        양액내 염류농도 증가에 의한 상추뿌리의 마이크로솜 ATPase 활성증가

        염류장애에 의한 작물의 생육저해 원인을 밝혀내기 위하여 상추를 대조양액과 대조양액에 30 mM 및 50 mM KNO_3를 첨가하여 염류농도를 높인 양액 등 3가지 조건에서 재배하였으며, 이때 이들 양액의 EC는 각각 1.0, 4.5, 6.5 dS/m 이었다. 뿌리세포의 원형질막 및 액포막에 위치한 H^+-ATPase 활성은 각각에 특이적 저해제인 vanadate와 NO_3를 이용하여 측정하였다. 대조양액에서 재배한 상추 뿌리의 ATPase 활성은 356±1.5 nmol/min/mg protein이었으며, 30 mM과 50 mM KNO_3를 첨가한 양액에서는 활성이 대조활성에 비하여 각각 1.6배, 1.9배 증가하였다. 이것은 상추가 염류장애시 뿌리조직의 ATPase 활성증가를 통하여 적응함을 보여주는 것이며, 활성증가는 주로 액포막 H^+-ATPase 활성증가에 의해 이루어짐을 확인하였다. 마이크로솜 ATPase 활성에 미치는 여러 가지 중금속 이온들의 효과를 측정하였으며, 중금속 이온은 100 μM 농도에서 종류에 따라 활성을 10~25% 저해하였다. 특히, Cu^(2+)는 주로 액포막 H^+-ATPase 활성을 저해함을 확인하였다. 본 연구의 결과로부터 상추는 염류집적 환경에서 뿌리의 ATPase 활성, 특히 액포막에 위치한 H^+-ATPase 활성을 증가시켜 적응하며, Cu^(2+)는 주로 액포막 H^+-ATPase 활성을 저해하는 성분으로 뿌리의 ATPase 활성변화 연구에 유용하게 이용될 수 있음을 확인하였다. In order to investigate the mechanism of growth inhibition by salt stress, lettuces were grown hydroponically in three different nutrient solutions, normal and 30 mM or 50 mM KNO_3-added nutrient solutions, and the electrical conductivities of these solutions were 1.0, 4.5, and 6.5 dS/m, respectively. The activities of plasma and vacuolar H^+-ATPases in the root tissue of lettuce were measured by specific inhibitors, 100 μM vanadate and 50 mM No_3^-, respectively. Microsomal ATPase activity of lettuce grown in the normal nutrient solution was 356±1.5 nmol/min/mg protein. When lettuces were grown in 30 mM and 50 mM KNO_3-added nutrient solutions, total activities of microsomal ATPases were increased by 1.6 and 1.9 times, respectively, and the increases were mainly mediated by vacuolar H^+-ATPase. These results show that lettuces adapt themselves to salt-stressed condition by increasing the activities of H^+-ATPases. Effects of various heavy metal ions were investigated on the microsomal ATPases and various metal ions at 100 pM inhibited the activities by 10~25%. Cu^2+ showed the highest inhibitory effect on the vacuolar H^+-ATPase. These results suggest that lettuce increases the activities of root ATPases, specially that of vacuolar H^+-ATPase, in salt-stressed growth conditions and Cu^(2+) could be a useful tool to control the activity of vacuolar H^+-ATPase.

      • 마산연안지역의 국지풍 순환 및 대기오염물질 확산에 관한 연구

        To investigate a horizontal windrose and the mixing height in Masan coastal area, a local atmospheric flow and the concentration of air pollutants were analyzed by abserved data from AWS, airsonde and air sampler. The land-sea breeze model and the ISCST2 model of U. S. EPA(Environmental Protection Agency) were used to predict the concentration of air pollutants such as SO2 and TSP. As a result, during spring the principal wind direction in the Masan region is mainly represented by windrose of the WNW and NW, which can be induced by land breeze and the SE and SSW of sea breeze even if weak wind speed. The mixing height during the measurement was marked with the range from 400 m(0900 LST) to 1450 m(1500 LST), and the height of an inverse layer was presented as the range from 30 m(2100 LST) to 150 m(0300 LST). It is very important to cosider the local wind field for the accurate diffusion and movement of air pollutants in coastal urban area. The atmospheric flow was simulated by the two-dimensional local wind model upon considering the orographic characteristics such as land-sea contrast, mountain valley and urban building. The result of air quality simulated by ISCST2 support that the dispersion pattern of the pollutants in that area was followed as local circulation system in coastal urban area.

        Development and effects of a high-risk pregnancy emotive role-play program for nursing students: a quasi-experimental study

        Purpose: This study aimed to develop an emotive role-play program for nursing students focusing on high-risk pregnancy and analyze its effects on communication skills, clinical performance, and emotional intelligence.Methods: A quasi-experimental nonequivalent control group design was adopted with 83 nursing students (experimental group, 45; control group, 38) who participated voluntarily in an extracurricular program. The preliminary survey was conducted on November 3 and November 4, 2020, and the follow-up survey was conducted on November 12, 2020, for the control group and on November 27, 2020, for the experimental group. A program that included five role-play scenarios related to induced labor, preeclampsia, premature rupture of membranes, preterm labor, and infertility was developed by a group of experts and presented to the experimental group over 11 total hours across 3 days. Each student participated in a role-play scenario as a patient, family member, or nurse and observed three other scenarios. The control group received a workbook after the follow-up evaluation. The independent t-test was performed to analyze changes in communication skills, clinical performance, and emotional intelligence.Results: Communication skills (t=1.84, p=.035) and clinical performance (t=2.75, p=.004) significantly increased in the experimental group compared to the control group. A significant difference was not observed between the experimental and control groups for emotional intelligence (t=1.36, p=.088).Conclusion: The emotive role-play program concerning high-risk pregnancy was effective in improving nursing students’ communication skills and clinical performance and can be used in nursing education related to high-risk pregnancy and childbirth.

        High-Angle Tilt Boundary Graphene Domain Recrystallized from Mobile Hot-Wire-Assisted Chemical Vapor Deposition System

        <P>Crystallization of materials has attracted research interest for a long time, and its mechanisms in three-dimensional materials have been well studied. However, crystallization of two-dimensional (2D) materials is yet to be challenged. Clarifying the dynamics underlying growth of 2D materials will provide the insight for the potential route to synthesize large and highly crystallized 2D domains with low defects. Here, we present the growth dynamics and recrystallization of 2D material graphene under a mobile hot-wire assisted chemical vapor deposition (MHW-CVD) system. Under local but sequential heating by MHW-CVD system, the initial nucleation of nanocrystalline graphenes, which was not extended into the growth stage due to the insufficient thermal energy, took a recrystallization and converted into a grand single crystal domain. During this process, the stitching-like healing of graphene was also observed. The local but sequential endowing thermal energy to nanocrystalline graphenes enabled us to simultaneously reveal the recrystallization and healing dynamics in graphene growth, which suggests an alternative route to synthesize a highly crystalline and large domain size graphene.         Study on the Hatching Characteristics and Diet of the Stick Insect, Baculum elongatum (Phasmida: Phasmatidae) for Artificial Mass Rearing

        Baculum elongatum has several peculiarities such as parthenogenesis, unique external features, changing body color, and dropping oviposition, which makes it a potential economically useful insect. Oviposition was performed by females without fertilization by the sperm of males. The oviposition period was $42.2{\pm}22.7$ days and the number of eggs per female was $109.5{\pm}70.5$ eggs. The hatch rate was 73.3% at $25^{\circ}C$ and 66.7% after low temperature treatment ($8^{\circ}C$ for 60 days). In nature, B. elongatum overwinters as an egg, but it can be assumed from the results that cold temperatures were not required for hatching. The hatch rate was 98.2% in the treatment using floral foam, fermented sawdust, and leaves. The developmental period was $100.9{\pm}4.2$ days for eggs, $55.3{\pm}4.6$ days for nymphs, and $49.7{\pm}16.0$ days for adults. The length of the eggs was $0.33{\pm}0.0$ cm and the lengths of the nymphs were clearly distinguishable according to the instar stage. Clover(Trifolium repens) was an excellent diet as it was similar to the host plant and could be used as an alternative diet. The rate of reaching adulthood for the insects was 66.7% on an artificial diet containing 25% acacia leaves. For sustainable mass rearing of nymphs or adults of B. elongatum, a natural diet could be used such as acacia (Robinia pseudoacacia), white oak (Quercus aliena), chestnut (Castanea crenata var. dulcis), and bush clover (Lespedeza bicolor), or an alternative diet such as clover or artificial diet.

        이광수의 동정과 루쉰의 수치 - 신기원 시대의 정동 초탐

        This essay explores the affects observable in colonial Korea and China of the late 1910s. Two kinds of affects can be taken from the media responses to the March First Movement and the May Fourth Movement, shame and sympathy respectively. The Chinese became shameful in the face of the March First and the colonial Koreans were sympathized with the Chinese in light of their similar historical experience. The two forms of affects are found in the fictional narratives of Yi Kwangsu and Lu Xun. Yi tried to call forth the changes to colonial Korea by illustrating “philanthropic sympathy” in Heartless in which the sympathy goes beyond its conventional notion. The “traveller’s” portrayal, however, arrives only at harmony, the collaboration for the benefits of social maintenance. On the other hand, the madman in Lu Xun’s “A Madman’s Diary” realizes shame, which is related to the fear of unreliable self-portrayal rather than the guilty consciousness by breaching norms. This shame enables a person to go beyond the “threshold” and to create new vision by being accompanied with a radical self-dichotomy such as a desire for matricide. Yi’s sympathy and Lu Xun’s shame differ in the faithful practice of colonial modernization and the radical self-destruction.
