      • Research and Development of Garlic Harvester

        Because the head of garlic can be prevented from heart disease, cancer, improving immunity function and sterilizing, it also is used as important cooking seasoning. Therefore, making use of the machinery to gather the head of garlic harvest homework seems to be extremely important. This machine belongs to a plant bed of three lines of self-propelled harvest machines. It can reap the head of garlic of field smoothly, and achieve to cut the function of stems within 5cm of pole measurement. Its principal apparatus is composed of plowing, picking up, dialing in, propping up send, cutting stem pole, behind transport by organization, collecting networks or container, engine and the power conveying, controlling system and running apparatus. Under the preliminary stage investigation and proceeding to the test in the field, the result shows that the high mean value of the plant is 493.6mm, the pole diameter mean value of the stem is 8.4mm, the high mean value of the garlic ball is 39.8mm and the long mean value of the root is 63.99mm. At present, the function to cut the pole size of stems has already reached to 2-4 cm. We are sure that this machine not only can solve the rural aging and fewer manpower dilemmas, but also can lower the production costs, raising peasant's income and developing the garlic industry forever.

        A Color-Reaction-Based Biochip Detection Assay for RIF and INH Resistance of Clinical Mycobacterial Specimens

        The widespread occurrence of drug-resistant Mycobacterium tuberculosis places importance on the detection of TB (tuberculosis) drug susceptibility. Conventional drug susceptibility testing (DST) is a lengthy process. We developed a rapid enzymatic color-reaction-based biochip assay. The process included asymmetric multiplex PCR/templex PCR, biochip hybridization, and an enzymatic color reaction, with specific software for data operating. Templex PCR (tem- PCR) was applied to avoid interference between different primers in conventional multiplex- PCR. We applied this assay to 276 clinical specimens (including 27 sputum, 4 alveolar lavage fluid, 2 pleural effusion, and 243 culture isolate specimens; 40 of the 276 were non-tuberculosis mycobacteria specimens and 236 were M. tuberculosis specimens). The testing process took 4.5 h. A sensitivity of 50 copies per PCR was achieved, while the sensitivity was 500 copies per PCR when tem-PCR was used. Allele sequences could be detected in mixed samples at aproportion of 10%. Detection results showed a concordance rate of 97.46% (230/236) in rifampicin resistance detection (sensitivity 95.40%, specificity 98.66%) and 96.19% (227/236) in isoniazid (sensitivity 93.59%, specificity 97.47%) detection with those of DST assay. Concordance rates of testing results for sputum, alveolar lavage fluid, and pleural effusion specimens were 100%. The assay provides a potential choice for TB diagnosis and treatment.

        CircularRNA_104670 plays a critical role in intervertebral disc degeneration by functioning as a ceRNA

        This study was carried out to explore the roles of circular RNAs (circRNAs) in nucleus pulposus (NP) tissues in intervertebral disc degeneration (IDD). Differentially expressed circRNAs in IDD and normal NP tissues were identified based on the results of microarray analysis. Bioinformatics techniques were employed to predict the direct interactions of selected circRNAs, microRNAs (miR), and mRNAs. CircRNA_104670 was selected as the target circRNA due to its large multiplier expression in IDD tissues. After luciferase reporter and EGFP/RFP reporter assays, we confirmed that circRNA_104670 directly bound to miR-17-3p, while MMP-2 was the direct target of miR-17-3p. The receiver-operating characteristic (ROC) curve showed that circRNA_104670 and miR-17-3p had good diagnostic significance for IDD (AUC circRNA_104670 = 0.96; AUC miRNA-17-3p = 0.91). A significant correlation was detected between the Pfirrmann grade and expression of circRNA_104670 (r = 0.63; p = 0.00) and miR-17-3p (r = −0.62; p = 0.00). Flow-cytometric analysis and the MTT assay showed that interfering with circRNA_104670 using small interfering RNA (siRNA) inhibited NP cell apoptosis (p < 0.01), and this inhibition was reduced by interfering with miR-17-3p. Interfering with circRNA_104670 suppressed MMP-2 expression and increased extracellular matrix (ECM) formation, which were also reduced by interfering with miR-17-3p. Finally, an MRI evaluation showed that circRNA_104670 inhibition mice had a lower IDD grade compared with control mice (p < 0.01), whereas circRNA_104670 and miRNA-17-3p inhibition mice had a higher IDD grade compared with circRNA_104670 inhibition mice (p < 0.05). CircRNA_104670 is highly expressed in the NP tissues of IDD and acts as a ceRNA during NP degradation.

        Insufficient radiofrequency ablation-induced autophagy contributes to the rapid progression of residual hepatocellular carcinoma through the HIF-1α/BNIP3 signaling pathway

        Currently speaking, it is noted that radiofrequency ablation (RFA) has been the most widely used treatment for hepatocellular carcinoma (HCC) occurring in patients. However, accumulating evidence has demonstrated that the incidence of insufficient RFA (IRFA) may result in the identified rapid progression of residual HCC in the patient, which can greatly hinder the effectiveness and patient reported benefits of utilizing this technique. Although many efforts have been proposed, the underlying mechanisms triggering the rapid progression of residual HCC after IRFA have not yet been fully clarified through current research literature reviews. It was shown in this study that cell proliferation, migration and invasion of residual HepG2 and SMMC7721 cells were significantly increased after the IRFA was simulated in vitro. In other words, it is noted that IRFA could do this by enhancing the image of autophagy of the residual HCC cell via the HIF-1 α/BNIP3 pathway. Consequently, the down-regulation of BNIP3 may result in the inhibition of the residual HCC cell progression and autophagy after IRFA. Our present study results suggest that IRFA could promote residual HCC cell progression in vitro by enhancing autophagy via the HIF-1 α/BNIP3 pathway. For this reason, it is noted that the targeting of the BNIP3 may be useful in preventing the rapid growth and metastasis of residual HCC after IRFA. [BMB Reports 2019; 52(4): 277-282]

        Evaluation of Liver Metastases Using Contrast-Enhanced Ultrasound: Enhancement Patterns and Influencing Factors

        Background/Aims: To evaluate the enhancement patterns of liver metastases and their influencing factors using dynamic contrast-enhanced ultrasound (CEUS). Methods: A total of 240 patients (139 male and 101 female; 58.5±11.2 years of age) diagnosed with liver metastases in our hospital were enrolled in this study to evaluate tumor characteristics using CEUS. A comparison of enhancement patterns with tumor size and primary tumor type was performed using the chi-square test. The differences between quantitative variables were evaluated with the independent-sample t-test and one-way analysis of variance. Results: The enhancement patterns of liver metastases on CEUS were categorized as diffuse homogeneous hyperenhancement (133/240, 55.4%), rim-like hyperenhancement (80/240, 33.3%), heterogeneous hyperenhancement (10/240, 4.2%), and isoenhancement (17/240, 7.1%). There were significant differences in the enhancement patterns during the arterial phase based on the nodule size (p=0.001). A total of 231 of the nodules showed complete washout during the portal phase, and 237 nodules were hypoenhanced during the delayed phase. The washout time was correlated with tumor vascularity, with a longer washout time observed in hypervascular metastases compared to hypovascular metastases (p=0.033). Conclusions: Diffuse homogeneous hyperenhancement followed by rapid washout was the most common enhancement pattern of liver metastases on CEUS and was affected by the nodule size and tumor vascularity. Small metastases were prone to show diffuse homogeneous hyperenhancement. Hypervascular metastases showed a significantly longer washout time compared to hypovascular metastases.         Three New 29 Carbon Skeletons Pentacyclic Triterpenoids and S-equol from Biogas Slurry

        Bioactive natural products were firstly obtained from biogas slurry. Three new 29 carbon skeletons of the pentacyclic triterpenoid compounds 24-norolean-12-ene-3,22-dione (1), 3β-hydroxy-24-norolean-12-ene-22-one (2), 3α-hydroxy-24-norolean-12-ene-22-one (3), as well as one known compound S-equol (4) were isolated and purified from the MeOH extract of chicken manure biogas slurry. The molecular structures of the four compounds were elucidated based on the extensive spectroscopic data analysis, and the structure of Compound 1 was further confirmed by single-crystal X-ray diffraction. The structures of Compounds 1, 2, and 3 are similar with oleanolicum and hederagenin that has excellent anti-tumor activities. The cytotoxicity against five cancer cell lines (Hela, A549, MCF7, PC3, and B16) of Compounds 1–4 was tested. Similar to Compound 4 (S-equol), Compounds 1–3 (pentacyclic triterpenoids) showed cytotoxicity activity against different tumor cell lines. Compounds 1–3 showed slightly lower cytotoxicity activities than Compound 4. The IC50 of Compound 4 was determined to be 9.7–27.6 μM, while the IC50 values of 1–3 were 17.6–65.3 μM. There are no significant differences in the cytotoxicity capacities between Compounds 1, 2, and 3.

        Inhibitory Effect of Melanoma Differentiation Associated Gene-7/Interleukin-24 on Invasion In Vitro of Human Melanoma Cancer Cells

        The acquisition of metastasis potential is a critical point for malignant tumors. Melanoma differentiation associated gene-7/interleukin-24 (mda-7/IL-24) is a potential tumor suppress gene and frequently down-regulated in malignant tumors. It has been implicated that overexpression of MDA-7 led to proliferation inhibition in many types of human tumor. Invasion is an important process which is potential to promote tumor metastasis. However, the role and potential molecular mechanism of mda-7/IL-24 to inhibit the invasion of human melanoma cancer is not fully clear. In this report, we identified a solid role for mda-7/IL-24 in invasion inhibition of human melanoma cancer LiBr cells, including decreasing of adhesion and invasion in vitro, blocking cell cycle, down-regulating the expression of ICAM-1, MMP-2/9, CDK1, the phosphorylation of ERK and Akt, NF-κB and AP-1 transcription activity. Meanwhile, there was an increased expression of PTEN in mda-7/IL-24 over-expression LiBr cells. Our results demonstrated that mda-7/IL-24 is a potential invasion suppress gene, which inhibits the invasion of LiBr cells by the down-regulation of ICAM-1, MMP-2/9, PTEN, and CDK1 expression. The molecular pathways involved were the MAPK/ERK, PI3K-Akt, NF-κB, and AP-1. These findings suggest that mda-7/IL-24 may be used as a possible therapeutic strategy for human melanoma cancer.

        Quantitative Assessment on the Variation of Compressibility of Wenzhou Marine Clay During Destructuration

        A series of oedometer test were performed on both undisturbed and remolded specimens of Wenzhou marine clay to quantitatively investigate the degradation of soil structure during compression. The laboratory tests show that the swell index of both natural and remolded Wenzhou marine clay increase with the increase in consolidation stress. Hence, the normalizing parameter ‘swell sensitivity’, termed as the ratio of the remolded to the natural swelling index, can only be regarded as a qualitative parameter. On the other hand, the normalized stress sensitivity has a good parabolic relationship with the consolidation stress during destructuration. This result indicates that the stress sensitivity can be used as a quantitative interpretation of the degradation of soil structure for natural Wenzhou marine clay. Comparison between stress sensitivity and additional void ratio during destructuration shows that the variation of soil structure during destructuration represented by stress sensitivity and additional void ratio are essentially the same. In addition, the parameter used in formulating the variation of stress sensitivity with the stress level is correlated with natural void ratio and void ratio at liquid limit.

        The influence of soil aggregation induced by water content variation on the engineering behaviors of cement-treated silt subgrade

        Water content is an important indicator of highway subgrade compaction, which has an impact on the engineering behaviors of pavement structures significantly. To study the influence of soil aggregation induced by water content variation on the engineering behaviors of silt subgrade, a series of laboratory experiments, including sieving analysis, unconfined compression tests and hydraulic conductivity tests, were conducted on the cement-treated silt subgrade. The influence of water content on the soil aggregation, unconfined compressive strength (UCS) and hydraulic conductivity were examined, respectively. The distribution range of UCS index (IUCS) and hydraulic conductivity index (Ik) were expressed accordingly. Test results demonstrate that when the water content lower than the optimum water content (wopt), the specimens are more homogeneous, the UCS increases regularly with the increase of curing time and water content, the hydraulic conductivity decreases regularly with curing time and water content increased. When the water content is higher than the wopt, the silt aggregation leads to the structural inhomogeneity of specimens, the strength decreases irregularly with the increase of curing time and water content, the hydraulic conductivity increases irregularly with the increase of curing time and water content accordingly. The soil aggregation induced by water content variation is proved to be a major reason for the ununiformly distribution of the UCS and hydraulic conductivity, the influence of which is more significant than that of binders when the water content higher than the wopt. Hence, controlling water content within 2% lower the wopt is strongly suggested to forming a more homogeneous structure of the subgrade.

        Effect of Cu doping on the SCR activity of CeO2 catalyst prepared by citric acid method

        CeO2–CuO catalyst prepared by citric acid method was investigated for selective catalytic reduction of NO with NH3. The activity of the CeO2 catalyst was enhanced about 8–27% in the temperature range of 125–225 ℃ at a space velocity of 28,000 h 1 by the addition of Cu. It was found that the state of Cu species had great impact on the SCR performance of CeO2–CuO catalyst. Cu2+ can enhance the low temperature activity of SCR reaction, while CuO would promote NH3 oxidation before SCR reaction at high temperature, which would cause the decrease of its high temperature SCR activity.
