        환경 오염물질의 진보된 독성 평가 기법

        Recently, several new methods for the detection of genetic damages in vitro and in vivo based on molecular biological techniques were introduced according to the rapid progress in toxicology combined with cellular and molecular biology. Among these methods, mouse lymphoma thymidine kanase (tk) gene forward mutation assay, single cell gel electrophoresis (comet assay) and transgenic animal and cell line model as a target gene of lac I (Big Blue) and lac Z (Muta Mouse) gene mutation are newly introduced based on molecular toxicological approaches. The mouse lymphoma tk^(+/-) gene assay (MOLY) using L5178Y tk+i- mouse lymphoma cell line is one of the mammalian forward mutation assays, and has many advantages and more sensitive than hprt assay. The target gene of MOLY is a heterozygous tk^(+/-) gene located in 11 chromosome, so it is able to detect the wide range of genetic changes like point mutation, deletion, rearrangement, and mitotic recombination within tk gene or deletion of entire chromosome 11. The comet assay is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakages in mammalian cells. Also, transgenic animal and cell line models, which have exogenous DNA incorporated into their genome, carry recoverable shuttle vector containing reporter genes to assess endogenous effects or alteration in specific genes related to disease process, are powerful tools to study the mechanism of mutation in vivo and in vitro, respectively. Also in vivo acridine orange supravital staining micronucleus assay by using mouse peripheral reticulocytes was introduced as an alternative of bone marrow micronucleus assay. In this respect, there was an International workshop on genotoxicity procedure (IWGTP) supported by OECD and EMS (Environmental Mutagen Society) at Washington D, C, in March 25-26, 1999. The objective of IWGTP is to harmonize the testing procedures internationally, and to extend to finalization of OECD guideline, and to the agreement of new guidelines under the International Conference of Harmonization (ICH) for these methods mentioned above. Therefore, we introduce and review the principle, detailed procedure, and application of MOLY, comet assay, transgenic mutagenesis assay and supravital staining micronucleus assay.

        도시지역 여자고등학생들의 체형과 체형에 대한 의식 및 자각증상과의 관계

        Purpose: This study investigated the relationship between body type subjective symptoms and health awareness in female high school students Methods: The subjects were 393 students at the female high school in Taejon City by self-recorded questionnaires in May, 2006. Results: In the classification of body type based on the degree of obesity low weight group was 20.9% normal weight group was 58.0% over weight group was 11.7% and obesity group was 9.4% In the level of body type awareness those who consider themselves to be obesity type 13.7% standard type were 43.8% and leptosomic type 8.1% but there was a clear tendency to be leptosomic type that the rate was 76.3% In the complaint rates of subjective symptoms "fatigue" was the highest followed by "neck pain(stiffness)", "dizziness", "stomachache" in the descending order In the complaint rates of subjective symptoms by the degree of obesity dizziness irregular menstruation and anorexia were higher according to the lower obesity but constipation and wearied eyes were the higher the more obesity Conclusion: The study results showed that the recent female high school students were clear tendency to be leptosomic type and leptosomic and obesity type were related with subjective symptoms Considering these findings it appeared that being excessively thin is related to symptoms and young women's thinness-oriented attitudes are unhealthful.

        The preconception is socalled the naive theories was made according to the general experience, direct investigation, wrong explanation on textbook or bibliography material and different of culture. By Champagne and klopfer(1983) find out that the preconception are latent in those blains long time, so they cause serious influences to the general teaching method without special healing. The researches on the air unit in primary school are first, the concept of air volume and its weight, secondly the difference of concepts between urban and local area, and third the distinction of sex. In this work, there are formed preconception in the third grade children primary school, urban children thought are more than local, and last male children have more right thought than female both urban and local area.

        A Repeated-dose Oral Toxicity Study of Orostachys japonicus Extract in Sprague-Dawley Rats

        A 12-week repeated-dose oral toxicity study of water-soluble Orostachys japonicus extract (WOJ) was performed in Sprague-Dawley (SD) rats of both genders. Each group of ten rats was orally administered in doses of either 0 or 250 mg/day over a 12-week period. As a result, no WOJ-related changes were observed in terms of survival rate, clinical signs, body weight, or food intake. In addition, no difference in organ weight between the control and treated groups was detected. Furthermore, serum biochemistry parameters revealed some changes within normal ranges although significant decreases in total-bilirubin in the females. In spite of some alterations in serum biochemistry, the clinical signs, body weight changes from food intake, and autoptical remarks indicated that WOJ was not toxic. This study suggests that repeated treatment of O. japonicus very low toxicity and the NOAEL (no observed adverse effect dose) of WOJ exceeds 250 mg/kg in the SD rats.

        Acute oral toxicity of the ethyl acetate fraction of <i>Orostachys japonicus</i> in mice

        <P><I>Context</I>: <I>Orostachys japonicus</I> (Crassulaceae) is referred to as Wa-song in Korea. It is used as an anti-inflammatory, antifebrile, hemostatic, and anti cancer agent, and as an antidote.</P><P><I>Objective</I>: The purpose of this study was to evaluate the acute toxicity of the ethyl acetate fraction of <I>O. japonicus</I> (OJE) after the oral administration in Balb/c mice of both sexes.</P><P><I>Materials and methods</I>: Mice were oral administered a single doses of 500, 1000, and 2000 mg/kg of body weight and were monitored for 14 d. Biochemical parameters [aspartate amino transferase (AST), alanine amino transferase (ALT), alkaline phosphatase (ALP), total protein (TP), globulin (GB), total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN), and creatinine (CR)] and histopathological examination of liver were performed.</P><P><I>Results and conclusion</I>: No animals died and no toxic changes were observed in clinical signs, body weight, and organ weight. The LD<SUB>50</SUB> of orally administered OJE was higher than 2000 mg/kg/d in both sexes. No toxicological findings were found in biochemical parameters. In histophathological examination, neutrophilic infiltration was observed at a dose of 2000 mg/kg group in both sexes. These finding suggest that oral administration of OJE does not produce acute toxicity. Therefore, these results could provide satisfactory preclinical evidence of safety to launch clinical trials on standardized formulation of OJE to be a biohealth product.</P>

        연평도 일대의 식물상 연구

        인천광역시 옹진군에 속한 서해 5도 중 한 곳인 연평도일대에 자생하는 관속식물들을 밝히고 주요 식물들을 파악하였다. 2010년 5월부터 2010년 10월까지 총 4차례 현지조사를 통해 밝혀진 소산식물은 90과 315속 459종 4아종 53변종 8품종의 524분류군으로 조사되었다. 이 중 한반도 고유종은 총 2분류군, 산림청 지정 한국 희귀식물은 8분류군이 조사되었다. 구계학적 특정식물은 Ⅴ등급이 2분류군, Ⅳ등급이 2분류군, Ⅲ등급이 4분류군, Ⅱ등급이 3분류군, Ⅰ등급에 27분류군 등 총 38분류군이 분포하고 있는 것을 알 수 있었다. 귀화식물은 총 66분류군으로 조사되었으며, 귀화율은 12.6%로 높게 나타났다. The vascular plants of Is. Yeonpyeongdo regions, located in Ongjin-gun, Incheon were surveyed from May to October 2010. The vascular plants collected 4 times were consisted of total 524 taxa; 90 families, 315 genera, 459 species, 4 subspecies, 53 varieties and 8 formas respectively. Among them, 2 taxa endemic species of Korea and 8 taxa of Korean rare plants were investigated in addition to 38 taxa of specially designated plants announced by the Ministry of Environment. The naturalized plants were 66 taxa, therefore naturalized ratio was12.6% highly.

        대학 내 학문 목적 한국어 학습자를 위한 교양한국어 교재 개발 기초 연구

        This study aimed to explore the direction of development of textbooks to improve Korean language competency of foreign students studying in Korean universities. For this, the author explored and identified opening of Korean language subjects in major Korean universities and the status of textbook development. The author conducted a demand survey on the development of textbooks of Korean language as liberal arts for 345 foreign undergraduates. Based on the results, the author could confirm the necessity for development of a textbook for step-by-step and concentrative Korean language education conducted within universities currently. It was found that the demand for development of textbooks by grades in consideration of differences of competency, functioning and linguistic knowledge-strengthening textbooks was high among undergraduates. Therefore, this paper suggested a direction for development of textbooks for Korean language as liberal arts, adaptation to university lives and support for studying as well as textbook contents and basic knowledge enhancement. It derived ‘communication-based linguistic knowledge strengthening’ that would enable the students to naturally acquire linguistic knowledge in actual communication tasks in the context of learning.

        Because of the environmental matters the importance of the city railroad is as time goes by increasing. The case of obstacle of the power equipment which supplies electric power to city railroad will occur social and economical enormous loss. Thus, I studied on the preventing method in advance which makes it possible for us to maintain facilities efficiently. The main check points of the power facilities are voltage, current, humidity, partial discharge, move current. These points are gathered by sensor and transmitting to data acquisition device. These data are used to check equipment status in real time. In this paper I described in brief test process and results of the detection system.

        Effects of High Protein Diet on Meat Productivity and Quality in Multiparous Hanwoo Cull Cows

        일반적으로 다산 한우암소는 미경산 혹은 어린 한우암소에 비하여 낮은 육질을 나타내는 것으로 알려져 있으며, 현재까지 다산 한우 암소 비육을 위한 사료급여방법에 대한 연구는 매우 미비하다. 본 연구는 두 개의 서로 다른 사료로 구성된 사료급여방법이 다산 한우 암소의 육생산과 육질에 미치는 영향을 조사하였다. 대조구로는 일반 비육우사료 급여를 사용하였다. 처리구의 사료는 두 개의 서로 다른 단백질 수준으로 구성되어 있으며 14.28%의 단백질 사료는 비육초기에 급여하였으며, 12.70%의 단백질 수준은 출하 전까지 급여하였다. 대조구는 일반비육우사료(12.39% CP)를 사용하였다. 사양실험은 3개의 농장에서 수행되었다. 농장A에서는 29두의 다산 한우암소를 처리구 그리고 3두를 대조구로 공시하였다. 농장B와 농장C에서는 처리구와 대조구에 각각 8두와 3두 그리고 11두와 4두를 공시하였다. 실험사료들은 총 211일 동안 급여하였고, 처리구의 비육전기사료는 4개월간 급여하였고, 나머지는 비육후기 사료를 급여하였다. 평균일당증체량, 등 지방두께, 등심면적, 도체중, 육량지수, 근내지방, 육색 및 조직감등을 분석하였다. 통계처리는 렌덤효과모형[8]을 사용하였다. 처리의 정적효과는 평균일당 증체량, 등지방두께, 등심면적, 도체중 및 근내지방에서 나타났으며, 육량지수, 육색 및 조직감은 처리구의 부적효과가 나타났다. 모든 분석항목에서 통계적 유의성이 발견되지 않았는데 이것은 농장별 실험결과의 분산이 매우 켰기 때문이다. 그러나 통합적 효과에 있어 평균일당증체량, 등지방두께, 등심면적 및 도체중에 대한 90% 신뢰구간은 정적효과 방향으로 나타났으며, 나머지 분석항목의 90% 신뢰구간은 부적효과의 방향으로 나타났다. 결과적으로 다산 한우암소비육에 있어 비육전기기간의 고단백질 사료 급여는 육생산성 향상 및 육질 개선의 효과를 나타낼 수 있다. Multiparous Hanwoo cull cows have been generally regarded to have poor meat quality compared to young and primiparous Hanwoo in Korea, and there have been few studies attempting to understand various feeding programs for the multiparous Hanwoo cull cow. In this study, the effects of a feeding program consisting of two different diets for multiparous Hanwoo cull cows on meat production and quality were tested in comparison to a commercially used diet. Diets for treatment consisted of two levels of crude protein contents, 14.28% and 12.70% for early fattening and finishing, respectively. For a control, commercially used fattening feed (12.39% crude protein) was used. Feeding trials were performed at three different farms. In farm A, 29 herds of multiparous Hanwoo cull cows were used for the treatment group and 3 herds for the control. In farms B and C, the number of animal herds for treatment and control were 8 vs. 3 and 11 vs. 4 herds, respectively. Experiment diets were fed for an average of 211 days and in treatment, early fattening diet was fed for 4 months and then finishing feed was fed until slaughter. Average daily gains, thickness of back fat, area of Longissimus dorsi, carcass weight, index of carcass weight, intramuscular fat, meat color and texture were analyzed after slaughter. Random effect model [8] was employed in effect analysis. Positive effects of treatment were found in terms of average daily gain, back fat thickness, Longissimus dorsi area, carcass weight, and intramuscular fat. Carcass yield index, meat color and texture showed a negative effect. In this study, significant results were not found in all factors analyzed because the variance between experiment farms was large, however the 90% confidence interval of summary effects of ADG, back fat thickness, Longissimus dorsi area and carcass weight were significant and that of carcass yield index, intramuscular fat, meat color and texture were less so. Conclusively, a high protein diet fed early during the fattening period in multiparous Hanwoo cull cows could have positive effects on meat production.
