Association of DOCK8, IL17RA, and KLK12 Polymorphisms with Atopic Dermatitis in Koreans

Background: Early-onset and severe atopic dermatitis (AD) in patients increase the probability of the development of allergic rhinitis or asthma. Treatment and prevention strategies in infants and young children with AD are targeted toward treating the symptoms, restoring skin barrier functions, and reducing the absorption of environmental allergens in an attempt to attenuate or block the onset of asthma and food allergy. Objective: Given that the initiating events in AD remain poorly understood, identifying those at risk and implementing strategies to prevent AD is necessary. Methods: Whole-exome sequencing (WES) was performed in a 43 control group and a disease group with 20 AD patients without atopic march (AM) and 20 with AM. Sanger sequencing was carried out to validate found variants in cohorts. Results: DOCK8, IL17RA, and KLK12 single-nucleotide polymorphisms were identified by WES as missense mutations: c.1289C>A, p.P97T (rs529208); c.1685C>A, p.P562G (rs12484684); and c.457+27>C, rs3745540, respectively. A case-control study show that total immunoglobulin E (IgE) level was significantly increased in the AA genotype of DOCK8 compared to the CA genotype in allergic patients. The rs12484684 of IL17RA increased risk of adult-onset AD (odds ratio: 1.63) compared to the control for (A) allele frequency. AD and AM Patients with the IL17RA CA genotype also had elevated IgE levels. rs3745540 of KLK12 was associated with AD in dominant model (odds ratio: 2.86). Conclusion: DOCK8 (rs529208), IL17RA (rs12484684), and KLK12 (rs3745540), were identified using a new WES filtering method. the result suggests that polymorphism of DOCK8 and IL17RA might be related to increase the total IgE level. The Lipopolysaccharide from Porphyromonas gingivalis Induces Vascular Permeability

Porphyromonas gingivalis, one of the major periodontal pathogens, is implicated in the initiation and progression of periodontal disease. The initial stages of periodontal inflammation are accompanied by vascular hyperpermeability. In our present study, we report that the P. gingivalis lipopolysaccharide (LPS) increases the mRNA expression of interleukin-8 (IL-8), a major inducer of vascular permeability, in vascular endothelial cells. P. gingivalis LPS also stimulated the induction of IL-8 secretion in endothelial cells. The P. gingivalis LPS-induced expression of IL-8 was primarily modulated by nuclear factor-κB (NF-κB). P. gingivalis LPS significantly enhanced the vascular permeability both in vitro and in vivo, and a blockade of the IL-8 receptor decreased the P. gingivalis LPS-induced vascular permeability. Taken together, these results suggest that P. gingivalis LPS increases vascular permeability through the NF-κBdependent production of IL-8 in vascular endothelial cells.

This paper presents an improved probabilistic formalization to the computational methods of the case-based reasoning parading. In particular, It is computationally very efficient. In contrast to earlier probabilistic methods, this method does not require a transformation step between the first case space and the distribution space. It is concentrated on applying the marginal distribution to the case matching problem, and proposed a probabilistic scoring metric for this problem. In the experiment, the probabilistic case is evaluated empirically by using the available real-world case bases. The result shows that when treated with cases where some of the features have been removed, a relatively small number of remaining cases is sufficient for retrieving the original case from the case base by using the proposed method.

The author treated 3 class I malocclusion patients with the lingual appliance followed by the extraction of the 4 bicuspids. One of them was finished with the labial appliance at the final stage. The treatment results were acceptable and the patients gad good tolerance to the lingual appliance without complaints in these cases. There were some problems in treatment on the lingual side both the patient and the practioner, but I think we can overcome them with the development of the orthodontic materials, the treatment technics & the increased clinical experience. Of course, we cannot treat all the patients with the lingual braces, but patients are carefully selected, lingual braces will be a valuable orthodontic appliance. In conclusion, the lingual braces have very good esthetic advantages, so the patients, especially in adults, who hesitate or refuse the orthodontic treatment due to the esthetic problem of the labial braces will get the motivation & the chance for the orthodontic treatment.

Neuro-fuzzy systems have been considered for several years already. However, the term neuro-fuzzy still lacks proper definition, and still has the flavour of a buzzword to it. Few neuro-fuzzy methods do actually employ neural networks, even though they are very often depicted in form of some kind of neural network structure. However, all methods display some kind of learning capability, as it is known from neural networks. This article reviews neuro-fuzzy systems, which combine methods from neural network theory with fuzzy systems. Further, it presents NEFCLASS, an approach to neuro-fuzzy classification, and is discussed how a neuro-fuzzy classification, and is discussed how a neuro-fuzzy classifier can be initialized by rules generated by initial rule bases.

Training the output units of a neural network into saturations means that the derivative will be zero and that the training is caught in a local minima. Adding a small offset term to the derivative in a backpropagation method is known to eliminate local minima and increase learning speed. The same simple trick cannot be directly applied to second methods as the scaled conjugate gradient, as they use not only the derivative but also the error function itself. By simple integrating the derivative which includes the offset term, a formulation of the error function that corresponds to this derivative is given. Being able to calculate the derivative as well as the actual error, the offset term is now directly applicable in second order methods.

In this paper, a new method capable of directly inputting English characters using the characteristic shapes of alphabets for a small keypad used in the imbedded systems has been introduced. The new method selects keypad buttons for each character by its ending and crossed points of the character line in order to input a specific character. This method is capable of directly inputting characters to the system through keypad since it utilizes the characteristics of each character and also the user does not need to read small character arrays written in keypad buttons, thus the rapid input of alphabets are possible and the fatigue in user's eyes can greatly be reduced, which leads to the improvement in overall embedded system performance.

ALOPEX 신경망은 계산이 간편하고, 가중치를 갱신하는 데 있어서 처리 요소들 간에 자체 연결을 요구하지 않는 빠른 방법에 속한다. 가중치 변화는 가체 출력에서 변화화 오차함수에서 변화간에 상관이라 할 수 있는 피드백을 통해 확률적으로 처리된다. 각 가중치 처리기는 전체 오차함수에만 종속되기 때문에 처리기들 간에 어떠한 교호작용도 요구되지 않지만 실제 적용에 있어서 back propagation을 사용하는 것과 유사한 속도로 수렴하게 된다. 이러한 원인들로서 네트워크 위상, 뉴런 형들의 선택도 있지만, 무엇보다도 random walk에 따른 지역적인 최소 점으로의 수렴을 들 수 있다. 특히 전달함수가 saturation 상태에서 학습될 경우에 쉽게 지역적인 최소 점에 빠지게 된다. 본 논문에서는 Moller 등에 의해 제안된 offset 항을 추가하는 방식으로 오차 함수의 적절한 수정을 통해 기존 ALOPEX 신경망을 개선하는 방법을 나타낸다.
