      • 영동지역 4개 저수지의 식물성 플랑크톤 분포에 의한 오수생물학적 연구

        영동지역에 위치하는 4개 저수지를 대상으로 1991년 9월에 수질조사 및 식물성 플랑크톤 분포에 의한 오수생물학적 수질평가 연구를 시도하였다. 수질평가를 위하여 수온, 수소이온농도, 총질소, 총인, 엽록소 a, 및 식물성 플랑크톤의 현존량을 조사하였다. 동정된 식물성 플랑크톤은 총 58종류로 남조류 10종, 황색편모조 1종, 와편모조 2종, 갈색편모조 1종 및 녹조류 37종류였다. 식물성 플랑크톤의 우점종은 저수지별로 달랐으며, 죽헌저수지는 Microsystis aeruginosa, 북동저수지는 Peridinium cunningtonii var. contactum, 달방댐은 Staurastrum simonyi var. simonyi, 초당저수지는 Chlamydomonas umbonata로 밝혀졌다. 수질 및 식물성 플랑크톤의 분포유형을 기준으로 호수 수질을 생물학적으로 판정할 때 죽헌, 북동 및 초당저수지는 β-중부수성 호수로 달방댐은 빈부수성 호수로 판정되었다. The survey of water quality and saprobiotic phytoplankton distribution was carried out in 4 reservoirs in eastern area of Kangwon Province at September 1991. Temperature, pH, total-N, total-P, chl.a, and standing crop of phytoplankton were measured for water quality examination. Total numbers of phytoplankton identified were 58 spp, consisted of 10 spp Cyanophyta, 1 spp Chrysophyta, 2 spp Dinophyta, 1 spp Crytophyta, and 37 spp Chlorophyta. The dominant species of phytoplankton in each reservoir was different as follows: Microsystis aeruginosa in Jukhean, Peridinium cunningtonii var. contactum in Bukdong, Staurastrum simonyi var. simonyi in Dalbang, and Chlamydomonas umbonata in Chodang. Estimated from the data of water quality and phytoplankton distribution, the reservoirs of Jukheon, Bukdong and Chodang were β-mesosaprobic whereas Dalbang was oligosaprobic.

      • Pocine Adenovirus-3의 E1B Region의 鹽基序列 分析

        돼지 아데노바이러스(PAV-3). 6618주의 EIB region이 包含되어 있는 map unit 4.0에서 9.7까지의 유전자에 대한 1,984 bp의 염기서열을 決定하였으며, 이 結果를 알려진 여러 아데노바이러스 유전자와 비교하여 다음과 같은 결과를 얻었다. 1. PAV-3의 EIB유전자는 10개의 ORF로 구성되어 있으며, 그 중 아데노바이러스의 단백질과 유사성이 있는 것은 ORF1, ORF2 및 ORF3이었다. ORF1은 Ad41의 19kd 와 BAV-2에서의 EIB ORF2에서의 아미노산의 一致率은 각각 32%와 31%이었다. 2. ORF2는 Ad2 55kd protein과 tupaia adenovirus 44kd protein가 각각 34%로 아미노산 一致率이 가장 높았으며, Ad41의 52kd protein. BAV-3의 EIB ORF3에서도 33%의 一致率을 보였다. 3. ORF1은 61-666 uncleotide (606 bp), ORF 2에서는 429-1,850 uncleotide (1,422 bp)의 부위로 각각 202, 474 a.a로 構成되었으며, 예상되는 분자량은 20 kd와 52 kd이었다. 4. ORF3는 hexon-associated pIX유전자로 추정되며 내부에 1개의 polyadenylation signal(ATAAA)이 1938-1942 uncleotide에 위치하였으며, 이 부위는 TATA box (1937-1942 uncleotide)와 중복되어 존재하였다. Porcine adenovirus type 3 (PAV-3) does not cause severe infection in pigs. Adenovirus has been suggestive of live vaccine vector carrying foreign gene. One of insertion regions is delayed early (EIB) region. However, EIB region of PAV-3 has not been molecularly characterized to date. Nucleotide sequence of EIB of PAV-3 was determined. The EIB region was composed of 1,984 bp and located between 4.0 and 9.7 map units. Three potential open reading frames(ORFs) with low level of homology to other adenoviruses and a polyadenylation signal were identified in the rightward direction of genome. The nucleotide and the predicted amino acid sequences of EIB were compared to those of human and animal adenoviruses. One of the three potential ORFs. ORF1 encoded a polypeptide homologous to bovine adenovirus type 2(BAV-2) ORF2 and human adenovirus type 41(Ad41) 19 kd protein. ORF2 encoded a polypeptide homologous to human adenovirus type 2(Ad2) 55 kd protein, bovine adenovirus type 3(BAV-3) ORF3 and porcine adenovirus type 4(PAV-4) ORF2. The predicted protein of ORF1 had homology to those of Ad41 and BAV-2 with 32 and 31% respectively, whereas the deduced protein of ORF2 had homology to those of Ad2. BAV-3 and PAV-4 with 34, 33 and 29%, respectively.

        En bloc transplantation of horseshoe kidney in Korea

        Transplantation of the horseshoe kidney can be performed en bloc or split into 2 grafts according to the vascular anomaly and the existence of the urinary collecting system in isthmus. From 2011 to 2014, there were 3 horseshoe kidney transplantations in Korea and transplantations were performed at 2 different centers. The transplantations were carried out successfully for all recipients without complications. All recipients have shown good graft kidney function after transplantation. No severe complication was revealed during follow-up period. We described the surgical technique used in the en bloc method to overcome various vascular anomalies and difficulties in choosing cannulation site and postoperative complications. En bloc transplantation of a horseshoe kidney is a useful strategy for patients with end-stage renal disease, and can provide favorable outcomes compared to the transplantation of a normal kidney.

        Mathematical model for early functional recovery pattern of kidney transplant recipients using serum creatinine

        Background: Commonly used equations for calculating estimated glomerular filtration rate (eGFR) are not applicable when serum creatinine (Scr) is rapidly changing like the post-transplant period. A new mathematical model applicable to the post-transplant period is required. Methods: All 623 patients who underwent kidney transplantation from January 2008 to June 2018 at a single institute were included to validate the Scr mathematical equations, and 14,360 Scr laboratory results from the time of re-perfusion to 30 days post-transplantation were analyzed. Results: In the validation of model equations, linear regression analysis yielded adjusted R2 values of 0.972 and 0.925 for equation 5 (applicable when renal function is changing) and equation 1 (applicable when renal function is unchanged), respectively. In selected cases, the population comprised individuals who presented an adjusted R2 value >0.95 with equation 5. Linear regression analysis showed that adjusted R2 values and Pearson's correlation coefficients for equation 5 and equation 1 were 0.994 and 0.997 (P<0.001) and 0.956 and 0.978 (P<0.001), respectively. Most of the eGFR formulas are mathematically applicable only if the creatinine input rate equals the creatinine output rate when comparing between commonly used eGFRs and creatinine clearance using the modeled equation. Conclusions: The proposed equations can provide a new perspective for calculating renal function during the early phase of kidney transplantation. A study of a correlation between the equations and long-term graft outcomes is required.

      • Investigation for Expression of the Porcine Pseudorabies Virus Glycoproteins in Bacteria and Insect Cells

        Aujeszky's disease (AD), also called pseudorabies, is an infectious viral disease caused by an alpha herpes virus and has domestic and wild pigs, as well as a wide range of domestic and wild animals, as the natural host. Aujeszky's disease virus (ADV) virions contain several envelope glycoproteins. Among them, glycoproteins gB, gC and gD are regarded as the major immunogenicity proteins and the antibodies induced by them can neutralize virus in vitro or in vivo. In this study, we investigated expression of these glycoproteins using the bacterial and baculovirus expressionn system. Successful expression of ADV glycoproteins in E. coli was confirmed by SDS-PAGE and Western blot analysis and their optimal expression condition was determined. However, the recombinant proteins generated in the bacterial expression system which lacks glycosylation process frequently lose their biological activity. We tried to express the ADV glycoproteins using the baculovirus expression vector system. The recombinant gB, gC and gD were detected at approximately 100, 60 and 50 kDa on SDS-PAGE and Western blotting, respectively. The optimal expression conditions were determined for MOI(multiplicity of infection) and post-infection days. One MOI and 4 or 5 days post-infection were the best conditions for the expression of the ADV glycoproteins in Sf21 cells. We are currently investigating the antigenicity of recombinant proteins using experimental animals.

        Efficient Production of Porcine Circovirus Type 2 Capsid Protein using Baculovirus

        Porcine circovirus type 2 (PCV2) is a single-stranded circular DNA virus associated with Postweaning multisystemic wasting syndrome (PMWS), which is considered to be an important infectious swine viral disease. PCV2 capsid protein encoded by ORF2 is a structural protein and expected as the high immunogenicity protein. In this study, we generated recombinant baculovirus containing ORF2 of PCV2 and analyzed the optimal conditions for the production of capsid protein in insect cell. Production and status of recombinant capsid protein in insect cell were confirmed by SDSPAGE and Western blot analysis using His tag antibody and anti-PCV2 serum. The yield of recombinant capsid protein was high like as shown visible on SDS-PAGE. Optimal multiplicity of infection (MOI) and infection time of recombinant virus were determined as 5 MOI and 4 days, respectively. ORF2 is known to have Nlinked glycosylation site, but we couldn`t detect the glycosylation of recombinant protein in insect cells.

        <P>We report an accurate extraction technique of effective mobility (mu(eff)) by considering gate-bias (V-GS) dependent effective inversion charges (Q(inv,eff)) as a normalized correction factor (sigma(V-GS)) when using the gate-to-source/drain capacitance-voltage (C-V) measurement in a p-channel Si/SiGe MOSFET with a floating body structure. In the proposed technique, two different capacitance-voltage configurations: a gate-to-source/drain (CG-SD) configuration without body contact and a gate-to-source/drain/body (CG-SDB) configuration with body contact are utilized for the accurate extraction of mu(eff).</P>

        Effects of hydrogen peroxide on voltage-dependent K<sup>+</sup> currents in human cardiac fibroblasts through protein kinase pathways

        Human cardiac fibroblasts (HCFs) have various voltage-dependent $K^+$ channels (VDKCs) that can induce apoptosis. Hydrogen peroxide ($H_2O_2$) modulates VDKCs and induces oxidative stress, which is the main contributor to cardiac injury and cardiac remodeling. We investigated whether $H_2O_2$ could modulate VDKCs in HCFs and induce cell injury through this process. In whole-cell mode patch-clamp recordings, application of $H_2O_2$ stimulated $Ca^{2+}-activated$ $K^+$ ($K_{Ca}$) currents but not delayed rectifier $K^+$ or transient outward $K^+$ currents, all of which are VDKCs. $H_2O_2-stimulated$ $K_{Ca}$ currents were blocked by iberiotoxin (IbTX, a large conductance $K_{Ca}$ blocker). The $H_2O_2-stimulating$ effect on large-conductance $K_{Ca}$ ($BK_{Ca}$) currents was also blocked by KT5823 (a protein kinase G inhibitor) and 1 H-[1, 2, 4] oxadiazolo-[4, 3-a] quinoxalin-1-one (ODQ, a soluble guanylate cyclase inhibitor). In addition, 8-bromo-cyclic guanosine 3', 5'-monophosphate (8-Br-cGMP) stimulated $BK_{Ca}$ currents. In contrast, KT5720 and H-89 (protein kinase A inhibitors) did not block the $H_2O_2-stimulating$ effect on $BK_{Ca}$ currents. Using RT-PCR and western blot analysis, three subtypes of $K_{Ca}$ channels were detected in HCFs: $BK_{Ca}$ channels, small-conductance $K_{Ca}$ ($SK_{Ca}$) channels, and intermediate-conductance $K_{Ca}$ ($IK_{Ca}$) channels. In the annexin V/propidium iodide assay, apoptotic changes in HCFs increased in response to $H_2O_2$, but IbTX decreased $H_2O_2$-induced apoptosis. These data suggest that among the VDKCs of HCFs, $H_2O_2$ only enhances $BK_{Ca}$ currents through the protein kinase G pathway but not the protein kinase A pathway, and is involved in cell injury through $BK_{Ca}$ channels.
