        Integration of Manufacture and Commerce for a Product Learning System in the Service Industry

        Purpose - The purpose of this thesis is to assess the product design digital learning status of universities that are currently involved in learning environment projects in manufacture and commerce integration (MCI). Thus, enterprises must keep learning and creating new inventions with revolutionary progress. Research design, data, and methodology - This study not only emphasizes the analysis of technical ability, course concepts, conducting models, and learning environments of every aspect, but also systematically probes the planning of learning, system framework, web learning, environmental activities, data statistics, and digitalized learning, among other aspects. Results - The results of this study help in finally understanding each school’s manufacture and commerce integration situation, in order to evaluate product design learning. Consequently, it is essential to evaluate computer learning at schools, thereby affecting communication and the requirements of business education training. Conclusions - It is essential to focus on MCI to promote web teaching to preserve and enhance knowledge disseminating technologies, and immediately share knowledge with learners, while improving work efficiency and cultivating the talent needed by industry.

        Stimulating Effect of a Novel Synthesized Sulfonamido-Based Gallate ZXHA-TC on Primary Osteoblasts

        Purpose: This study is intended to investigate the effects of plants or plant-derived antioxidants on prevention of osteoporosis through the maintenance of reactive oxygenspecies (ROS) at a favorable level. Materials and Methods: In this study, a novel antioxidant, namely 3,4,5-Trihydroxy-N-[4-(5-hydroxy-6-methoxy-pyrimidin-4-ylsulfamoyl)-phenyl]-benzamide (ZXHA-TC) was synthesized from gallic acid and sulfadimoxine. Its effect on osteoblast metabolism was investigated via the detection of cell proliferation, cell viability, production of ROS, and expression of osteogenic-specific genes including runt-related transcription factor 2 (RUNX2), bone sialoprotein (BSP), osteocalcin (OCN), alpha-1 type I collagen (COL1A1), and osteogenic-related proteins after treatment for 2, 4, and 6 days respectively. Results: The results showed that ZXHA-TC has a stimulating effect on the proliferation and osteogenic differentiation of primary osteoblasts by promoting cell proliferation, cell viability, and the expression of genes BSP and OCN. Productions of bone matrix and mineralization were also increased by ZXHA-TC treatment as a result of up-regulationof COL1A1 and alkaline phosphatase (ALP) at the early stage and down-regulationof both genes subsequently. A range of 6.25×10-3 μg/mL to 6.25×10-1 μg/mL is the recommended dose for ZXHA-TC, within which 6.25×10-2 μg/mL showed the best performance. Conclusion: This study may hold promise for the development of a novel agent for the treatment of osteoporosis.

        AsJAZ1 represses the expression of the sesquiterpene synthase gene based on the JA signaling pathway in Aquilaria sinensis (Lour.) Gilg

        Aquilaria sinensis is a species of plant endemic to China, which is typical wound-induced economic plant. It is widely used in the production of perfume, incense and traditional Chinese medicine. A healthy A. sinensis tree cannot produce agarwood. It forms agarwood only when wounded. Sesquiterpenes and phenylethyl chromone derivatives have been demonstrated to be the main active compounds in agarwood, but little is known about jasmonate (JA) signaling that regulates the biosynthesis of agarwood sesquiterpenes. In this work, we firstly cloned the full-length cDNA sequence of a JA signaling suppressor gene (JAZ) from A. sinensis, and then characterized its roles through bioinformatics prediction, tissue expression, wounding-induced expression, subcellular localization, protein interaction and overexpression analysis in Arabidopsis thaliana. The results showed that the full-length cDNA sequence of JAZ gene (termed as AsJAZ1, GenBank accession number: KP677281) is 1507 bp, containing a complete open reading frame (ORF) of 990 bp. The bioinformatics analysis shows that the deduced amino acid residues contain a tify (ZIM) domain, the same as A. thaliana JAZ3 protein. Mechanical wounding and MeJA were found to up-regulate the expression level of the AsJAZ1 gene. The AsJAZ1 could interact with the MYC2 transcription factor in vitro. The MYC2 transcription factor in Arabidopsis thaliana (AtMYC2) was minimally expressed in transgenic plants and had a weak response to MeJA treatment. The AsJAZ1 overexpression in A. thaliana was observed to down-regulate the expression of sesquiterpene synthase genes (TPS21 and TPS11), suggesting that AsJAZ1 might serve as a repressor to regulate the expression of sesquiterpene synthase genes in Aquilaria plants. Taken together, this work may lay a foundation for identifying the JA signaling pathway and exploring the role of JA signaling in agarwood sesquiterpene biosynthesis in A. sinensis trees.

        Kindergarten space design based on BP (back propagation) neural network

        과거에 설계자는 주로 과거의 경험과 설계 공간에 대한 산업 표준 임계값에 대한 참조에 의존했습니다. 이러한 설계는 종종 사용자의 요구를 충족하지 않는 공간을 초래합니다. 공간설계를 위한 BP신경망 알고리즘을 구축해 설계 매개변수를 생성하는 과정과 방법을 조사하는 것이 목적이다. 그런 관점에서. 본 논문은 공간 내 복잡한 욕구가 많은 유치원을 연구 대상으로 삼고 있으며, 기계학습의 BP신경망 알고리즘을 통해 환경행동변수와 공간설계변수의 상관관계를 각인하고 있다. 공간 설계 매개변수를 생성하는 방법을 연구합니다. 미래에는 특정 환경행동영향요소를 대체하여 해당 공간설계 매개변수를 도출할 수 있어 보다 광범위한 시나리오에 적용할 수 있고 설계자의 효율성을 높일 수 있다. In the past, designers relied primarily on past experience and reference to industry standard thresholds to design spaces. Such design often results in spaces that do not meet the needs of users. The purpose of this paper is to investigate the process and way of generating design parameters by constructing a BP neural network algorithm for spatial design. From the perspective. This paper adopts an experimental research method to take a kindergarten with a large number of complex needs in space as the object of study, and through the BP neural network algorithm in machine learning, the correlation between environmental behavior parameters and spatial design parameters is imprinted. The way of generating spatial design parameters is studied. In the future, the corresponding spatial design parameters can be derived by replacing specific environmental behavior influence factors, which can be applied to a wider range of scenarios and improve the efficiency of designers.

        Preparation and photoluminescence properties of the Eu2+, Sm3+ co-doped Li2SrSiO4 phosphors

        A series of Eu2+ and Sm3+ co-doped Li2SrSiO4 phosphors are prepared by the high temperature solidstate reaction. The morphology, structure and spectroscopic properties of the prepared samples are characterized by scanning electron microscopy, X-ray diffraction, diffuse reflection spectra, photoluminescence spectra and electron paramagnetic resonance spectra, respectively. The effect of Sm3þdoping concentration on the photoluminescence intensity of the prepared samples is also investigated. The results indicate that the crystal structure of Li2SrSiO4 is not changed with the Eu2+, Sm3+ co-doping. The spherical-like particle size of the obtained product is about 20-30 nm in diameter. When the Sm3+ concentration is 0.3 mol% and the Eu2+ concentration is 0.7 mol%, the phosphors show the maximum emission intensity, which is 50% higher than that of Eu2+ doped Li2SrSiO4. Excited at 420 nm, the phosphor presents a single broad emission band peaking at 558 nm, which is ascribed to the 4f65d1 / 4f7 transitions of Eu2+ and 4G5/2 / 6H5/2 and 4G5/2 / 6H7/2 transitions of Sm3+. The Commission International de I0Eclairage chromaticity coordinates of Li2SrSiO4:0.7 mol% Eu2+, 0.3 mol%Sm3+ are x = 0.28, y = 0.28.         Characterization, Expression Profiling, and Functional Identification of a Gene Encoding Geranylgeranyl Diphosphate Synthase from Salvia miltiorrhiza

        Geranylgeranyl diphosphate synthase (GGPPS,EC: 2.5.1.29) catalyzes the biosynthesis of geranylgeranyl diphosphate (GGPP), which is a key precursor for diterpenes including tanshinone. In this study, a full-length cDNA encoding GGPPS was isolated from Salvia miltiorrhiza by rapid amplification of cDNA ends (RACE) for the first time, which was designated as SmGGPPS (GenBank Accession No. FJ643617). The full-length cDNA of SmGGPPS was 1,234 bp containing a 1,092 bp open reading frame (ORF) encoding a polypeptide of 364 amino acids. Analysis of SmGGPPS genomic DNA revealed that it contained 2 exons and 1 intron. Bioinformatics analyses revealed that the deduced SmGGPPS had extensive homology with other plant GGPPSs contained all 5 conserved domains and functional aspartate-rich motifs of the prenyltransferases. Molecular modeling showed that SmGGPPS is a new GGPPS with a spatial structure similar to other plant GGPPSs. Phylogenetic tree analysis indicated that SmGGPPS belongs to the plant GGPPS super-family and has the closest relationship with GGPPS from Nicotiana attenuate. The functional identification in Escherichia coli showed that SmGGPPS could accelerate the biosynthesis of carotenoid, demonstrating that SmGGPPS encoded a functional protein. Expression pattern analysis implied that SmGGPPS expressed higher in leaves and roots, weaker in stems. The expression of SmGGPPS could be up-regulated by Salicylic acid (SA) in leaves and inhibited by methyl jasmonate (MeJA) in 3 tested tissues, suggesting that SmGGPPS was elicitor-responsive. This work will be helpful to understand more about the role of SmGGPPS involved in the tanshinones biosynthesis pathway and metabolic engineering to improve tanshiones production in S. miltiorrhiza.

        Thioredoxin A of Streptococcus suis Serotype 2 Contributes to Virulence by Inhibiting the Expression of Pentraxin 3 to Promote Survival Within Macrophages

        Streptococcus suis serotype 2 (SS2) is an important zoonotic pathogen that can infect humans in contact with infected pigs or their byproducts. It can employ different types of genes to defend against oxidative stress and ensure its survival. The thioredoxin (Trx) system is a key antioxidant system that contributes adversity adaptation and pathogenicity. SS2 has been shown to encode putative thioredoxin genes, but the biological roles, coding sequence, and underlying mechanisms remains uncharacterized. Here, we demonstrated that SSU05_0237-ORF, from a clinical SS2 strain, ZJ081101, encodes a protein of 104 amino acids with a canonical CGPC active motif and an identity 70–85% similar to the thioredoxin A (TrxA) in other microorganisms. Recombinant TrxA efficiently catalyzed the thiol-disulfide oxidoreduction of insulin. The deletion of TrxA led to a significantly slow growth and markedly compromised tolerance of the pathogen to temperature stress, as well as impaired adhesion ability to pig intestinal epithelial cells (IPEC-J2). However, it was not involved in H2O2 and paraquat-induced oxidative stress. Compared with the wild-type strain, the ΔTrxA strain was more susceptible to killing by macrophages through increasing NO production. Treatment with TrxA mutant strain also significantly attenuated cytotoxic effects on RAW 264.7 cells by inhibiting inflammatory response and apoptosis. Knockdown of pentraxin 3 in RAW 264.7 cells was more vulnerable to phagocytic activity, and TrxA promoted SS2 survival in phagocytic cells depending on pentraxin 3 activity compared with the wild-type strain. Moreover, a co-inoculation experiment in mice revealed that TrxA mutant strain is far more easily cleared from the body than the wild type strain in the period from 8–24 h, and exhibits significantly attenuated oxidative stress and liver injury. In summary, we reveal the important role of TrxA in the pathogenesis of SS2.

        Effect on Zn on Microstructures and Mechanical Properties of Mg–Gd–Y–Zn LPSO Alloys

        In this study, the microstructures and mechanical properties of Mg–1Gd–1Y–xZn alloys (x = 0.2, 0.4, 0.6, and 0.8) wereinvestigated. Microstructural evolution was tracked during the process of casting, homogenization, and extrusion. Long-periodstacking-ordered (LPSO) and β phases were formed in the as-cast alloys. The volume fraction of the LPSO phase increasedwith increasing Zn content, whereas that of the β phase decreased. Although the homogenization dissolved the β phase intothe Mg matrix, there was no significant change in the volume fraction of the LPSO phases. Increasing the Zn content led toa slight reduction in grain size and a simultaneous strengthening of texture. The Mg–1Gd–1Y–0.2Zn alloy possessed a highdensity of fine LPSO phases and excellent reinforcement capacity, thereby exhibiting excellent mechanical properties atroom temperature. In addition, the low volume fraction of the LPSO phases in this alloy led to suppressed yield asymmetry.
