        LncRNA SNHG6 promotes glycolysis reprogramming in hepatocellular carcinoma by stabilizing the BOP1 protein

        Metabolic reprogramming is an important feature in tumor progression. Long noncoding RNA’s (lncRNA) small nucleolar RNA host gene 6 (SNHG6) acts as a proto-oncogene in hepatocellular carcinoma (HCC) but its role in glycolysis is mostly unknown. The role of SNHG6 and Block of proliferation 1 (BOP1) on glycolysis is assessed by glucose uptake, lactate production, oxygen consumptive rate (OCR) and extracellular acidification rate (ECAR) and glycolytic enzyme levels. The regulatory effect of SNHG6 on BOP1 protein was confirmed by Western blotting, MS2 pulldown, RNA pull-down, and RIP assay. SNHG6 and BOP1 levels were increased in HCC tissues and cells. SNHG6 and BOP1 were prognostic factors in HCC patients and significantly correlated to TP53 mutant and tumor grade. SNHG6 promoted proliferation, inhibited apoptosis, enhanced glucose uptake and lactate production, decreased OCR, and increased ECAR in HCC cell lines. SNHG6 could bind the BOP1 protein and enhance its stability. BOP1 overexpression rescued the change of proliferation, apoptosis, and glycolysis in HCCLM3 and SMMC-7721 cells. Our data indicate that SNHG6 accelerates proliferation and glycolysis and inhibits the apoptosis of HCC cell lines by binding the BOP1 protein and enhancing its stability. Both SNHG6 and BOP1 are promising prognostic and therapeutic markers in HCC.

        RNA sequencing reveals lncRNA-mediated non-mendelian inheritance of feather growth change in chickens

        Background: Long non-coding RNAs (lncRNAs) play an essential role in biological processes. However, the expression patterns of lncRNAs that regulate the non-Mendelian inheritance feather phenotypes remain unknown. Objective: This study aimed to compare the expression profiles of lncRNAs in the follicles of the late-feathering cocks (LC) and late-feathering hens (LH) that followed genetic rules and the early-feathering hen (EH) and early-feathering cock (EC) that did not conform to the genetic laws. Methods: We performed RNA sequencing and investigated the differentially expressed lncRNAs (DElncRNAs) between the early- and late-feathering chickens, which function by cis-acting or participate in the competing endogenous RNA (ceRNA) network. Results: A total of 53 upregulated and 43 downregulated lncRNAs were identified in EC vs. LC, and 58 upregulated and 109 downregulated lncRNAs were identified in EH vs. LH. The target mRNAs regulated by lncRNAs in cis were enriched in the pentose phosphate pathway, TGF-β signaling pathway and Jak-STAT signaling pathway in EC vs. LC and were associated with the TGF-β signaling pathway, Wnt signaling pathway, p53 signaling pathway and Jak-STAT signaling pathway in EH vs. LH. In addition, the lncRNA-mediated ceRNA regulatory pathways of hair follicle formation were mainly enriched in the TGF-β signaling pathway, Wnt signaling pathway, melanogenesis, and calcium signaling pathways. The levels of ENSGALG00000047626 were significantly higher in the late-feathering chickens than in the early-feathering chickens, which regulated the expression of SSTR2 by gga-miR-1649-5p. Conclusion: This study provides a novel molecular mechanism of lncRNA's response to the feather rate that does not conform to the genetic laws in chickens.

        Synthesis and Properties of Benzodithiophene-Based Donor-Acceptor Metallo-Supramolecular Polymers

        Two new conjugated donor-acceptor (D-A) supramolecular building blocks, which contain benzo[1,2-b:4,5-b']dithiophene (BDT) electron-donating cores modified with alkoxy (BDT-OR) or thiophene (BDT-Th) groups and electron-accepting 2,2':6',2''-terpyridine moieties, were synthesized via Pd-catalyzed Stille coupling. Under the introduction of transition metal ion Ru2+, the novel metallo-supramolecular polymers PBDT-OR and PBDT-Th were obtained by self-assembly polymerization of the monomeric building blocks BDT-OR and BDT-Th, respectively. The resulting polymers exhibit broad absorption band from 300 to 600 nm. The embedding of the transition metal ions Ru2+ into the backbones of the supramolecular polymers increases the electron-withdrawing capacity of the terpyridine moieties. Consequently, the absorption bands of both PBDT-OR and PBDT-Th exhibit red-shift at the longer wavelength compared with the corresponding monomeric building blocks, 622 nm for PBDT-OR and 625 nm for PBDT-Th, which results from intramolecular charge transfer (ICT). The LUMO energy levels of PBDT-OR and PBDT-Th are similar, -3.44 and -3.43 eV, respectively, while their HOMO energy levels are different, -5.25 and 5.17 eV, respectively, due to distinct electron-donating abilities of the BDT cores modified with alkoxy and thiophene groups. PBDT-OR and PBDT-Th show reduced electrochemical energy gaps, 1.81 and 1.74 eV, respectively. Meanwhile, the resulting polymers PBDT-OR and PBDT-Th are thermally stable under 300 and 389 oC, respectively.

        Comparison between laparoscopic and abdominal radical hysterectomy for stage IB1 and tumor size <2 cm cervical cancer with visible or invisible tumors: a multicentre retrospective study

        Objective: To compare 5-year disease-free survival (DFS) and overall survival (OS) rates oflaparoscopic radical hysterectomy (LRH) and abdominal radical hysterectomy (ARH) forstage IB1 and tumor size <2 cm with visible or invisible tumors. Methods: We retrospectively compared the oncological outcomes of 1,484 cervical cancerpatients with IB1 and tumor size <2 cm on final pathology, who received ARH (n=899) or LRH(n=585) between January 2004 and December 2016. Patients were divided into visible tumorsubgroup (ARH: n=668, LRH: n=444) and invisible tumor subgroup (ARH: n=231, LRH:n=141) according to tumor type. Results: LRH and ARH showed similar 5-year DFS and OS rates (93.3% vs. 93.1%, p=0.997;96.2% vs. 97.5%, p=0.351) in total study population. LRH was not associated with worse5-year DFS rate (hazard ratio [HR]=0.96; 95% confidence interval [CI]=0.58–1.58; p=0.871)or OS rate (HR=1.37; 95% CI=0.65–2.89; p=0.409) by multivariable analysis. In the visibletumor subgroups, LRH and ARH showed similar 5-year DFS and OS rates (91.9% vs. 91.9%,p=0.933; 95.0% vs. 96.9%, p=0.276), and LRH was not associated with worse 5-year DFS orOS rate (p=0.804, p=0.324). In the invisible tumor subgroups, LRH and ARH also showedsimilar 5-year DFS and OS rates (97.3% vs. 97.1%, p=0.815; 100% vs. 99.5%, p=0.449), andLRH was not associated with worse 5-year DFS rate (p=0.723). Conclusions: Among patients with stage IB1 and tumor size <2 cm, whether the tumor isvisible or not, the oncological outcomes of LRH and ARH among cervical cancer patients arecomparable. This suggests that LRH may be suitable for stage IB1 and tumor size <2 cm withvisible or invisible tumors.Trial Registration: International Clinical Trials Registry Platform Identifier: CHiCTR180017778

        Intercellular Trafficking of Homeodomain Proteins

        Homeotic proteins have pivotal roles during the development of both plant and animals. Many homeotic proteins exert control over cell fate in cells where their genes are not expressed, i.e., in a non-cell autonomous manner. Cell-to-cell communication, which delivers critical information for position-dependent specification of cell fate, is an essential biological process in multicellular organisms. In plants, there are two pathways for intercellular communication that have been identified: the ligand/receptor-mediated apoplastic pathway and the plasmodesmata-mediated symplasmic pathway. Regulatory proteins and RNAs traffic symplasmically via plasmodesmata and play a critical role in intercellular communication. Thus, the non-cell autonomous function of homeotic proteins can be explained by the recent discovery of cell-to-cell trafficking of proteins or RNAs. This article specifically focuses on understanding the intercellular movement of homeodomain proteins, a family of homeotic proteins.

        Molecular cloning and expression analysis of the highly conserved eukaryotic translation initiation factor 5A (eIF-5A) from Antheraea pernyi

        Eukaryotic initiation factor 5A (eIF‐5A) is a highly conserved protein found in all eukaryotic organisms that plays a key role in the regulation of many cellular processes including translation elongation, cell proliferation, programmed cell death, mRNA turnover and decay, and abiotic stress responses. In this study, the eIF‐5A gene from the Chinese oak silkworm Antheraea pernyi (Lepidoptera: Saturniidae) was characterized. The full‐length ApeIF‐5A cDNA of 1056 bp includes a 5′‐untranslated region (UTR) of 138 bp, a 3′‐UTR of 435 bp, and an open reading frame of 483 bp encoding a polypeptide of 160 amino acids. The deduced ApeIF‐5A protein shares 99 %, 82 %, and 72 % sequence identity with orthologs in Bombyx mori, Drosophila melanogaster and Homo sapiens, indicating high conservation during animal evolution. Real‐time quantitative reverse transcription PCR revealed expression in all four developmental stages and in all nine tissues tested, consistent with an important role in development. After challenge with lipopolysaccharide, the expression levels of ApeIF‐5A were markedly upregulated. Phylogenetic analysis of amino acid sequences revealed A. pernyi eIF‐5A was closely related to B. mori eIF‐5A, consistent with traditional classification and other molecular data. The results indicate the potential value of eIF‐5A in phylogenetic analysis.

        Characterization of Hot Deformation Behavior and Processing Map of As‑Cast H13 Hot Work Die Steel

        The hot-working behavior of as-cast H13 hot work die steel was investigated in the method of isothermal compression testinvolving the wide deformation temperatures of 900–1150 °C and strain rates of 0.01–10 s−1, with the true strain to 0.8, onthe MMS-200 thermo-mechanical simulator. Two characteristic parameters involving the critical strain for DRX initiation(c ) and the strain for peak stress (p ) were identified. The ratio of critical strain to peak strain ranged from 0.26 to 0.6, whichdecreased with the increase in temperature and decrease in strain rate. Processing maps were established using dynamicmaterial model at strains of 0.2, 0.4, 0.6, 0.8. The power dissipation maps were not significantly affected by the strain, whilethe instability maps were sensitivity with the strain when it was over 0.4. The area of instability domain at strain of 0.8 wasthe largest. The instable characteristics contained the mixed grain structure, adiabatic shear band, intense deformation inserious deformation area and brittle elemental segregation area. The chief effect on the power dissipation was the strain rate,the optimum hot working parameters at strain of 0.8 (910–985 °C, 1010–1150 °C and 0.01–0.05 s−1) were determined. Inthis filed, the original coarse as-cast grains were gradually refined by dynamic recrystallization mechanism and the DRXgrain numbers had a significant increase with the increase of power dissipation efficiency.
