        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

        <P>Natural phytochemicals of plant origin, including flavonoids, have been found to be potent antioxidants providing beneficial effects against oxidative stress-related diseases. The present study was carried out to investigate the antioxidant properties of morin, a flavonoid originally isolated from the flowering plants of the Moraceae family. Superoxide dismutase (SOD)-like activity and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) radical scavenging activity were determined. We also investigated the cytoprotective effects of morin against hydrogen peroxide (H2O2)-induced DNA damage and apoptosis in V79-4 Chinese hamster lung fibroblasts. Our results demonstrated that morin had strong scavenging effects against ABTS' radicals with enhanced SOD activity, which varied in a dose-dependent manner. Morin was found to reduce H2O2-induced intracellular reactive oxygen species generation and nuclear DNA damage, and it recovered cell viability damaged by H2O2 via inhibition of mitochondrial dysfunction-mediated apoptosis. Notably, the treatment of V79-4 cells with morin markedly enhanced the expression of heme oxygenase-1 (HO-1) but not quinone oxidoreductase-1, which was associated with the increased expression and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the downregulation of Kelch-like ECH-associated protein 1 expression. Based on our findings, we conclude that morin effectively ameliorated oxidative stress-induced DNA damage through intrinsic free radical scavenging activity and activation of the Nrf2/HO-1 pathway.</P>

        During high-speed running of machine tools, serious vibrations often may occur in their feed-drive system. In this paper, a 4-D.O.F. planar motion model is proposed in order to analyze a ball screw feed-drive system. The equations of motion are derived for the proposed model and natural frequencies of the system are calculated. Also its transient response due to driving motor speed control input are calculated. The theoretical analysis results showed good agreement with experimental ones measured from a laboratory feed-drive test system. Consequently we concluded that the proposed 4-D.O.F model can be well applicable to the vibration analysis and dynamic design of machine tool feed-drive system.

      • 체중이동 과제 학습시 효과적인 운동학적 되먹임 유형과 상대적 빈도

        Physical therapists have been using balance and weight shifting training to induce improvements in standing and walking. This study compared the effects of kinetic feedback frequency and concurrent kinetic feedback on the performance and learning of a weight shifting skill in young, nondisabled adults. Sixteen young adults without known impairment of the neuromusculoskeletal system volunteered for the study. Subjects in each of three kinetic feedback groups performed a weight shifting task in an attempt to minimize error between their effort and a center of pressure (COP) template for a 12 second period. Feedback was provided: 1) concurrently (concurrent feedback), 2) after each trial (100% feedback), 3) after ever other trial (50% feedback). Immediate and delayed (24 hous) retention tests were performed without feedback. During ascquistion phase, the concurrent feedback group exhibited less error than either of the post response feedback group. For the immediate retention test, the 50% feedback group exhibited less error than did the 100% feedback and concurrent feedback. During the delayed retention, 50% feedback group displayed less error than did the other groups. But no significant differences were found between groups. these results suggest that practice with concurrent feedback is beneficial for the immediate performance, but not for the learning of this weight shifting skill. Lower frequency of feedback resulted in more permanent changes in the subject's ability to complete the task.

      • 韓國 高等學生들의 生殖과 發生에 대한 誤槪念

        구성주의자들은 학생들이 수업전에 수업내용과 관련된 선입관을 가지고 있는데, 이 선입관중에 서도 당대의 과학적 지식과는 다른 개념들을 오개념(misconception)이라고 하였다. 또 이 오개념은 전통적인 수업방법에 의해서는 쉽사리 바뀌지 않는다고 하였다. 본 연구는 우리나라 고등학교 3학년 학생들이 보편적으로 생식과 발생에 대한 오개념을 가지고 있는지를 알아보기 위해 수행되었다. 선행연구로써 먼저 학생들이 가지고 있는 생식과 발생에 대한 오개념들을 생물교사와 학생들에게 자유응답형 질문지를 통하여 이미 조사하여, 본 연구를 위한 객관식 질문지를 만드는 자료로 이용하였다. 생식과 발생에 대한 학생들의 오개념을 알아보기 위해 특별히 제작된 질문지는 모두 10개 문항으로 구성되어 있으며, 연구대상으로 선정된 541명의 학생들은 대도시ㆍ중소도시ㆍ시골 지역별, 남ㆍ녀 성별, 일반ㆍ과학고 능력별로 유층표집하였다. 우리나라 고등학생들의 생식과 발생에 대한 오개념을 알아보기 위한 객관식 문항들을 분석한 결과, 시험관 아기가 수정되는 장소와 자라는 장소, 수정되지 않은 달걀의 부화와 달걀 세표, 수정란의 난할, 식물의 수정과 수분의 차이, 고등동물에서 암, 수의 성 결정 시기, 사람의 정자가 난자와 만나 수정이 이루어진 순간부터 아기가 태어나기까지 걸리는 시간, 생식과정에서 감수분열이 필요한 이유, 배낭과 배낭세포, 일란성 쌍생아의 수정원리 등에 대한 개념에서 오개념을 가진 학생들이 많이 나타났다. 정답률은 일반ㆍ과학 고등학생간에 유의한 차가 없었으며, 일반학생들의 성별, 지역별 차이도 없었다. In order to find out the Korean high school students' misconception about reproduction and development, a paper and pencil test form of the questionnaire was developed which was constructed of 10 items, and administered to 541 Korean high school students. Their responses were collected and analyzed. According to the analysis of their responses, students misunderstood the concepts of the fertilizing site and incubating region of human embryo, development of unfertilized egg, a location of cell in an egg, cleavage of fertilized egg, korean terminology about fertilization and pollination of plant, sex determination time of higher organisms, the period of pregnancy, significance of meiosis, an embryo sac and principle of fertilization of identical twins. For the students' conceptions about reproduction and development, the general lower academic level students didn't misunderstand statistically compared to higher level students' group significantly, and there was not the statistically significant difference for sex and residential locality.

      • HIV-IN과 NaeIR의 chimeric protein의 제작 및 activity 측정

        The retroviral integrase (IN) mediates a sequence of cleavage and joining reaction in integration of viral DNA copy into host chromosome in the retroviral life cycle. C-terminal domain of IN has DNA-binding activity. NaeIR, a type-Ⅱ restriction system from the bacterium Nocardia aerocolonigenes, recognizes the sequence 5'-GCCGGC-3'. N-terminal domain of NaeIR has catalytic activity. We construct chimeric protein with catalytic domain of HIV-1 integrase and binding domain of NaeIR. All chimeric proteins overexpressed in Escherichia coli and purified by nikel affinity chromatography and SP-Sepharose column. All chimeric protein did not display detectable endonucleolytic activity. 3-D structure change of protein is reason of non-detectable activity.

      • 高等學生들의 生態學的 誤槪念

        Since the research findings about ecological misconception of korean pupils have not represented until now, the primary purpose of this research was to identify high school students' misconceptions on ecology. At first, in order to find out high school students' misconceptions on ecology, a free-answer form of questionnaire, specially designed through analysis of high school biology curriculum, particularly ecological content, was developed and sent to ninety five high school students who had lived in Chungju area.Their responses were collected and analyzed qualitatively. As the results of the analysis of the data, the summary of this research are as follows: 1. High school students have remembered at the point of ecological terminology or simple knowledge than understood dynamic relations among ecosystem about ecology. These results were same with foreign pupils'. 2. They confused the output of plant respiration to oxygen. Especially, they recognized that invisible microorganisms, example for zoo plankton or fungi, didn't do gas exchange. 3. Although many students usually misundertood that bacteria is toxic organism, they thought correctly the bacteria in ecosystem as decomposer was not toxic. 4. There were several misconceptions arose from biological terminology such as red blooming, red algae and winter migratory birds.

      • 한국기업의 해외기업인수 결정요인에 관한 연구

        본 연구는 한국기업의 해외기업인수의 증대에 부응하여 이의 결정요인을 분석하고자 하였다. 해외기업인수 결정요인은 모기업의 국제화 경험, 인수지역의 진출경험, 인수기업의 규모, 모기업의 연구집약도, 제품수명주기 등의 여섯 가지로 파악되었다. 모기업의 국제화경험은 그 정도가 높을수록 해외기업에 대한 인수성향이 높은 것으로 나타났다. 인수지역에 대한 진출경험은 해외기업인수시 현지의 기업환경 또는 기업문화에 생소하지 않고 적응가능하게 하여 기업인수에 대한 불확실성을 제거해준다. 특히 한국기업과 같은 개도국의 경우 초기 인수전략의 특성상 해당 투자동일 지역에 이미 자화사가 진출해 있는 경우 현지기업의 매각정보입수 및 기존의 사업관계등으로 신설보다는 인수전략을 선택할 가능성이 높아진다. 또한 과거국내기업의 인수경험이 있는 기업은 국내기업의 인수를 통해 인수작업을 학습할 수 가 있어 새로운 조직적변화에 보다 효과적으로 대처할 수 있다. 인수기업의 규모는 그 정도가 클 수록 상대적으로 많은 경영자원과 생산설비를 투입할 수 있는 여력이 있는 것으로 보아 인수행위에 보다 적극적이라 하겠다. 한국기업의 해외기업인수의 대표적인 사례의 분석에서는 두 기업모두 선진시술의 취득, 기존시장의 보호 및 개척, 원자재 확보의 동기로 투자가 되었다. 이는 한국기업의 해외투자의 특성을 잘 대변해 주는 것이라 하겠다. 특히 선진기술의 취득을 위한 해외기업인수와 해외시장 확보 및 개척을 위한 해외기업인수의 중요성은 최근들어 가격경쟁력의 약화, 선진각국의 기술보호조치로 인하여 기술도입이 점차어려워지도 있다는 점에서 볼 때 해외기업인수가 갖는 의미는 매우 크다고 하겠다.

        상악 전치부 견인시 치근 흡수 발생 양상에 관한 연구

        본 연구는 상악 전돌증을 보이는 환자에게서 상악 제 1 소구치를 발치한 후 견인시 bracket 종류에 따른 치근흡수 차이, 이동양상에 따른 치근 흡수 차이, 각 치아간에 따른 전치부의 치근 흡수 정도를 연구하기 위해 단국 대학교 부속 치과 병원 교정과에 내원한 남녀 환자 32명을 대상으로 연구하여 다음과 같은 결과를 얻었다. 1. 상악 전치 후방 견인시 치근 흡수를 보이지 않는 것은 31.77%, 약간의 흡수는 33.33%, 치근의 1/4이하 흡수는 23.96%, 1/4이상은 10.94%를 보였다. 2. Apical tipping, Lingually bodily movement, ant-post apical displacement의 세가지 이동 양상은, 치근 흡수와의 상관 관계에 유의한 차이를 보이지 않았다. 3. Standard br.을 사용한 환자는 많은 치축 변화를 보이고 치근첨의 이동은 적었으나, straight br.을 사용한 환자는 적은 치축 변화를 보이고 치근첨의 이동량은 많았다. 4. Standard br.과 straight br.의 치근흡수에서 standard br.을 사용한 환자가 더 많은 치근 흡수를 보였다. 5. Standard br.을 사용시 치근 흡수정도는 2>1>3=0순을 보이고, straight br.을 사용시에는 0>1>2>3으로 나타났다. This study was carried out in order to find out the pattern of the root resorption following maxillary incisor retraction after maxillary 1st bicuspid extration in maxillary protrusion patients. For this study, thirty two patient who received orthodontic treatment were chosen. The results were as follows; 1. Of the total 192 teeth, 61 teeth(31.77%) showed no apical root loss, 64 teeth(33.33%) fell into the class "slight", 46 teeth(23.96%) fell into the class "moderate". Only 21 teeth(10.94%) were classified as "excessive". 2. No correlation was noted between the amount of apical root loss and the types of tooth movement of the maxillary central incisors. 3. The patients who were treated with standard brackets had more changes in tooth axis and less movement of root apexes, but the patients with straight brackets has less changes in tooth axis and more movement of root apexes. 4. Comparing the degree of root resorption between bracket types, patients who had used standard brackets showed more apical root loss than patients who had used straight brackets. 5. The most frequent degree of root resorption observed in standard bracket patients was second degree, followed by first degree and third degree. The most frequent degree of root resorption observed in straight bracket patients was zero degree, followed by first, second degree and third degree.
