      • Effect of Ionizing Radiation on Rat Tissue: Proteomic and Biochemical Analysis

        <P>Reactive oxygen species (ROS), generated by ionizing radiation, has been implicated in its effect on living tissues. We confirmed the changes in the oxidative stress markers upon irradiation. We characterized the changes in the proteome profile in rat liver after administering irradiation, and the affected proteins were identified by MALDI-TOF-MS and ESI-MS/MS. The identified proteins represent diverse sets of proteins participating in the cellular metabolism. Our results demonstrated that proteomics analysis is a useful method for characterization of a global proteome change caused by ionizing radiation to unravel the molecular mechanisms involved in the cellular responses to ionizing radiation.</P>

      • Pocine Adenovirus-3의 E1B Region의 鹽基序列 分析

        돼지 아데노바이러스(PAV-3). 6618주의 EIB region이 包含되어 있는 map unit 4.0에서 9.7까지의 유전자에 대한 1,984 bp의 염기서열을 決定하였으며, 이 結果를 알려진 여러 아데노바이러스 유전자와 비교하여 다음과 같은 결과를 얻었다. 1. PAV-3의 EIB유전자는 10개의 ORF로 구성되어 있으며, 그 중 아데노바이러스의 단백질과 유사성이 있는 것은 ORF1, ORF2 및 ORF3이었다. ORF1은 Ad41의 19kd 와 BAV-2에서의 EIB ORF2에서의 아미노산의 一致率은 각각 32%와 31%이었다. 2. ORF2는 Ad2 55kd protein과 tupaia adenovirus 44kd protein가 각각 34%로 아미노산 一致率이 가장 높았으며, Ad41의 52kd protein. BAV-3의 EIB ORF3에서도 33%의 一致率을 보였다. 3. ORF1은 61-666 uncleotide (606 bp), ORF 2에서는 429-1,850 uncleotide (1,422 bp)의 부위로 각각 202, 474 a.a로 構成되었으며, 예상되는 분자량은 20 kd와 52 kd이었다. 4. ORF3는 hexon-associated pIX유전자로 추정되며 내부에 1개의 polyadenylation signal(ATAAA)이 1938-1942 uncleotide에 위치하였으며, 이 부위는 TATA box (1937-1942 uncleotide)와 중복되어 존재하였다. Porcine adenovirus type 3 (PAV-3) does not cause severe infection in pigs. Adenovirus has been suggestive of live vaccine vector carrying foreign gene. One of insertion regions is delayed early (EIB) region. However, EIB region of PAV-3 has not been molecularly characterized to date. Nucleotide sequence of EIB of PAV-3 was determined. The EIB region was composed of 1,984 bp and located between 4.0 and 9.7 map units. Three potential open reading frames(ORFs) with low level of homology to other adenoviruses and a polyadenylation signal were identified in the rightward direction of genome. The nucleotide and the predicted amino acid sequences of EIB were compared to those of human and animal adenoviruses. One of the three potential ORFs. ORF1 encoded a polypeptide homologous to bovine adenovirus type 2(BAV-2) ORF2 and human adenovirus type 41(Ad41) 19 kd protein. ORF2 encoded a polypeptide homologous to human adenovirus type 2(Ad2) 55 kd protein, bovine adenovirus type 3(BAV-3) ORF3 and porcine adenovirus type 4(PAV-4) ORF2. The predicted protein of ORF1 had homology to those of Ad41 and BAV-2 with 32 and 31% respectively, whereas the deduced protein of ORF2 had homology to those of Ad2. BAV-3 and PAV-4 with 34, 33 and 29%, respectively.

        Training is very important for health, physical fitness and prevention of various kinds of disease. Recently, hypokinesis from automation, excessive caloric intake and stresses from personal relations increase and systematic physical exercise are keenly felt. In hypertensive, static or anaerobic exercise are apt to increase blood pressure too much, so moderate or light whole body exercise which intensity can be easily controlled are encouraged. Proper intensity for hypertensive are thought to be within the range of 60% of maximal oxygen consumption (HP 120-135 beats/min). ​

      • 力道 選手들의 筋力과 心肺機能에 관한 硏究

        The purpose of this study was to observe the effects of Strength and cardiorespiratory function of man weight lifter. Athletes group was that ten weight lifters of H University in Seoul and Non-Athletes group was that ten male students of H University did not especially player activity. Weight lifters career was about four years. The result of statistics data were as follows. 1. Comparison of Strength Back strength showed significant difference (p<0.001) between groups. Grip strength, Bench press and Squat showed significant diffence(p<0.001) between groups. 2. Comparison of Body weight. Heart Rate. HR max, VE max. VO, max, and 0.pulse. Body weight and rest heart rate showed significant difference (p<0.01) between groups and V0,max showed significant difference (p<0.001) between groups. But 0,pulse showed significant difference (p<0.001) between groups. 3. Comparison of Blood pressure During rest systolic blood pressure and diastolic blood pressure did not showed significant difference between groups. During exercise systolic lood pressure showed significant difference (p<0.001) but during exercise diastolic blood pressure did not showed between groups.​

      • 태권도 선수의 최대운동부하 시 혈중 백혈구 수와 호중구 활성산소 산생능에 미치는 영향

        Twenty endurance-trained Teakwondo ran on a treadmill by a protocol of incremental graded increase in workload until exhaustion during an endurance training period in off-season summer. Immediately after exercise, all developed peripheral leukocytosis (1.9 times; p<.01) due mainly to lymphocytosis (2.6 times; p<.01) with a predominant effect on large granular lymphocyte{natural killer cell) count(5.9 times; p<.01). Monocyte count was also enhanced 2.3 times (p<.01). These increases were transitory and returned to the pre-exercise levels lh later. Peripheral neutrophilia was also observed by 43%(p<.01) immediately after exercise and remained elevated by 25% (p<.01) lh after exercise, but a shift to the left did not take place. The capacity of isolated neutrophlis to produce reactive oxygen species was assessed by luminol-dependent chemiluminescene which detects mainly myeloperoxidase (MPO)-mediated formation of such hyperreactive oxidants as HOCI. The maximam intensity of chemiluminescence(peak height) upon stimulation with opsonized zymosan was significantly enhanced following exercise (p<.05). Similar results were obtained when phorbol myristate acetate was employed as nonphagocytic solubic stimulus(p<.01), suggesting that the capacity of neutrophils to degranulate MPO rather than phagocytosis was enhanced following exercise. In addition, the enhancements of chemiluminescence were positively correlated with the increase in segmented neutrophil count. These data indicate that maximal exercise not only mobilized mature neutrophilis from the marginated pool into the circulation, but also augmented their capacity to generate reactive oxygen species of higher reactivity.

      • 국내에서 분리된 canine parvovirus의 구조유전자 cloning과 염기서열 분석

        In this study gene encoding structural proteins of a CPV isolate was cloned and saquenced to elucidate the molecular genetical properties of the canine parvoviruses isolated from the field. Six recombinant plasmids of pEP3, p1471, p2070, pEP069, pEP338 and p14711p were constructed from the map positions 22 to 98 of RF DNA to clone the VP1 and VP2 genes of CPV-V20. Sequentialy the gene comprising 3780 nucleotides were sequenced by dideoxy chain termination method. When nucleotide sequence of gene encoding the structural proteins of CPV-V20 was compared with those of other strains, CPV-N, CPV-d and CPV-780929 published previously. DNA homologies to CPV-V20 were 99.87% with CPV-NM, 99.73% with CPV-d, 96.85% with CPV-780929 AND 98.4% with FPLV-Carl, respectively. The DNA sequence data of CPV-V20 showed seven point mutations and also deletion of 135 nucleotides from the nucleotide position 4745 to 4879 located in the 3-noncoding region of CPV-N.

      • 인체구강상피세포의 배양조건에 대한 연구

        Human epithelial cell cultures were initiated from explants of normal human oral mucosa. Growth parameter and cell type were assessed by DNA concentration and morphology of the cell lines. Tissues were excised and epithelial cells isolated by collagenase digestion. Spermine was used in order to inhibit overgrowth of the epithelial cells by fibroblast. The effects of different medium formulations were compared. Cultures maintained on the same volume of RPMI 1640 and Dulbecco's modified minimum essential medium containing 10 % fetal bovine serum, 10 ?g/ml of insulin, lng/ml of somatomedin and 10 ng/ml of epidermal growth factor showed optimal and maximal growth. For each cultured human oral epithelial cell the DNA content was in the- range of 5.7 pg to 6.6 pg. This culture system provides epithelial cells that can be used for a variety of dental and biochemical studies.

        만성적 소음폭로가 근로자의 혈압에 미치는 영향

        The question of an association between occupational noise exposure and blood pressure has important public health implications. The harmful effects of hypertension are well known and noise is considered the most pervasive problem of all occupational exposures in Taegu City, Korea. A cross-sectional study on the effect of long-term noise exposure on blood pressure was done for 276 noise exposed workers(203 male workers, 73 female workers). Long-term noise exposure was measured by cumulative noise exposure level and noise-induced hearing loss. Hearing loss = 500Hz+2×1,000Hz+2×2,000Hz+4,000Hz -------------------------------------- 6 Questionnaire was administered to the workers, which includes age, sex, occupational history on the noise exposure, habits on alcohol drinking and smoking, practice of regular exercises, family history of hypertension. Type A behavior, question on stress, height, weight and blood pressure were measured. Serum triglyceride and low density lipoprotein(LDL) were also measured. Audiometry was done in the closed booth at 1,000, 2,000, 3,000, 4,000, 6,000, 8,000, 1,000 and 500Hz in order. Multiple logistic regression analysis revealed that cumulative noise exposure level(p〈0.05), serum triglyceride(p〈0.01), age(p〈0.01) and family history of hypertension(p〈0.05) predict hypertension (systolic blood pressure : 160㎜Hg or above, or diastolic blood pressure : 95㎜Hg or above) in male workers. In female workers, serum triglyceride(p〈0.01), LDL(p〈0.01) and family history of hypertension(p〈0.05) were predictors of hypertension (systolic blood pressure : 160㎜Hg or above, or diastolic blood pressure : 95㎜Hg or above). As the number of female workers with hypertension was too small, the multiple logistic regression was done according to hypertension criteria of systolic blood pressure greater than or equal to 140㎜Hg, or diastolic blood pressure greater than or equal to 90㎜Hg. Based on this criteria, cumulative noise exposure level(p=0.055) and age(p=0.057) predict hypertension. It is suggested that long-term noise exposure which was calculated by cumulative noise exposure level was a significant predictor of hypertension in noise exposed workers.

        The purpose of this study is investigated the effect factors of the long-term tendency between consumers and distributers in the moving communication industry. Two hypotheses were generated regarding the effect factors among these structure. The results of this study were following. First, relationship formation factors had an effect on the long-term tendency between consumers and distributers. Second, among the distributer's character factors, consumer oriented consideration and discriminated poduct feature had an effect on the long-term tendency between consumers and distributers.
