        A Novel Function of Interleukin-10 Promoting Self-Renewal of Hematopoietic Stem Cells

        <P>Self-renewal of hematopoietic stem cells (HSCs) is key to their reconstituting ability, but the factors regulating the process remain poorly understood. Here, we show that Interleukin-10 (IL-10), a pleiotropic immune modulating cytokine, can also play a role in regulating HSC self-renewal. First, a quantitative decrease of primitive hematopoietic cell populations, but not more matured cells, was observed in the bone marrows of IL-10 disrupted mice as determined by long-term in vitro cultures or in vivo competitive repopulation assays. In contrast, normal HSCs from 5-fluorouracil treated marrows cultured on the IL-10 secreting stroma displayed an enhanced repopulating activity compared with cells grown on control stroma, with ninefold higher numbers of donor-derived HSCs in the reconstituted recipient marrows. Moreover, limiting dilution transplantation assay demonstrated that exogenous addition of IL-10 in the stroma-free cultures of purified Lin- Sca-1+ c-kit+ cells caused three- to fourfold higher frequencies of HSCs in the 5-day short-term culture without indirect inhibitory effect of IL-10 on tumor necrosis factor-alpha or interferon-gamma secretion. Interestingly, primitive hematopoietic cells, including Lin- Sca-1+ c-kit+ or side population cells, expressed the surface receptor for IL-10, and microenvironmental production of IL-10 was sharply increased in the osteoblasts lining the trabecular regions of the radiation-stressed marrow but not in the steady-state marrows. These results show that IL-10 may be a ligand that can stimulate self-renewal of HSCs to promote their regeneration in addition to being a ligand for immune regulation. Disclosure of potential conflicts of interest is found at the end of this article.</P>

      • <i>Davallia mariesii</i> Moore Improves Fc <i>ε</i> RI-Mediated Allergic Responses in the Rat Basophilic Leukemia Mast Cell Line RBL-2H3 and Passive Cutaneous Anaphylaxis in Mice

        <P><I>Davallia mariesii</I> Moore (Drynaria rhizome extract (DRE)) is widely known for its efficacy in treating inflammation, arteriosclerosis, and bone injuries. This study evaluated whether treatment with DRE inhibited Fc<I>ɛ</I>RI-mediated allergic responses in the RBL-2H3 mast cells and investigated the early- and late-phase mechanisms by which DRE exerts its antiallergic effects. IgE anti-DNP/DNP-HSA-sensitized RBL-2H3 mast cells were tested for cytotoxicity to DRE, followed by the assessment of <I>β</I>-hexosaminidase release. We measured the amounts of inflammatory mediators (e.g., histamine, PGD<SUB>2</SUB>, TNF-<I>α</I>, IL-4, and IL-6) and examined the expression of genes involved in arachidonate and Fc<I>ε</I>RI signaling pathways. In addition, we confirmed the antiallergic effects of DRE on passive cutaneous anaphylaxis (PCA) in mice. DRE inhibited RBL-2H3 mast cell degranulation and production of allergic mediators in them. In early allergic responses, DRE reduced expression of Fc<I>ε</I>RI signaling-related genes (e.g., Syk, Lyn, and Fyn) and extracellular signal-regulated kinase phosphorylation in mast cells. In late allergic responses, DRE reduced PGD<SUB>2</SUB> release and COX-2 expression and cPLA<SUB>2</SUB> phosphorylation in Fc<I>ɛ</I>RI-mediated mast cells. Lastly, 250–500 mg/kg DRE significantly attenuated the IgE-induced PCA reaction in mice. These findings provide novel information on the molecular mechanisms underlying the antiallergic effects of DRE in Fc<I>ɛ</I>RI-mediated allergic responses.</P>

        식육중 잔류항균물질 비교 조사 -서울지역 도축 소와 돼지를 중심으로-

        This study was carried out to compare the residual antibiotic materials in muscles of slaughter cattle and swine from slaughterhouses in Seoul from 2000 to 2001 by EEC-4-plate method, Charm Il and HPLC method. 1. Residual antibiotic materials were detected from 95 samples(0.8%) by EEC-4-plate and 57 samples(10.2%) by Charm Il. The final HPLC method determined the positives are 43(45.3%) and 27(47.3%) respectively. 2. The detection ratios were 45% by EEC-4-plate and 47% by Charm Il. 3. Seventy samples were classified as tetracyclines 56(75.7.4%), sulfonamides 10(14.9%), β-lactam 6(8.1%) chloramphenicol 1(1.4%). Three of them were confirmed to be positive simmultaneously for tetracyclines, sulfonamides and chloramphenicol. 4. The highest residual concentration of chlortetracycline, oxytetracycline, sulfamethazine, sulfadimethoxine, sulfaquinoxaline, penicillin, ampicillin and chloramphenicol were 0.34, 11.29, 68.16, 0.13, 4.0, 0.12, 0.4 and 0.04ppm, respectively.

      • 송아지고기 소시지와 시중에 판매되는 우육과 돈육 소시지 제품의 저장특성 비교

        This study was undertaken to compare the storage characteristics between veal sausage and commercial pork and beef sausages in Korean domestic market. We manufactured 3_types of sausages and purchased 2_types of sausages in the market: T1(B-company product), T2(O-company product) and we made T3(smoked veal sausage), T4(smoked veal sausage with pepper) and T5(cooked veal sausage) products. During 20 days of storage at 4oC, T5 showed significantly higher in TBA value and VBN value (p<0.05). But TBA and VBN values showed normal range in all products. T3, T4, and T5 showed higher total microbial count than T1 and T2 (p<0.05). According to the result, beef and pork sausages in the market showed different total microbial count and VBN values from those made in laboratory. This result could be come from the different additives used and manufacturing processes. However, all products can be stored up to 20 days in the cooler.

        We extract hand region by using color information, because it is an important feature for human vision to distinguish objects. Because pixel values in images are changed according to the luminance and lighting source, it is difficult to extract hand region exactly without previous knowledge. We generate a hand skin model at leaning stage, and extract hand region from images by using the model. We also use Kalman filter to consider the change of pixel values in hand skin model. Kalman filter restricts the search area for extracting hand region at next frame also.

      • Treadmill을 이용한 중년 여성들을 혈중지질의 변화

        The Purpose of the study is to know the Relation of the disease of adult people. The plasma Lipoprotein after treadmill exercise routines. The subjects were 15 healthy woman in their 40s and proceeded the exercise 12 weeks at the 70%of their maximal heart rate. the training frequency was 5 times a week and exercise time the 30 minutes. 1. T.G are reduced significantly after 12-week training 30.4mg/dℓ comparing with them before training. (P< 0.05) 2. LDL-C are reduced signigicantly after 12-week 29.5mg/dℓ training comparing with them before training. (P< 0.05) 3. HDL-C are reduced significantly after 12-weektraining. (P< 0.05) 4. Hb are reduced significantly after 12-week 6.4mg/dℓ training comparing with them before training. (P< 0.05) In conclusion, when the subjects continued doing exercise for 12 weeks at the intensity of 70% their maximal heart rate, the serum lipid was chanced reducing significantly. Accordingly, the training intercity and duration of this study sill natice the importance if chronic exerese in their 40s woman.

      • 육우 비선호 부위를 활용한 육우햄의 품질특성

        This study was undertaken to compare the quality characteristics of dairy beef ham. Dairy beef hams were manufactured using non-preferred portion (top round). 5 treatments were prepared: T1(beef 50% + pork emulsion 50%), T2(beef 65%emulsion 35%), T3(beef 80%emulsion 20%), T4(beef 90%emulsion 10%), T5(beef 100%emulsion 0%). In chemical composition, dairy beef content increases, moisture content was increased, fat content was decreased. In the meat quality characteristics, T5 showed higher (p<0.05) water holding capacity value than the others. Dairy beef content increases, product loss and cooking loss were decreased. In hunter color, dairy beef content increases, redness was increased. In texture profile analysis, T4 and T5 were significantly higher than the others. As a result, dairy beef ham with 80~90% dairy beef and 10~20% pork emulsion has low fat content, high product yield and superior texture profile, which could be developed as a dairy beef hams using non-preferred portion.

      • NOD/SCID 마우스 모델을 이용한 인간 제대혈 혼합이식에서의 생착 양상 분석

        연구배경: 제대혈은 골수에 비해 많은 장점을 가지고 있음에도, 이식 후 성적에 있어 총세포수에 의해 좌우되며, 한 개의 태반으로부터 얻을 수 있는 총세포수의 한계가 있어 주로 소아에 국한되고 있다. 따라서 제대혈 응용의 확대를 위해 생착량을 늘이기 위한 방법으로 다중공여자에 의한 제대혈 혼합이식을 늘일 수 있는지 연구하기 위하여, 본 연구에서는 주조직적합성 형별로 조합된 제대혈을 NOD/ SCID 생쥐에 이식하는 방법으로, 혼합생착의 가능성을 연구하였다. 방법: 조합된 2개 공여자로부터 얻어진 제대혈들을 단일이식 및 혼합이식하여, 혼합이식된 숙주에 생착된 세포를 PCR-SSOP 방법을 사용하여 공여자 별로 추적하였다. 또한 각 공여자세포의 상대적 생착비율을 대조군인 단일제대혈 이식에서 얻어진 생착량과 비교하는 정량적 비교도 함께 하였다. 결과: 총단핵구를 혼합이식한 실험군에서는 주조직적합성의 일치도에 관계없이 한쪽공여자의 세포가 다른쪽에 비해 우세한 편향생착을 보였다. 그러나 임파구를 비롯한 Lineage 양성세포를 제거한 실험군에서는 주조직적합성의 불일치에도 불구하고 뚜렸한 생착의 공존이 관찰되었다. 결론: 본 연구는 제대혈의 생착량을 증가를 목적으로 총단핵구를 혼합이식할 경우 한쪽공여자의 세포에 의해 편중되며, 이것을 이식 후 생착과정에서의 두 종류의 세포의 경쟁에 의해 초래된다는 것을 보이고 있다. 또한 이러한 경쟁은 제대혈에 포함되어 있던 임파구들에 의한 것이며, 생착공존을 위하여는 공여자간 주조직적합성의 일치도 보다, 임파구의 제거가 더 중요하다는 것을 보여주고 있다. Background: Although umbilical cord blood (UCB) has been an attractive alternatives for bone marrow cells, application is limited to pediatric case due to limitation in total number of nucleated cells, that is a single most important factor for outcomes of UCB transplantation. Therefore, increasing overall engraftment by mixed transplantation of UCB derived from multiple donor should comprise one strategy to circumvent the posed limitation of UCB transplantation. In order to investigate the feasibility of establishing co-engraftment by multi-donor UCB cells, we carried out a xenotransaplantation study using NOD/SCID mice for systemic analysis of results on the input-based control of single unit transplantation. Methods: UCB units with various extent of HLA-matchings were co-transplanted into NOD/SCID mice along with single unit transplantation control. The relative contributions of engraftment by cells from each donor-derived were analyzed by HLA polymorphism using PCR-SSOP. Results: In all HLA-based mixed transplantation of total nucleated UCB cells, engraftment of one donor predominated over the other despite that equivalent amount of engraftments were achieved by single donor transplantation. When lineage depleted UCB units were co-transplanted, significant degree of co-engraftments were observed regardless of HLA disparity. Conclusion: Our result show that one donor cells dominate over the other in mixed UCB transplantation and that it is due to competition between donor cells during post-transplantation process. Our results suggest that immune cells contained in UCB unit mediate such competitioin and that 6 locus HLA matching would not be sufficient to prevent the competition.

        교잡종 찰옥수수에 대한 연구 III : Ⅲ. 국내 옥수수 유전자원 수집종에 대한 주요 특성 분포

        2006년 국내 각 지역으로부터 수집된 재래종옥수수 유전자원을 관행방법에 의해 파종하고 주요특성별로 분포도를 살펴본 결과는 다음과 같다. 1. 수집된 유전자원에 대한 발아율은 대부분 80∼100%이상의 분포를 보였으나, 일부 수집계통에는 30% 미만의 극히 저조한 발아율을 보였다. 2. 수집자원에 대한 초기생육을 살펴보면 극히 생육이 저조한 것이 40% 정도를 보였고, 극히 우수한 자원은 3%미만으로 나타났다. 3. 간장은 대부분이 150∼199cm범위에 55%가 분포하고 있었으나, l00cm이하가 5%, 250cm이상이 8%로 나타나 수집종간에 큰 차이를 보였다. 4. 주당 분얼수는 1개 미만이 40%의 분포를 보였으나, 일부 수집종에서 2 내지 3개의 분얼수를 보이는 계통이 확인되었다. 5. 개화소요일수는 61∼64일의 범위에서 40%, 57∼60일의 범위에서 30%의 분포를 보였으며, 특히 일부 수집종의 경우 55일 이하의 극조생종도 10% 정도의 분포를 보였다. 6. 이삭길이는 대부분 11∼15cm범위에 60%가 포함되어 있으나, 일부계통에서 5cm미만이 2%, 16∼20cm의 이삭길이도 10%가 분포하였다. Germination ratio on the 300 collected genetic resources showed 80 to 100 percentage, but those of some accessions were very low as below 30 percentage. Early growth phase of most of these lines were very poor while some collected lines were very vigor. Range of stem height showed from 150 to 199cm, but that of some accessions showed below 100cm as five percentage and some of them was over 250cm as eight percentage. Number of tillers per plant appeared less one but some collected lines have two to three tillers per plant. In days to tasseling, some accessions showed more four per plant. Ear length of these accession showed above 60 percentage as 11 to 15cm, while that of some collected lines showed over 10 percentage as 16 to 20cm.

      • 뇌기저동맥 첨단부 폐쇄증후군 후 지연성 올리브 비대에 동반된 눈떨림

        눈입천장떨림(Oculopalatal tremor)은 치아핵, 적핵 및 올리브핵을 연결하는 신경로(dentatorubro-olivary pathway, Guillain-Mollaret triangle)의 손상 이후에 발생하는 비대성올리브핵 변성(hypertrophic inferior olivary degeneration)에 의한 지연성(delayed) 합병증이다. 다리 뇌출혈(pontine hemorrhage) 또는 뇌줄기경색(brainstem infarction)후 아래 올리브핵비대(inferior olivary hypertrophy)가 발생한 환자에서 입천장떨림(palatal tremor) 없이 눈떨림이 보고된 경우가 있으나, 뇌바닥동맥끝증후군(top of the basilar syndrome) 후 아래올리브핵비대가 발생한 환자에서 눈떨림이 보인 경우는 드물다. 뇌바닥동맥끝증후군 후에 아래올리브핵비대가 발생한 환자에서 지연성 수평 눈떨림 보고하고자 한다. 이것은 양측 소뇌다리의 손상 때문에 눈떨림의 수직 성분이 동반되지 않은 수평눈떨림을 보인 것으로 유추할 수 있다. Oculopalatal tremor(OPT) is a delayed complication of hypertrophic inferior olivary degeneration after the lesion in the dentato-rubro-olivary pathway, the Guillain-Mollaret triangle. Some cases of ocular tremor without palatal tremor associated with inferior olivary hypertrophy after pontine hemorrhage or brainstem infarction have been reported, but ocular tremor with inferior olivary hypertrophy after top of the basilar syndrome has rarely been described. We describe a patient showing delayed-onset horizontal ocular tremor with inferior olivary hypertrophy after top of the basilar syndrome. It can be postulated that the vertical component of ocular tremor could have not developed because of the lesions in bilateral cerebral peduncles.
