Effect of secretory leukocyte protease inhibitor on migration and invasion of human KB oral carcinoma cells

Guanlin Wang,임도선,최백동,박진주,정순정,김진수,김재덕,박정수,김응권,김병훈,함주현,정문진 한국통합생물학회 2011 Animal cells and systems Vol.15 No.2

Secretory leukocyte protease inhibitor (SLPI) plays an important role in promoting the invasion and metastasis of a range of cancer cells. However, there are no reports of the expression and function of SLPI in oral carcinoma cells. In this study, the oral carcinoma cell line KB was used to determine whether SLPI affects the proliferation, migration and invasion of oral carcinoma cells. RT-PCR and Western blotting revealed high levels of endogenous SLPI expression in KB cells as well as a strong increase in SLPI secretion after wounding compared to immortalized normal oral keratinocytes (INOK). The wound healing assay revealed more migration of KB cells than INOK cells, and the SLPI treatment increased the migration of KB cells. KB cell proliferation was increased significantly by the SLPI protein but decreased by SLPI-siRNA. SLPI strongly increased the migration and invasion of KB cells. On the other hand, SLPI-siRNA decreased the migration and invasion of KB cells. This suggests that SLPI plays an important role in the metastasis of oral carcinoma cells.

Integrated Guidance and Control for the Fixed-trim Vehicle against the Maneuvering Target

        Guanlin Li,Tao Chao,Songyan Wang,Ming Yang 제어·로봇·시스템학회 2020 International Journal of Control, Automation, and Vol.18 No.6

From the view of the fixed-trim reentry vehicle (FTRV) application, the integrated guidance and control (IGC) design is a key challenge. In order to improve the performance of the FTRV against the maneuvering target, an IGC system combining with the virtual target and the filter is proposed in this paper. To investigate the dynamics of the FTRV against the maneuvering target, a 7-DOF mathematical model is established and the error angle is employed to describe the relative motion between the FTRV and the target. Considering the controllability of theFTRV, a nonlinear differentiable error angle command is presented and its influence on the terminal velocity is discussed. To deal with the contradiction between the limited maneuverability and the demand of high guidance accuracy, the actual target is taken place with the virtual target, of which acceleration is estimated by the ESO. For improving the performance of the virtual target position prediction, a rolling updating strategy is proposed and derived analytically. Then, the back-stepping based IGC system is designed with the virtual target and the filter. The finite-time convergence of the IGC system is proved via the Lyapunov stability theorem. The numerical simulation results show the effectiveness of the proposed IGC system for the FTRV against the maneuvering target with time-varying acceleration.

Bacterial Fruit Rot of Apricot Caused by Burkholderia cepacia in China

        Fang, Yuan,Li, Bin,Wang, Fang,Liu, Baoping,Wu, Zhiyi,Su, Ting,Qiu, Wen,Xie, Guanlin The Korean Society of Plant Pathology 2009 Plant Pathology Journal Vol.25 No.4

An unreported disease of apricot was observed in orchards in Zhejiang province, China. Symptoms started as water soaked lesions on the fruit surface. Later, water-soaked areas developed and spread to the entire fruit, resulting in soft rot of the whole fruit. The causal organism isolated from symptomatic fruits was identified as Burkholderia cepacia based on its biochemical and physiological characteristics and confirmed by the cellular fatty acid composition and Biolog data as well as 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms when inoculated onto fruits of apricot. In addition, European plum, Japanese plum, nectarine and kiwifruit were susceptible to the B. cepacia pathogen. However, the B. cepacia pathogen failed to cause any visible symptoms when it was inoculated onto 16 other fruits. This is the first report of a bacterial disease of apricot caused by B. cepacia in China.

      • Secretory Leukocyte Protease Inhibitor (SLPI) Increases Focal Adhesion in MC3T3 Osteoblast on Titanium Surface.

        Jeong, Soon-Jeong,Wang, Guanlin,Choi, Baik-Dong,Hwang, Young-Hyun,Kim, Byung-Hoon,Ko, Yeong-Mu,Jeong, Moon-Jin American Scientific Publishers 2015 Journal of Nanoscience and Nanotechnology Vol.15 No.1

An appropriate interaction between implanted materials and the surrounding tissue is essential for successful implantation. Titanium (Ti) and some of its alloys have been used in dentistry and orthopedics as a substitutive material for hard tissue, such as teeth or natural bone. Nevertheless, metal ions released from titanium and alloy implants have adverse biological effects on biological tissues or cells. Secretory leukocyte protease inhibitor (SLPI) promotes cell migration, proliferation and wound healing. FAK and ERK1/2 signaling regulate cell adhesion and proliferation for cell survival. This study evaluated the potential of SLPI as a molecule to increase the cell adhesion on the Ti surface. Compared with the untreated cells, SLPI increased the adhesion of MC3T3-E1 cells to Ti discs, formation of actin stress fibers, paxillin expression and the phosphorylation of FAK. Moreover, SLPI enhanced the level of Grb2 and Ras expression and ERK1/2 phosphorylation in the MC3T3-E1 cells on Ti discs. These results suggest that SLPI can increase the interaction between the implanted Ti material and surrounding bone in orthodontic and dental surgery, making an effective nanomolecule for successful implantation.

Resveratrol raises in vitro anticancer effects of paclitaxel in NSCLC cell line A549 through COX-2 expression

        Kong, Fanhua,Zhang, Runqi,Zhao, Xudong,Zheng, Guanlin,Wang, Zhou,Wang, Peng The Korean Society of Pharmacology 2017 The Korean Journal of Physiology & Pharmacology Vol.21 No.5

The aim of this study was to determine the raising anticancer effects of resveratrol (Res) on paclitaxel (PA) in non-small cell lung cancer (NSCLC) cell line A549. The $10{\mu}g/ml$ of Res had no effect on human fetal lung fibroblast MRC-5 cells or on A549 cancer cells and the 5 or $10{\mu}g/ml$ of PA also had no effect on MRC-5 normal cells. PA-L ($5{\mu}g/ml$) and PA-H ($10{\mu}g/ml$) had the growth inhibitory effects in NSCLC cell line A549, and Res increased these growth inhibitory effects. By flow cytometry experiment, after Res ($5{\mu}g/ml$)+PA-H ($10{\mu}g/ml$) treatment, the A549 cells showed the most apoptosic cells compared to other group treatments, and after additional treatment with Res, the apoptosic cells of both two PA concentrations were raised. Res+PA could reduce the mRNA and protein expressions of COX-2, and Res+PA could reduce the COX-2 related genes of VEGF, MMP-1, MMP-2, MMP-9, $NF-{\kappa}B$, Bcl-2, BclxL, procollagen I, collagen I, collagen III and CTGF, $TNF-{\alpha}$, $IL-1{\beta}$, iNOS and raise the TIMP-1, TIMP-2, TIMP-3, $I{\kappa}B-{\alpha}$, p53, p21, caspase-3, caspase-8, caspase-9, Bax genes compared to the control cells and the PA treated cells. From these results, it can be suggested that Res could raise the anticancer effects of PA in A549 cells, thus Res might be used as a good sensitizing agent for PA.
