      • 유기질문화재 보존을 위한 천연 추출물의 항균·살충활성 연구

        This study evaluated the activity of biocidal compounds isolated from natural products and confirmed applicable possibility as conservative agents for orgainic cultural heritages damaged by biological degradation. we used methanol extracts of 11 Korean trees and 17 medicinal plants as natural resources. Among these extracts, Phellodendron amurense was investigated on antifungal activity against Coniophora puteana, Lentinus lepideus, Tyromyces palustris and Aspergillus niger. Asarum sieboldii was investigated on insecticidal activity against adult of Lasioderma serricorne. First, the extract of Phellodendron amurense was partitioned into hexane, dichloromethan and ethylacetate fraction. Among them, the ethylacetate fraction had the most significant antifungal activity. In addition, Asarum sieboldii essential oil showed an effective antifungal and insecticidal activity. As a result, the extract of Phellodendron amurense and Asarum sieboldii essential oil could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and Lasioderma serricorne.

        The objective of this study is to develope an interface of digital house which could be interacted between human and space. In here, digital house has dual meanings of indicating a virtual space itself and the extension of a physical space. The fundamental steps for this study are as 1) identify concept of digital house, 2) developing human sensibility database, 3) implementation an user interlace system for digital house. Specially, this paper focused on developing virtual model house and constructing the interactive user interlace environment to control and manage. The results of this study were as follows; Sensing data as a human sense and living environment was converted color and then expressed building components of the virtual model house such as 'wall', 'floor', 'ceiling' and so on. Since the color appeal to human sense, user could be have more comfortable feeling.

        고려시대의 청동 주물에서 관찰되는 불순물(Cu<sub>2</sub>S) 특성 연구

        청동으로 주조된 공예품의 금속 조직학적 연구를 통해 그 내부에 관찰되는 불순물을 추측하고, 그 물질의 특성을 파악하여 향후 제련 및 과거 청동 주물 제조 방법 및 재료 연구의 기초데이터를 제공하는 것을 목적으로 한다. 분석에는 SEM-EDS, XRD, 광학현미경을 이용하였다. 분석 결과, 3점의 시료는 ${\alpha}$상, ${\delta}$상, Pb, 불순물 입자인 $Cu_2S$의 4개의 상으로 이루어져 있으며, $Cu_2S$입자는 황동광을 제련하였을 때 생기는 제련 중간 발생물로 추측된다. 이 입자는 Pb와 인접하여 주로 관찰되며, ${\delta}$상 내부에서 관찰되는 경우도 있었다. $Cu_2S$의 입자 특성을 파악하기 위하여 1) 고려경 파편 및 2) $Cu_2S$와 Pb을 1273 K에서 녹여 $10^{\circ}C/min$의 속도로 냉각시켰다. 두 가지 실험으로 공통적으로 검은색 연기가 발생하였으며, 이 연기를 분석한 결과 PbS이었다. $Cu_2S$와 Pb을 반응 시킨 실험에서는 $Cu_2S+Pb{\rightarrow}PbS{\uparrow}+2Cu$의 반응식을 얻었다. The aim of this study was to obtain information on the ancient material of cast bronze through an investigation of the microstructure and impurity characteristics of the casting. Three Koryo bronze coins were analyzed using an optical microscope, scanning electron microscope, and electron dispersive X-ray analyses were used to determine the composition of the specimens. The three coins had 4 phases: ${\alpha}phase$, ${\delta}phase$, Pb, and impurities ($Cu_2S$). $Cu_2S$ was found to exist near Pb or in ${\delta}phase$. $Cu_2S$ is the inter mediate product of copper ore refining. Therefore, the copper ore was not completely refined. To find out the characteristic of $Cu_2S$, we melt 1)Koryo bronze coin and 2)$Cu_2S$ and Pb powder at 1273 K. The reaction between $Cu_2S$ and Pb at 1273 K yielded fine Cu and black gas, which was identified to be PbS and is presented below: $Cu_2S+Pb{\rightarrow}PbS{\uparrow}+2Cu$.

        인간 정체성에 관한 사회문화적 패러다임의 체현으로서의 사이보그

        매튜 비로는 그의 저서『다다사이보그』에서 사이보그가 베를린 다다의 실행에 있어 중요한 역할을 담당하였다고 주장한다. 그의 연구에서는, 베를린 다다의 포토몽타쥬에서부터 "뉴맨"의 구상까지 현대적 사이보그이론의 영향력 안에서 재고된다. 사이보그의 개념과 베를린 다다를 연결시키면서 파괴적이고 무정부적으로만 여겨졌던 베를린 다다의 건설적인 측면을 재발견 했다는 점에서 그의 연구는 학술적 가치를 지닌다. 하지만, 비로가 다다의 실행들을 현대 사회의 사회문화적 관념들이 함축된 사이보그의 개념 안에서 포괄적으로 분석하는 동안, 그의 연구에서 다다의 당대적 개입과 사회적 실행들은 주목받지 못한다. 따라서 이 논문은 베를린 다다 예술가들과 현 시대의 퍼포먼스 예술가 (올랑과 스텔락)들의 작업들을 분석함으로써, 각각의 예술적 실행에 다르게 드러나는 사이보그 이미지들을 비교 대조하려고 한다. 특히, 이 연구는 라울 하우스만의 구상과, 하나회크의 포토몽타쥬에서 드러난 "뉴맨"의 개념과, 그들의 혼성적이라기 보다는 분열되어있고, 갈등상황에 놓여진 근대적 인간을 표현하는 방법들을 살펴본다. 베를린 다다이스트들의 다양한 실행을 관통하는 "갈등"은 그들의 바이마르 공화국에 대한 경멸과, 전후 사회의 혼란의 반영이다. 현대의 사이보그 이론이 인간의 신체를 차이들이 결합하여 자아를 구성하는 제3의 공간으로 규정하는 데에 반하여, 베를린 다다의 실행에서 나타나는 분열된 자아는 자아와 타자 (사회적 권력등) 사이의 갈등의 유지를 인간 의식 구성의 중요한 부분으로 나타낸다. 이 논문은 또한 베를린 다다이스트들과 현 시대의 사이보그적 실행들이 기술에 대하여 취하는 다른 입장들을 논의한다. 이러한 여정을 통해서, 이 연구는 각각의 실행에서 드러난 혼성성은 작가들의 당대의 이데올로기를 반영하는 인간 정체성의 다른 관점들을 체현한다고 제안한다. In his book The Dada Cyborg, Matthew Biro argues that the cyborg played a central role in many important Dada productions in Berlin. From photomontage to envisioning the "New Man," the practices of the Berlin Dadaists are examined under the shadow of contemporary cyborg theories, particularly those of Donna J. Haraway. Biro's attempt to rediscover a constructive side of Berlin Dada by linking its practices to the notion of the cyborg has contributed a new scholarly approach to existing scholarship on Berlin Dada, which had previously been labeled as deconstructive and nihilistic. However, while he provides a comprehensive view of Dada practices under the notion of the cyborg, which is fully embedded within present-day complex social and cultural connotations, he fails to invite us to look at Dada's engagement with its own time and social practices. This paper, thus, will carefully analyze practices of Berlin Dadaists and contemporary performance artists (e.g., Orlan and Stelarc) to compare and contrast the notions of human identity represented in their practices. In particular, this paper examines Raoul Hausmann's notion of the New Man, as well as in photomontages made by Hanna Höch, and the ways in which they represent a modern man whose identity remains fragmented and conflicted rather than hybridized. The conflict that penetrates Berlin Dadaists' various practices reflects their condemnation of the Weimar Republic and the chaos of the post-war period. While contemporary cyborg theory regards the body as the third space in which differences are interlaced together to construct the idea of the self as in a perpetual state of becoming, the fragmented identity represented in Berlin Dada's practices hold on to the struggle between the self and the Other (e.g., social authority) as an integral part of constructing one's consciousness. The paper also addresses different attitudes toward technology in the Berlin Dadaists' practices and contemporary Cyborgian practices. Through these explorations, the study suggests that hybridity, which is represented in each practice, indeed embodies different perspectives on human identity that reflect certain ideologies of the artists' own time.

      • 사상자탕과 가미사상자탕의 항염증 및 항산화 효과 비교 연구

        Objectives : The anti-inflammatory and antioxidant effects of water extracts of Sasangja-tang(SSJ) and Gami-sasangja-tang(GSJ) were investigated. The effects of SSJ and GSJ were compared. Methods : We performed cell viability assay in HaCaT cells and RAW 264.7 cells using 3- (4,5-dimethylthiazol-2-yl)-2, 5 diphenyltetrazolium bromide(MTT) assay. We measured chemokines( regulated on activation normal T-cell expression and secreted ; RANTES/CCL5, interferon-inducible protein; IP-10/CXCL10, macrophage-derived chemokine; MDC/CCL22) in HaCat cells, also we measured cytokines (tumor necrosis factor-α; TNF-α, interleukin-6; IL-6) and nitric oxide(NO) production in RAW 264.7 cells using enzyme-linked immunosorbent assay(ELISA) and NO assay. Western blot assay was used to evaluate the expression for inducible nitric oxide synthase(iNOS) in RAW 264.7 cells. Results : SSJ and GSJ did not affect the cell viability at the concentrations treated (0-800 μg/ml). As a result of SSJ and GSJ treatment in HaCat cells stimulated by TNF-α(10 ng/ml) and interferon(IFN)-γ(10 ng/ml), the production of RANTES and IP-10 was inhibited significantly. However there was no significant difference in the secretion of MDC. And in RAW 264. 7 cells stimulated by lipopolysaccharide(LPS, 1 μg/ml), SSJ and GSJ treatment significantly inhibited the secretion of TNF-α and IL-6 and the production of NO. The expression of iNOS was also decresed by SSJ and GSJ treatment in RAW 264. 7 cells. Compared with SSJ, GSJ was superior to SSJ in inhibition of RANTES, IP-10, TNF-α, IL-6 and NO production at the concentration of 200 μg/ml. Conclusion : Both SSJ and GSJ have anti-inflammatory and antioxidant effects. And GSJ has better effects than SSJ.

        국내에서 유통 중인 축산환경개선제의 표기사항 및 항생제 내성 현황조사

        축산환경개선용 미생물 제품 중 시장점유율을 기준으로 상위제품 A, B,하위 제품 E, F와 현재 개발완료 되어진 C, D를 선정하여 생균수 측정과, 사료첨가용으로 사용이 금지된 항생제에 대한 내성 테스트를 실시하였다. 생균수 측정 결과 상위 점유율의 A, B사 제품은 표기상의 생균수와 일치하였으나, 하위 점유율을 차지하고 있는 E제품의 경우 함유균주나 효능에 대한 정보를 제공하고 않고 있을뿐더러 F사의 제품은 균수가 적게 측정되어 실제 표기사항과 일치하지 않았다. 항생제 내성실험결과 점유율에 따른 상관관계는 없었으나, B사를 제외한 제품의 대부분에서 내성균이 존재함을 확인하였다. Lincosimides 계열의 Lincomycicne과 Clindamycin의 경우 B사를 제외한 제품에서 내성균이 존재하였다. Penicillins 계열의 Amoxicillin, Ampicillin, Penicillin 및 Macrolide 계열의 Erythromycin 항생제는 B사와 E를 제외한 제품에서 내성균이 존재함을 확인하였다. Quinoline계열 Norploxacin 또한 B사와 E사를 제외한 제품에서 내성균을 보였으며 Neomycin의 항생제 또한 유사한 내성분포 결과를 나타내었다. Neomycin과 같은 Aminiglycosides계열의 Gentamycin, Streptomycin은 B사제품을 제외한 제품에서 내성을 나타내었다. 마지막으로 내성-균으로 사용 금지된 Tetracycline계열의 Oxytetracyclin은 12가지 항생제중 가장 높은 비율의 감수성을 나타내었으나 B, E사를 제외한 제품에서 내성균의 존재를 확인하였다. 실험결과 사용 금지된 항생제 품목임에도 불구하고 많은 제품에서 때 성균이 존재하였으며, 이에 따라 환경개선제 특성의 정확한 표기와 체계적인 유통 체계와 검증 과정이 필요하다고 사료된다. A study on livestock environment improving agents was conducted; top two brands (A and B) in the market, bottom two brands (E and F) based on market shares and two newly developed agents (C and D) were measured for viable count and tested for resistance towards antibiotics prohibited against livestock feeds. Test results revealed that the measured viable count of agents A and B matched those on the labels were identical; however agent E lacked information on viable counts nor the intended usage, while the measured viable count of agent F was less than the label-stated count. No correlation was found between the antibiotic-resistance test and market share, and most of the agents excluding B were found to display resistance case of Lincosimides such as Lincomycine and Clindmycin, resistant bacteria were found, with the except of agent B. Amoxicillin, Ampicillin and Penillin (type-Penecillins) and Erythromycin (type-Macrolide) were shown to contain resistant bacteria, with the except of agents Band E; the same for Norploxacin (type-Quinoline) and Neomycin antibiotics. Aminoglycosides such as Gentamycin and Streptomycin contained resistant bacteria, excluding agent B. Oxytetracyclin (type-Tetracycline), which is banned for use as resistant bacteria showed the highest sensitivity among the 12 antibiotics, revealed positive results in the test for resistant bacteria; again excluding of agents Band E. These results reveal that many agents contained resistant bacteria despite the fact that they were prohibited; this calls for a more accurate display of the facts and specifications, systematic distributions and strict verification processes of environment improving agents.

        불멸화된 일차 간세포를 이용한 C형 간염바이러스의 in vitro 배양

        Background/Aims: It is essential to develop an in vitro culture model of primary hepatocytes for the study of hepatocellular function and the pathogenesis of hepatitis C virus (HCV) infection. In this study, we have established the immortalized primary human hepatocyte (IPHH) and performed in vitro culture of HCV derived from human patient. Methods: Primary human hepatocytes were isolated from surgically resected liver tissue and then were immortalized by transfection with the SV40 large T antigen. The characterization of the IPHH during culture was analyzed by immunocytochemistry, RT-PCR, Western blot, ELISA, and soft agar assay. Next, sera and/or liver tissue homogenates from surgically resected liver tissues of patients with HCV infection were inoculated for the culture of HCV in IPHH. After HCV RNA extraction from IPHH and culture media, positive or negative stranded HCV RNA was examined by specific nest RT-PCR. Results: IPHH expressed liver-associated proteins but did not express alpha-fetoprotein. Also IPHH showed ammonia removal activity. With regard to its malignant potential, colony formation in soft agar assay was not observed. Next, positive and negative stranded HCV RNAs in IPHH infected with patient`s sera plus liver tissue homogenates were clearly detected whereas those in IPHH infected with only patient`s sera were not detected. Conclusions: These results demonstrated the phenotypic characteristics of IPHH and the feasibility in vitro culture system of HCV infected human samples. This system might be useful for study of pathogenesis of HCV infection or hepatocyte-based applications. (Korean J Gastroenterol 2008;52:150-160)

        한양방치료를 병행 후 호전된 진성적혈구증가증 환자 1례

        Objective: This study reports one case of polycythemia vera patient treated with oriental medicine in conjunction with conventional medicine. Methods: One patient who was diagnosed with polycythemia vera was treated using different modalities of herbs, acupuncture, and pharmacopuncture. An herbal formula, Yongdamsagan-tang was given three times a day with daily acupuncture treatment session, in addition to Soyeom and Jahageo pharmacopuncture injection which were administered three times a week. After treatment, patient's symptoms and blood test results were observed and analyzed. Results: After ten days of treatment, the patient had shown improvements in symptoms of headache, dizziness, tinnitus, and itchiness. Two months later, laboratory findings continued to be kept within the normal range and the intervals of phlebotomy therapy has extended to three months. Conclusions: This case study shows minimum evidence of safety and efficacy of oriental medicine for managing symptoms of polycythemia vera. Considering this is only a short term, single case study, further research is needed to confirm these results.
