CD44v3 and VEGF-C were found to be associated positively with lymph node metastasis and TNM classification (both p=0.000). Neither CD44v3 nor VEGF-C was found to be associated with patient age, histological differentiation, formation of tumor emboli in lymphoid vessels and FIGO staging. CD44v3 was found to be associated with VEGF-C positively (p=0.000). Conclusions: Abnormal expression of CD44v3 and VEGF-C is associated closely with the lymph node metastasis in cervical SCC, and these agents may cooperate in carcinogenesis and development of metastatic lesions.

Berries of six Vitis davidii Foex (spine grape)cultivars (‘Baiputao’, ‘Gaoshan 1’, ‘Gaoshan 2’, ‘Seputao’,‘Miputao’, and ‘Tianputao’) were harvested from a commercialvineyard in Hunan Province in China. Free andbound volatile compounds and fatty acids were analyzedby GC–MS, and amino acids were analyzed by HPLC. ‘Tianputao’ and ‘Miputao’ were characterized by relativelyhigher concentrations of aromatic amino acids and lowerconcentrations of branched-chain amino acids. The majorfree volatile compounds of spine grapes were hexanal, (E)-2-hexenal, 1-hexanol, (E)-2-hexenol, (E)-b-damascenone,and benzeneacetaldehyde. The major glycosidically boundvolatile compounds identified were 1-hexanol, menthol,nerol, 1-butanol, 3-methyl-3-butenol, benzenemethanol, bphenylethanol,eugenol, and guaiacol. (E)-b-damascenone,benzeneacetaldehyde, guaiacol, and eugenol had odoractivity values (OAVs)[1 in all cultivar grapes. Partialleast squares discriminant analysis (PLS-DA) revealed‘Tianputao’ to be distinct from the other cultivars due to itsrelatively higher concentrations of major terpenoids,norisoprenoids, higher alcohols, and aromatic amino acids.

Background: The meniscus injury is a common disease in the area of sports medicine. The main treatment for this disease is the pain relief, rather than the meniscal function recovery. It may lead to a poor prognosis and accelerate the progression of osteoarthritis. In this study, we designed a meniscal scaffold to achieve the purposes of meniscal function recovery and cartilage protection. Methods: The meniscal scaffold was designed using the triply periodic minimal surface (TPMS) method. The scaffold was simulated as a three-dimensional (3D) intact knee model using a finite element analysis software to obtain the results of different mechanical tests. The mechanical properties were gained through the universal machine. Finally, an in vivo model was established to evaluate the effects of the TPMS-based meniscal scaffold on the cartilage protection. The radiography and histological examinations were performed to assess the cartilage and bony structures. Different regions of the regenerated meniscus were tested using the universal machine to assess the biomechanical functions. Results: The TPMS-based meniscal scaffold with a larger volume fraction and a longer functional periodicity demonstrated a better mechanical performance, and the load transmission and stress distribution were closer to the native biomechanical environment. The radiographic images and histological results of the TPMS group exhibited a better performance in terms of cartilage protection than the grid group. The regenerated meniscus in the TPMS group also had similar mechanical properties to the native meniscus. Conclusion: The TPMS method can affect the mechanical properties by adjusting the volume fraction and functional periodicity. The TPMS-based meniscal scaffold showed appropriate features for meniscal regeneration and cartilage protection.

When low-speed rolling bearings fail, it is hard to diagnose the extent of their damage. We developed a test rig to simulate the lowspeed rolling bearing operating condition, where bearings with various fault states are installed on the test wheelset and subjected to the same external loading condition. The collected bearing box acceleration time histories are processed with the Empirical mode decomposition (EMD) method combined with kurtosis criterion to filter the trend and noise components. Five characteristic parameters of Alpha stable distribution (ASD) are identified by fitting the ASD distribution to the vibration acceleration signals and computing the Probability density function (PDF). To highlight the advantage of ASD method in feature extraction, kurtosis also has be calculated. Through sensitivity and stability analysis of the six parameters and utilization of Least squares support vectors machine (LSSVM) with Particle swarm optimization (PSO), three most sensitive and stable feature parameters including the characteristic exponent a, the scale factor g and the peak value of the PDF h are located and applied to evaluate the low-speed rolling bearings’ damage position and damage extent. The proposed method was validated by test data, and the results demonstrated that the ASD characteristics combined with PSO-LSSVM can not only achieve fault diagnosis of low-speed rolling bearings' damage position and damage extent, but also have better diagnosis accuracy and operational efficiency than other methods.
InSAR-based Glacier Velocity Mapping in the Parlung Zangbo River Basin, Tibetan Plateau, China

By applying the method of SAR interferometry to X-band synthetic aperture radar (SAR) image of COSMO-SkyMed, detailed motion patterns of five glaciers in the Parlung Zangbo River basin, Tibetan Plateau, in January 2010 have been derived. The results indicate that flow patterns are generally constrained by the valley geometry and terrain complexity. The maximum of 123.9 ma-1 is observed on glacier No.1 and the minimum of 39.4 ma-1 is found on glacier No.3. The mean values of five glaciers are between 22.9 and 98.2 ma-1. Glaciers No.1, No.2, No.4 and No.5 exhibit high velocities in their upper sections with big slope and low velocities in the lower sections. A moraine lake accelerates the speed of mass exchange leading to a fast flow at the terminal of glacier No.3. These glaciers generally move along the direction of decreased elevation and present a macroscopic illustration of the motion from the northwest to the southeast. The accuracy of DEM and registration conditions of DEMsimulated terrain phases has certain effects on calculations of glacier flow direction and velocity. The error field is relatively fragmented in areas inconsistent with the main flow line of the glaciers, and the shape and uniformity of glacier are directly related to the continuous distribution of flow velocity errors.

Background: Propofol has been reported to protect vascular endothelial cells against oxidative stress. In this study we investigated its effect on hydrogen peroxide (H2O2)-induced apoptosis of human umbilical vein endothelial cells (HUVECs) and examined the possible signaling pathways. Methods: HUVECs were pretreated with propofol (1, 5, 25, and 50 μM) for 30 min and then co-incubated with 0.4 mM H2O2 for 4 h. Cell viability was assessed using a Cell Counting Kit-8. Cell apoptosis was analyzed using flow cytometry with annexin V/propidium iodide staining, and evaluated by quantifying caspase-3, Bax, and Bcl-2 expression levels. The expression levels of p38 mitogen activated protein kinase (MAPK), phosphorylated (p)-p38 MAPK, cJun-N-terminal kinases (JNK), phosphorylated (p)-JNK, Akt and phosphorylated Akt [(p)-Akt] (Ser473) were measured by western blotting. Results: H2O2 treatment induced the activation of caspase-3, downregulated Bcl-2 expression, and up-regulated Bax expression, all of which were dose-dependently attenuated by propofol pretreatment. Furthermore, propofol significantly ameliorated H2O2-induced phosphorylation of p38 MAPK, JNK, and Akt in HUVECs. Conclusions: Propofol can protect HUVECs against H2O2-induced apoptosis via a mechanism that may involve p38 MAPK, JNK, and Akt signaling pathways.

Background: We examined changes in hepatitis B core-related antigen (HBcrAg) during the four sequential phases of chronic hepatitis B virus (HBV) infection: hepatitis B e antigen (HBeAg)-positive chronic infection (EPCI) and hepatitis (EPCH), followed by HBeAg-negative chronic infection (ENCI) and hepatitis (ENCH). We compared the performance of serum HBcrAg, hepatitis B surface antigen (HBsAg), and HBV DNA in predicting EPCH and ENCH.
