        Cloning and characterization of the cardiac-specific Lrrc10 promoter

        Leucine-rich repeat containing protein 10 (LRRC10) is characterized as a cardiac-specific gene, suggesting a role in heart development and disease. A severe cardiac morphogenic defect in zebrafish morphants was recently reported but a contradictory result was found in mice, suggesting a more complicated molecular mechanism exists during mouse embryonic development. To elucidate how LRRC10 is regulated, we analyzed the 5`enhancer region approximately 3 kilo bases (kb) upstream of the Lrrc10 start site using luciferase reporter gene assays. Our characterization of the Lrrc10 promoter indicates it possesses complicated cis-and trans-acting elements. We show that GATA4 and MEF2C could both increase transcriptional activity of Lrrc10 promoter individually but that they do not act synergistically, suggesting that there exists a more complex regulation pattern. Surprisingly, knockout of Gata4 and Mef2c binding sites in the 5`enhancer region (-2,894/-2,889) didn`t change the transcriptional activity of the Lrrc10 promoter and the likely GATA4 binding site identified was located in a region only 100 base pair (bp) upstream of the promoter. Our data provides insight into the molecular regulation of Lrrc10 expression, which probably also contributes to its tissue-specific expression. [BMB reports 2011; 44(2): 123-128]

        Low-speed rolling bearing fault diagnosis based on EMD denoising and parameter estimate with alpha stable distribution

        When low-speed rolling bearings fail, it is hard to diagnose the extent of their damage. We developed a test rig to simulate the lowspeed rolling bearing operating condition, where bearings with various fault states are installed on the test wheelset and subjected to the same external loading condition. The collected bearing box acceleration time histories are processed with the Empirical mode decomposition (EMD) method combined with kurtosis criterion to filter the trend and noise components. Five characteristic parameters of Alpha stable distribution (ASD) are identified by fitting the ASD distribution to the vibration acceleration signals and computing the Probability density function (PDF). To highlight the advantage of ASD method in feature extraction, kurtosis also has be calculated. Through sensitivity and stability analysis of the six parameters and utilization of Least squares support vectors machine (LSSVM) with Particle swarm optimization (PSO), three most sensitive and stable feature parameters including the characteristic exponent a, the scale factor g and the peak value of the PDF h are located and applied to evaluate the low-speed rolling bearings’ damage position and damage extent. The proposed method was validated by test data, and the results demonstrated that the ASD characteristics combined with PSO-LSSVM can not only achieve fault diagnosis of low-speed rolling bearings' damage position and damage extent, but also have better diagnosis accuracy and operational efficiency than other methods.

        Diagnosing axle box bearings’ fault using a refined phase difference correction method

        The wheelset treads and axle box bearings of railway vehicles often suffer from fatigue failures. Their regular maintenance highly depends on manual off-line inspection with low working efficiency and poor precision for early failure detection. This study proposes a fault diagnosis method by band-pass filtering and by enveloping the accelerations collected from the axle box bearing on the underfloor wheelset lathe to improve the maintenance efficiency. This process is followed by the refined phase difference correction using the fourterm third derivative Nuttall-windowed fast Fourier transform (RPNWF) to extract accurate amplitudes of the fault characteristic frequency and its harmonics. The integration scheme, work flow, and application examples of the fault diagnosis system are presented. Simulation analysis and results show that the developed method can achieve effective diagnosis of the fault and fault degree of axle box bearings as well as yield better correction accuracy than the commonly used discrete spectrum correction methods.

        Regional Differences and Variability on Enterprise Management Performance: A case in China

        This paper measures variation in the economic performance of firms using the entropy weight method and cluster analysis. Regional differences and changes in the trend of listed companies' management performance are analyzed in China's four economic regions and 31 provinces from 2006 to 2010, the Eleventh Five Year period. We show that the listed companies' management performance in four economic regions presents a ladder type distribution with large swings and different trends over time. Moreover, there is a large difference in listed companies' management performance across provinces and some provinces have abnormal fluctuations. It is indicated that differences in industrial structures across regions are the main factor determining regional differences in economic development and the variability of enterprise management performance. So from the perspective of policy and institution, some suggestions are advanced to promote integrated development of all regions. In addition, the research results can provide governments at all levels with systematic reference information to formulate a coordinated, sustainable development policy for different regional economies.

      • Metformin Inhibits Growth of Hepatocellular Carcinoma Cells by Inducing Apoptosis Via Mitochondrion-mediated Pathway

        Recently, population-based studies of type 2 diabetes patients have provided evidence that metformin treatment is associated with a reduced cancer incidence and mortality, but its mode of action remains unclear. Here we report effects of metformin on hepatocellular carcinoma (HCC) Hep-G2 cells and details of molecular mechanisms of metformin activity. Our research indicates that metformin displays anticancer activity against HCC through inhibition of the mTOR translational pathway in an AMPK-independent manner, leading to G1 arrest in the cell-cycle and subsequent cell apoptosis through the mitochondrion-dependent pathway. Furthermore, we showed that metformin strongly attenuated colony formation and dramatically inhibited Hep-G2 tumor growth in vivo. In conclusion, our studies suggested that metformin might have potential as a cytotoxic drug in the prevention and treatment of HCC.

        Model Identification and Human-robot Coupling Control of Lower Limb Exoskeleton with Biogeography-based Learning Particle Swarm Optimization

        Lower limb exoskeleton is a typical wearable robot to assist human motion and improve physiological power. However, the control performance and stability are affected by some unknown model parameters and control algorithms. Therefore, it is necessary to investigate the model parametric identification and the control design of lower extremity exoskeleton. Firstly, the two degree-of-freedom (DoF) exoskeleton model is constructed by the Lagrange technique. Then the biogeography-based learning particle swarm optimization (BLPSO) is used to optimize the B-spline function parameters and the smooth stimulated trajectories is designed. Meanwhile, the BLPSO is also adopted to identify unknown model parameters of the exoskeleton based on the torques and the joint angles. To decline the negative effect of parametric identification error of exoskeleton, the passive backstepping controller is proposed to improve the tracking performance of human-robot motion. Furthermore, the active admittance controller is adopted to improve the motion comfort of tester. Finally, the comparative experimental results are verified on the platform, which show the BLPSO algorithm has better parametric identification accuracy than PSO and GA. Furthermore, the comparative results have verified that the proposed controller can improve the tracking behavior and reduce the human-robot interaction torque in wearable motion.

        Experimental Study on Shear Transfer Performance of Uncracked Monolithic Steel Fiber Concrete

        Shear transfer performance at interface of steel fiber reinforced concrete (SFRC) is of great importance that many factors influence the shear transfer performance of SFRC such as concrete strength and aggregate interlock, reinforcement dowel action and steel fiber. Many studies have focused on part of the factors. The shear transfer performance of uncracked monolithic SFRC is studied in this paper by considering the combined effects of cohesion, clamping stress, confined stress, reinforcement, and steel fiber. Thus, this study presents a series of direct shear tests by Z-type push-off specimens. The direct shear strength, failure mode and failure process of plain SFRC and reinforced SFRC are studied, and their shear stress-slip and shear stress-crack width responses of the tested specimens are obtained. Test results showed that reinforcement and steel fibers can improve the ductile behavior and shear strength of the shear interface simultaneously. And shear stress-slip and –crack curves of the test specimens with those combined effects are strongly nonlinear. Finally, based on the existing shear strength calculation model and the test results of this paper, a shear transfer calculation formula of SFRC interface is proposed and verified according to the Mohr Coulomb failure criteria.

      • A Data Placement Algorithm for Data Intensive Applications in Cloud

        Data layout is an important issue which aims at reducing data movements among data centers to improve the efficiency of the entire cloud system. This paper proposes a data-intensive application oriented data layout algorithm. It is based on hierarchical data correlation clustering and the PSO algorithm. The datasets with fixed location have been considered, and both the offline strategy and the online strategy for data layout have been given. As this proposed strategy is aimed at reducing the global amount of data transmissions, and the special permission of the datasets has been introduced, the cost of data transmission can be measured more reasonable. Simulation results show that compared with two classical strategies, our algorithm can reduce the amount of data transmission more effectively.

        Exact Error Rate of Dual-Channel Receiver with Remote Antenna Unit Selection in Multicell Networks

        The error rate performance of circularly distributed antenna system is studied over Nakagami-m fading channels, where a dual-channel receiver is employed for the quadrature phase shift keying signals detection. To mitigate the Co-Channel Interference (CCI) caused by the adjacent cells and to save the transmit power, this work presents remote antenna unit selection transmission based on the best channel quality and the maximized path-loss, respectively. The commonly used Gaussian and Q-function approximation method in which the CCI and the noise are assumed to be Gaussian distributed fails to depict the precise system performance according to the central limit theory. To this end, this work treats the CCI as a random variable with random variance. Since the in-phase and the quadrature components of the CCI are correlated over Nakagami-m fading channels, the dependency between the in-phase and the quadrature components is also considered for the error rate analysis. For the special case of Rayleigh fading in which the dependency between the in-phase and the quadrature components can be ignored, the closed-form error rate expressions are derived. Numerical results validate the accuracy of the theoretical analysis, and a comparison among different transmission schemes is also performed.

        Feature analysis on air quality in the main urban area of Nanchong City in 2015–2018

        This study evaluates the air quality in Nanchong City in 2015–2018 based on automatic monitoring data and meteorological data in the main urban area. Results show that overall air quality in the main urban area of Nanchong was standard and improved from 2015 to 2018. SO₂, NO₂, and CO had slight influences on air quality in the study area, whereas excessive annual average PM2.5 and increasing O₃ concentration challenged the air quality. The monthly average mass concentrations of particulate matters (PM2.5 and PM10), NO₂, and CO generally presented a “U-shaped” distribution pattern while O₃ showed the opposite. The diurnal variation of PM, NO₂, and CO has two peaks related to the morning and evening rush hours. Vehicle exhaust is the main source of local pollution. Over-standard PM2.5 can significantly inhibit O₃ production while O₃ can easily promote the secondary generation of PM2.5 due to its strong oxidization. PM2.5 emission should be controlled strictly in winter, while in summer, the sources of O₃ should be controlled vigorously. Air pollution is comprehensive and complicated and is influenced by various factors, mainly meteorological and human factors. Measures such as artificial rainfall should be included at the appropriate time to cope with adverse meteorological conditions when heavy pollution occurs in winter.
