      • Chitosan-graft-polyethylenimine as a gene carrier

        Jiang, H.L.,Kim, Y.K.,Arote, R.,Nah, J.W.,Cho, M.H.,Choi, Y.J.,Akaike, T.,Cho, C.S. Elsevier Science Publishers 2007 Journal of controlled release Vol.117 No.2

Chitosans have been proposed as biocompatible alternative cationic polymers that are suitable for non-viral delivery. However, the transfection efficiency of chitosan-DNA nanoparticles is still very low. To improve transfection efficiency, we prepared chitosan-graft-polyethylenimine (CHI-g-PEI) copolymer by an imine reaction between periodate-oxidized chitosan and polyethylenimine (PEI). The molecular weight and composition of the CHI-g-PEI copolymer were characterized, using multi-angle laser scattering (GPC-MALS) and 1H nuclear magnetic resonance (1H NMR), respectively. The copolymer was complexed with plasmid DNA (pDNA) in various copolymer/DNA (N/P) charge ratios, and the complex was characterized. CHI-g-PEI showed good DNA binding ability and high protection of DNA from nuclease attack. Also, with an increase in charge ratio, the sizes of the CHI-g-PEI/DNA complex showed a tendency to decrease, whereas the zeta potential of the complex showed an increase. The CHI-g-PEI copolymer had low cytotoxicity, compared to PEI 25K from cytotoxicity assays. At high N/P ratios, the CHI-g-PEI/DNA complex showed higher transfection efficiency than PEI 25K in HeLa, 293T and HepG2 cell lines. Our results indicate that the CHI-g-PEI copolymer has potential as a gene carrier in vitro.

The 8th International Comparison of Absolute Gravimeters 2009: the first Key Comparison (CCM.G-K1) in the field of absolute gravimetry

        Jiang, Z,,linká,š,, V,Arias, F E,Liard, J,Merlet, S,Wilmes, H,Vitushkin, L,Robertsson, L,Tisserand, L,Pereira Dos Santos, F,Bodart, Q,Falk, R,Baumann, H,Mizushima, S,,kinen, J Springer-Verlag 2012 METROLOGIA -BERLIN- Vol.49 No.6

<P>The 8th International Comparison of Absolute Gravimeters (ICAG2009) took place at the headquarters of the International Bureau of Weights and Measures (BIPM) from September to October 2009. It was the first ICAG organized as a key comparison in the framework of the CIPM Mutual Recognition Arrangement of the International Committee for Weights and Measures (CIPM MRA) (CIPM 1999). ICAG2009 was composed of a Key Comparison (KC) as defined by the CIPM MRA, organized by the Consultative Committee for Mass and Related Quantities (CCM) and designated as CCM.G-K1. Participating gravimeters and their operators came from national metrology institutes (NMIs) or their designated institutes (DIs) as defined by the CIPM MRA. A Pilot Study (PS) was run in parallel in order to include gravimeters and their operators from other institutes which, while not signatories of the CIPM MRA, nevertheless play important roles in international gravimetry measurements. The aim of the CIPM MRA is to have international acceptance of the measurement capabilities of the participating institutes in various fields of metrology. The results of CCM.G-K1 thus constitute an accurate and consistent gravity reference traceable to the SI (International System of Units), which can be used as the global basis for geodetic, geophysical and metrological observations of gravity. The measurements performed afterwards by the KC participants can be referred to the international metrological reference, i.e. they are SI-traceable.</P><P>The ICAG2009 was complemented by a number of associated measurements: the Relative Gravity Campaign (RGC2009), high-precision levelling and an accurate gravity survey in support of the BIPM watt balance project. The major measurements took place at the BIPM between July and October 2009. Altogether 24 institutes with 22 absolute gravimeters (one of the 22 AGs was ultimately withdrawn) and nine relative gravimeters participated in the ICAG/RGC campaign.</P><P>This paper is focused on the absolute gravity campaign. We review the history of the ICAGs and present the organization, data processing and the final results of the ICAG2009.</P><P>After almost thirty years of hosting eight successive ICAGs, the CIPM decided to transfer the responsibility for piloting the future ICAGs to NMIs, although maintaining a supervisory role through its Consultative Committee for Mass and Related Quantities.</P>

Detection of 881A→881G Mutation in Tyrosinase Gene and Associations with the Black Ear Coat Color in Rabbits

        Jiang, Y.L.,Fan, X.Z.,Lu, Z.X.,Tang, H.,Xu, J.-Q.,Du, L.-X. Asian Australasian Association of Animal Productio 2002 Animal Bioscience Vol.15 No.10

The tyrosinase gene was selected as a candidate for uncovering genetic mechanism causing 'black ear' coat color in rabbits. A PCR-SSCP detection method was established for the $881^A{\rightarrow}881^G$ mutation located in the central region of the tyrosinase gene between the CuA and CuB binding region signatures, and this was confirmed by sequencing and alignment. Fully consistent associations between the SNP and 'black ear' coat color were observed by analysis in a "black ear" pedigree and on 61 unrelated individuals. This SNP can serve as a molecular marker for use in "back ear" wool rabbit breeding.

      • Genotypexenvironment interactions for chilling tolerance of rice recombinant inbred lines under different low temperature environments

        Jiang, W.,Lee, J.,Chu, S.H.,Ham, T.H.,Woo, M.O.,Cho, Y.I.,Chin, J.H.,Han, L.,Xuan, Y.,Yuan, D.,Xu, F.,Dai, L.,Yea, J.D.,Koh, H.J. Elsevier Scientific Pub. Co 2010 Field crops research Vol.117 No.2

Chilling injury is one of the major environmental stresses in rice cultivation in high-latitude and high-altitude regions. In this study, we cultivated a set of recombinant inbred lines (RILs) derived from Milyang 23 (indica)/Tong 88-7 (japonica) crosses in Kunming (high-altitude location), Yanji (high-latitude location), Chuncheon (cold water irrigation), and Suwon (normal) to evaluate the genotypexenvironment (GxE) interactions for chilling tolerance. RILs were the most severely damaged under the natural chilling air temperatures in Kunming. Significant GxE interactions in all measured agronomic traits were detected, and thus, the additive main effects and multiplicative interaction (AMMI) statistical model was applied to dissect the GxE interactions. The biplots of grand mean and IPCA1 (interaction principal component axes) of chilling-related traits accounted for most of the total treatment sums of squares. The IPCA scores of spikelet fertility and phenotypic acceptability were relatively smaller in Chuncheon than in Yanji and Kunming, implying that the screening for chilling tolerance with cold water irrigation in Chuncheon was more stable, whereas the Yanji and Kunming plantings were more sensitive to GxE interactions for chilling tolerance. These results demonstrate that multi-locational screening should be the best strategy for developing widely adaptable chilling-tolerant varieties in rice.

Galactosylated chitosan-graft-polyethylenimine as a gene carrier for hepatocyte targeting

        Jiang, H-L,Kwon, J-T,Kim, Y-K,Kim, E-M,Arote, R,Jeong, H-J,Nah, J-W,Choi, Y-J,Akaike, T,Cho, M-H,Cho, C-S Nature Publishing Group 2007 Gene Therapy Vol.14 No.19

Chitosans have been proposed as alternative, biocompatible cationic polymers for nonviral gene delivery. However, the low transfection efficiency and low specificity of chitosan need to be addressed before clinical application. We prepared galactosylated chitosan-graft-polyethylenimine (GC-g-PEI) copolymer by an imine reaction between periodate-oxidized GC and low-molecular-weight PEI. The molecular weight and composition were characterized using gel permeation chromatography column with multi-angle laser scattering and 1H nuclear magnetic resonance, respectively. The copolymer was complexed with plasmid DNA in various copolymer/DNA (N/P) charge ratios, and the complexes were characterized. GC-g-PEI showed good DNA-binding ability and superior protection of DNA from nuclease attack and had low cytotoxicity compared to PEI 25K. GC-g-PEI/DNA complexes showed higher transfection efficiency than PEI 25K in both HepG2 and HeLa cell lines. Transfection efficiency into HepG2, which has asialoglycoprotein receptors, was higher than that into HeLa, which does not. GC-g-PEI/DNA complexes also transfected liver cells in vivo after intraperitoneal (i.p.) administration more effectively than PEI 25K. These results suggest that GC-g-PEI can be used in gene therapy to improve transfection efficiency and hepatocyte specificity in vitro and in vivo.Gene Therapy (2007) 14, 1389–1398; doi:10.1038/sj.gt.3302997; published online 19 July 2007

      • A Phase 3 Evaluation of Daclatasvir plus Asunaprevir in Treatment-Naive Patients with Chronic HCV Genotype 1b Infection

        ( L. Wei ),( F. Wang ),( M. Zhang ),( J. Jia ),( A.A. Yakovlev ),( W. Xie ),( E.Z. Burnevich ),( J. Niu ),( Y.J. Jung ),( X. Jiang ),( M. Xu ),( X. Chen ),( Q. Xie ),( J. Li ),( J. Hou ),( H. Tang ),( 대한간학회 2017 춘·추계 학술대회 (KASL) Vol.2017 No.1

Background/Aims: Treatment-naive GT 1b-infected patients from mainland China, South Korea and Russia were assessed for SVR at follow-up week 12 (SVR12) after receiving daclatasvir (60 mg, QD) and asunaprevir (100 mg, BID) (DCV+ASV). Methods: Patients were randomized 3:1 to receive DCV+ASV (24 weeks; immediate treatment [IM]) or 12 weeks of placebo followed by DCV+ASV (24 weeks; placebo-deferred treatment [PD]). The primary endpoint was to evaluate SVR12 in the IM arm to the historical rate for peginterferon/ribavirin (70%). Secondary endpoints included overall safety and safety comparisons between the treatment arms during the first 12 weeks. Results: 207 patients were randomized to IM (n=155) or PD (n=52); Asian (86%), female (59%), IL28B CC genotype (68%) and median age 49 (range 18-73) years; cirrhosis (13%), HCV RNA ≥6x106 IU/mL (53%). SVR12 in the IM arm was 92% and broadly unaffected by most baseline factors assessed (Figure); SVR12 was higher in patients without (96%) baseline NS5A-L31M/V or Y93H polymorphisms. There were 6 virologic breakthroughs, 6 relapses and 1 detectable HCV RNA at end-of-treatment in the IM arm. Safety was mostly comparable between the two arms during the first 12 weeks. The most frequent adverse events (AEs; ≥5%) during DCV+ASV (24 weeks) treatment in both arms were aminotransferase, bilirubin and INR elevations, hypertension, fatigue and respiratory tract infections; the most frequent treatment-emergent grade 3/4 laboratory abnormalities were aminotransferase (≤4.5%) and hematologic, lipase and total bilirubin abnormalities (≤2%); one patient (IM) discontinued DCV+ASV for aminotransferase elevations, nausea and jaundice (all reversible); one patient PD) discontinued DCV+ASV for a fatal AE unrelated to treatment. Conclusions: These data demonstrate that DCV+ASV is a highly efficacious and well tolerated treatment for treatment-naive HCV GT 1b-infected patients. Those treated immediately with DCV+ASV achieved a 92% SVR12 rate which was unaffected by factors known to attenuate response to interferon.

Effects of Alfalfa Meal on Growth Performance and Gastrointestinal Tract Development of Growing Ducks

        Jiang, J.F.,Song, X.M.,Huang, X.,Zhou, W.D.,Wu, J.L.,Zhu, Z.G.,Zheng, H.C.,Jiang, Y.Q. Asian Australasian Association of Animal Productio 2012 Animal Bioscience Vol.25 No.10

A study was conducted to evaluate effects of alfalfa meal on growth performance and gastrointestinal tract development of growing layer ducks to provide evidence for application of alfalfa meal in the duck industry. Two hundred and fifty-six healthy Shaoxing 7-wk old growing layer ducks were selected and randomly allocated to 1 of 4 dietary treatments based on corn and soybean meal and containing 0, 3, 6, and 9% of alfalfa meal for 8 wks. Each treatment consisted of 4 replicates of 16 ducks each. Briefly, birds were raised in separate compartments, and each compartment consisted of three parts: indoor floor house, adjacent open area and a connecting water area. The results showed: i) Growing ducks fed alfalfa meal diet were not significantly different in average daily gain, feed intake and gain-to-feed ratio from those fed no alfalfa diet (p>0.05). ii) Alfalfa meal increased the ratio crop, gizzard to live weight, caecum to live weight, the caecum index of growing ducks (p<0.05). iii) Villus height in duodenum and jejunum of growing ducks increased significantly with the increase of alfalfa meal levels (p<0.05). Crypt depth in duodenum and jejunum of growing ducks decreased significantly with the increase of alfalfa meal levels (p<0.05). This experiment showed that feeding of alfalfa meal to growing layer ducks could improve gastrointestinal tract growth and small intestinal morphology without effect on performance. This experiment provides evidence that alfalfa meal is a very valuable feedstuff for growing layer ducks.

New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        Chen, J.F.,Dai, L.H.,Peng, J.,Li, J.L.,Zheng, R.,Zuo, B.,Li, F.E.,Liu, M.,Yue, K.,Lei, M.G.,Xiong, Y.Z.,Deng, C.Y.,Jiang, S.W. Asian Australasian Association of Animal Productio 2008 Animal Bioscience Vol.21 No.4

The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.
