      • Quantifying herbicide dose-response and resistance in <i>Echinochloa</i> spp. by measuring root length in growth pouches

The aim of the presented study was to develop a bioassay for rapid diagnosis of herbicide dose-response and resistance in Echinochloa. Pre-germinated seeds of Echinochloa spp. were incubated in growth pouches (18 cm×16.5 cm) containing herbicide solutions in a range of concentrations. Shoot and root lengths were measured after 6 d of incubation. Dose-responses estimated by measuring root lengths in the growth pouches were well-described by the log-logistic dose-response model and similar to those estimated by a whole-plant assay. Accurate dose-response curves were successfully generated for several herbicides with different modes of action, suggesting that the growth pouch method can be used for herbicide bioassays. The suitability of the growth pouch method for rapid diagnosis of acetyl coenzyme-A carboxylase (ACCase) and acetolactate synthase (ALS) inhibitor resistance in Echinochloa spp. was also tested. For cyhalofop-butyl, resistant and susceptible biotypes were discriminated at 180-300 mg a.i. L<SUP>−1</SUP> and 80-120 mg a.i. L<SUP>−1</SUP> for barnyardgrass (E. crus-galli) and late watergrass (E. oryzicola), respectively. For penoxsulam, the discriminatory dosage was 350-500 mg a.i. L<SUP>−1</SUP> for barnyardgrass and 650-1000 mg a.i. L<SUP>−1</SUP> for late watergrass. The method was further used to identify late watergrass biotypes resistant and susceptible to two other ALS inhibitors, azimsulfuron and bispyribac-sodium. Our results show that the growth pouch method can be reliably used in herbicide dose-response studies and to diagnose herbicide resistance in Echinochloa spp., with significant time and cost savings compared with conventional whole-plant assays. </P>

        Objectives: The purpose of this research was to identify the effect of mental health, parenting attitude and attachment on gambling addiction severity among male college students. Methods: Subjects of the research were 292 students recruited from colleges in Seoul, Korea. The questionnaires were composed of CPGI, DASS-21, PBI, ECR-R. The data was statistically analyzed by Chi-square, ANOVA, Scheffe test and Pearson’s correlation. Results: The proportion of moderate-risk gambling and high-risk gambling group was reported 28.1%. There a statistically significant difference between gambling addiction severity groups in Anxiety(F=3.19, p<.05). Statistically significant linear relationship was found between Anxiety and gambling addiction score(r=.15, p<.05). There was statistically significant difference between gambling addiction severity groups in Caring(F=4.74, p<.01). Statistically significant linear relationship was found between Caring and gambling addiction score(r=-2.14, p<.01). There was statistically significant difference between gambling addiction severity groups in Overprotection(F=4.74, p<.01). Statistically significant linear relationship was examined between Overprotection and gambling addiction score(r=.20, p<.01). Significant difference was examined between gambling addiction severity in Attachment Anxiety(F=3.76, p<.05). Attachment Anxiety showed significant linear relationship with gambling addiction score(r=-.17, p<.01). Significant differences were examined between gambling addiction severity in Attachment avoidance (F=3.46, p<.05). Attachment avoidance showed no significant relationship with gambling addiction score(r=-.09, p=.12). Conclusions: This study showed the effect of mental health, parenting attitude and attachment on gambling addiction severity among male college students. Based on the result, it is recommend for nurses to develop intervention to prevent gambling addiction among college students by lowering anxiety, stabilizing attachment through desirable caring attitude from parents.

      • Alleviation of temperature-sensitive secretion defect of Pseudomonas fluorescens ATP-binding cassette (ABC) transporter, TliDEF, by a change of single amino acid in the ABC protein, TliD

        <P>An ABC transporter, TliDEF, from Pseudomonas fluorescens SIK W1, mediates the secretion of its cognate lipase, TliA, in a temperature-dependent secretion manner; the TliDEF-mediated secretion of TliA was impossible at the temperatures over 33 degrees C. To isolate a mutant TliDEF capable of secreting TliA at 35 degrees C, the mutagenesis of ABC protein (TliD) was performed. The mutated tliD library where a random point mutation was introduced by error-prone PCR was coexpressed with the wild-type WE, tliF and tliA in Escherichia con. Among approximately 10,000 colonies of the tliD library, we selected one colony that formed transparent halo on LB-tributyrin plates at 35 degrees C. At the growth temperature of 35 degrees C, the selected mutant TliD showed 1.75 U/ml of the extracellular lipase activity, while the wild-type TliDEF did not show any detectable lipase activity in the culture supernatant of E. coli. Moreover, the mutant TliD also showed higher level of TliA secretion than the wild-type TliDEF at other culture temperatures, 20 degrees C, 25 degrees C and 30 degrees C. The mutant TliD had a single amino acid change (Ser287Pro) in the predicted transmembrane region in the membrane domain of TliD, implying that the corresponding region of TliD was important for causing the temperature-dependent secretion of TliDEF.         한국잔디 수집계통들 중에서 우수계통들의 생육특성 비교

        2001년 수집된 한국잔디 133계통들 중에서 우수한 계통들을 선발하기위하여 밀도(품질),<br/> 피복성, 녹색기간, 내병성, 출수유무(종자수) 등의 주요특성과 그 외 생육특성들이 조사되었다. 생육특성이 우수한 6계통이 선발되었으며 그 결과를 요약하면 다음과 같다.<br/> 수집된 133계통들 중에서 가장 녹색기간이긴 계통은 J01067으로 11월초까지 녹색도를 유지하였다. J01106 및 J01129 계통들은 엽폭이 각각 1.5mm 및 2mm 정도로 매우 좁으며, 밀도가 높고, 피복성도 좋은 것으로 나타났으며, 내병성도 강하였다. J01122 계통은 들잔디 계통들 중에서 유일하게 내병성이 강한 계통으로 선발되었다. J01128 계통은 엽폭이 3.2mm로 중엽에 속하였으며, 대비품종인 Sunburst 와 비슷하였다. 이 계통의 경우도J01106, J01129 계통들과 마찬가지로 밀도가 높고 내병성이 강한 것으로 나타났다.<br/> 수집된 133계통들의 형태적 분류를 위하여 5가지의 생육특성을 사용하여 크게 세가지 군집<br/> 으로 분류하였으며, 그 중에서 대비품종으로 사용된 6품종들과 선발된 우수 6계통의 분류는<br/> 다음과 같다. 제 1군집에 속하는 품종 및 계통은 Belare, Meyer, 안양중지(Anyang-jungji), J01067, J01112 등이 포함되었으며, 제 2군집으로는 S-94, J01105가 포함되었고, 제 3군집에는 Sunburst, 건희(Konhee), J01106, J01128, J01129 등이 포함되었다. Growth characteristics such as density(quality), covering speed, green period, disease resistance, heading existence(number of seeds) and other characteristics were examined for selection of superior lines among the 133 zoysiagrass lines collected in 2001. Six superior lines were selected and the results were summarized as follows.<br/> Superior line, J01067 was longest for green period among the 133 zoysiagrasses and stayed green until the beginning of November. Leaf width of J01106 and J01129 lines was very narrow as 1.5㎜ and 2㎜, respectively and superior for density(quality), covering speed and disease resistance. J01122 line was selected for strongest disease resistance among the lines of Zoysia japonica. Leaf width of J01128 was 3.2㎜ showing midium type and similar to standard cultivar, Sunburst. This line also showed higher density and strong disease resistance like J01106 and J01129.<br/> Five growth characteristics for morphological classification of 133 zoysiagrass lines<br/> was used and divided into 3 cluster groups. Of 133 lines, 6 standard cultivars and 6 superior lines were classified as follows. First cluster group contained Belare, Meyer, Anyang-jungji, J01067, J01122, and second group contained S-94 and J01105, and third group contained Sunburst, Konhee, J01106, J01128 and J01129.<br/> <br/>

        Phenotypic Diversity of Escherichia coli O157:H7 Strains Associated with the Plasmid O157

        Escherichia coli O157:H7, a food-borne pathogen, causes hemorrhagic colitis and the hemolytic-uremic syndrome. A putative virulence factor of E. coli O157:H7 is a 60-MDa plasmid (pO157) found in 99% of all clinical isolates and many bovine-derived strains. The well characterized E. coli O157:H7 Sakai strain (Sakai) and its pO157-cured derivative (Sakai-Cu) were compared for phenotypic differences. Sakai-Cu had enhanced survival in synthetic gastric fluid, did not colonize cattle as well as wild-type Sakai, and had unchanged growth rates and tolerance to salt and heat. These results are consistent with our previous findings with another E. coli O157:H7 disease outbreak isolate ATCC 43894 and its pO157-cured (43894-Cu). However, despite the essentially sequence identical pO157 in these strains, Sakai-Cu had changes in antibiotic susceptibility and motility that did not occur in the 43894-Cu strain. This unexpected result was systematically analyzed using phenotypic microarrays testing 1,920 conditions with Sakai, 43894, and the plasmid-cured mutants. The influence of the pO157 differed between strains on a wide number of growth/survival conditions. Relative expression of genes related to acid resistance (gadA, gadX, and rpoS) and flagella production (fliC and flhD) were tested using quantitative real-time PCR and gadA and rpoS expression differed between Sakai-Cu and 43894-Cu. The strain-specific differences in phenotype that resulted from the loss of essentially DNA-sequence identical pO157 were likely due to the chromosomal genetic diversity between strains. The O157:H7 serotype diversity was further highlighted by phenotypic microarray comparisons of the two outbreak strains with a genotype 6 bovine E. coli O157:H7 isolate, rarely associated with human disease.

      • Inhibitory effects of triptolide on titanium particle-induced osteolysis and receptor activator of nuclear factor-κB ligand-mediated osteoclast differentiation

        <P>Purpose We examined the effects of triptolide on receptor activator of nuclear factor-kappa B ligand (RANKL)-induced osteoclast differentiation and on titanium (Ti) particle-induced osteolysis. Methods To examine the effect of triptolide on osteoclast differentiation, bone marrow macrophages (BMMs) were treated with 100 ng/mL of RANKL and 30 ng/mL of macrophage-colony stimulating factor, or co-cultured with osteoblasts stimulated with 10 nM vitamin D3 and 1 mu M prostaglandin E2 in the presence or absence of triptolide (2.8-14 nM). Osteoclast differentiation and activation were assessed using tartrate-resistant acid phosphatase staining, reverse transcriptase-polymerase chain reaction analysis to determine differentiation marker gene expression and pit formation assays. To examine the effect of triptolide on wear debris-induced osteolysis, titanium (Ti) particles were injected into the calvaria of ICR mice. Then, the mice were divided into three groups and were orally administered vehicle, or 16 or 32 mu g/kg/day triptolide for ten days, followed by histomorphometric analysis. Results Triptolide suppressed RANKL-mediated osteoclast differentiation of BMMs in a dose-dependent manner. In a co-culture system, osteoblasts treated with triptolide could not induce osteoclast differentiation of BMMs, which was accompanied by down-regulation of RANKL and up-regulation of osteoprotegrin. Moreover, triptolide significantly inhibited bone resorption, and expression of the bone resorption marker genes. RANKL-induced activation of p38, ERK, and JNK was substantially inhibited by triptolide. Further, in a Ti-induced mouse calvarial erosion model, mice perorally administrated with triptolide showed significant attenuation of Ti-mediated osteolysis. Conclusion Our data indicated that triptolide had an anti-osteoclastic effect and significantly suppressed wear debris-induced osteolysis in mice.</P>

        β-Galactosidase Gene of Thermus thermophilus KNOUC112 Isolated from Hot Springs of a Volcanic Area in New Zealand: Identification of the Bacteria, Cloning and Expression of the Gene in Escherichia coli

        To isolate the $\beta$-galactosidase producing thermophilic bacteria, samples of mud and water were collected from hot springs of avolcanic area near Golden Springs in New Zealand. Among eleven isolated strains, the strain of KNOUC112 produced the highest amounts of $\beta$-galactosidase at 40 h incubation time (0.013 unit). This strain was aerobic, asporogenic bacilli, immobile, gram negative, catalase positive, oxidase positive, and pigment producing. Optimum growth was at 70-72$^{\circ}C$, pH 7.0-7.2, and it could grow in the presence of 3% NaCl. The main fatty acids of cell components were iso-15:0 (30.26%), and iso-17:0 (31.31%). Based on morphological and biochemical properties and fatty acid composition, the strain could be identified as genus Thermus, and finally as Thermus thermophilus by phylogenetic analysis based on 16S rRNA sequence. So the strain is designated as Thermus thermophilus KNOUC112. A gene from Thermus thermophilus KNOUC112 encoding $\beta$-galactosidase was amplified by PCR using redundancy primers prepared based on the structure of $\beta$-galactosidase gene of Thermus sp. A4 and Thermus sp. strain T2, cloned and expressed in E. coli JM109 DE3. The gene of Thermus thermophilus KNOUC112 $\beta$-galactosidase(KNOUC112$\beta$-gal) consisted of a 1,938 bp open reading frame, encoding a protein of 73 kDa that was composed of 645 amino acids. KNOUC112$\beta$-gal was expressed as dimer and trimer in E. coli JM109 (DE3) via pET-5b.

        The LIM-only transcription factor LMO2 determines tumorigenic and angiogenic traits in glioma stem cells

        Glioblastomas (GBMs) maintain their cellular heterogeneity with glioma stem cells (GSCs) producing a variety of tumor cell types. Here we interrogated the oncogenic roles of Lim domain only 2 (LMO2) in GBM and GSCs in mice and human. High expression of LMO2 was found in human patient-derived GSCs compared with the differentiated progeny cells. LMO2 is required for GSC proliferation both in vitro and in vivo, as shRNA-mediated LMO2 silencing attenuated tumor growth derived from human GSCs. Further, LMO2 is sufficient to induce stem cell characteristics (stemness) in mouse premalignant astrocytes, as forced LMO2 expression facilitated in vitro and in vivo growth of astrocytes derived from Ink4a/Arf null mice and acquisition of GSC phenotypes. A subset of mouse and human GSCs converted into vascular endothelial-like tumor cells both in vitro and in vivo, which phenotype was attenuated by LMO2 silencing and promoted by LMO2 overexpression. Mechanistically, the action of LMO2 for induction of glioma stemness is mediated by transcriptional regulation of Jagged1 resulting in activation of the Notch pathway, whereas LMO2 directly occupies the promoter regions of the VE-cadherin gene for a gain of endothelial cellular phenotype. Subsequently, selective ablation of human GSC-derived VE-cadherin-expressing cells attenuated vascular formation in mouse intracranial tumors, thereby significantly prolonging mouse survival. Clinically, LMO2 expression was elevated in GBM tissues and inversely correlated with prognosis of GBM patients. Taken together, our findings describe novel dual roles of LMO2 to induce tumorigenesis and angiogenesis, and provide potential therapeutic targets in GBMs.

      • Base tolerant polybenzimidazolium hydroxide membranes for solid alkaline-exchange membrane fuel cells

        Poly(dibenzylated benzimidazolium) bromides (Bz-PBI-Br) were converted successfully to OH<SUP>-</SUP> ion conducting poly(dibenzylated benzimidazolium) hydroxides (Bz-PBI-hydroxides) by the treatment of KOH. The Bz-PBI-hydroxides obtained in this study showed an excellent alkali tolerance compared to previously synthesized poly(dimethylated benzimidazolium) hydroxides (Me-PBI-hydroxides). According to <SUP>1</SUP>H-NMR analysis, Me-PBI-hydroxides were decomposed during KOH treatment. In order to find out the reason, density functional theory (DFT) calculations of two benzimidazolium structures, e.g., dimethylated benzimidazolium (Me-BI<SUP>+</SUP>) and dibenzylated benzimidazolium (Bz-BI<SUP>+</SUP>), were performed. Bz-BI<SUP>+</SUP> showed lower electron affinity and OH<SUP>-</SUP>-binding energies at the C2 position of the benzimidazolium ring than Me-BI<SUP>+</SUP>. These DFT results strongly confirm that Bz-BI<SUP>+</SUP> is less vulnerable to an OH<SUP>-</SUP> attack than Me-BI<SUP>+</SUP>; this contributes to the enhanced stability and OH<SUP>-</SUP> ion conductivity of the Bz-PBI-hydroxides.
