        A Synthetic Naringenin Derivative, 5-Hydroxy-7,4′-diacetyloxyflavanone-<i>N</i>-phenyl Hydrazone (N101-43), Induces Apoptosis through Up-regulation of Fas/FasL Expression and Inhibition of PI3K/Akt Signaling Pathways in Non-Small-Cell Lung Cancer Cells

        <P>Naringenin, a well-known naturally occurring flavonone, demonstrates cytotoxicity in a variety of human cancer cell lines; its inhibitory effects on tumor growth have spurred interest in its therapeutic application. In this study, naringenin was derivatized to produce more effective small-molecule inhibitors of cancer cell proliferation, and the anticancer effects of its derivative, 5-hydroxy-7,4′-diacetyloxyflavanone-<I>N</I>-phenyl hydrazone (N101-43), in non-small-cell lung cancer (NSCLC) cell lines NCI-H460, A549, and NCI-H1299 were investigated. Naringenin itself possesses no cytotoxicity against lung cancer cells. In contrast, N101-43 inhibits proliferation of both NCI-H460 and A549 cell lines; this capacity is lost in p53-lacking NCI-H1299 cells. N101-43 induces apoptosis via sub-G<SUB>1</SUB> cell-cycle arrest in NCI-H460 and via G<SUB>0</SUB>/G<SUB>1</SUB> arrest in A549 cells. Expression of apoptosis and cell-cycle regulatory factors is altered: Cyclins A and D1 and phospho-pRb are down-regulated, but expression of CDK inhibitors such as p21, p27, and p53 is enhanced by N101-43 treatment; N101-43 also increases expression levels of the extrinsic death receptor Fas and its binding partner FasL. Furthermore, N101-43 treatment diminishes levels of cell survival factors such as PI3K and p-Akt dose-dependently, and N101-43 additionally induces cleavage of the pro-apoptotic factors caspase-3, caspase-8, and poly ADP-ribose polymerase (PARP). Cumulatively, these investigations show that the naringenin derivative N101-43 induces apoptosis via up-regulation of Fas/FasL expression, activation of caspase cascades, and inhibition of PI3K/Akt survival signaling pathways in NCI-H460 and A549 cells. In conclusion, these data indicate that N101-43 may have potential as an anticancer agent in NSCLC.</P>

      • A sub-1-volt nanoelectromechanical switching device.

        <P>Nanoelectromechanical (NEM) switches have received widespread attention as promising candidates in the drive to surmount the physical limitations currently faced by complementary metal oxide semiconductor technology. The NEM switch has demonstrated superior characteristics including quasi-zero leakage behaviour, excellent density capability and operation in harsh environments. However, an unacceptably high operating voltage (4-20 V) has posed a major obstacle in the practical use of the NEM switch in low-power integrated circuits. To utilize the NEM switch widely as a core device component in ultralow power applications, the operation voltage needs to be reduced to 1 V or below. However, sub-1 V actuation has not yet been demonstrated because of fabrication difficulties and irreversible switching failure caused by surface adhesion. Here, we report the sub-1 V operation of a NEM switch through the introduction of a novel pipe clip device structure and an effective air gap fabrication technique. This achievement is primarily attributed to the incorporation of a 4-nm-thick air gap, which is the smallest reported so far for a NEM switch generated using a 'top-down' approach. Our structure and process can potentially be utilized in various nanogap-related applications, including NEM switch-based ultralow-power integrated circuits, NEM resonators, nanogap electrodes for scientific research and sensors.</P>

        Inkjet‐Printable Nanoporous Ag Disk Arrays Enabling Coffee‐Ring Effect‐Driven Analyte Enrichment Towards Practical SERS Applications

        To make surface enhanced Raman scattering (SERS) sensors more practical, we propose nanoporous Ag disks as SERSactive plasmonic structures that can be readily inkjet-printed just before use to avoid degradation of SERS enhancement. Together with the aid of the enhanced plasmonic fi elds from the nanoporous Ag (confirmed by electromagnetic simulation), we utilize a coffee-ring effect to concentrate target analytes, which is demonstrated by confocal Raman measurements. By using the proposed SERS sensor, Raman signals of TiO 2 nanoparticles with a concentration of ppm to sub-ppb have been successfully measured. TiO 2 in commercial consumables has been also detected by distinguishing its crystalline phase.

        2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242) induces apoptosis via modulating E7 expression and inhibition of PI3K/Akt pathway in SiHa human cervical cancer cells.

        <P>The Maillard reaction is a chemical reaction occurring between an amino acid and a reducing sugar, usually requiring thermal processing. Maillard reaction products (MRPs) have antioxidant, antimutagenic, and antibacterial effects, and although 2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242), a fructose-tyrosine MRP, appears to inhibit proliferation of cancer cells, its mechanism of action has not been studied in detail. We found that HPB242 treatment modulated expression of cyclins and tumor suppressor genes in SiHa human cervical cancer cell lines: cyclins and phospho-pRB were downregulated, whereas the expression of CDK inhibitors and p53 was enhanced. HPB242 induced apoptosis dose-dependently by suppressing E7 expression and leading to sub-G1 cell-cycle arrest in SiHa cell lines; treatment also led to the proteolytic cleavage of caspase-3, -9, and poly (ADP-ribose) polymerase. Moreover, HPB242 upregulated Fas expression, altered expressions of pro- and antiapoptotic factors, and also inhibited nuclear translocation of nuclear factor κB and phosphorylation of IκB. HPB242 treatment decreased phosphatidyl inositol-3 kinase and p-Akt expression levels, demonstrating that this survival pathway may also be inhibited by HPB242. Cumulatively, HPB242 promotes apoptosis by influencing E7 expression, inducing cell-cycle arrest at sub-G1 phase, and promoting both intrinsic (mitochondrial) and extrinsic (Fas-dependent) apoptosis in SiHa human cervical cancer cells.</P>

        Currently the evaluation of high school science and mathematics learning has been too much knowledge-centered without concerning the inquiring ability or scientific attitude. This study, therefore, was made for the purpose of establishing of evaluation model for high school science (biology and earth science) and mathematics learning and its evaluation items. In this paper, the research was done about three fields, biology, earth science and mathematics. 1. For the biology, the evaluation methods of inquiry learning were studied with the foreign references, and proper evaluation items were suggested. 2. For the earth science, after analizing 5 kinds of earth science textbook of current using in high school, essential 19 items were selected. 5 of them were tested in test group and control group, then, the results were analized. 3. For the mathematice, the teaching elements of「Set」and「Flow chart」in mathematics (Ⅰ) were selected and structured. Detailed teaching objects were established and evaluation items were introduced.

        브라켓 제거에 따른 입술과 주위 연조직의 변화

        교정치료에 따른 연조직 변화 양상은 개개인의 차이가 심하므로 교정치료 중 흔히 재평가가 필요하다. 이를 위해서 고정식 교정장치가 부착되어 있는 상태의 입술의 위치와 교정치료 종료 후의 위치에 대한 비교 예측이 필요하다. 이 연구는 교정치료가 종료되어 고정식 교정장치를 제거하는 52명을 대상으로 T1(장치제거 전), T2(장치제거 직후), T3(장치제거 1.5개월 후)의 측모두부방사선사진상에서 입술과 그 주위 연조직을 분석한 결과 다음과 같은 결론을 얻었다. 장치제거 직후(T2-T1) 하순이 후퇴하며 장치제거 직후부터 1.5개월 후(T3-T2)까지도 계속 후퇴하였다. 장치제거 1.5개월 후(T3-T1), 기준선(G-perpendicular line)에 대한 입술의 평균 후퇴양은 상순(Ls)에서 0.38 mm, 하순(Li)은 0.88 mm를 보였다. 주로 장치제거 직후(T2-T1)에 하순이 후퇴하는 여자군과 달리 남자군은 장치제거 직후부터 1.5개월 후(T3-T2)까지 하순이 주로 후퇴하는 양상을 보였으나, 장치제거 1.5개월 후(T3-T1)에는 남자군과 여자군 간의 뚜렷한 차이를 보이지 않았다. Objective: Soft tissue changes due to orthodontic treatment has large individual variation. Therefore continuous evaluation during treatment is required. Patients with fixed orthodontic appliances often wonder if their lip positions will change after the removal of brackets, but only a few studies exist on this topic. The objective of this study was to evaluate the changes of the lips and perioral soft tissue after bracket removal. Methods: The sample used in this study was 19 males and 33 females. Cephalometric X-rays were taken at 3 stages - T1 (before debonding), T2 (just after debonding), T3 (1.5 months after debonding). Results: The lower lip was retruded immediately after debonding (T2-T1), and 1.5 months after debonding (T3-T2). The mean amounts of retrusion from the vertical reference plane (sG perpendicular line) were about 0.38 mm for the upper lip and 0.88 mm for the lower lip. Immediately after debonding, lip retrusion of females was greater than that of males. During the post-debonding period, lower lip of males was retruded more than that of females. Conclusion: Lips are retruded after bracket removal, and there is no gender difference 1.5 months after debonding.

        급성 충수돌기염 의증 환자의 진단에서 컴퓨터 단층촬영과 초음파검사의 비교 분석

        Background: This study was conducted to compare the accuracy of computered tomography(CT) and sonography(US) in diagnosing acute appendicitis and to analyze the radiologic factors which influence the result. Methods: We retrospectively studied the cases of 53 patients who were suspected of having appendicitis and who were examined by CT and US covered a three-year period. The final diagnosis was determined from the surgico-pathologic diagnosis and clinical follow-up for two months, as well as from the discharge diagnosis. Results: Thirty-eight(38) patients had acute appendicitis; 15 patients did not. In the analysis, CT and US, respectively, revealed sensitivities of 89.5% and 73.7%, specificities of 93.3% and 60.0%, positive predictive values of 97.1% and 82.4%, negative predictive values of 77.8% and 47.4%. Among patients diagnosed with acute appendicitis, 10 patients were discordant in the US and the CT examinations; CT findings were correct in 8 and US findings in 2. Among non-appendicitis patients, 5 patients were discordant in the US and the CT examinations; CT was correct in 5 and US in 0. Nighteen(19) patients had complications such as an abscess or a perforation. Among these patients, the degree and the extent of the complication were accurately evaluated by CT in 15 patients and by US in 5 patients. Conclusion: CT is more accurate than US in diagnosing and excluding acute appendicitis. The use of CT will substantially decrease unnecessary laparotomies and post-operation complications.

        To investigate the effect of Shudihuang(熟地黃) aqua-acupuncture at the meridian points BL 23 on the renal function in normal and two kidney one clip Goldblatt hypertensive rats. The results of this study were as follows : 1. Urine volume, free water clearance rate were significant increased after Shudihuang aqua-acupuncture in normal rats. 2. Urinary excretion on potassium and chloride, glomerular filtration rate were significant decreased after Shudihuang aqua-acupuncture in normal rats. 3. Urine volume, ruinary excretion of electrolytes were significant decreased after Shudihuang aqua-acupuncture in two kindney one clip Goldblatt hypertensive rats. 4. Free water clearance, glomerular filtration rate were significant increased after Shudihuang aqua-acupuncture in two kidney one clip Goldblatt hypertensive rats.

        2 차원 선형탄성 균열문제에 적합한 파괴기준을 알아보기 위해, 응력확대계수 K과 K 균열파단각과 파괴하중을 최소 변형에너지밀도 기준에 의해 구하는 유한요소 프로그램, SED를 개발하였다. 이 프로그램에서는, 6절점 삼각형 특이요소를 사용한 균열선단을 제외한 다른 부분에서는 고전적인 8절점 사변형 등매개변수 요소를 사용하였다. SED의 결과를 최대 원주방향 인장응력 기준과 최대 에너지 해방률 기준에 의한 프로그램의 결과차, 그리고 Jenq & Shah의 실험결과와 동일한 조건에서 비교하였다. 본 연구는 혼합모드 균열전파 모델의 계속적인 연구로서 이전의 최대 원주방향 인장응력 기준과 최대 에너지 해방률 기준에 의한 해석치와는 다소 차이가 있으나 전반적으로 Jenq 와 Shah의 실험치와 비슷한 양상을 보였다. To find out an adequate failure criterion in two-dimensional linear elastic crack problems, finite element program, SED, which determines stress intensity factors K, K, crack angle and peak load by the minimum strain energy density failure criterion were developed. In this program, the conventional quadratic isoparametric elements were used in all regions except the crack tip zone where triangular singular elements with 6 nodes were used. The results of SED were compared with the results of those which followed by the maximum circumferential tensile stress criterial and those by the maximum energy release rate criteria and those by Jenq and Shah's experiments of the same geometry and material properties. This study which has kept continuously as mixed mode crack propagation models is somewhat different from previous results, maximum circumferential tensile stress criterial and the maximum energy release rate criteria, but SED's result is generally similar to Jenq & Shah's experiments.

        This study is to obtain a basic data for a better environment and visual design improvement for the sound barrier tunnel located along the overpasses which passed through downtown areas. For this purpose, the traffic noise was measured in Daegu. In addition. using the sound analysis software, equivalent sounds were checked and analyzed. Finally, the alternatives of sound barrier tunnel were proposed.
